Abstract
Durch das erfindungsgemaesse Verfahren werden monoklonale Antikoerper gegen humanes Interleukin-2 mit hoher Affinitaetskonstante und hoher Spezifitaet hergestellt. Milzzellen von Maeusen, die mit humanem Interleukin-2 immunisiert wurden, werden mit Maus-Myelomzellen fusioniert. Die Hybridomzellen werden mit Hilfe immunchemischer Screeningverfahren selektioniert und kloniert. Hinsichtlich ihrer Reaktivitaet mit dem Interleukin-2 werden die Eigenschaften der erhaltenen monoklonalen Antikoerper bestimmt. Die Produktion der Antikoerper durch die Hybridomklone wird entweder in vitro mit biotechnologischen Verfahren der Zellkultur oder in vivo in Ascitesform in Maeusen durchgefuehrt. Die spezifischen Hybridome wurden im ZI fuer Molekularbiologie (Liste 1987) unter den Nummern 0173 bis 0192 hinterlegt. Anwendungsgebiete sind die Medizin und Immunologie.By means of the method according to the invention, monoclonal antibodies to human interleukin-2 are produced with high affinity constant and high specificity.
