Abstract
Verfahren zur Herstellung von 3-Morpholinoformyloxy-cardenoliden und 3-Morpholinoformyloxy-cardenolidanalogen, geeignet, als Herz-Kreislauf-Pharmaka. 3-Hydroxy-cardenolide bzw. 3-Hydroxy-cardenolidanaloge werden mit einem reaktiven Derivat der Kohlensaeure zu Chlorameisensaeureestern der 3-Hydroxy-steroide bzw. zu reaktiven gemischten Estern der 3-Hydroxy-steroide umgesetzt und anschliessend die reaktiven Reste gegen Morpholin ausgetauscht.Process for the preparation of 3-morpholinoformyloxy-cardenolides and 3-morpholinoformyloxy-cardenolide analogues suitable as cardiovascular drugs. 3-hydroxy-cardenolides or 3-hydroxy-cardenolide analogues are reacted with a reactive derivative of carbonic acid to form chloroformates of 3-hydroxy-steroids or to reactive mixed esters of 3-hydroxy-steroids and then the reactive radicals are replaced by morpholine.

Die erfindungsgemäßen Verbindungen zeichnen sich durch gute Herzwirksamkeit, insbesondere durch eine gute Kontraktilitätssteigerung bei gleichzeitiger Erhöhung der therapeutischen Breite im Vergleich zu den Standardglykosiden Digitoxin und Digoxin aus. Die positiv inotrope Wirkung (Verbesserung der Kontraktionskraft des Herzens) läßt sich an verschiedenen Tierspezies (Meerschweinchen, Katzen, Schweinen und Hunden) nachweisen. Der Rückstand (1 g) wird einer Gradientenchromatographie an Kieselgel mit Benzen-Ether und Benzen-Ether-Aceton unterworfen, man erhält 470 mg ^-Nor-digitoxigenin-S-morpholinoformiat, das man aus Acetonitril umkristallisiert.
