Description

从浸渍胶乳橡胶制品去除蛋白质的方法
本发明涉及一种从浸渍胶乳橡胶制品,特别是橡胶手套中去除蛋白质的方法。
巴西三叶胶胶乳(即天然橡胶或NR胶乳)是用于检查用手套、手术用手套和许多其它种类浸渍制品的优选材料。每年有250-300亿双天然橡胶胶乳手套供应到全球市场上。手套占所有天然橡胶胶乳消费的45%,而气球、粘合剂、泡沫塑料、地毯背材和其它应用占约50%,避孕套占2%。
近年来,天然橡胶胶乳的应用受到出现蛋白质变态反应的严重影响。实际上,有三种健康问题与NR胶乳的应用有关,分别是产生刺激物、亚硝胺和胶乳蛋白质变态反应。后一种变态反应因素是巴西三叶胶(NR)胶乳所独有的。变态反应是个体免疫系统对接触或暴露于其它方面可能无害的物质时的过激反应。正是个体的基因构成使个人易于对NR胶乳蛋白质有变态反应。例如,特应性的人通常对胶乳蛋白质有变态反应。在过去的15年中,蛋白质变态反应的问题急剧增多。
浸渍制品工业已经进行了多种尝试来减少胶乳蛋白质变态反应的问题。这包括通过浸滤来减少可萃出蛋白质;氯化;在手套和人体之间构造阻挡层;以及在某些情况下放弃NR胶乳而代之以合成橡胶胶乳。然而,天然橡胶胶乳具有综合了强度、柔软性和弹性的独特性能平衡。没有任何替代性的合成材料具备这种性能平衡。天然橡胶胶乳是一种非常“宽容”的材料,人们例如可以舒适地佩带小号的医用手套。而且,在可灭菌性、安全性、成本和有效性方面,天然橡胶胶乳是非常具有竞争力的材料。
商品NR胶乳的检查用手套中的蛋白质含量随生产日期和生产批次而改变,其可以低至50微克/克直到高达1000微克/克。
根据本发明,提供了一种从浸渍胶乳橡胶制品除去蛋白质的方法,该方法包括:
(A)通过用热水处理所述制品将预先施用于制品的玉米淀粉粉末溶解,以除去所有的玉米淀粉粉末;
(B)用含有碱金属氢氧化物的水溶液处理该制品,除去蛋白质;和
(C)在水溶液中用一种或多种表面活性剂处理该制品,除去残余的蛋白质。
本发明还提供了由巴西三叶胶(NR)胶乳制成的浸渍胶乳橡胶制品,其可萃出蛋白质含量低于20微克/克。
本发明的方法可以成功地减少浸渍胶乳橡胶制品中的蛋白质,使蛋白质含量低于现有用于检测可萃出蛋白质含量的分析技术的灵敏度含量,所述灵敏度含量为20微克/克。
本发明方法是一系列简单处理,其有效除去吸附在天然橡胶胶乳手套和任何其它胶乳浸渍产品之上及吸收在其内部的可萃出蛋白质。经本发明方法处理的浸渍胶乳产品,特别是手套,可以被安全地分类为NEP(无可萃出蛋白质)。本发明方法可以处理蛋白质含量超过1500微克/克的打粉(powdered)手套,可使其基本上不含可萃出蛋白质。
本发明方法可以并入生产浸渍胶乳制品的常规工厂工艺中,在这种情况下,打粉步骤可以任选省略,但优选其作为单独的离线操作来进行。它的成本经济,并可使胶乳橡胶工业能出售没有可萃出蛋白质,即蛋白质含量低于20微克/克的打粉手套、硅化手套或氯化手套。
本发明的分步骤方法包括如上所述的第一步,即步骤(A),如果待处理的浸渍胶乳橡胶制品是“打粉的”制品,才需要进行此步骤;也就是说,如果先将一层玉米淀粉颗粒用于该制品来处理以改进其表面润滑性,才需要进行此步骤。步骤(A)可使蛋白质含量降到约50微克/克。步骤(B)进一步降低蛋白质含量,步骤(C)使制品没有可萃出蛋白质。
下面通过本发明方法的一个实施例来说明。
步骤(A)
将蛋白质含量为800-1500微克/克的打粉手套浸在水温维持在80-98℃的水中长达10分钟。水和高温共同作用使玉米淀粉粉末溶解在水中,从而从手套上除去该粉末。除去玉米淀粉粉末带走了手套表面吸附的蛋白质。表1说明了效果。

步骤(B)
将热水处理过、已无粉末的手套浸渍装有0.5-2.0%的氢氧化钾(也可以使用氢氧化钠)溶液的槽内10-30分钟,然后用水清洗。这能除去手套表面上或表面正下浅表层的残余游离蛋白质。表2说明了这一效果。

步骤(C)
将步骤B处理过的手套浸在装有含0.1-0.5%非离子表面活性剂,例如已知的Lubrol LW或Vulcastab LW(鲸蜡醇-油醇的环氧乙烷缩合物的商标)和0.1%十二烷基硫酸钠或任何其它织物洗涤剂的水的槽中,并搅拌。10-20分钟后取出手套,清洗,并可以以常规方式进行打粉、硅化或氯化。

*这些数值无关紧要,因为它们已经低于试验方法的灵敏度含量,即20微克/克。
对于一个自动化的方法,在用热水除去粉末之后的所有这些处理均可以用市售氯化器来进行。每一处理后的溶液均可以泵出,并再使用至少两次。最后的清洗之后可以进行氯化。在进行打粉或硅化的情况下,其也可以在另一台氯化器中进行以避免污染。
本发明方法对于医用的浸渍胶乳橡胶制品,特别是手套特别有用,也可用于其它此类制品,例如导管及其它医用器件或部件。
当然,也应理解,用本发明方法处理过的浸渍胶乳橡胶制品特别适用于已知对蛋白质有变态反应的人,以及适用于不希望引起任何可能存在的蛋白质变态反应的场合。

Claims (16)

1.一种从浸渍胶乳橡胶制品去除蛋白质的方法,该方法包括:
(A)通过用热水处理所述制品将预先施用于制品的玉米淀粉粉末溶解,以除去所有的玉米淀粉粉末;
(B)用含有碱金属氢氧化物的水溶液处理该制品,以除去蛋白质;和
(C)在水溶液中用一种或多种表面活性剂处理该制品,以除去残余的蛋白质。
2.权利要求1的方法,其中浸渍胶乳橡胶制品是橡胶手套。
3.权利要求1或2的方法,其中制品是打粉的制品,并且其中用热水处理的步骤包括将该制品浸在80-98℃的水中长达10分钟。
4.前述权利要求任一项的方法,其中碱金属氢氧化物水溶液是氢氧化钾水溶液。
5.权利要求4的方法,其中制品浸在0.5-2.0%的氢氧化钾水溶液中10-30分钟。
6.前述权利要求任一项的方法,其中在用碱金属氢氧化物水溶液处理后用水清洗。
7.前述权利要求任一项的方法,其中所述一种或多种表面活性剂是非离子表面活性剂,或包括非离子表面活性剂。
8.权利要求7的方法,其中非离子表面活性剂是鲸蜡基油基醇一环氧乙烷缩合物。
9.权利要求7或8的方法,其中将非离子表面活性剂与离子表面活性剂一起使用。
10.权利要求9的方法,其中离子表面活性剂是十二烷基硫酸钠。
11.前述权利要求任一项的方法,在该方法后进行打粉、硅化或氯化处理。
12.权利要求1的方法,其基本上如上文的具体描述。
13.由巴西三叶胶(NR)胶乳制成的浸渍胶乳橡胶制品,其可萃出蛋白质含量低于20微克/克。
14.权利要求13的浸渍胶乳橡胶制品,其用权利要求1-12任一项的方法处理过。
15.权利要求13或14的浸渍胶乳橡胶制品用于医疗目的的用途。
16.权利要求13或14的浸渍胶乳橡胶制品由具有蛋白质变态反应的人来使用的用途。
