Abstract

市场背景
爱美之心人皆有之,俗话说得好:一白遮百丑,尤其是对东方女性来说,拥有白晰透嫩的皮肤更是梦寐以求的心愿。肤色是先天决定的,目前市面上大部分的所谓增白化妆品,不过是起到覆盖或漂白的功能,只是治标不治本。随着生物科技的发展,尤其是在全世界科学家联手合作完成了人类全部基因序列测定之后,科学家们已经发现一种新的方法,可用来抑制那些人体皮肤细胞中使皮肤变黑的黑色素和其它色素的基因表达,减少色素的含量和沉着(变白),去除皮肤上的斑点,如老年斑,色素斑,雀斑,妊辰斑,或其他由阳光,炎症,药物等因素引起的色素变化(变美),从而使皮肤变白变美成为可能。

皮肤颜色的来源:人体的肤色是由四种不同的基本颜色所组成,它们是白色,红色,黄色和黑色。白色是一种脂肪和脂质所具有的自然色。红色是由血管中红细胞所携带的一种呼吸色素的颜色,它由亚铁血红素所产生。黄色来自于一种称为胡罗卜素的物质,这种物质有三种异构体,它们在肝脏里能转变成维生素A。黑色是由皮肤中黑色素细胞所产生的黑色素的颜色,此色素的高低决定了皮肤的颜色深浅。

现有的漂白方法:最早使用让皮肤变白的方法是一种脱皮漂白法。不同的脱皮漂白的配方能进行不同程度的脱皮,通常可分为三种,即深、中、浅脱皮法。常用的配方是一些酸和醇类物质。前两种有一定的危险性,不合适作为常用化妆品的制作方法。而第三种是安全的并能有效的去除局部的高色素的班快。继而,人们逐渐认识到深浅肤色的差异与黑色素细胞中洛氨酸酶的活性的高低有关,于是,含有各种不同的洛氨酸酶的抑制剂的皮肤变白的方法和相应的产品不断出现。常用的洛氨酸酶的抑制剂有对笨二酚(hydroquinone),曲酸(Kojic Acid),熊果苷(Arbutin),Mulberry,抗坏血酸(Ascorbic Acid),LicoriceExtract,Burner Root Extract,Scutellaria Extract等,和最新的生物制品,如Melanostat。然而,对笨二酚具有细胞毒性,曲酸、熊果苷、和抗坏血酸等的增白效果并不明显,含有Melanostat活性成分的产品正在研发当中。此外,新的思路开始出现,如用还原剂来增强洛氨酸酶的抑制剂和同时干扰黑色素形成过程的多个环节从而使皮肤增白程度加大。

2.有机地整合传统的化学美白,新近的草木提取液美白和最新的基因美白等方法为一体,产生一种明显的使人满意的美白效果。同时,也具有保护皮肤,抗炎症,防衰老等功能。最终体现出新产品的用量小,见效快,无毒副作用的优良特征。

4 发明的具体技术方案
名词术语
如这里所用的术语“皮肤美白“是指皮肤中的黑色素明显减少,皮肤的质地更富有弹性,皮肤表面变得更为光滑均匀,皮肤上的斑点,如老年斑,色素斑,雀斑,妊辰斑,或其他由阳光,炎症,药物等因素引起的色素变化和斑块得到有效的去除。
在本发明的上下文中,术语“抑黑素基因链”是指一种或多种不同量的双链寡聚核苷酸(double strandoligonucleotide)和/或单链寡聚核苷酸(single-stranded oligonucleotide)及相应的能将这些寡聚核苷酸分子导入一个动物尤其是人的特异的皮肤细胞并在药效上能够接受的一种生物膜载体的混合物。术语“抑黑素基因链”进一步也包括那些裸露的寡聚核苷酸分子和/或其它的辅助成份的混合物。它们可直接用于临床治疗,保健美容和基础研究。

5.2 设计能有效抑制与黑色素形成有关的基因的mRNA的寡聚核甘酸
本发明中作为皮肤美白的重要活性成分的寡聚核苷酸主要来源于下述与黑色素形成有关的基因:
洛氨酸酶基因(Tyrosinase gene)编码一条55KD大小的能与铜离子结合的多肽链。洛氨酸酶是催化洛氨酸转变成黑色素的系列反应中的一个关键酶,洛氨酸酶不但存在于酚黑色素小体中,而前也存在于真黑色素小体中,前者与酚黑色素(为红色或黄色)的形成有关,后者与真黑色素(为棕色或黑色)的形成有关。真黑色素小体含有大的椭圆形的黑色素颗粒,这些棕色或黑色颗粒与一种高度有序的糖蛋白结合在一起,而酚黑色素小体却含有小的圆形的黑色素颗粒,这些红色或黄色颗粒与一种疏松而无序的糖蛋白结合在一起。深色皮肤的角质细胞中含有许多大而致密的黑色素小体,而浅色皮肤的角质细胞中含有小而疏松的黑色素小体。当此基因的突变时,将会引起一种遗传病,叫做OCA-1型白化症。

本发明中作为皮肤美白的重要活性成分的寡聚核苷酸包括所有的自然的和/或人工修饰单链或双链的RNA,DNA or RNA/DNA的杂合体。寡聚核苷酸包括但不局限于那些来自于上述与黑色素形成有关基因的任何部位的核苷酸序列,这些核苷酸序列的长度通常为10-29个核苷酸,最好的长度范围是:19-23个核苷酸。所有来自上述不同基因的双链寡聚核苷酸和单链的反意寡聚核苷酸的特异性和有效性是根据专利(专利申请号:01144177.1)中所描述的方法来设计,选择和合成。
本发明中作为皮肤美白的重要活性成分的寡聚核苷酸的百分构成通常为,0.0000001%-10%,较好的范围是0.000001%-0.1%,更好的范围是0.00001%-0.001%,最好的范围是0.00005%-0.0005%。
本发明中作为皮肤美白的重要活性成分的寡聚核苷酸可单独与合适的载体组成新型的皮肤美白化妆品,也能与其它的多种皮肤美白成分一起组成一种复合型的皮肤美白化妆品。
鉴定抑制洛氨酸酶的有效成分
熊果苷(Arbutin)是从熊梅(Bearberry)里提炼出来得一种活性物质。它具有抑制洛氨酸酶的作用,使皮肤变白。它能对抗自由基,抗老年化和抗紫外线,并具有防止黑色素细胞癌的效应。此物质没有毒付作用,无皮肤激若效应,无不良气味。
Glycyrrhetinic acid是从植物(Glycyrrihiza Glabra)里提炼出来得一种活性物质。它能有效的抑制洛氨酸酶的活性,消除由紫外线引起的色素沉着和红斑形成。它还有一定的抗炎作用。

大鹾尿素(Diazolidinyl Urea)是一种还原剂,加强熊果苷的作用,是皮肤更易增白。
抗坏血酸(Ascorbic Acid)()是一种还原剂,可引起黑色素中间体的还原,从而阻止从洛氨酸到黑色素的氧化反应过程的发生。抗坏血酸还有促进皮肤胶原的合成,从而减少皮肤的皱纹和增加皮肤的弹性。抗坏血酸还是一种抗氧化剂的电子供体,从而抗氧化剂的作用加强并持久。

润肤剂能使皮肤变得柔软而润滑.这些试剂包括Cetyl alcohol,Fatty acid,Stearic acid,Fatty alcohol,Isocetyl alcohol,Olive oil,Oleyl alcohol,Stearyl alcohol,Siliconeoil,Glyceryl monostearate,Mink oil,Soybean oil,Safflower seed oil,和它们可能的衍生物及混合物。
防晒剂(Sunscreening agents).许多防晒剂可与皮肤增白成分一起使用来对抗由紫外线引起的皮肤变黑。防晒剂包括:Zinc oxide,Titanium dioxide,P-aminobenzoic acid,Benzophenones,Quinine salts,Dihydoxycinnamic acid,Trihydroxycinnamic acid,2-ethylhexyl-p-methoxycinnamate,Butylmethoxydibenzoylmethane,Cinnarnic acid,2-hydroxy-4-methoxybenzophenone,Octyldimethyl-p-aminobenzoic acid,Anthranilates,Salicylates,和它们可能的衍生物及混合物。
抗氧化剂(Anti-Oxidants/Radical Scavengers).Tocopherol,Butylated hydroxy benzoic acid,Gallic acid,Uric acid,Sorbic acid,Dihydroxy fumaric acid,Sulfhydryl compounds,和它们可能的衍生物及混合物。
本发明中通常也包括但不局限于其它已知的传统化妆品成分,如酒精,脂肪,矿物油,植物油,动物油,合成油,表面活化剂,湿润剂,增厚剂,乳化剂,稳定剂,颜料,香料,等等。上述所有成分或一种成分都可以与一种和/或数种抑制黑色素形成的基因的寡聚核苷酸片断组成不同类型的基因皮肤美白产品。
挑选合适的载体
由于在水溶液中裸露的寡聚核苷酸很难进入细胞,有效的传递寡聚核苷酸是能否美白成功的一个重要环节。寡聚核苷酸的传递系统分为两大类,即生物的和机械的传递途径。生物传递可分为两种,一种是病毒性的,一种是非病毒性的,而机械传递可分为手工注射法和基因枪注射法。本发明主要采用但不局限于一种由脂质体和多聚体组成的复合物。

一些脂肪酸,脂肪酸脂,鳌合物和表面活化物质,也许都可用来作为载体,协助寡聚核苷酸进入细胞,常被使用的脂肪酸和各种脂,包括但不局限于1-dodecylazacycloheptan-2-one,arachidonic acid,羊脂酸(caprylic acid),山羊脂酸(capric acid),甘油二月桂酸脂(dilaurin),甘油二酯(diglyceride),dicaprate,花生酸(eicosanoic acid),glyceryl 1-monocaprate,lauric acid,linoleic acid,linolenic acid,单酸甘油酯(monoglyceride)甘油一油酸(monoolein)肉豆冠酸(myristic acid),油酸(oleic acid),棕酸(palmiticacid),硬脂酸(stearic acid),和tricaprate.等。
阳离子脂质体是用于人体基因治疗的最佳载体之一,因为它们是非感染性的,几乎没有免疫原性和毒性。在形态学方面,阳离子脂质体可分为三个主要类型,即:小型的单层小泡,大型的单层小泡和多层的小泡。常用的脂质和脂质体包括中性脂,即DLPE(1,2-dilauroyl-sn- glycero-3-phosphoethanolamine)、DiPPE(1,2-diphytanoyl-sn-glycero-3-phosphoethanolamine)和DOPE。这些脂质和脂质体能够协助入胞后的内涵体的破裂。阳离子脂质如DOTAP(dioleoyltetramethylaminopropyl)、DOTMA(the cytofectinN-[1-(2,3-dioleoyl)phosphatidyl]-N,N,N trimethyl ammonium chloride)和TMAG(N-(α-trimethylammonioacetyl)-didodecyl-D-glutamate chloride)。理想的脂质载体通常是一种等比例的阳离子脂质和中性脂质(Lecithin,Phosphoserine)的混合物或中性脂质。
另一种阳离子脂质是阳离子扑啉。tetra(4-methylpyridyl)porphyrin(TMP)和tetraanilinium porphyrin(TAP)能比裸露的寡聚核苷酸本身更有效地协助寡聚核苷酸进入细胞。此外,阳离子扑啉不仅能帮助寡聚核苷酸进入细胞,也能将其传递到细胞核内,以便寡聚核苷酸在那里能与相应的mRNA分子和RNase III核酸酶相作用。人造的病毒体是另一类传递媒介,它能够利用一个病毒进入细胞的那种天然能力来帮助寡聚核苷酸进入细胞,重组的流感病毒胞膜是一种著名的病毒体,它能通过受体介导的内吞作用而进入细胞,并与内噬体膜合为一体。近来研究己将阳离子脂质与病毒体溶合为一体来作为一种新型的运载工具。
多聚阳离子体是另一类有用的载体,可用来增强阳离子脂质体介导的入胞作用,尤其实用于单连的反意寡聚核苷酸。可用的阳离子多聚体包括:多聚L型赖氨酸(poly-L-lysine),硫酸鱼精蛋白(protamine sulfate),重组人体组蛋白H1(recombinant human H1 histone protein),精氨酸(spermidine)和PEI(polyethylenimine)。聚氮丙啶(PEI)已被证明是一种有效的非病毒性载体,可将基因递送进不同类型的细胞,和促进寡聚核苷酸进入哺乳类细胞的胞核,PEI其他的特征包括:能与核酸结合,将核酸压缩,具有高效的缓冲能力和内在的溶解内噬体的活性。此外,它还能保护核酸对抗核酸酶的降解。在外用或注射线状的22KDa大小的PEI和寡聚核苷酸复合物后,在细胞内可以看到其传递的标志基因能够高效地表达。进一步的研究发现,分支状的25KDa的PEI除了与线状的PEI在体外转染实验中有相似的效果外,它还被证明其体内的基因传递效果远远好于线状的PEI。当结合所有这些PEI的特点与受体介导的基因传递机制在一起时,与配体结合的PEI能够更有效地将基因导入各种不同的肿瘤细胞。
此外。还需提及的其它方法包括结合肽和基因枪。结合肽能够在寡聚核苷酸周围形成一个多肽圈,有利于寡聚核苷酸被细胞摄入。许多含有多聚赖氨酸的结合肽可引起膜的去稳定性。一般来说,这些结合肽比脂质有较低的细胞毒性,并具有相似的传递效果。除了老式的手工注射法外,一种新型的基因枪也已宣告诞生,其原理是高压的氮气能够产生一种超声的速度,从而通过极高的加速度将金颗粒包裹的DNA分子打入活的细胞中,此方法可以用来有效地并直接地传递寡聚核苷酸进入细胞。
基因皮肤美白化妆品组份的标准
本发明中所用的各种皮肤美白成分都必须满足下速标准;属于药物学上或化妆品上能接受的物质,这些物质需没有毒性,没有不可忍受的气味,没有皮肤激若效应,没有不稳定性,和一个合理的受益与危害的比例。
基因皮肤美白化妆品寡聚核苷酸分子的合成
合成一个单链或双链寡聚核苷酸分子的方法在文献上已有许多介绍(Needham-VanDevanter et al.1984,Nucleic Acids Res.,12:6159-6168,Beaucage and Caruthers,1981,Tetrahedron Letts,22:1859-1862)。大体上可分为仪器合成法和生物合成法。用仪器合成的一个寡聚核苷酸分子在合成后需经进一步的纯化,常用的方法是阴离子交换树脂或聚丙稀酰胺电泳(Pearson and Regnier,1983,J.Chrom.255:137-149.)。纯化后的寡聚核苷酸分子可用化学降解法和核酸序列分析仪来加以鉴定(Maxam and Gilbert,1980,in Grossmanand Moldave(eds.)Academic Press,New York,Methods in Enzymology 65:499-560)。生物合成法又包括体外(如用T7多聚酶介导的生物合成)和体内合成法(如用含有人U6促进子的质粒为媒介的生物合成)两种。本发明包括但不局限于用所有这些方法来合成一个单链或双链寡聚核苷酸分子。
基因皮肤美白化妆品的配制方法
一个单纯基因皮肤美白化妆品的组装涉及到方方面面许多因素,包括寡聚核苷酸分子、与脂质和/或阳离子多聚体的比例、它们的浓度、缓冲液的PH值、离子强度和其增加稳定性的物质,主要需注意的事项是避免或减少混合物的沉淀,保护寡聚核苷酸分子以免被降解和提高转化的效益。一个复合皮肤美白化妆品除了上述的因素外,还涉及到许多其他的成分,如一种洛氨酸酶抑制剂,一种还原剂,一种脱和新皮,一种鏊合剂,一种乳化剂,一种防晒剂,一种抗氧化剂等。当配制本发明的一个单纯基因皮肤美白化妆品时,应注意下述的诸条件。

此外,理想的通过皮肤上皮途径所需的转化复合物的大小为30-100nm(纳米)。活性成份是一种或一组不同的特异的寡聚核苷酸分子,他们能有效地抑制相关的靶RNA分子,根据不同肤色细胞中一组基因表达的程度和美白的需要,来调节寡聚核苷酸分子的种类、剂量和组合的方式,以便获得最大的美白效果和最低的毒副效应。
一个复合基因皮肤美白化妆品主要被配制成水包油状的悬浮液,具体配制如下:
(1)0.01-10g的不同的寡聚核苷酸分子(p-蛋白基因,Kinesin基因,和mcir基因)溶于20ml的无离子双蒸水中,
(2)将1g的磷脂和0.5g的DOTAP溶于50%的乙醇(无离子双蒸水配制)中,
(3)将溶液(1)滴入溶液(2),轻轻混合后置于室温下30分钟。
(4)将1-10%曲酸和1-10%乳酸溶加温到100C,与(5)充分混合,
(5)1-10%熊果苷和1-10%抗坏血酸于20ml的无离子双蒸水中,
(6)将溶液(4)慢慢滴入溶液(3)后,轻轻摇晃使它们充分混合,然后置室温中40分钟。
(7)将溶液(5)慢慢滴入溶液(6)后,用超声使它们充分混合,
(8)配制其他成份,
(9)最终的混合溶液可用于皮肤细胞的转染和美白。
一个单纯基因皮肤美白化妆品的配制:
●0.01-10g的不同的寡聚核苷酸分子(p-蛋白基因,Kinesin基因,和mcir基因)和0.1g PEI溶于20ml的无离子双蒸水中,滴入10%的甘油。
·将1g的磷脂和0.5g的DOTAP溶于乙醇和乙醇酸中,
·将溶液(1)滴入溶液(2),轻轻混合后置于室温下30分钟。
·将溶液(4)慢慢滴入溶液(3)后,轻轻摇晃使它们充分混合,然后置室温中40分钟。
·将溶液(5)慢慢滴入溶液(6)后,用超声使它们充分混合,
·配制其他成份,
·最终的混合溶液可用于皮肤细胞的转染和美白。
不同类型的单链或双链寡聚核苷酸
本发明的另一实例是可采用不同类型的单链和/或双链寡聚核苷酸作为一种抑黑素基因链分子,并且这些不同类型的抑黑素基因链分子以相同或不同的剂量,组成一种混合物。这些不同类型的抑黑素基因链分子的不同配伍也许可用来设计产生不同长短疗效的治疗方案,就象传统的药物的配伍那样。例如,每种类型的单链或双链寡聚核苷酸分子的反意链可产生立即的功效。由于它们均能与相应的mRNA分子杂交,从而阻断其生物学功能。类型siRNA和sRNA-cDNA中的正意链也许能作为模板产生更多的反意链从而扩大双链寡聚核苷酸分子的效能。或者它们可能与甲基转移酶结合,对同源的DNA片段进行甲基化修饰,导致相应基因的活性的下降或完全关闭。此外,当双链siRNA和单链cDNA合用时却能激活两种核酸酶的活性。这样如混合配伍这些不同的单双链寡聚核苷酸就能使酶切效益大大增加,因为,他们能产生协同效应。

单链或双链寡聚核苷酸的用量

Claims (13)
- 本发明要申明的权利是:1.基因皮肤美白产品的配方,它包括但不局限于下述的所有或部分成份:●一个或多个抑制黑色素形成的寡聚核苷酸成份;●一个或多个洛氨酸酶的抑制成份;●一个或多个药效上能接受的载体;●一种或多种还原剂;●一种或多种促进老批脱落的弱酸;●一种或多种抗氧化和防紫外线物质;●一种或多种有益于皮肤的中草药提取液;●其它的活性成份,辅助成份或添加剂。
- 2.在申明(1)中的抑制黑色素形成的寡聚核苷酸成份,它是一个19-25个核苷酸长的双链寡聚核苷酸(dsRNA,dsDNA,或RNA/DNA)或是一个10-30个核苷酸长的单链寡聚核苷酸,如RNA,DNA,或RNA/DNA的杂合体,并能有效地抑制人体内的与黑色素形成有关的基因的表达。
- 3.在申明(1和2)的配方中,寡聚核苷酸成份是指任何一段与黑色素形成有关的基因的同源序列和/或反意序列,这些基因的产物包括但不局限于:Gp75/Tyrp1,Tyrp2,Tyr,P-protein,AIM1,MC1R,PAR2,Myosin Va,Rab27a,OA1 protein,MITF,gp100/pMel17/Silver,Kinesin,Agouti signal protein和POMC。
- 4.在申明(1,2和3)中的寡聚核苷酸成份,它是一组由一种或多种单链和/或双链寡聚核苷酸组成,能与相应的RNA分子中的特异靶序列杂交的分子。
- 5.在申明(1和2)中的寡聚核苷酸成份,它是一系列的能有效地灭活相应的mRNA分子的10-30个核苷酸长的寡聚核苷酸。它们包括但不局限于下列序号的单和/或双链寡聚核苷酸分子。它们是:1,2,3,4,5,6,7,8,9,10,11,12,13,14,15,16,17,18,19,20,21,22,23,24,25,26,27,28,29,30,31,32,33,34,35,36,37,38,39,40,41,42,43,44,45,46,47,48,49,50,51,52,53,54,55,56,57,58,59,60,61,62,63,64,65,66,67,68,69,70,71,72,73,74,75,76,77,78,79,80,81,82,83,84,85,86,87,88,89,90,91,92,93,94,95,96,97,98,99,100一直到248。
- 6.在申明(1)的配方中,寡聚核苷酸成份是一种混合物,由一种或多种不同形式的单链和/或双链寡聚核苷酸分子,以等量或不等量的方式,或以任何其它的配伍形式所组成,特别适用于美白不同类型的肤色。
- 7.申明(1,2,3,4,5和6)中的寡聚核苷酸分子,它是一种自然的单链链和/或双链寡聚核苷酸分子,它也可是一种杂合式的含有修饰核苷酸成分的寡聚核苷酸分子。
- 8.申明(1,2,3,4,5,6和7)中的寡聚核苷酸分子,它们包括但不局限于下列模序(Motif),CGGA,CGGG,CGGT,CGGC,GCCA,GCCC,GCCG,GCCT,TCCC,GATA,GATG,GATC,GATT,CCCA,CCAC,CCGC,和CCTC。
- 9.在申明(2和7)中的杂合式寡聚核苷酸分子,它是指核苷酸与脱氧核苷酸形成的杂合式的单链或双链寡聚核苷酸分子,或是指单链或双链寡聚核苷酸分子中的单核苷酸或单脱氧核苷酸是自然的和/或经不同方法修饰过的单体或双体。
- 10.在申明(1)的配方中,寡聚核苷酸成份的组成可为最终皮肤美白产品总量的0.0000001%到99%。
- 11.一种皮肤美白产品的方法,它是指将单纯的或复合的基因皮肤美白产品用于人的皮肤上
- 12.一种能有效地抑制人的皮肤黑色素细胞中的黑色素的产生的方法,它是指用基因皮肤美白产品的一定量的抑制黑色素形成的寡聚核苷酸成份来转染黑色素细胞从而抑制黑色素的形成。
- 13.在申明(10)的方法中,单纯的基因皮肤美白产品主要由一定量的抑制黑色素形成的寡聚核苷酸成份和一种药效上可接受的载体或一种稀释剂组成的复合物,而复合的基因皮肤美白产品可由多种成份组成,如●一个或多个抑制黑色素形成的寡聚核苷酸成份;●一个或多个洛氨酸酶的抑制成份;●一个或多个药效上能接受的载体;●一种或多种还原剂;●一种或多种促进老批脱落的弱酸;●一种或多种抗氧化和防紫外线物质;●一种或多种有益于皮肤的中草药提取液;●其它的活性成份,辅助成份或添加剂。
