Abstract
Das Verfahren zur Herstellung eines Eiweisshydrolysates ist anwendbar in bakteriologischen Laboratorien und Impfstoffwerken, in denen Bakterien fuer diagnostische Zwecke oder zur Gewinnung spezifischer Immunogene gezuechtet werden. Bei diesem Verfahren wird durch die Vorspaltung mit Pankreas die anschliessende Hydrolysezeit wesentlich verkuerzt. Die Entfernung der Schwefelsaeure wird umgangen, indem ein Anteil Konzentrattrypton mit Salzsaeure und ein anderer gleicher Teil mit Kalilauge hydrolysiert wird. Nach der Hydrolyse erfolgt die Neutralisation durch Mischen beider Anteile. Bei hoher Caseinkonzentration kann das entstandene Kaliumchlorid im Caseinhydrolysat verbleiben, da der Anteil die uebliche Konzentration im Naehrboden nicht uebersteigt. DD 159844 verwendeten durch schwefelsaure Druckhydrolyse aus Casein gewonnene Aminosäuregemische in Kombination mit anderen Eiweißkomplexen in bakteriologischen Nährmedien. Totalhydrolysate haben gegenüber enzymatischen Hydrolysaten den Nachteil, daß sie allein den Bakterien nicht ausreichend Bedingungen zur Vermehrung bieten. Säurehydrolysate enthalten kein Tryptophan, während basische Hydrolysate kein Arginin und keine Vitamine mehr enthalten. Außerdem liegen auf Grund der langen Einwirkungszeit alle Aminosäuren als DL-Verbindungen vor, die für Bakterien wenig geeignet sind. Durch die lange Einwirkungszeit von Basen oder Säuren auf Proteine zurtotalen Spaltung (bis 24 Stunden) steigen die Produktionskosten durch den hohen Energiebedarf stärker als bei enzymatischen Hydrolysen (REISSBRODT u. Die so hergestellten Eiweißhydrolysate sollen die für die Bakterienernährung nötigen Substanzen in ausreichenden Mengen enthalten. Erfindungsgemäß wird die Aufgabe dadurch gelöst, daß das Protein, vorzugsweise Casein, Gluten und/oder Keratin erst nach bekannten Verfahren mit den Enzymen der Pankreasdrüse oder anderen Proteasen gespalten wird und anschließend die Totalhydrolyse mit Säuren bzw. Laugen in den bekannten Konzentrationen erfolgt. Damit durch die Totalhydrolyse der Verlust an Aminosäuren möglichst klein gehalten wird, wird ein Teil mit Lauge und ein andererTeil mit Säure gespalten. Die Spaltung mit Säure erfolgt durch Salzsäure und mit Lauge durch Kalilauge. Nach der Hydrolyse werden beide Ansätze so gemischt, daß sie neutral reagieren. Durch Verwendung von Kalilauge zur Hydrolyse entsteht durch Neutralisation ein Kaliumsalz. Das Kaliumsalz, z. B. Kaliumchlorid braucht aus dem Hydrolysat nicht entfernt zu werden, da KCI ohnehin den Nährböden zugesetzt wird. Um den Gehalt an KCI im Verhältnis zum Eiweißhydrolysat gering zu halten, wird eine 30%ige Tryptonkonzentratlösung zur Hydrolyse verwendet.The invention has for its object to provide a method for producing a protein hydrolyzate with little technical effort that provides hydrolyzates with high nutritional value.
