Verfahren zur Herstellung eines neutralen Piperaziniumsalzes Bisher
sind neutrale Salze der Citronensäure mit organischen Aminen, die also für jedes
Mol Citronensäure drei Äquivalente Amin enthalten, noch nicht als chemisch einheitliche
Verbindungen hergestellt und beschrieben worden. Im Zentralblatt, 1903, 1I, S. 566,
wird aus den Berichten der Deutschen pharmazeutischen Gesellschaft, Bd. 13, S. 151
bis 153 eine Arbeit von A n s e 1 m i n o referiert, in der festgestellt wird, daß
Citronensäure mit aliphatisch gebundenen Aminogruppen einfach saure Salze bildet,
in denen je zwei Äquivalente eines aliphatischen Amins mit jeweils 1 1-Iol Citronensäure
reagiert haben. Aromatische Amine hingegen, in denen die Aininogruppe unmittelbar
an einen aromatischen Kern gebunden ist, ergeben zweifach saure Citrate, in denen
auf 1 Mol Citronensäure ein Äquivalent eines aromatischen Amins entfällt. Pvridin
verhält sich wie ein aromatisches Amin und bildet nur ein zweifach saures Citrat.
Stöchiometrisch neutrale Citrate von irgendwelchen Aminen wurden in keinem Falle

Versuche, aus einer höchstkonzentrierten wäßrigen Lösung, die auf ein Mol Citronensäure
anderthalb Mole Piperazin enthält, das stöchiometrisch neutrale Piperaziniumcitrat
etwa durch Zusatz eines finit Wasser mischbaren organischen Lösungsmittels, z. B.
Alkohol oder Aceton, herauszudrücken. Auch in diesem Falle scheidet sich bei Zugabe
einer sehr großen Menge eines der genannten wassermischbaren organischen Lösemittel
nur das einfach saure Piperaziniuincitrat aus, während das dazu nicht benötigte

sich merkliche Mengen des flüchtigen Piperazins ebenfalls verflüchtigen. Man erhält
bei Verwendung entsprechend reiner Ausgangsmaterialien dann ein schneeweißes Salz,
das bei der Analyse Werte ergibt, die einem stöchiometrisch neutralen Piperaziniumcitrat
mit 1,5 Mol Kristallwasser entsprechen. Dieses bisher noch unbekannte Salz ist an
der Luft mit normalem Feuchtigkeitsgehalt nur sehr wenig hygroskopisch, nimmt dagegen
aus feuchtigkeitsgesättigter Luft Feuchtigkeit auf und wird schmierig. Es löst sich
spielend in Wasser gegen Lakmus mit nur schwach saurer Reaktion, schmeckt angenehm
säuerlich und verursacht weder Reizungen der :Mundschleimhaut und Schluckbeschwerden

zwischen 35 und 55° C, zweckmäßig eine solche zwischen 40 und 50° C, ist optimal,
wobei mit dem verdampfenden Wasser kaum wägbare Mengen Piperazin sich verflüchtigen,
falls man keinen Piperazinüberschuß genommen hat. Beispiel 1 Zu 194 Gewichtsteilen
reinem Piperazinhydrat gibt man bei Zimmertemperatur unter gutem Rühren eine Lösung
von 140 Gewichtsteilen reiner kristallisierter Citronensäure, die 1 Mol Kristallwasser
enthält, in 500 Gewichtsteilen destilliertem Wasser. Die entstandene völlig klare,
farblose homogene Lösung wird dann unter 10 bis 20 mm Druck bei einer 55° C nicht
überschreitenden Temperatur zur Trockne gebracht, worauf man den Rückstand unter
3 bis 5 mm Druck noch weitere 8 Stunden bei 50° C hält. Das stöchiometrisch neutrale
Piperaziniumcitrat fällt dann als schneeweiße, porzellanartige spröde Masse an,
die sich leicht pulverisieren läßt. Die Analyse zeigt, daß es sich um das stöchiometrisch
neutrale Salz handelt mit einem Verhältnis von 12 Kohlenstoffatomen zu 3 Stickstoffatomen,
das 1,5 Mol Kristallwasser enthält. Das Produkt zieht an trockener Luft kaum Feuchtigkeit
an, wird jedoch an feuchter Luft bald klebrig. Es löst sich spielend mit nur schwach
lakmussaurer Reaktion in Wasser, ist dagegen in Äther und Kohlenwasserstoffen unlöslich.
In feinstgepulvertem Zustand wird es durch Kochen mit Alkohol in sich lösendes Piperazin
und ungelöst bleibendes einfach saures Piperaziniumcitrat zersetzt, so daß die alkoholische
Lösung dann gegen Phenolphthalein sehr stark alkalisch reagiert. Im Schmelzpunktsröhrchen
bleibt das Produkt bis etwa 130° C unverändert, zersetzt sich dann aber langsam
und ist bei 195° C völlig geschmolzen. Es hat einen angenehmen, schwach säuerlichen
