Abstract

采用萃取方法从姜黄植物中提取有效成分制成姜黄活性物质稳定性制剂的方法。把姜黄块茎打成粉后用CO2超临界法对姜黄中防碍姜黄类分离的非极性油溶物质进行分离获取姜黄素和姜黄油,再用多元醇对姜黄素或姜黄油或两者混合物加以保护生产成制剂。本发明方法操作简单,不含有害残留物,不易褪色,保存期延长。

一种生产姜黄稳定制剂的方法
技术领域:
本发明涉及从植物中提取活性物质制取制剂的方法,具体是从姜黄属植物块茎中提取有效成分制成姜黄活性物质稳定性制剂的工艺方法。
背景技术:
姜黄属植物Curcuma块茎中含极性黄色素即具酚性的β-二酮等化合物和非极性的油脂类有效成分,其酚类物质为稀有的莪术醇,莪术酮,莪术二酮等,它们可强烈捕捉自由基,非极性油亦有药用效果。这些有效成分已在抗癌、抗炎、抗病毒、降血脂、美容及食品着色之类扮演重要角色,极具经济价值。
然而姜黄素与果胶、淀粉、纤维素及非极性油之间结合紧密,难以分离。用传统的有机溶剂分离方法残留溶剂多有害,而得到的色素及非极性油易于在光和常态下氧化失效,尤其姜黄素。即使是在干燥避光器皿中也难于久存,会褪色而影响效用。
发明内容:
本发明目的在于克服传统有机溶剂分离法生产姜黄制剂存在的缺陷,采用新方法萃取姜黄素和生产姜黄制剂,既防止有害物质的残留,又保证了有效成分的稳定。
本发明所提供生产姜黄稳定制剂的方法,是采用CO2超临界法,添加乙醇一次将姜黄油、姜黄素液同时获取,用分液漏斗使它们分开,再用多元醇对姜黄素或姜黄油或两者混合物加以保护生产成制剂,微波短时灭菌得成品。
本发明采用CO2超临界法,是把姜黄块茎打成粉后放入萃取釜中,于萃取釜套壁中加入乙醇,用压缩机把CO2引入萃取釜,再经节流阀减压通入分离釜偱环1~5小时,然后将气体从分离釜顶部排空,从分离釜底部取出萃取物姜黄油和姜黄素混和液;再用分液漏斗分离两者.得率达90%以上。
本发明姜黄油或姜黄素或两者混合物的重量加入5%~90%的多元醇制成乳液或片剂或粉剂或胶囊。
本发明所采用的多元醇可以在工艺过程中任一步骤加入做成功能制剂。
本发明在生产片状制剂时,需加入被膜以防止黄色物质渗出,该被膜可用甲壳素复合物或聚丙烯酸酯及其复合物。
本发明采用CO2超临界萃取,缩短了操作过程,萃取物既防止了有害物质的残留,又保证了有效成分的稳定。经加与不加多元醇的姜黄素置常态观察,用多元醇对姜黄素液活性物质加以保护的200天颜色变化不大,而不加多元醇的姜黄色100天基本褪色。
具体实施方式:
将姜黄块茎清洗干净,切成约厚度0.2cm薄片于70℃左右热风烘干,打成20-80目粉。将粉放入萃取釜中,于萃取釜套壁间加入乙醇,量约为物料的5%-6%,使CO2从钢瓶以压缩机压缩引入萃取釜,再经节流阀减压通入分离釜,CO2通过压缩机偱环运动,经1~6小时将CO2气体从分离釜顶部排空,萃取物从釜底部取出,此时得到的是姜黄油和姜黄素混合物,两液并不相溶,用分液漏斗分开.。
实例1:
5kg姜黄烘干粉,置萃取釜中,于萃取釜套壁间加入乙醇250克,通过压缩CO2,压力20-30Mpa,温度31℃~55℃,再将CO2经节流阀减压进入分离釜,通过压缩机循环运作5小时,将CO2从分离釜中排空,萃取物自分离釜下部放出,得到约290克姜黄油和姜黄素混和物,将混合物通入分液漏斗,取出上层,得175克姜黄素,用825克环状糊精吸附,干燥,加入多元醇500克压成片剂,入滚筒以聚丙烯酸脂成膜,微波调100度50秒灭菌,得成品。

Claims (3)

1.一种生产姜黄稳定制剂的方法,其特征是用CO2超临界法,即把姜黄块茎打成粉后放入萃取釜中,于萃取釜套壁间加入乙醇,用压缩机把CO2引入萃取釜,再经节流阀减压通入分离釜循环1~6小时,然后将气体从分离釜顶部排空,从分离釜底部取出萃取物姜黄素和姜黄油混合液,或用分液漏斗使它们分开,再用多元醇对姜黄素或姜黄油或两者混合物加以保护,经微波灭菌,生产成制剂。
2.按照权利要求1所述生产姜黄稳定制剂的方法,其特征是按姜黄油或姜黄素或两者混合物的重量加入5%~90%的多元醇制成乳液或片制或粉剂或胶囊。
3.按照权利要求1或2所述生产姜黄稳定制剂的方法,其特征在于生产片剂时采用被膜防止黄色物质渗出,该膜可用甲壳素复合物或聚丙烯酸酯及其复合物。
