Verfahren zur Herstellung von Salpetersäureestern von Dialkylxanthin-carbonsäureoxyalkylamiden
Die Umsetzung von Alkanolaminen mit rauchender Salpetersäure führt unter bestimmten
Bedingungen zu den entsprechenden Salpetersäureestern, die sowohl sprengstofftechnisch
als auch pharmakologisch interessante Verbindungen darstellen. Es wurde nun gefunden,
daß die noch nicht bekannten Salpetersäureester der Dialkylxanthin-carbonsäureoxyalkylamide
ebenfalls physiologisch interessante Verbindungen sind. Sie haben gegenüber den
Salpetersäureestern der Alkanolamine den Vorzug, daß sie so geringe Sprengstoffeigenschaften
haben, daß sie sich leicht handhaben lassen und daß sie gut kristallisierte, auch

Die nach dem erfindungsgemäßen Verfahren hergestellten Nitroxyalkylcarbonsäureamide
haben die allgemeine Formel
in der A einen niedrigmolekularen Alkylenrest, R einen Xanthinrest, dessen 1-, 3-
und 7-Stellung durch zwei Alkylgruppen und die Carbonsäurenitroxyalkylamidgruppe
besetzt sind, R, ein Wasserstoffatom, eine Alkyl-oder Nitroxyalkylgruppe und R2
ein Wasserstoffatom oder einen Carbonsäurerest der allgemeinen Formel
bedeutet. Die Verbindungen können in an sich bekannter Weise erhalten werden durch
direkte Nitrierung der entsprechenden Dialkylxanthin-carbonsäureoxyalkylamide. Zu
den gleichen Verbindungen gelangt man in an sich bekannter Weise durch Umsetzung
eines entsprechenden Dialkylxanthin-carbonsäurechlorids mit Nitroxyalkylaminen der
allgemeinen Formel H N (R1) - A - O N 02. Die Verfahrensprodukte sind ebenso coronarwirksam
wie die bekannten Salpetersäureester des 7-ß-Oxyäthyltheophyllins, sie sind jedoch,

5 g rauchender Salpetersäure bei -10° C umgesetzt und das Nitriergemisch 12 Stunden
im Kühlschrank aufbewahrt. Man trägt anschließend die Salpetersäurelösung nach und
nach unter Rühren in tiefgekühlten absoluten Äther ein, wobei der Ester als weiße,
zum Teil schmierige Substanz ausfällt. Man dekantiert die ätherische Schicht ab
und kristallisiert den Rückstand aus absolutem Alkohol um. Der Salpetersäureester
des Theophyllin-7-essigsäure-ß-oxyäthylamids schmilzt bei 159° C. Ausbeute: 6911f,

Ester durch Eintragen des Reaktionsgemisches in Äther ausgefällt. Man trennt die
Kristalle ab, reinigt sie mit viel Wasser von anhaftender Salpetersäure und kristallisiert
aus absolutem Alkohol um. Der Disalpetersäureester des Theophyllin-7-essigsäure-bis-(ß-oxyäthyl)-amids
schmilzt bei 124° C. Ausbeute: 55 bis 67°(o der Theorie.

Beispiel 3 5 g Theophyllin-7-malonsäure-bis-(ß-oxyäthylamid) werden
bei -10°C in 15 ccm rauchende Salpetersäure nach und nach eingetragen und 12 Stunden
in der Kälteder Esterbildung überlassen. Man fällt den entstandenen Ester durch
Eingießen des Reaktionsgemisches in eine größere Menge tiefgekühlten Äthers, dekantiert
anschließend den Äther ab und wäscht den Rückstand mit Wasser salpetersäurefrei.
Nach Umkristallisation aus Alkohol schmilzt der reine Disalpetersäureester von Theophyllin-7-malonsäure-bis-(ß-oxyäthylamid)
bei 166 bis 168° C. Salpetersäure hergestellt. Umkristallisiert aus absolutem Alkohol hat das Verfahrensprodukt
einen Schmelzpunkt von 194 bis 196° C. Ausbeute: 43 °% der Theorie. Beispiel 5 Der
Disalpetersäureester von Theobromin-l-malonsäure-bis-(ß-oxyäthylamid) wird analog
Beispie13 aus Theobrominmalonsäure-bis-(ß-oxyäthylamid) und rauchender Salpetersäure
hergestellt. F. über 300'C, umkristallisiert aus Acetonitril. Ausbeute: 510/,
der Theorie. Beispiel 6 Aus 16,9 g ß-Nitroxyäthylaminnitrat wird durch Zersetzen
mit einer Lösung von 4 g Natriumhydroxyd und 30 ml Wasser bei 0° C eine ätherische
Lösung der freien Base bereitet und mit 6,4 g Theophyllinessigsäurechlorid in wasserfreiem
Dioxan umgesetzt. Der ausfallende Niederschlag wird isoliert und aus wenig Alkohol
umkristallisiert und mit 1 °/»iger wäßriger Ammoniaklösung und anschließend mit
Wasser gewaschen. Der Salpetersäureester des Theophylün-7-essigsäure-ß-oxyäthylamids
kristallisiert in weißen Nadeln vom F. 157 bis 159° C.
