具体实施方式
实施例1、构建本发明的重组质粒pETDuet-CYP-POR表达的超级工程菌
本发明提供的含有细胞色素氧化还原酶P450 CYP9G2和细胞色素P450还原酶的基因的超级工程菌的克隆如下:
1.1质粒和受体菌
小菜蛾CYP9G2基因的克隆
1)总RNA的提取
1-1)按TRIzol试剂盒操作程序,提取小菜蛾幼虫总RNA:2RT-PCR反应
1-2)用反转录的第一链cDNA作为模板进行PCR反应,向PCR反应管中加入以下试剂,轻轻混匀反应体系;

50mM MgCl2 0.75μl
10mM dNTPs mixture 0.5μl
Forward primer(10μM) 0.5μ1
Reverse primer(10μM) 0.5μl
cDNA 1μl
Taq DNA polymerase(5U/μl) 0.5μl
autoclaved distilled water 18.75μl
1-3)PCR反应的反应程序为:95℃热变性5min,然后94℃30s,60℃退火1min,72℃延伸2min,共30个循环,最后进行72℃延伸10min;
1-4)用1%的普通琼脂糖电泳,检查PCR扩增产物。
2)cDNA文库的构建、扩增、筛选等参照Sambrook等的方法进行,具体步骤如下:
2-1)用序列特异性引物筛选基因文库,根据RT-PCR产物测序的结果,设计一对序列特异性引物,用逐步富集文库中阳性克隆的方法,利用变性的文库作为模板,进行PCR(Lardelli et al,1994);

反应条件:
95℃热变性5min,然后94℃保持30s,62℃保持30s,72℃延伸1min,共30个循环,最后进行72℃延伸10min;
PCR反应体系如下:

2-2)阳性噬菌体插入序列的测序:
为了对阳性的噬菌体文库进行测序,必须将阳性的噬菌体载体(λTriplEx)转换为可以测序的质粒载体(pTriplEx)。
首先将重组的阳性噬菌体和大肠杆菌BM25.8共同培养,让噬菌体转染BM25.8,而BM25.8在31℃生长时,可以产生一种Cre重组酶,该酶能使噬菌体的插入片段经过位点特异性的重组,使插入片段准确地重组到质粒载体的loxP位置上。这样转染后重组的质粒载体就可以在大肠杆菌中大量繁殖,因此可以得以测序。
2-3)cDNA和λTripIEx2载体左右臂的连接
取1μl载体,1μl对照插入片段,1.5μl去离子水及其他相关试剂。16℃连接过夜。包装并测定滴度,滴度应不小于107pfu/μg;按下表(cDNA与载体连接反应的优化方案)在3个0.5ml离心管中加入试剂,轻轻混匀混合物,稍离心收集内容物于管底,16℃连接过夜即可得到噬粒λTriplEx2-CYP9G2。
将此噬粒λTriplEx2-CYP9G2用作P450基因的模板,质粒pGEM-POR作为P450还原酶基因来源,构建方法如上。Escherichia coli strain BL-21(DE3)用作表达菌,质粒pETDuet购自Novagen公司用作表达CYP基因和POR基因的载体,该质粒在大肠杆菌E.coli.DH5α增殖,所有宿主菌株BL-21(DE3)。
所述的细胞色素氧化还原酶P450 CYP9G2为具有图1所示的核苷酸序列,也可以是从抗有机磷磷的小菜蛾中克隆得到。构建对农药具有抗性的小菜蛾(diamondbackmoth)cDNA文库,从文库中钓取细胞色素氧化还原酶P450,CYP9G2,提交GenBank注册(Accession number:AB096739),命名为CYP9G2。
所述的细胞色素P450还原酶的基因为具有图2所示的核苷酸序列,也可以从家蝇中克隆得到。根据家蝇的NADPH-细胞色素P450还原酶的cDNA序列(GenBank登录号:L19897)设计引物,从单个家蝇提取的mRNA合成cDNA,以此cDNA为模板,用RT-PCR的方法获得家蝇细胞色素P450还原酶基因,命名为POR。
1.2工具酶及试剂
Ex Taq DNA聚合酶、各种限制性内切酶BamHI、PstI、BglII、XhoI和T4 DNA连接酶均为TaKaRa产品;连二亚硫酸钠、NADPH和DTT(1,4-二硫代苏糖醇)购自上海生物工程有限公司;δ氨基γ酮戊酸(δ-Aminolevulinic acid,δ-ALA)购自百灵威公司。
1.3培养基和培养条件
细菌增殖培养所使用培养基为LB培养基,含质粒菌体的培养基中加氨苄青霉素终浓度为50μg/ml,经转化重组质粒的细菌培养至菌体密度为OD600=0.6时加入终浓度为1mM IPTG,同时加入δ-ALA(500mM)50μl,使终浓度为0.5mM在30℃继续诱导培养。
1.4表达载体的构建
根据本研究组在GenBank(登录号:AB096739)注册的小菜蛾细胞色素P450基因的cDNA序列,设计了一对特异引物,并在引物上引入了不同的限制性内切酶位点。以噬粒λTriplEx2-CYP9G2为模板作PCR,扩增CYP 9G2基因。引物由上海博雅生物工程公司合成,序列为:

上游引物引入BamHI位点,下游引物引入PstI位点,5`端各引入两个酶切位点保护碱基。
根据家蝇的还原酶cDNA序列(GenBank登录号:L19897)设计引物,以本研究室保存的pGEM-POR载体为模板作PCR,扩增POR基因。引物由上海博雅生物工程公司合成,序列为:

上游引物引入BglII位点,下游引物引入XhoI位点,5`端各引入两个酶切位点保护碱基。
PCR扩增体系:取模板2μl加入上游引物1μl、下游引物1μl、2.5mM dNTP2μl、10×buffer 2.5μl、ExTaq DNA聚合酶0.25μl,补充去离子水至总反应体系为25μl,进行PCR扩增,PCR反应条件:
扩增CYP9G2基因PCR反应条件:
95℃ 3min 1个循环
94℃ 1min
58℃ 1min 30个循环
72℃ 1min
72℃ 7min 1个循环
扩增POR基因PCR反应条件:

经PCR扩增得到的CYP 9G2基因和POR基因分别插入双表达载体pETDuet的第一和第二多克隆位点,并且保证两个基因阅读框正确。CYP 9G2基因自身具有完整的开放阅读框,并且与载体上的His-Tag融合表达。POR基因自带起始密码子,它的终止密码子克隆时被缺失,在载体上与S-Tag序列正确通读,使用载体上自带的终止密码子,使表达的B1蛋白携带有S-Tag标签。两个基因均带有融合标签,以利于表达产物的检测和纯化。
构建程序如图3所示,经过克隆程序,得到融合表达载体pETDuet-CYP-POR。
1.5转化大肠杆菌BL-21(DE3)
将构建得到的表达载体pETDuet-CYP-POR转化大肠杆菌BL21(DE3),得到重组菌,即本发明的超级工程菌,已于2005年11月10日保藏于《中国微生物菌种保藏管理委员会普通微生物中心》,其保藏号为CGMCC No.1529。
1.6解毒酶的制备与提取
从新鲜转化的平板挑取单克隆菌落,加入含50μg/ml氨苄青霉素的2ml LB培养基37℃震荡培养8-10hr,然后将2ml培养液注入50ml含氨苄青霉素的培养液继续培养。当细胞的浓度OD600=0.6时加入500μl IPTG(浓度为100mM)使IPTG终浓度为1mM,加入δ-ALA(500mM)50μl,使终浓度为0.5mM,用异丙基硫代半乳糖苷(IPTG)诱导表达,在25℃诱导,继续培养22小时。5000×g离心5min收集经诱导的细菌,重新用PBS调节细菌的浓度为OD600=1,5000×g离心5min重新收集细胞,再次加入1/10体积(OD600=1)的PBS,使菌液浓缩倍数为10×,细菌菌液中加入溶菌酶使其终浓度为100μg/ml,加入1/10体积的1%Triton X-100,30℃温浴15min,在冰浴条件下超声波裂解处理,得到本发明的解毒酶。
以类似的方法,构建融合表达载体pETDuet-CYP,并转化到大肠杆菌BL-21(DE3),得到CYP9G2 P450单表达基因工程菌。以同样条件进行诱导表达,得到以CYP9G2P450单表达P450 9G2的酶。
用12%SDS-PAGE进行分析,电泳完毕后将凝胶用考马斯亮蓝染色,然后用脱色液脱色,同时用薄层扫描仪分析目的蛋白的相对含量。结果如图4所示,由各种菌在E.Coli BL21(DE3)中表达的SDS-PAGE分析图可知本发明的超级工程菌诱导表达产生的解毒酶的效率与细胞色素氧化还原酶P450 CYP9G2(CYP9G2)单独表达的效率是一致的。
实施例2、本发明的解毒酶对杀螟硫磷(fenitrothion)、乐果(dimethoate)、甲基对硫磷(methyl parathion)等的降解作用的测定
在100ml三角瓶中加入20ml水,1ml 2%TritonX-100,加入适量农药的乙醇溶液,使农药的终浓度为10ppm,加入0.02M,pH7.0磷酸缓冲液补齐体积,使总体积为10mL。将其充分混匀后加入4mL实施例1制备的解毒酶粗酶液(约合30mg总蛋白),置30℃-32℃摇床摇动。分别在0min,20min,40min,60min,80min和120min取样。每个时间点取两个重复,实验以实施例1制备的CYP9G2 P450单表达P450 9G2的酶为对照组。取出1ml处理液置于5ml试管中,加入1ml重蒸正己烷,在混旋器上振荡3min充分混匀、提取,然后于6500rpm离心15min,取上清液,重复提取一次,将取出的上清液合并后使用气相色谱仪和氮磷检测器测定不同时间正己烷提取液中杀螟硫磷的残留量的含量,以检测解毒酶对杀螟硫磷的降解速率,具体测试条件如下:使用惠普5890II型气相色谱仪,配自动进样器(HP 7673 autosampler),载气:N2气,1mL/min流量,检测器:氮磷检测器(GC-NPD)和色谱柱:fused silica capillary column(0.53mm i.d.×30m×0.5μm film thickness,Supelco Corp.USA),进样口温度:300℃,检测气温度:300 ℃,柱温:220℃,分流不分流:不分流进样,进样量:1μL。其实验结果列于表1。

1.本实验以实施例1制备的CYP9G2 P450单表达P450 9G2的酶作为对照组。
2.降解效果中,“+”为与对照组相比较有显著降解效果,“-”为与对照组相比较降解效果不显著。
3.其中,敌敌畏(dichlorovos)在对照组中有降解,为41%,与以CYP9G2 P450基因和NADPH双表达在E.coli中的实验组降解数据39%,相接近,刨除误差等因素,视为无降解效果。原因可能有两点:a)单表达的P450酶或E.coli中原有的酶对敌敌畏有降解作用。b)敌敌畏在实验水溶液所给定的条件下,自身能迅速降解。
图5为本发明的解毒酶对杀螟硫磷的降解曲线。如图所示的结果可知,本发明的解毒酶在2小时可以降解64%杀螟硫磷,而同样条件下测得对照样品没有任何降解。从而可以可知:超级工程菌用一次诱导表达发酵培养的时间同时得到2种酶,p450氧化酶和p450氧化还原酶,该2种基因共表达后得到的2种酶同时都具有活性,如图显示的那样只使用p450氧化酶对杀螟硫磷没有任何降解效果,说明该氧化酶的降解活性必须有p450 NADPH还原酶的参与才能使反应进行。
本研究可以通过1次发酵过程得到具有生物活性的酶,大大减少了诱导表达发酵培养生产工程菌所需要的各种原材料,缩短时间,节约能源,降低成本,这样便可以提高在单位时间内大大降解毒物的速率,在处理水污染时将可以有效降低运行的成本,减少开支。
实施例3、本发明的解毒酶对氨基甲酸酯类杀虫剂的的降解作用的测定

由表2中的数据可知,本发明提供的解毒酶对氨基甲酸酯类的农药的降解效果不够理想,在所测定的农药中只对克百威和抗蚜威的效果较明显,对其他的降解效果较差。
