Abstract
本发明公开了一种近红外荧光磁性微乳纳米粒子及其制备方法和在肿瘤治疗中的应用,本发明将磁性纳米粒子与近红外荧光量子点或与近红外荧光有机染料分子一起包埋到油包水的微乳中,微乳中还可包埋抗癌药物,通过磁性纳米粒子的磁导向作用,将微乳包埋的近红外荧光物质靶向到肿瘤部位或固定在肿瘤部位,在近红外光的激发下,通过近红外荧光物质发射的近红外荧光所产生的热效应来杀伤肿瘤细胞,利用近红外荧光量子点还可以通过光激发产生的具有高活性的·OH和·O2自由基与热效应一起来协同摧毁肿瘤细胞。热效应和抗癌药物的毒杀作用以及量子点的光催化活性来协同摧毁肿瘤细胞。本发明对于临床上恶性肿瘤的治疗具有重要的意义,应用前景广阔。
Description
技术领域
本发明涉及一种纳米粒子及其制备和在肿瘤治疗中的应用,尤其涉及的是近红外荧光磁性微乳纳米粒子及其制备和在肿瘤靶向治疗中的应用,属于纳米技术与生物医学的交叉领域。
发明背景
恶性肿瘤是一类严重威胁人类健康的多发病和常见病。根据世界卫生组织2003年公布的数据,2000年全球共有恶性肿瘤患者1000万,其中男性530万,女性470万;因恶性肿瘤死亡者高达620万,占总死亡人数的12%,在多数发达国家这一数字可达25%。在发展中国家,由于城市化进程的加快,饮食习惯及与之密切相关的肿瘤的发生均将逐渐转变成经济发达国家的类型。如果这一趋向得不到改善,预期到2020年,全球每年新发病例将达1500万;肿瘤病人总数,在发展中国家将增长73%,而发达国家增长29%。这很大程度上是老年人口增加的结果。我国2002年公布的发病情况(全国12市县1993~1997年登记资料统计),发病率男性为143.9/10万~213.1/10万;女性为112.9/10万~157.2/10万。面对这一严峻的局面,全球应当联合行动抗击癌症,救治千百万癌症患者的生命,各国应当采取必要的防治措施。
可见,探索肿瘤的治疗方法是医学工作者和所有相关科研人员责无旁贷的职责。多年来,人们为战胜肿瘤作出了坚持不懈的努力,然而,迄今为止,仍然没有十分理想的方法来有效遏制肿瘤的发生和发展。
研究表明,癌细胞在温度达到43℃时即呈现死亡,而人体正常的细胞加热到48℃亦能健康生存。因此,利用正常细胞与癌细胞之间的耐热差别,将癌细胞部位加热到43℃左右的温热疗法引起了研究者们极大的兴趣。肿瘤热疗的方法包括组织间射频消融热疗、高能聚焦超声、微波热疗、以及通过全身加热使体温升高到39.5℃~41.5℃维持2~4小时来进行热疗等。
纳米技术的出现给肿瘤的治疗带来了全新的思路。如可以利用纳米磁性粒子在交变磁场作用下产生的热来杀伤肿瘤。最近,美国赖斯大学和得克萨斯大学的L.R.Hirsch等研究人员将金纳米颗粒吸附到硅纳米颗粒上,形成以110±11nm的硅为核,10nm厚度的金为壳的核-壳式纳米复合粒子。他们将这种纳米颗粒注入实验鼠肿瘤部位,并采用近红外光进行辐射,结果纳米颗粒在吸收近红外光后很快产生热量,在4到6分钟之内,肿瘤就因内部温度过高而致死,而肿瘤周围的健康组织并未受损[L.R.Hirsch,et al.Nanoshell-mediatednear-infrared thermal therapy of tumors under magnetic resonaceguidance.PNAS,2003,100(23):13549-13554]。L.R.Hirsch等人的方法可使肿瘤细胞在很短的时间内就因热而受到遭受到杀伤,而正常细胞可免受伤害,这是纳米技术与生物医学非常好的结合的典范,对于肿瘤的治疗具有重大意义。然而,L.R.Hirsch等人制备的纳米复合粒子所产生的热效应是其在吸收光能后自身发热所形成的,其热量需要通过肿瘤细胞及其间隙的传递才能达到较远的地方,而且,这种纳米复合粒子不具备良好的靶向治疗肿瘤的功能。此外,杀伤肿瘤的主要是热效应作用,没有像半导体纳米粒子那样还因在光激发下产生具有强氧化性和还原性的物质来杀伤肿瘤。
如果能利用近红外荧光物质发射的近红外光所产生的热来杀伤肿瘤细胞,并使其具有靶向治疗的功能,则无疑对肿瘤的治疗具有重要意义。能产生近红外荧光的物质主要有半导体量子点无机纳米粒子和近红外有机荧光染料等。利用半导体量子点还能通过光激发产生的·OH和·O2自由基即光催化活性来协同摧毁肿瘤细胞。
近红外荧光量子点是指在光的激发下能发射近红外荧光的量子点,包括由单一半导体材料组成的近红外荧光量子点纳米粒子,以一种半导体纳米粒子为核,另一种无机材料为壳的核-壳型近红外荧光量子点纳米复合粒子,以及近红外荧光量子点量子阱结构的纳米复合粒子等。这些量子点纳米粒子具有显著的量子尺寸效应,应用领域涉及药学、细胞生物学、基因组学、蛋白质组学、分子生物学、疾病诊断与治疗等,其研究将对生物医学领域产生巨大而深远的影响。虽然组成量子点的元素如Cd、Hg等是有毒物质,但研究表明,当它们为纳米颗粒时,则基本不存在毒性或毒性很小。
近红外荧光有机染料是生物学上常用的标记示踪材料,它包括罗丹明类染料,花箐类染料,方酸箐类染料,噻嗪类和口恶嗪类染料,酞箐类及其络合物染料,BODIPY类染料等,这些近红外有机染料在DNA杂交测序、免疫检测、基因重组检测、肿瘤的早期诊断等方面有着广泛的应用。
纳米磁性金属粒子在近红外光的照射下,也能因为吸收光子而发热,如果再施加一个交变磁场而使磁性纳米粒子产生热,则当将近红外荧光量子点纳米粒子或者是近红外荧光有机染料分子与磁性纳米金属粒子一起用于肿瘤的治疗,利用它们在光的激发下共同产生的热以及磁性粒子在磁场作用下产生的热来杀伤肿瘤细胞,同时磁性纳米粒子还可以将微乳固定在肿瘤部位。因此,这种思路将比单独一种物质产生的热效应可能会更好。
发明内容
针对已有技术存在的不足,本发明的目的之一在于提供一种近红外荧光磁性微乳纳米粒子;
本发明的目的之二在于提供这种纳米粒子的制备方法;
本发明的目的之三在于提供这种纳米粒子在肿瘤治疗中的应用。
实现本发明的发明目的的技术方案如下:
一种近红外荧光磁性微乳纳米粒子,是将近红外荧光材料与磁性粒子包埋到药用乳剂中,其微乳水相的组成及配比(质量百分数)为:纳米磁性粒子0.1~30w%,近红外荧光量子点为0.01~30w%或近红外荧光有机染料为0.01~5w%,其余为水相。
所述的纳米磁性粒子为纳米四氧化三铁或纳米三氧化二铁中的一种,粒度在1nm~50nm之间。纳米磁性粒子表面可修饰能增强粒子稳定性和亲水性的高分子或表面活性剂等分子。
所述的近红外荧光材料为近红外荧光量子点纳米粒子中的任意一种或几种与近红外荧光有机染料分子中的任意一种或一种以上的组合;所述近红外荧光量子点是一种半导体纳米粒子,是由II-VI族或III-IV族元素组成的化合物,包括1、由单一半导体材料组成的近红外荧光量子点纳米粒子,如InP,InAs,CdTe,HgS,HgSe,等;2、以一种半导体纳米粒子为核,另一种无机材料为壳的核-壳型近红外荧光量子点纳米复合粒子,如CdS/HgS,HgS/CdS,CdTe/HgTe,CdSe/HgSe,InAs/ZnSe等;3、以及近红外荧光量子点量子阱结构的纳米复合粒子如CdSe/HgSe/CdSe,CdS/HgS/CdS等中的一种或任意几种的组合。近红外荧光量子点纳米粒子表面可修饰能增强粒子稳定性的分子,如亲水性高分子、疏水性高分子、表面活性剂等。所述的近红外荧光量子点纳米粒子是能在水相中稳定分散的。
所述有机染料为罗丹明类染料、花箐类染料、方酸箐类染料、噻嗪类和口恶嗪类染料、酞箐类及其络合物染料或BODIPY类染料型中的任意一种或一种以上的组合。
本发明将近红外荧光材料与磁性粒子包埋到药用乳剂中,形成具有磁性的近红外荧光物质的乳剂,所述的乳剂是指油包水型(W/O)微乳,在所述微乳的水相中还可以包埋抗癌药物。抗癌药物是在近红外荧光磁性微乳制备过程中加入的,或在近红外荧光磁性微乳制备结束后加入的,或是在临床使用前临时加入的。
本发明将近红外荧光量子点纳米粒子或近红外荧光有机染料分子与磁性纳米粒子一起包埋到油包水的微乳中,通过磁性纳米粒子的磁导向作用,将微乳包埋的近红外荧光物质靶向到肿瘤部位并固定在肿瘤部位,在近红外光的激发下,通过近红外荧光物质发射的近红外荧光所产生的热来治疗肿瘤,或同时利用近红外荧光所产生的热和磁性纳米粒子在交变磁场下产生的热共同杀伤肿瘤,或在微乳中进一步包埋抗癌药物,使其与纳米粒子产生的热效应一起来治疗肿瘤。
本发明的具体方法可选用以下任意一种:
(1)将近红外荧光材料和磁性粒子一起分散到去离子水或分散到蒸馏水或分散到0.9%的生理盐水或分散到缓冲溶液中,形成近红外荧光材料和磁性粒子的混合水溶液,然后与乳化剂和油进行混合,即形成近红外荧光材料和磁性粒子混合物的W/O型微乳液。
(2)将近红外荧光材料分散到去离子水或分散到蒸馏水或分散到0.9%的生理盐水或分散到缓冲溶液中,然后与乳化剂和油进行混合,形成近红外荧光材料的W/O型微乳液。将磁性粒子分散到去离子水或分散到蒸馏水或分散到0.9%的生理盐水或分散到缓冲溶液中,然后与乳化剂和油进行混合,形成磁性粒子的W/O型微乳液。然后将近红外荧光材料的微乳液与磁性粒子的微乳液进行混合,即得到近红外荧光材料和磁性粒子混合物的微乳液。
(3)将近红外荧光材料分散到去离子水或分散到蒸馏水或分散到0.9%的生理盐水或分散到缓冲溶液中,然后与乳化剂和油进行混合,形成近红外荧光材料的W/O型微乳液。配制磁性粒子的水溶液,将该磁性粒子的水溶液滴加到近红外荧光材料的W/O型微乳液中,振荡,即得到近红外荧光材料和磁性粒子混合物的微乳液。可以在临床使用前临时向近红外荧光材料的W/O型微乳液中滴加无菌的磁性粒子的水溶液。
(4)将磁性粒子分散到去离子水或分散到蒸馏水或分散到0.9%的生理盐水或分散到缓冲溶液中,然后与乳化剂和油进行混合,形成磁性粒子的W/O型微乳液。配制近红外荧光材料的水溶液,将该近红外荧光材料的水溶液滴加到磁性粒子的W/O型微乳液中,振荡,即得到近红外荧光材料和磁性粒子混合物的微乳液。
本发明对获得的近红外荧光磁性微乳或近红外荧光材料的微乳进行灭菌,包括加热灭菌或热压灭菌或滤过除菌或紫外灯辐照或其他无菌操作中的一种。对临床使用前临时加入的磁性粒子的水溶液或抗癌药物水溶液,亦是无菌状态。
所述的微乳是由水相液滴分散在另一种油相液体中形成的由乳化剂为界面膜的热力学稳定的胶体分散体系,粒径为10~100nm。水相液滴内部包含着近红外荧光材料和纳米磁性粒子的去离子水或蒸馏水或0.9%的生理盐水或缓冲溶液,水相液滴外部是由油组成的连续相。所述的乳化剂是由表面活性剂与助表面活性剂或者单独由表面活性剂组成。在微乳的水相中,纳米磁性粒子浓度为0.1~30w%,近红外荧光量子点浓度为0.01~30w%或近红外荧光有机染料浓度为0.01~5w%,其余为水相。
所述的乳化剂是指药剂中常用的天然乳化剂、合成乳化剂、固体乳化剂和混合型乳化剂,包括表面活性剂、亲水性高分子或固体微粒中的一种或任意几种的组合。所述的助表面活性剂是指含2~8个碳原子的短链醇中的任意一种或几种的组合。所述的油是指药剂中常用的植物油(如花生油、玉米油、芝麻油、十四酸异丙酯等)、矿物油(如液状石蜡)以及有机溶剂如三氯甲烷、二氯甲烷、乙酸乙酯等中的一种或任意几种的组合。所述的微乳的油、乳化剂以及水溶液的组成比例是落在微乳体系的微乳相区内,是根据组成成分不同来确定的。
本发明近红外荧光磁性微乳纳米粒子应用于肿瘤的治疗,具体是指利用纳米磁性粒子在磁场的导向下将近红外荧光磁性微乳定向(或固定)在肿瘤部位,在肿瘤部位放置激发光源,通过近红外荧光物质在激发光的激发下发射出的近红外荧光所产生的热效应或同时利用近红外荧光产生的热效应和抗癌药物的毒杀作用来治疗肿瘤。利用近红外荧光量子点还可以通过光激发产生的具有高活性的·OH和·O2自由基与热效应一起来协同摧毁肿瘤细胞。本发明还可以利用纳米磁性粒子在磁场的导向下将近红外荧光磁性微乳定向(或固定)在肿瘤部位,通过近红外荧光磁性微乳中的纳米磁性粒子在交变磁场下产生的热效应以及近红外荧光物质在光的激发下所产生的近红外光的热效应来治疗肿瘤,或同时利用这些热效应和抗癌药物以及量子点的光催化活性的毒杀作用来治疗肿瘤。
本发明将近红外荧光材料产生的热效应、近红外荧光量子点的光催化活性、磁性纳米粒子产生的热效应以及抗癌药物的毒杀作用进行有机的结合,用于肿瘤的治疗尤其是肿瘤的热疗中,在恶性肿瘤的治疗领域具有重大意义。
具体实施方式
实施例1
在氮气保护下,以氯化隔和碲氢化纳以及一种保护剂为原料按一定的摩尔比在pH=10.0的水溶液中反应获得棕色透明的碲化镉(CdTe)纳米粒子溶液,进一步熟化和钝化处理,获得的CdTe的发射光谱的最大发射波长约在700nm处,荧光拖尾到840nm。
在氮气保护下,将FeCl3·6H2O和FeCl2·4H2O共同溶于50mL含浓度为0.3w%的聚乙二醇(PVA)水溶液中,于80℃和磁力搅拌下向混合液中滴加氢氧化钠溶液至反应液pH达11。反应液自然冷却后,采用磁分离并洗涤黑色产物,然后再将产物分散到0.5w%的PVA水溶液中,超声分散,得到粒度在3~15nm之间的黑色球形四氧化三铁(Fe3O4)。
将适量的蛋黄卵磷脂溶解到5mL的乙醇溶液中,再将该溶液溶解到12mL十四酸异丙酯中,根据绘制的微乳相图,向该混合溶液中滴加CdTe和Fe3O4纳米粒子的混合水溶液,振荡。通过0.22μm的微孔滤膜过滤除菌,即得到同时包埋CdTe和Fe3O4的微乳,氮气密封保存。结果获得分散均匀的乳液,透射电子显微镜下可见纳米粒子的团簇,用磁铁可将微乳液中的磁性粒子吸引到试管壁一侧。
实施例2
以实施例1获得的CdTe和Fe3O4为纳米粒子原料。
将适量的蛋黄卵磷脂溶解到5mL的乙醇溶液中,再将该溶液溶解到15mL十四酸异丙酯中,然后向该混合溶液中滴加CdTe纳米粒子水溶液,振荡,结果得到澄清透明的棕色CdTe微乳液。
另将适量的蛋黄卵磷脂溶解到5mL的乙醇溶液中,再将该溶液溶解到12mL十四酸异丙酯中,然后向该混合溶液中滴加Fe3O4水溶液,振荡,结果得到均匀的Fe3O4微乳液。
然后将CdTe的微乳液与Fe3O4的微乳液进行等体积混合,振荡,即得到同时包埋CdTe和Fe3O4的微乳。通过0.22μm的微孔滤膜过滤除菌,氮气密封保存。结果获得的产物在外观和微观结构上与实施例1相似。
实施例3
以适量浓度的近红外荧光的硅酞菁衍生物(LaJolla Blue,最大发射波长为680nm)代替实施例1中的CdTe,按实施例1相同的方法制备同时包埋LaJolla Blue和Fe3O4的微乳。结果亦得到均匀的乳液,有较强的磁性。
实施例4
以实施例1获得的CdTe和Fe3O4为纳米粒子原料。
将适量的蛋黄卵磷脂溶解到5mL的乙醇溶液中,再将该溶液溶解到12mL十四酸异丙酯中,然后向该混合溶液中滴加CdTe纳米粒子水溶液。振荡,得到透明的CdTe微乳液。
配制Fe3O4水溶液,在手摇振荡的情况下向CdTe微乳液中逐滴滴加一定体积的Fe3O4水溶液。滴完后继续振荡10~15min。即得到同时包埋CdTe和Fe3O4的微乳。通过0.22μm的微孔滤膜过滤除菌。结果得到在外观和磁性等方面与实施例1相似的微乳液。
实施例5
按实施例1制备同时包埋CdTe和Fe3O4的微乳液约22mL。
配制适量浓度的阿霉素水溶液,在手摇振荡的情况下向微乳液中逐滴滴加适量体积的阿霉素水溶液。滴完后继续振荡10~15min。即得到同时包埋CdTe和Fe3O4以及阿霉素的微乳。通过0.22μm的微孔滤膜过滤除菌。结果获得红色的微乳,具有较强的磁性和较强的荧光。
实施例6
取Balb/C小鼠20只,在小鼠的左后腿部种上S180细胞,当肿瘤大小约为1cm时,将小鼠随机分5组,每组4只。
对第1组小鼠:剃去小鼠肿瘤部位的体毛,取按实施例1制备的同时包埋CdTe和Fe3O4的微乳0.2mL,进行瘤中注射,在肿瘤上方放置一块圆盘形磁铁,使微乳固定在肿瘤部位,然后采用近红外光激发肿瘤部位。
对第2组小鼠:剃去小鼠肿瘤部位的体毛,取按实施例1制备的同时包埋CdTe和Fe3O4的微乳0.2mL,进行瘤中注射,将小鼠麻醉后固定在交变磁场的装置中,同时采用近红外光激发肿瘤部位。
对第3组小鼠:剃去小鼠肿瘤部位的体毛,取按实施例4制备的同时包埋CdTe和Fe3O4以及阿霉素的微乳0.2mL,进行瘤中注射,并采用与第2组小鼠相同的方法进行光激发和交变磁场的作用。
对第4组小鼠:剃去小鼠肿瘤部位的体毛,取按实施例1制备的同时包埋CdTe和Fe3O4的微乳0.3mL,进行瘤中注射,除在肿瘤上方放置一块圆盘形磁铁外,不作其它处理。
对第5组小鼠:剃去小鼠肿瘤部位的体毛,然后不经过任何其他处理。
结果,第1~3组小鼠的肿瘤生长明显受到抑制,第2组和第3组最为显著,个别小鼠的肿瘤已显著缩小,接近消失。第4组小鼠肿瘤有一定缩小,而第5组小鼠肿瘤的大小最大。
Claims (10)
1、一种近红外荧光磁性微乳纳米粒子,是将近红外荧光材料与磁性粒子包埋到药用微乳中,其微乳水相的组成及配比(质量百分数)为:纳米磁性粒子0.1~30w%,近红外荧光量子点为0.01~30w%或近红外荧光有机染料为0.01~5w%,其余为水相。
2、根据权利要求1所述的一种近红外荧光磁性微乳纳米粒子,其特征在于:所述的纳米磁性粒子为纳米四氧化三铁或纳米三氧化二铁中的一种,粒度在1nm~50nm之间。
3、根据权利要求1所述的一种近红外荧光磁性微乳纳米粒子,其特征在于:所述的近红外荧光材料为近红外荧光量子点纳米粒子中的任意一种或几种与近红外荧光有机染料分子中的任意一种或一种以上的组合;所述近红外荧光量子点是一种半导体纳米粒子,是由II-VI族或III-IV族元素组成的化合物;所述有机染料为罗丹明类染料、花箐类染料、方酸箐类染料、噻嗪类和口恶嗪类染料、酞箐类及其络合物染料或BODIPY类染料型中的任意一种或一种以上的组合。
4、根据权利要求3所述的一种近红外荧光磁性微乳纳米粒子,其特征在于:所述的近红外荧光量子点纳米粒子表面修饰能增强粒子稳定性的亲水性高分子、疏水性高分子或表面活性剂中的一种或一种以上混合物。
5、根据权利要求1所述的一种近红外荧光磁性微乳纳米粒子,其特征在于:所述的微乳为油包水型微乳;在所述微乳的水相中包埋抗癌药物。
6、根据权利要求1、2、3、4或5所述的一种近红外荧光磁性微乳纳米粒子,其制备方法如下:将近红外荧光材料和磁性粒子一起分散到去离子水或分散到蒸馏水或分散到0.9%的生理盐水或分散到缓冲溶液中,形成近红外荧光材料和磁性粒子的混合水溶液,然后与乳化剂和油进行混合,即形成近红外荧光材料和磁性粒子混合物的油包水型微乳液。
7、根据权利要求1、2、3、4或5所述的一种近红外荧光磁性微乳纳米粒子的制备方法,其特征在于:将近红外荧光材料分散到去离子水或分散到蒸馏水或分散到0.9%的生理盐水或分散到缓冲溶液中,然后与乳化剂和油进行混合,形成近红外荧光材料的油包水型微乳液,将磁性粒子分散到去离子水或分散到蒸馏水或分散到0.9%的生理盐水或分散到缓冲溶液中,然后与乳化剂和油进行混合,形成磁性粒子的油包水型微乳液,再将近红外荧光材料的微乳液与磁性粒子的微乳液进行混合,即得到近红外荧光材料和磁性粒子混合物的微乳液。
8、根据权利要求1、2、3、4或5所述的近红外荧光磁性微乳纳米粒子的制备方法,其特征在于:将近红外荧光材料分散到去离子水或分散到蒸馏水或分散到0.9%的生理盐水或分散到缓冲溶液中,然后与乳化剂和油进行混合,形成近红外荧光材料的油包水型微乳液,配制磁性粒子的水溶液,将该磁性粒子的水溶液滴加到近红外荧光材料的油包水型微乳液中,振荡,即得到近红外荧光材料和磁性粒子混合物的微乳液,在临床使用前临时向近红外荧光材料的油包水型微乳液中滴加无菌的磁性粒子的水溶液。
9、根据权利要求1、2、3、4或5所述的近红外荧光磁性微乳纳米粒子的制备方法,其特征在于:将磁性粒子分散到去离子水或分散到蒸馏水或分散到0.9%的生理盐水或分散到缓冲溶液中,然后与乳化剂和油进行混合,形成磁性粒子的油包水型微乳液,配制近红外荧光材料的水溶液,将该近红外荧光材料的水溶液滴加到磁性粒子的油包水型微乳液中,振荡,即得到近红外荧光材料和磁性粒子混合物的微乳液。
10、根据权利要求1-5项权利要求所述的近红外荧光磁性微乳纳米粒子,应用于肿瘤的治疗。
