Abstract

Description
本发明涉及包含药物物质的粒状缓释(sustained release)颗粒。
发明背景
缓释药物制剂的早期公开出现在Keating的美国专利2990332中,其中使用了磺酸阳离子交换树脂。药物被离子化结合或吸附到离子交换树脂颗粒上。药物的一个要求是它包含碱性官能团。
此后,离子交换树脂药物复合物具有扩散阻碍包衣,其通过用药物治疗的人的胃液提供延缓作用。这种制剂的早期例子是Raghunathan的美国专利4,221,778,其中提供的树脂为各种聚合物基质,包括AMBERLITE IR120、阳离子交换树脂和AMBERLITE XE69,其是从AMBERLITE IR120得到的较小尺寸的树脂颗粒。提到的其它离子交换树脂有甲基丙烯酸、丙烯酸、苯酚甲醛离子交换剂与纤维素或右旋糖酐聚合物基质和无机离子交换基质。在’778专利中,使用乙基纤维素作为在离子交换树脂颗粒上的可透水的扩散阻碍包衣。

在Oshlack和Chassin的美国专利5,968,551中找到得到缓释药物的另一方法,其中通过构建一系列的大小为0.1mm至3mm的不同大小颗粒来提供阿片类物质的单位剂量。在一些实施方案中,提供球形颗粒(sphericalparticles),但然后用材料如乙基纤维素或水溶性纤维素如羟基低级烷基纤维素来包衣。公开了各种溶剂包衣方法。另外,公开了熔化制粒(meltpelletization)方法,其中阿片类物质与粘合剂和其它任选成分混合。然后用高速剪切混合器对包含阿片类物质的粘合剂材料进行造粒以得到所需的大小。
美国公开的申请2002/0031490描述了另一方法。该方法基于可用少量水水合的树脂,由此树脂吸收相对不溶于所用水量中的活性物质。在这种体系中,活性物质至少部分溶解在溶剂中。因此,还公开了水合树脂分散在活性物质用溶剂中的溶剂体系。溶剂包括有机溶剂如乙醇、二氯乙烷和1,1,2,2-四氟乙烷。
涉及包衣树脂颗粒的各种缓释制剂的其它例子包括美国专利6,001,392、6,228,398、4,996,047、4,959,219、4,847,077、4,762,709、4,690,933和EP911039。
现有技术已公开了缓释组合物和包括包衣或包衣步骤的方法。由于这类方法时间长且昂贵,因此通过不需要聚合物薄膜包衣来控制药物输送的方法提供缓释药物颗粒将是有利的。
发明概述
根据本发明,提供一种通过不需要包衣的方法生产的包含缓释离子交换树脂颗粒物质的药物。本发明的缓释颗粒包含与药物物质复合的离子交换树脂,其中药物物质嵌入(embed)在通过造粒方法结合的粒状颗粒扩散阻碍物质(diffusion barrier material)的内部和表面上。
胜于复合物的连续包衣,本发明的缓释颗粒包含扩散阻碍物质,该物质包括具有药物树脂复合物颗粒的粒状扩散物质颗粒,其中药物树脂复合物嵌入到扩散阻碍物质的内部和表面上。扩散阻碍物质与离子交换树脂复合物在造粒方法中结合,造粒工艺依赖作用在复合物和形成扩散阻碍物质的颗粒上的剪切力。通过典型手段使药物物质与合适的离子交换树脂颗粒结合,然后利用使用造粒载体的造粒机沉积这些颗粒到粒状颗粒扩散阻碍物质的内部和表面上来提供本发明的粒状颗粒缓释物质。在本发明的一种实施方案中,在造粒结束后可提供球形的缓释药物颗粒,不用进行球形化(spheronization)的进一步步骤。本领域中已知的合适筛选技术可提供所需的颗粒大小。
本发明的缓释颗粒能在哺乳动物体内提供长期(extended period)如约8小时时间的药物,但也可提供其它释放时间。通过改变树脂的药物装填量和改变颗粒中使用的扩散阻碍物质的类型和数量,可通过本发明的方法得到各种释放时间,这将在下文更充分地描述。
发明详述
如本领域中已知的,离子交换树脂颗粒可与活性药物物质反应形成复合物。阳离子交换树脂可与包含碱性成分的药物形成复合物,而阴离子交换树脂可与包含酸性成分的药物形成复合物。通常,使药物与离子交换树脂的水性悬浮液混合,并干燥复合物。复合到树脂的药物量可通过溶液pH的变化、通过复合物物理性质的其它变化或通过水相中溶解的药物浓度降低来检测。
阳离子药物带有正电荷,并往往取代阳离子基团,一般通过离子键变得复合到树脂上。由于碱性药物一般为阳离子的,因此经常使用阳离子交换树脂制备具有碱性药物的药物-树脂复合物。形成水不溶性药物-树脂复合物的一般方法是使阳离子交换树脂的钠盐与阳离子药物反应或使药物的碱形式与阳离子交换树脂的酸形式反应。
在本发明的新型实施中,可成功地使用大量不同的离子交换树脂。选择的离子交换树脂应对人无毒,并通常不应妨碍药物物质的药效。已知的用于本发明的离子交换树脂有AMBERLITE IRP69(Rohm & Haas Chemical Co.的商标)等。这种树脂为凝胶型二乙烯基苯磺酸阳离子交换树脂。在本发明的产品和方法中可既使用阳离子交换树脂又使用阴离子交换树脂。用于实施本发明的合适树脂包括由以下得到的官能化树脂:二乙烯基苯、苯乙烯、甲基丙烯酸、异丁烯酰胺、丙烯酸、丙烯酰胺、carbacrylic、苯酚甲醛、聚羟基树脂、聚羧酸、羧乙烯、纤维素和右旋糖苷聚合物树脂。也可使用两性树脂,即从上述单体得到但在同一聚合物中同时包含阴离子和阳离子位的那些。在本发明的实施中,还可使用两性离子树脂。本发明中使用的树脂颗粒的尺寸范围随所用树脂的类型变化。这种树脂尺寸范围典型为US.Std.Mesh100-400(150-37微米)。
可在半极性或极性溶剂如水中以离子形式存在的任何药物活性成分都是用于本发明的潜在备选对象。合适的药物物质包括所有的酸性和碱性药物。例子包括具有碱性基团如氨基、酰胺基、胍基和杂环基的药物。其它例子包括为羧酸或酰胺或具有羰基或其它酸性基团的药物。
大部分可用药物物质都能与离子交换树脂形成复合物。典型的药物包括但不限于盐酸羟考酮、对苯二甲酸羟考酮、马来酸氯苯那敏、可待因、吗啡、右美沙芬(Dextromethorphan)、苯丙醇胺、伪麻黄碱、酸性酒石酸氢可酮、二氢可待因、吗啡的盐和衍生物、甲基麻黄碱、盐酸曲马多、麻黄碱、对氨基水杨酸、芬特明、毛果芸香碱、甲氧氯普胺和茶碱。用于本发明的其它可能药物包括α-肾上腺素能激动剂和阻断剂;β-肾上腺素能激动剂和阻断剂;麻醉性和非麻醉性止痛药;减食欲剂;抗过敏药;抗阿米巴药;抗心绞痛药;平喘药;抗菌素如氨基糖苷类、碳头孢烯类、碳青霉烯类、头孢菌素类、头霉素类、青霉素类、多肽类、四环素类、喹诺酮类和磺胺类药;抗胆碱能药;抗抑郁药;抗真菌药;非甾体类抗炎药;解痉药;抗溃疡药;抗病毒药;抗焦虑药;钙通道阻断剂;多巴胺受体激动剂和拮抗剂;麻醉拮抗剂;蛋白酶抑制剂;呼吸兴奋剂;逆转录病毒蛋白酶抑制剂;逆转录酶抑制剂;镇静剂如苯二氮类衍生物;和脑、冠状和外周血管扩张剂。当然,根据药物的pKa,将选择阴离子或阳离子交换树脂。在一些情况下,根据药物的物理化学性质即pKa以及结合常数,可使用两性树脂。
在本发明的缓释颗粒和方法中可使用许多其它药物物质。在本领域中尤其在Keating的上述US2,990,332中能得知大量这类例子,本文引入US2,990,332作为参考。
本发明的缓释颗粒典型地包含约5%~约80%重量的药物物质。本发明颗粒的更优选药物含量为约10%~约60%重量,本发明颗粒药物含量的更优选范围为约10%~约50%重量。
在本发明的方法中,形成离子交换树脂的悬浮液,并施加药物物质到树脂颗粒悬浮液中。或者,可将离子交换树脂加入到药物物质的悬浮液或溶液中。离子力提供了药物物质与树脂的复合。
然后以使用造粒载体的低或高剪切造粒方法准备将制备的树脂-药物复合物引入到扩散阻碍物质中。造粒载体包括水以及水和醇的混合物。可通过改变混合物中醇的数量调整本发明缓释颗粒的释放曲线。在一种实施方案中,首先用含水造粒载体将扩散阻碍物质引入到造粒装置中,其中发生扩散阻碍物质的部分水合。
在将扩散阻碍物质加入到造粒设备中后,使上述树脂-药物复合物与扩散阻碍物质结合。尽管不受任何理论约束,但认为剪切力使树脂-药物复合物结合到颗粒扩散阻碍物质的内部和表面上。扩散阻碍物质不需要完全覆盖树脂-药物复合物的颗粒。
优选地,使用高剪切造粒方法。本发明方法中使用的典型高剪切造粒机为VG-5型造粒机或VG-25型;两者都由Powrex Corp制造。
在本发明的缓释颗粒中可使用任何扩散阻碍层材料。当然这类材料必须对药物物质惰性并且是无毒的。典型的扩散阻碍物质包括但不限于乙基纤维素和微晶纤维素或它们的混合物、聚甲基丙烯酸酯和聚丙烯酸酯和它们的共聚物、壳聚糖、淀粉和乳糖或淀粉或乳糖与微晶纤维素的组合物。聚甲基丙烯酸酯的例子为由Rohm Pharma GmbH以商标名Eudragit出售的物质。在一种实施方案中,扩散阻碍物质为乙基纤维素和微晶纤维素的组合物。典型地,扩散阻碍物质的标称的(nominal)平均粒度在约20至约180微米的范围内,但可使用其它粒度范围。
造粒后,干燥产品并通过典型方法分级。可使用任何合适的干燥装置如流化床干燥器。如果需要可进行产品的进一步处理。该造粒方法可提供具有不规则形状的颗粒。在一些情况下,需要球形。为了使产品成为球形,可将来自造粒工艺的产品挤成小细棒。棒典型地通过强制材料通过具有约0.5mm至约5mm范围内孔的冲模来生产,并通过典型方法制成球。提供球形颗粒的一种这类手段是将棒引入到制丸机(spheronizer)内。然后将球形物过筛至所需的粒度范围。在另一实施方案中,可在造粒结束后提供球形的缓释药物颗粒,不用进行球形化的进一步处理。例如,在用高剪切造粒机造粒时,通过颗粒进一步致密化和树脂-药物复合物在扩散阻碍物质内嵌入,颗粒生长得到控制。在颗粒干燥后,不需要进一步处理,就提供药物物质的缓释。
可在各种剂型如粉剂、胶囊、液体混悬剂或其它常规剂型中包括本发明的缓释颗粒。具体的应用为组合带有不同药物物质的缓释颗粒组合的硬或软明胶胶囊。

高剪切造粒机装备有5升的滚筒,并装有来自上述的100g HC-树脂酸盐。喷洒去离子水(100g)到复合物上,并混合约5分钟。向造粒滚筒中加入100克乙基纤维素粉末,并混合5分钟,然后加入200克微晶纤维素,其以商标名Avicel PH-101(FMC Corp.的产品)出售,并再混合6分钟。用339.9g去离子水造粒湿混合物。将湿颗粒分成两部分(A和B部分)。
通过US.Std.Mesh#6筛子筛选A部分并流化床干燥。通过US.Std.Mesh#16和#20筛子筛选干颗粒并标记为A样品。这部分的HPLC分析表明,以酸性酒石酸氢可酮重量计,氢可酮含量等于8.9%。在0.1N HCl中进行溶出试验(桨:100rpm;500ml;N=3),数据记录在下面表3中。
使用利用0.8mm冲模的LCI Laboratory Dome Granulator(DG-L1型)将未过筛的部分B挤成细棒。使用Marumerizer(Fuii Paukal Co.,Ltd制造)将细棒制成球形物。利用流化床干燥器干燥球形物。将这样产生的球形颗粒过筛,并且用HPLC分析US.Std.Mesh-16+20部分,表明颗粒包含8.9%重量的酸性酒石酸氢可酮。B部分被标记为B样品。通过上述步骤进行这部分的溶出试验,得到的数据提供在下面表1中。

实施例2
按实施例1制备一批氢可酮-树脂复合物,除了在引入到造粒工艺中时湿滤饼未被干燥而是处于湿状态。用200g乙基纤维素(Dow Chemical Co.出售的Ethocel Standard 10FP Premium)、100g微晶纤维素(Avicel PH-101)和228.42g去离子水造粒水分含量为45.43%的183.3g氢可酮-树脂复合物。通过US.Std.Mesh#10筛子将湿颗粒过筛,在流化床干燥器中干燥至最终4.2%的水分含量,并标记为C样品。然后通过US.Std.Mesh#16和#20筛子将干颗粒过筛。按实施例1通过HPLC和通过溶出试验来分析-16+20级分。数据记载在下面的表2中。
实施例3
重复实施例2的程序,除了造粒基质为只有300g Avicel PH-101和181.8g湿氢可酮-树脂复合物。最终产品标记为D样品。按实施例1通过HPLC和通过溶出试验来分析-16+20级分。数据记载在下面的表2中。
实施例4
重复实施例2的程序,除了造粒基质为200g Avicel PH-101、200g乙基纤维素和181.8g湿氢可酮-树脂复合物。最终产品标记为E样品。按实施例2将干颗粒过筛,并按实施例1所述通过HPLC和通过溶出试验来分析-16+20级分。数据提供在下面的表2中。

将粒度范围为US.Std.Mesh#100-#400的1.5kg离子交换树脂AMBERLITE IRP69的颗粒悬浮在6kg USP水中。在搅拌下向树脂悬浮液中一次加入900g酸性酒石酸氢可酮。在室温下搅拌混合物1小时,通过烧结玻璃漏斗过滤,并用USP水洗涤三次,每次1.5kg。
向Powrex高剪切造粒机的25升滚筒中装入2.55kg微晶纤维素AvicelPH-101。用0.96kg USP水润湿Avicel。造粒参数设置如下:主叶片(mainblade):200rpm,交叉螺杆(cross-screw):400rpm,加水速率:97g/min。加入全部水后,加入上述的0.806kg湿HC-树脂酸盐滤饼,并混合6分钟。用1.316kg USP水造粒混合物。造粒参数为:主叶片:60rpm,交叉螺杆:600rpm,加水速率:133g/min。在加入全部水后,再使混合物混合6分钟。通过US.Std.Mesh#4筛子过筛得到的湿颗粒,并在流化床干燥器中干燥至4.70%的最终水分含量。首先通过US.Std.Mesh#16筛子然后用#40筛子将干颗粒过筛。-16+40级分的产量为2.081kg,通过HPLC分析合并级分的氢可酮含量。产品标记为F样品。进行如上面实施例1所述溶出试验的六次重复试验,该产品的数据提供在下面的表3中。
实施例6
将离子交换树脂AMBERLITE IRP69的颗粒(1.5kg;US.Std.Mesh#100-#400)悬浮在6kg USP水中。在搅拌下向树脂悬浮液中一次加入900g酸性酒石酸氢可酮。在室温下搅拌混合物1小时,通过烧结玻璃漏斗过滤,并用USP水洗涤三次,每次1.5kg。

实施例7
将离子交换树脂AMBERLITE IRP69的颗粒(1.5kg;US.Std.Mesh#100-#400)悬浮在4kg USP水中。升温悬浮液至30℃-35℃。在搅拌下向树脂悬浮液中一次加入300g马来酸氯苯那敏。在30℃-35℃下搅拌混合物1小时,通过烧结玻璃漏斗过滤,并用USP水洗涤三次,每次1.5kg,得到湿滤饼(CP-树脂酸盐)。
向Powrex Hi-Shear造粒机的25升滚筒中装入1.5kg微晶纤维素AvicelPH-101。用0.570kg USP水润湿Avicel。造粒参数为:主叶片:80rpm,交叉螺杆:400rpm,加水速率:58g/min。加入全部水后,加入上述的3.151kg湿滤饼CP-树脂酸盐,并混合6分钟。用1.651kg USP水造粒混合物。造粒参数为:主叶片:60rpm,交叉螺杆:600rpm,加水速率:102g/min。在加入全部水后,再使混合物混合6分钟。通过US.Std.Mesh#4筛子将得到的湿颗粒过筛,并在流化床干燥器中干燥至5.31%的最终水分含量。首先通过US.Std.Mesh#16筛子然后用#40筛子将干颗粒过筛。-16+40级分的产量为1.450kg,水分含量为5.31%。产品标记为H样品,通过HPLC分析-16+40级分的氯苯那敏含量。该产品溶出速率的六次重复试验的数据显示在下面的表3中。按上面实施例1所述进行试验。
实施例8

实施例9
在约70℃下通过使4.00kg酸性酒石酸氢可酮和悬浮在USP水中的4.76kg离子交换树脂颗粒(AMBERLITE IRP69;US.Std.Mesh#100-#400)混合来制备氢可酮polistirex。将得到的悬浮液离心,产品用USP水洗涤。干燥湿滤饼,并通过US.Std Mesh#30目筛子过筛。以酸性酒石酸氢可酮重量计,干燥产品的氢可酮含量等于大约56%。

实施例10
在约30℃下通过使3.12kg马来酸氯苯那敏和悬浮在USP水中的26.00kg离子交换树脂颗粒(AMBERLITE IRP69;US.Std.Mesh#100-#400)混合来制备氯苯那敏polistirex。将得到的悬浮液离心,产品用USP水洗涤。干燥湿滤饼,并通过美国标准#30目筛子过筛。干燥产品的氯苯那敏含量等于大约11%重量。
