Abstract
Description
Způsob výroby 3-karbonylandrostadien-17-karboxamidů a nový meziproduktA process for the preparation of 3-carbonylandrostadiene-17-carboxamides and a novel intermediate
Oblast technikyTechnical field
Vynález se týká nového postupu pro přeměnu substituovaných steroidních 3-halogen-3,5-dienových derivátů na substituované deriváty steroidních 3,5-dien-3-karboxylových kyselin Uvedené sloučeniny byly popsány v US patentovém spisu č.The present invention relates to a novel process for converting substituted steroid 3-halo-3,5-diene derivatives into substituted steroid 3,5-diene-3-carboxylic acid derivatives.
No. 017,568 of May 21, 1991 (Holt et al.) As useful for inhibiting steroid 5-alpha-reductase.
Dosavadní stav technikyBACKGROUND OF THE INVENTION
Uvádí se, že výhodnou baží pro tvorbuNumerous processes have been described for producing substituted steroid 3,5-diene-3-carboxylic acid derivatives from substituted intermediates, steroid 3-halo-3,5-diene derivatives. In particular, the carboxylation of steroid 3-bromo-3,5-diene intermediates is used catalytically or in the presence of alkyllithium to form steroid 3,5-diene-3-carboxylic acid derivatives. The products are obtained in a yield of 15% when N-butyllithium is used, the procedure described in U.S. Patent No. 5,017,568. It has been shown that using a basic medium for selective deprotonation of hydrogen acid atoms in the thickened intermediate in the above reaction before adding the dehalogenation The yield of the resulting steroid 3,5-diene-3-carboxylic acid derivative may be increased, as described in U.S. Patent Application Serial No. 07 / 817,179, filed Jan. 6, 1992, the yield being 63% for N-tertiary acid production. Butylandrost-3,5-diene-17β-carboxamide-3-carboxylic acid from N-tert-butlandrost-3,5-diene-3-bromo-17β-carboxamide. It is said to be a preferred base for formation
2bazického prostředí je ethylmagnesiumbromid a ethylmagnesiumchlorid. The 2-base medium is ethylmagnesium bromide and ethylmagnesium chloride.
In addition to the overall low yield, a major disadvantage of these processes is the fact that N-butyllithium, ethylmagnesium bromide and ethylmagnesium chloride are expensive reagents that make the process uneconomical. In addition, N-butyllithium is flammable and the carboxylation reaction must be carried out at dilute concentrations. Thus, there is a need for a safe, cost-effective and rapid process for converting substituted steroid 3-halo-3,5-diene derivatives to substituted steroid 3,5-diene-3-carboxylic acid derivatives.

The most preferred value for R 1 when performing the above 2 3 2 3 group of said process is a beta-CONR R, wherein R and R are independently hydrogen, alkyl of 1-8 carbon atoms, cycloalkyl of 3 to 6 carbon atoms or phenyl.
Sloučeniny obecného vzorce I obsahují R1 nebo skupiny, které je chemicky možno na skupinu Rx převést známými reakcemi , například těmi, které jsou popsány v publikaci Derek Bartoň a U. D. Reactions to convert some of the groups to the R group can be carried out on the resulting products or on some of the intermediates during the process. For example, carboxylic acid substituents can be converted to carboxamides by conversion to an acid halide, which is then reacted with an amine. The esters can be converted to the acid and worked up as described above. Nitriles can be converted to carboxamides by reaction with an alkylating agent, for example tert-butyl acetate or tert-butanol, catalyzed using an acidic medium.

Tato koncentrace se následným doplněním objemu opakuje.100 ml of methylene chloride and 6.12 ml, 2.5 molar equivalents of dimethylformamide were charged to the flask under nitrogen. The solution is cooled to 0-5 ° C and then 6.9 ml (2.5 molar equivalents) of oxalyl chloride are added, maintaining the temperature in the range of 0-10 ° C. A white precipitate formed. The mixture was stirred for one hour and 50.1 g (19.6 molar equivalents) of hydrogen bromide gas was bubbled through the solution while maintaining the temperature between 0 and 10 ° C. The suspension becomes a clear colorless solution. The solution is degassed by vacuum distillation, reducing the volume of the solution by approximately one-half, then bringing the solution to its original volume by adding methylene chloride. This concentration is repeated by replenishing the volume.
Výtěžek je 89 %, teplota tání 181 ažl83 °C.To the resulting white suspension was added 10.0 g, 1 molar equivalent of androst-4-en-3-one-17-beta-carboxylic acid, and the mixture was warmed to room temperature and stirred for 2 hours. The reaction mixture was then poured into a vessel containing 100 mL of methylene chloride and 23.1 g, 10 molar equivalents of tert-butylamine, maintaining the temperature in the range of 0-10 ° C. The mixture was stirred for 30 minutes, then about 100 ml of water was added and the biphasic mixture was filtered through a pad of celite. The organic phase was separated and the volume reduced by half by distillation in vacuo, then the solution was made up to the original volume with acetone. This concentration followed by volume replenishment was repeated two more times. Do baňky s objemem 5 litrů, se třemi hrdly (Morton), : opatřené mechanickým míchadlem, teploměrem a zpětným chladičem se vloží 250 g N-terc.butylandrost-3,5-dien-3-brom13In a 5-liter, three-necked (Morton) flask equipped with a mechanical stirrer, a thermometer and a reflux condenser was charged 250 g of N-tert-butyllandrost-3,5-diene-3-bromo13
-17beta-karboxamidu, připraveného způsobem podle příkladu 1 i), 55 g kyanidu měďného, 29 g kyanidu sodného a 1 litr dimethylformamidu. Pevný podíl se oddělí, promyje se 75 ml 50% vodného alkoholu a suší při 60 až 65 °C ve vakuu, čímž se ve výtěžku 88 % získá 191,3 g produktu s teplotou tání 190 až 192 °C.17beta-carboxamide, prepared according to the method of Example 1 i), 55 g of copper (I) cyanide, 29 g of sodium cyanide and 1 liter of dimethylformamide. The reaction mixture is heated to reflux at about 152-153 ° C for at least 12 hours. The reaction mixture was slowly cooled to 25-30 ° C in a cold water bath, then quenched by the rapid addition of 1 liter of aqueous ammonium hydroxide (50:50 concentrated ammonia / water) with vigorous stirring. The mixture was stirred for a further 15-20 minutes and then 1 liter of methylene chloride was added and the two-phase system was allowed to separate into two phases. The phases are separated and the aqueous phase is extracted again with 2 x 500 ml of methylene chloride. The methylene chloride extracts were combined and passed through a pad of celite to remove insoluble copper salts. The celite pad was washed with methylene chloride (150 ml). The methylene chloride phases were combined and washed with 3 x 500 mL ammonium hydroxide to remove the last traces of copper salts. The organic phase is then concentrated by distillation at atmospheric pressure, leaving approximately 1.5 liters of methylene chloride solution. 600 ml of ethanol are added to the reaction vessel and the solution is again concentrated to remove methylene chloride by distilling off an additional 500 ml of solvent. An additional 600 ml of ethanol was then added to the reaction mixture and distillation at atmospheric pressure was continued until the vapor temperature reached 82-84 ° C. 60 ml of water are then added and the resulting suspension is cooled to 0-5 ° C for at least two hours. The solid was collected, washed with 75 mL of 50% aqueous alcohol, and dried at 60-65 ° C under vacuum to give 191.3 g (88%), m.p. 190-192 ° C.
