Description
Process for the preparation of solutions for blood plasma replacement It is known to use gelatin solutions as solutions for blood plasma or serum substitutes. However, the Gela.tine solutions used so far have some disadvantages: Either they are solid at room temperature - then they have to be liquefied by heating before use and the associated loss of time can be decisive in the desired effect endanger - or one has to get a solution that is liquid at ordinary temperature obtained, the gelatin, with chemicals, e.g. B. with hyd, rolyzing agents, degraded. So far, however, this has always resulted in such a strong reduction in the average molecular weight associated with gelatin, the retention period of such a solution is intolerable was shortened. You have already degraded gelatine through a secondary one Condensation process (e.g. with succinic anhydride or the anhydride or chloride another polycarboxylic acid) built up again into larger molecules or gelatin z. B. condensed with glyoxal, acetaldehyde or quinone at pH 7 to 4.5 and then oxidized with hydrogen peroxide at pI 7.5 to 5.5 or treated with formaldehyde Gelatine partially degraded with protein-splitting agents; these procedures are however, all very cumbersome. In addition, such solutions are often exposed Table salt is used for isotonic reasons, so that such solutions are suitable for certain cases (e.g. in nephrotic diseases) cannot be used because table salt is used here is contraindicated.
The invention is concerned with the task of producing a plasma or serum substitute suitable gelatin solution by a simple process, that meets the following requirements: The gelatine may only be used to a limited extent be reduced so that the retention period is not significantly reduced; she must also not require the addition of table salt: it should be physiologically well tolerated and be liquid at room temperature, and their stability should also after addition therapeutically valuable additives common to blood substitute fluids are obtained stay.
According to the invention, these conditions are met in that one an aqueous gelatin solution with glucamine, which gelatinizes at room temperature, of low molecular weight N-alkyl or N-aralkyl glucatnin alone or in the presence of about 2 equivalents (based on glucamine) a physiologically indifferent, preferably dibasic Acid or treated with tatirin, preferably with warmth. This treatment is supposed to be continued at least until a liquid remains liquid at room temperature Solution has been reached, but no longer than the point in time at which the average molecular weight of gelatin - measured by conventional methods - is reduced by about 20%.
The 1 \ T-alkylated and N-aralkylated glucamines can, for. B. by Hydrogenation of a mixture of glucose and the corresponding primary amines after by Ka r r e r and H e r k e n r a t h (Helv. Chim. Acta, vol.20, p.83 [1937]) described method. Glucamine itself can be obtained through hydrogenation Gain cleavage of its N-benzyl derivative.
Als physiologisch indifferente Säuren kommen z. B. H" S 04, Adipinsäure, Bernsteinsäure usw. in Frage. As physiologically indifferent acids come z. B. H "S 04, adipic acid, Succinic acid etc. in question. A gelatin solution treated as described above you can still use other therapeutically valuable substances, such as. B. Vitamins, Hormones, Rutin or its water-soluble derivatives, such as. B. rutin-sulfuric acid ester sodium salt, Add glucose, fructose, etc. without affecting the stability of the solution will. They have a very wide range of indications, because they are preferred Amounts of chemicals (glucamine etc) used make the final solution isotonic and therefore an addition of common salt is not necessary.
The method according to the invention has the particular advantage that the added substances can remain in the solution and thus no separation processes are necessary.
The gelatin solutions obtained thereafter have correspondingly The gelatin has a sufficiently high average molecular weight long retention period; on the other hand, they are liquid and at room temperature prove z. B. also remains liquid. often used as a blood substitute Superior to dextran solutions. This can be seen e.g. B. clearly when examining the dog according to S c h e g a. In this examination (Contributions to Clinical Surgery, Vol. 188, P. 109 [1954]) is after Replacement of the withdrawn blood by the Blood plasma replacement fluid the edema readiness on a mobilized intestinal loop tested, the gelatin solutions according to the invention proved to be advantageous.
