In einem 2 mL-Eppendorf-Reaktionsgefäß werden zu 1 mL Puffer (50 mM; pH 7.0) nacheinander 0.392 U der Monooxygenase mit der Sequenz ID NO. 2 (als lyophilisierter Rohextrakt; 0.0098 U/mg bezogen auf Cyclooktan), 0.1 mmol Cyclooktan (entsprechend einer Substratkonzentration von 0.1 M), 10 μL Isopropanol (0.13 mmol), (ca.) 1 mg Magnesiumchlorid und 20 U einer Alkoholdehydrogenase aus Lactobacillus kefir (20 statt 0 μL) gegeben und danach wird die Reaktion durch Zugabe von 0.01 mmol des Cofaktors NADP+ (6.5 mg) gestartet. Nachdem das Reaktionsgemisch mit Hilfe eines Thermomixers für 67 Stunden bei einer Reaktionstemperatur von 25°C geschüttelt wurde fügt man 1 mL Dichlormethan hinzu und extrahiert. Anschließend wird im Vakuum die organische Phase vom Lösungsmittel befreit und aus dem erhaltenen Rohprodukt direkt der Umsatz mit Hilfe der 1H-NMR-Spektroskopie bestimmt. Der Umsatz liegt bei 79% bezogen auf die Bildung von Cyclooktanon, wobei die Menge an gebildetem Cyclooktanol bei < 5% liegt.In a 2 mL Eppendorf reaction vessel, 0.392 U of the monooxygenase with the sequence ID NO.
