发明背景

发明概述

在制造过程中,通过分步梯度离心从人供体“白细胞层”分离单核细胞,用PHA培养以增加培养物中来自供体白细胞的IL-2和其它细胞因子的产生和分泌,如在美国专利5,093,479、4,390,623、4,388,309、4,406,830、4,661,447、4,681,844和4,464,355中公开的,上述文献通过引用合并在本文中。随后,无菌地收获培养物上清液,澄清和进行批量的病毒排阻过程。然后通过超滤作用和微滤将上清液进一步浓缩约10倍。

施用前进行单次环磷酰胺的静脉内输注,在第一次 施用前三天输注300mg/m2。自己口服施用(与食物一同)吲哚氯甲酰(25mg),每天三次,在环磷酰胺施用后3天开始,直到手术前24小时。在环磷酰胺施用后3天直到手术前24小时,自己施用硫酸锌(50mg)和多种维生素补充物,每天一次。建议并鼓励病人手术后继续自我施用多种维生素和锌的服用。这些药剂对肿瘤细胞周 期没有任何影响,以低于这些药物的正常癌症治疗3-5倍的剂量给药。
结果

比例范围3.5到4.5的GM-CSF比IL-2。

比例范围3.2-11.3的TNF-α比IL-2;
比例范围1.5-10.9的IFN-γ比IL-2;和
比例范围2.2-4.8的GM-CSF比IL-2,
