发明摘述
本发明部份取决于此发现,也就是已被基因修饰而增强免疫刺激性细胞因子表达的职业抗原呈递细胞,在没有以抗原使APCs预装载(pre-loading)或兴奋的情况下,可直接注射进入感染或肿瘤的位置或其附近,以诱导对抗与感染或肿瘤相关的抗原的专一性免疫反应。特别地,已发现树突状细胞(DCs),及较佳的是骨髓衍生的树突状细胞(BM-DCs)或CD34+衍生的树突状细胞(CD34+-DCs),其已被基因修饰以增强免疫刺激性细胞因子的表达,较佳地是白介素-12(IL-12),可注射进入感染或肿瘤的位置或其附近,以诱导对抗与注射位置相关的抗原的专一性免疫反应。
因此,本发明的一个观点是提供用以治疗具有感染或肿瘤的个体的方法,包括以一有效量的职业抗原呈递细胞(PAPCs)注射个体的感染或肿瘤的位置或其附近,而此职业抗原呈递细胞已被基因修饰,以增强免疫刺激性细胞因子的表达。
在较佳具体实施例中,基因修饰的APCs是职业抗原呈递细胞,最佳地,PAPCs是选择自CD34+衍生的树突状细胞、骨髓衍生的树突状细胞、单核细胞衍生的树突状细胞、脾脏细胞衍生的树突状细胞、皮肤衍生的树突状细胞、滤泡衍生的树突状细胞、以及胚种中心(germinal center)衍生的树突状细胞所组成的族群中的树突状细胞。在特别较佳的具体实施例中,树突状细胞是CD34+衍生的树突状细胞,其在至少一个因子的存在下培养,此因子是选择自G-CSF、GM-CSF、TNF-α、IL-4、Flt-3配体(ligand)以及kit配体所组成的族群中。

此外,在较佳具体实施例中,APCs已藉由转导编码免疫刺激的该细胞因子的病毒载体而基因修饰,最佳的是,以反转录病毒载体而基因修饰。然而,在其它的具体实施例中,APCs可藉由腺病毒载体或腺病毒相关的病毒载体而基因修饰,或藉由脂质感染(lipofection)、弹道注射(ballistic injection),或在此领域中所熟知的其它基因修饰的方法而修饰。

为了更清楚地以及更简洁地指出并说明申请人所考虑成为本发明的主题,提供以下的定义,以用于所撰写的说明以及附随的申请专利范围所使用的特定的名词。
如同此处所使用的,名词“抗原呈递细胞”或缩写“APC”意指可处理蛋白质抗原、将其破坏成肽、并连同MHC分子在细胞表面上呈递的细胞,它可在细胞表面上与适当的T细胞受体相互作用。名词,抗原呈递细胞,如同此处所使用的,是意指包含职业以及非职业的抗原呈递细胞两者。

I.处理方法
本发明部份衍生自此发现,也就是已被基因修饰而增强免疫刺激性细胞因子表达的抗原呈递细胞,在没有以抗原使APCs兴奋或预装载的情况下,可直接注射进入感染或肿瘤的位置或其附近,以诱导对抗与感染或肿瘤相关的抗原的专一性免疫反应。特别地,已发现职业抗原呈递细胞,例如,树突状细胞(DCs),以及更佳地是骨髓衍生的树突状细胞(BM-DCs)或CD34+衍生的树突状细胞(CD34+-DCs),其已被基因修饰以增强免疫刺激性细胞因子的表达,较佳地是白介素-12(IL-12),可注射进入感染或肿瘤的位置或其附近,以诱导对抗与注射位置相关的抗原的专一的免疫反应。显著的是,在牵涉到注射进入肿瘤的实验中,已发现职业抗原呈递细胞,例如,工程化而原构性地(constitutively)表达IL-12的DCs,在注射的位置以及在远离的位置皆可引起肿瘤的退化,并且在区域性淋巴结以及脾脏皆可诱导肿瘤相关的抗原(TAA)专一性Th1细胞的反应。
因此,一般而言,本发明提供对于患者的治疗的方法,特别是对于具有感染或肿瘤的人类患者,其中,将基因修饰以增强表达免疫刺激性细胞因子的APCs,较佳地是职业抗原呈递细胞,注射进入感染或肿瘤的位置或其附近。因此,此方法包括下列步骤:获得APCs的样本;基因修饰APCs以增强表达免疫刺激性细胞因子;以及将基因修饰的APCs,较佳地是职业抗原呈递细胞,注射进入感染或肿瘤的位置或其附近。在基因修饰APCs之前或之后,以及在将它们注射之前,藉由在此领域中所熟知的细胞培养的标准技术,可将它们选殖地扩增。如果APC是自体的,则获得及修饰细胞的步骤,以及可视需要地,选殖地扩增细胞的步骤,较佳地是在尽可能接近注射的时间执行。然而,如果是使用异质(heterologous)但同基因的APCs,则细胞可在早于注射之前而获得并基因修饰,并且可在使用之前无限期地维持。
以本发明的方法治疗的患者,包括癌症病患以及具有难以治疗的感染的患者。因为较佳的是基因修饰的APCs,较佳的是职业抗原呈递细胞,是直接注射于肿瘤或感染的位置,所以预期本发明的方法将更有助益于患者,其具有至少一种身体上充分定义的肿瘤,或一种身体上充分定义的感染位置,而非一种扩散的、高度转移的癌症,或扩散的感染。另一方面,如同在以下实施例中所显示的,注射进入肿瘤(或感染)存在的位置,可导致全身性免疫反应的发展。因此,本发明的方法,在治疗扩散的、高度转移的癌症,或扩散的感染上是有效的,如果可鉴定至少一个肿瘤或感染的位置,而在这个位置可注射APCs,较佳地是职业抗原呈递细胞,并且可有效地装载肿瘤相关或感染相关的抗原。
在目前较佳的具体实施例中,使用本发明的方法以治疗具有固体肿瘤的病患,将本发明的基因修饰的APCs,较佳的是职业抗原呈递细胞,直接注射进入肿瘤。适当的固体肿瘤可包括,黑色素瘤、肝癌、腺癌、基细胞癌、口腔癌、鼻咽癌、喉癌、膀胱癌、头颈部癌、肾细胞癌、胰脏癌、肺癌、子宫颈癌、卵巢癌、食道癌、胃癌、前列腺癌、睪丸癌以及乳癌。对于许多的这些癌症,已建立DC浸润(infiltration)以及预后之间的相关性。
使用对于皮下、皮内、经皮、肌肉内、腹膜内、或其它的注射形式的标准灭菌技术,注射本发明的基因修饰的APCs。细胞可以生理上可接受的溶液或缓冲液而给药,并且可与其它试剂结合而给药,特别是细胞因子,其可促进APCs存活、装载抗原、通行至引流淋巴结(draining lymph node)或脾脏以及呈递抗原以活化免疫反应的能力。打算引导的细胞数目取决于许多因素,包括打算注射的位置的数目、在期间内打算执行注射的次数、肿瘤或感染损害的大小、以及肿瘤或感染的性质。虽然打算使用的细胞数目随着这样的因素而将有所不同,目前预期将在每个位置、每次治疗,注射104-108个,较佳地105-107个细胞。
有关APCs的选择及分离、免疫刺激细胞因子的选择、以及APCs的基因修饰的细节以及目前较佳的具体实施例,分别说明如下。
II.APCs的选择以及分离
本发明使用抗原呈递细胞APCs,较佳地是职业抗原呈递细胞,最佳地是树突状细胞,其已被基因修饰以增强免疫刺激细胞因子的表达。
较佳地,APCs的原始来源是打算被治疗的患者,使得APCs是自体移植的。从其它个体获得的同种异基因的(allogenic)APCs,也可使用于本发明中,但较佳地,此APCs是衍生自组织兼容的(histocompatible)或同基因的个体,以便提供对于患者的同源的、抗原专一性的T细胞受体的适当的MHC呈递。此外,可创造基因工程化的动物,例如,老鼠或猪,其呈递人类或人类化的MHC蛋白质,以及可视需要地,共同刺激的分子,并且可使用这些动物,以作为对于患者同源的、抗原专一性的T细胞受体,可以有适当的MHC呈递的可更新的APCs来源。
较佳的PAPCs是树突状细胞,特别地是从流通的周围血液采集的CD34+衍生的DCs(CD34+-DCs),以及从骨髓采集的骨髓衍生的树突状细胞(BM-DCs)。其它有助益于本发明的DCs,包括从血液采集的单核细胞衍生的树突状细胞、从骨髓采集的CD34+-DCs、从脾脏采集的脾脏细胞衍生的DCs、皮肤衍生的DCs、滤泡的树突状细胞(FDCs)、以及胚种中心衍生的树突状细胞(GCDCs)。从它们产生及/或局部化的组织中分离这些树突状细胞的方法,是在此领域中所熟知的。
例如,分离BM-DCs的方法是说明于Inaba等人(1992),J.Exp.Med.,卷176:页1693-1702。另外,CD34+祖细胞可从人类脐带或成人血液中获得,并可以细胞因子刺激而分化进入树突状细胞(参见,例如,Caux等人(1996)J.Exp.Med.,卷184:页695-706;Romani等人(1994)J.Exp.Med.,卷180:页83-93)。Flt-3配体在产生树突状细胞的有效度是说明于,例如,Shurin等人(1997),CellImmunol.,卷179:页174-184)。特别较佳的是,与GM-CSF以及IL-4一起培养数天(例如,5天)的BM-DCs或CD34+-DCs。TNF-α以及kit配体,对于增加生长于培养中的DCs的产量,也已显示是有效的(参见,例如,Mayordomo等人(1997),Stem Cells,卷15:页94-103,以及在那里所引用的参考文献),并且可使用TNF-α以及kit配体,以得到本发明的DCs。根据先前的报导(Pierre等人,(1997),Nature,卷388:页787-792;Inaba等人(1993),J.Exp.Med.,卷178:页479-488),如同藉由流动细胞计数法(flow cytometry)以及MLR分析所测定的,大多数这样的DCs可显现未成熟的表现型。DCs只在它们未成熟阶段的期间,具有抓取抗原和加工,以及通行的能力(Pierre等人,(1997),Nature,卷388:页787-792;Inaba等人(1993),J.Exp.Med.,卷178:页479-488;Cella等人(1997),Nature,卷388:页782-787)。
III.细胞因子的选择

本发明的APCs也可基因地修饰,以表达这些细胞因子的变异体(variant)。例如,对于那些具有前形式(pro-forms)以及成熟形式的细胞因子(例如,在讯息肽的切割之前以及之后,或在有限的蛋白质分解而产生一活性的片段之前及之后),本发明的APCs可基因地修饰以表达前形式或成熟形式。其它的变异体,例如,也可使用在细胞因子的活性片段以及异质序列(例如,异质的讯息肽)之间的融合蛋白质。物种的变异体,也可以用它们在人类患者中保有活性的程度而使用。因此,例如,人类APCs可基因地修饰以表达鼠、牛、马、绵羊、猫、狗、非人类灵长动物或人类细胞因子的其它哺乳动物变异体,如果这些物种变异体保有本质上相似于它们的人类同源物的活性。
IV.APCs的基因修饰
本发明的APCs可藉由在此领域中所熟知的任何标准技术而基因地修饰,以导引编码所需的细胞因子的核酸。例如,人类树突状细胞藉由反转录病毒的转导作用而基因修饰的方法,说明于Reeves等人(1996),Cancer Res.,卷56:页5672-5677,以及Specht等人(1997),J.Exp.Med.,卷186:页1213-1221。相似地,骨髓细胞藉由反转录病毒调节的IL-4基因转移而基因修饰的方法,说明于Chambers等人(1992),J.Immunol.,卷149(9):页2899-2905。此外,人类CD34+祖细胞的反转录病毒的转导作用,伴随后续的细胞因子刺激而促进分化并成熟,以离心或不以离心而进入树突状细胞的方法,已说明于Henderson等人(1996),Cancer Res.,卷56:3763-3770,以及Reeves等人(1996),Cancer Res.,卷56:页5672-5677。使用反转录病毒的上清液,代替与反转录病毒的制造者细胞共同培养(Specht等人(1997)),具有许多胜过其它策略的优点,包括:(1)对于细胞没有直接的毒性;(2)获得稳定的基因表达;(3)具有最小的病毒专一性CTL反应,不像以腺病毒载体的方法(参见,例如,Smith等人(1996),J.Virol.,卷70:页6733-6740);以及(4)有较大规模的使用反转录病毒上清液的临床经验。
本发明的APCs的基因修饰可以是短暂的或稳定的。也就是,导引进入APCs的编码细胞因子的核酸序列,可与细胞的基因组DNA分开地存在,并且只短暂地表达,或它们可合并进入细胞的基因组内,并在细胞的生命中持续地表达。例如,使用腺病毒载体的IL-6的短暂表达,说明于Richards等人(1995),Ann.NY.Acad.Sci.,卷762:页282-292。对于短暂地表达的序列,较佳的是,免疫刺激细胞因子的表达至少持续1天,较佳的是数天,以提供足够的时间,使APCs在感染或肿瘤的位置获得抗原、迁移至区域性淋巴结,并且藉由抗原的呈递而活化同源T细胞。
较佳地,用以增强免疫刺激细胞因子表达的基因构筑,包括,可操作地连接至编码细胞因子的序列的原构性激活子,以使基因修饰的APCs原构性地表达细胞因子。然而,另外,如果用于诱导的条件,可与具有或不具有额外的患者的治疗的生理状况下相符合,则可使用可诱导的激活子(例如,给药一诱导物(inducer))。

实施例
反转录病毒载体:反转录病毒DFG-mIL-12、TFG-mIL-12以及DFG-hCD80-neo的构筑,已说明于先前的研究中(Tahara等人,(1995);Zitvogel等人(1996),Eur.J.Immunol.,卷26:页1335-1341)。藉由次选殖从pEGFP-N1(Clontech,Palo Alto,加州)以及pcDNA3.1/Zeo(-)(Invitrogen,Carlsbad,加州)所获得的各别片段,产生MFG-EGFP以及MFG-Zeo(Cormack等人,(1996),Gene,卷173:页33-38)。藉由将这些前病毒的(proviral)构筑转染进入BOSC23或BING包装细胞系(packing cell lines)中,而产生反转录病毒的上清液(Tahara等人,(1995))。藉由将BING或BOSC23所制造的反转录病毒感染这些包装细胞,而产生制造DFG-hCD80-neo反转录病毒的CRE以及CRIP细胞,分别伴随着以G418(Geneticin;Life Technologies公司,Grand Island,纽约)的后续筛选。在转导进入NIH3T3细胞之后,从存活于G418细胞群体中,计算反转录病毒的上清液的效价。

APCs的培养以及基因修饰:使用先前说明的方法(Mayordomo等人,(1995),Zitvogel等人,(1996);Inaba等人(1992),J.Exp.Med.,卷176:页1693-1702)而得到BM-DC培养物。简言之,从牺牲的老鼠的股骨以及胫骨采集老鼠骨髓细胞。在第0天,以0.83摩尔浓度的氯化铵缓冲液,将污染的红血球溶解,以抗体混合物(RA3-3A1/6.1、抗B220;2.43、抗-Lyt 2;GK1.5、抗L3T4;全部来自美国标准菌种中心(ATCC),Rockville,马里兰)以及兔子补体(AccurateChemical and Science Corp.,Westbury,纽约),而将淋巴细胞耗尽。这些细胞在CM中培养隔夜,以移除附着的巨噬细胞,并接着在第1天,将无附着的细胞放置在含有rmGM-CSF(1000单位/毫升)以及rmIL-4(1000单位/毫升)(DC培养液)的新鲜CM中。一般而言,在第6天采集细胞。藉由形态学、表现型以及强烈混合的淋巴细胞反应刺激的活性,以鉴定BM-DCs。藉由流动细胞计数仪的表现型分析显示,在多数的培养细胞中(60-95%),CD11b、CD11c、CD80、CD86以及MHC第I型和第II型的高度的表达。
对于反转录病毒的转导作用,将DC培养液中培养24小时的1×106个BM细胞分装至14毫升的圆底试管,并且悬浮在具有8微克/毫升的聚凝胺(polybrene)、1000单位/毫升的rmGM-CSF以及1000单位/毫升的rmIL-4的1毫升反转录病毒上清液中。将这些细胞在30-32℃、以2500xg的转速,离心2小时(Kotani等人,(1994),Hum.Gene Ther.,卷5:页19-28;Bahnson等人(1995),J.Virol.Methods,卷54:页131-143)。在离心之后,将细胞培养于DC培养液中。在第3天和第4天,重复转导作用的过程。当在可比较的效价的时候,与两性病毒(amphotropic viruses)比较,来自外生性(ectropic)制造者细胞(BOSC23以及CRE)的反转录病毒上清液,可更有效地转导老鼠BM-DCs。因为它们制造最高效价的病毒(2-8×106菌落形成单位(cfu)/毫升),所以使用来自BOSC23细胞的反转录病毒上清液。为了检查老鼠BOS-DCs的转导作用效率,我们做出具有插入的人类CD80(B7.1)或EGFP基因,作为转导作用的标记的反转录病毒载体,并且藉由流动细胞计数仪测定转导作用的效率。在高的转导作用效率时(22-75%),反转录病毒修饰的DCs,可在最后一次转导作用(在第4天)后至少12天,在培养物中表达转殖基因。转导作用的效率与所使用的反转录病毒上清液的效价适切地相关。二种有颜色的免疫萤光染色显示,显著数量的标记(hCD80)-阳性细胞也表达高程度的mCD80以及CD86、MHC第II型以及DEC-205。
以EGFP反转录病毒载体的转导作用显示,可以高效率反转录病毒地转导老鼠BM-DCs。此外,也可用修饰而表达两个IL-12基因(DFG-mIL-12)的反转录病毒载体,而转导BM-DCs。在第4天,完成转导作用的步骤之后,在培养液中的mIL-12p70异质二聚体的浓度,可藉由酶连结免疫吸附分析(ELISA)而测定。如同在第1A图中所显示,在DFG-mIL-12所转导的BM-DC的培养物中,观察到异质二聚体IL-12(p70)的累积,但是在未转导的培养物或是以标记基因转导的细胞中,是没有观察到的。在第6天,采集DCs,清洗两次并转移至一个新的培养盘。在这个时候,IL-12所转导的DCs产生大约80纳克(ng)/106个细胞/48小时的异质二聚体IL-12(第1B图)。来自IL-12所转导的DCs的IL-12制造范围是8-80ng/106个细胞/48小时,并且与每次实验所使用的反转录病毒上清液的效价有关。由基因修饰的DCs所制造的IL-12蛋白质,证实具有生物活性,并且有能力刺激来自Con A处理的脾脏细胞的IFN-γ制造。为了检查IL-12转导作用在DC表现型上的作用,使用流动细胞计数法检查各种细胞表面分子。除了它们表达较多程度的MHC第I型以及第II型分子之外,IL-12所转导的DCs与不转导的或Zeo转导的DCs并无不同
成纤维细胞的培养以及基因修饰:为了比较本发明的基因修饰的APCs的治疗有效度,制备相似修饰的成纤维细胞。简言之,从B6老鼠的肺脏获得同基因的成纤维细胞的初代培养。用剪刀将肺脏的小片块切碎,并且在胶原酶IV、玻尿酸酶V以及去氧核糖核酸酶IV(Sigma,St Louis,密苏里)的三种酶溶液中,在室温下搅拌3小时。以HBSS清洗二次之后,将细胞悬浮液培养于CM中,以获得成纤维细胞的初代培养。藉由感染以G418筛选之后的CRIP-TFG-mIL-12-neo的上清液,而产生IL-12所转导的成纤维细胞。
肿瘤内注射基因修饰的APCs:为了制造癌症病患的动物模式,在第0天,以1×105个细胞的MCA205、B16以及D122肿瘤细胞系,注射到老鼠(每群四或五只)的右腰窝。在第7天,当肿瘤的大小变成10-20平方毫米的时候,肿瘤内注射106个无转导的或转导的BM-DCs。如同先前所说明的,在辐射(5000拉德(rad))之后,使用IL-12所转导的同基因的成纤维细胞以用于肿瘤内注射(Zitvogel等人,(1995))。当老鼠排斥所建立的肿瘤时,在对面的腰窝上,再以较大量的细胞(2×105)激发,以评估对抗肿瘤的保护性的全身免疫的诱导。
所建立的肿瘤的抑制及/或排斥:为了检查肿瘤内注射APCs的抗肿瘤效果(APCs是被基因地修饰以增强免疫刺激细胞因子的表达),将106个无转导或IL-12转导的BM-DCs注射进入所建立的第7天的肿瘤(MCA205、B6以及D122)(肿瘤直径:3-5毫米)。如同第2图所显示的,IL-12转导的DCs显著地抑制这些所建立的肿瘤的生长,导致在5只注射MCA205的老鼠中的2只有最终的排斥。无转导的或Zeo所转导的DCs都不具有抗肿瘤的效果。在这些实验中,基因修饰的DCs的IL-12的制造是29ng/106个细胞/48小时。总计,以IL-12所转导的DCs的单一治疗,导致在14只老鼠中的5只(36%),排斥所建立的MCA205肿瘤。这些没有肿瘤的老鼠排斥后续两次相同量的MCA205的激发,暗示获得对于所注射的肿瘤的免疫记忆。如同在第3图中所显示的,使用皮耳森氏(Pearson’s)线性回归,IL-12所转导的DCs的抗肿瘤效果,与在第28天IL-12的产量相关连(r=-0.80,p<0.05)。在另一个实验中,老鼠一开始以1×105个MCA205细胞皮内注射,接着,在第7天,将HBSS(空心圆圈)、106个Zeo所转导的BM-DCs(三角形)、IL-12所转导的BM-DCs(方块)或IL-12所转导的同基因成纤维细胞(实心圆圈)注射进入所建立的肿瘤。如同在第4图中所显示的,IL-12所转导的DCs的抗肿瘤效果也与IL-12所转导的成纤维细胞的抗肿瘤效果比较。如同先前报导的(Zitvogel等人(1995)),IL-12所转导的成纤维细胞抑制MCA205的生长,然而,它们的单一注射并没有显示任何的肿瘤排斥。然而,当与IL-12所转导的成纤维细胞比较时,IL-12所转导的DCs更有效率地抑制肿瘤生长,而IL-12所转导的成纤维细胞以相似的,但稍微较高的程度表达IL-12(基因修饰的DCs以及成纤维细胞的IL-12产量,分别是13以及22ng/106个细胞/48小时)。也检查每周肿瘤内注射IL-12所转导的DCs的连续治疗。简言之,老鼠一开始以1×105个MCA205细胞皮内注射。在第7天以及第14天,将HBSS(空心圆圈)、106个IL-12所转导的BM-DCs(实心圆圈)、或IL-12所转导的同基因成纤维细胞(三角形),注射进入所建立的肿瘤。如同在第5图中所显示的,当与单一注射比较时,重复注射IL-12所转导的DCs,导致肿瘤的更显著以及延长的抑制(超过60天)。
全身性肿瘤专一的免疫反应:如同以上所提及的,IL-12所转导的DCs的局部抗肿瘤效果,比以IL-12所转导的成纤维细胞所观察到的抗肿瘤效果更为深刻。为了测定是否IL-12所转导的DCs的肿瘤内注射,可诱导对于肿瘤专一的显著的全身性免疫反应,在注射DCs后7天(在肿瘤接种后14天),从具有肿瘤的老鼠中,采集在接种的肿瘤的同一侧鼠蹊部区域中的皮下淋巴结(引流淋巴结)以及脾脏。将这些淋巴细胞与辐射的肿瘤细胞(MCA205)在活体外共同培养,并检查在培养上清液中的IFN-γ以及IL-4的产量。有趣地,当与IL-12所转导的成纤维细胞比较时,注射无转导的或Zeo所转导的DCs,增强从引流淋巴结以及脾脏采集而来的淋巴细胞的肿瘤专一的IFN-γ产量。此外,肿瘤内注射IL-12所转导的DCs,对于肿瘤的再刺激的反应,导致这些淋巴细胞的IFN-γ产量的更大的增强。有趣地,注射DC也增强较少程度的IL-4产量。IFN-γ的产量在MCA205刺激之后专一地释放,但不包含B16或MCA207肿瘤。这些结果暗示,肿瘤内注射的IL-12所转导的DCs流通至引流淋巴结,并且在原位有效地刺激淋巴细胞,以产生IFN-γ。为了证实这个假设,以萤光染剂(RKH-26)将IL-12所转导的DCs染色,并且肿瘤内地注射,并在注射后24小时检查引流淋巴结。在注射后24小时,在引流淋巴结侦测到显著数量的DCs。
此外,也评估来自治疗的老鼠的脾脏细胞的CTL活性。在肿瘤内注射BM-DCs后7天,从每一群的二只老鼠中,采集脾脏细胞并放在一起。在25国际单位/毫升的rhIL-2的存在下,这些细胞(2×106)在活体外以2×105个辐射的(5000拉德)MCA205再刺激。5天后,在标准的51Cr释放分析中,测试再刺激的细胞。CTL的对抗MCA205的活性,藉由来自以HBSS、Zeo所转导的以及IL-12所转导的BM-DCs、以及IL-12所转导的成纤维细胞而处理的族群中的脾脏细胞而测试(第6A图)。在诱导CTL的活性上,Zeo所转导的DCs比IL-12所转导的成纤维细胞更为有效。与使用任何其它策略所观察到的CTL活性比较,IL-12所转导的DCs显著地诱导更高的CTL活性。CTL的对抗MCA205、YAC-1以及同基因成纤维细胞的活性,藉由注射IL-12所转导的BM-DCs的老鼠脾脏细胞而测试(第6B图)。这个活性显露对于MCA205肿瘤是专一性的,并且可用抗CD8的抗体,阻断40-50%,以及可用抗H-2Kb的抗体,阻断24-35%。
为了进一步证实全身性免疫的诱导,检查未处理的肿瘤的相反侧的生长。将1×105个MCA205以及B16肿瘤细胞皮内注射至老鼠的两侧,并且在第7天,当肿瘤面积到达13-20平方毫米时,将IL-12所转导的DCs注射于右侧的肿瘤中。监测两侧的肿瘤生长。第7图显示,肿瘤内注射IL-12所转导的DCs,不只显著地抑制注射的肿瘤的生长(第7A图),也抑制相反侧的未注射的肿瘤的生长(第7B图)。在这些实验中,注射IL-12所转导的DCs后2天,在老鼠血清中并没有侦测到IL-12。
流动细胞计数法:对于BM-DCs的表现型分析,使用连结PE或FITC的对抗老鼠细胞表面分子(CD11b、CD11c、CD80、CD86、Gr-1、H-2Kb、I-Ab以及适当的同型物(isotype)控制组,全部来自PharMingen,圣地亚哥,加州)的单克隆抗体。藉由以NLDC-145抗体的染色,而侦测DEC-205(Serotec公司,牛津,英国)。转导作用的标记hCD80以FITC连结的抗hCD80抗体(PharMingen,圣地亚哥,加州)染色,其并未与老鼠的CD80交叉反应。
活体外细胞因子释放分析:从二只老鼠的一只所采集的输纳(鼠蹊)淋巴结以及脾脏细胞获得淋巴细胞,这些老鼠7天前已接受BM-DCs的肿瘤内注射。如同先前所描述的(Zitvogel等人(1996)),在25国际单位/毫升的rhIL-2(Chiron,Emeryville,加州)的存在下,这些细胞(2×106)与2×105个辐射的(5000拉德)MCA205,在24槽孔的培养盘中共同培养36小时。收集上清液,并在ELISA中估算mIFN-γ以及mIL-4的表达(PharMingen,圣地亚哥,加州)。对于每个分析的敏感度的下限(lower limit)分别是18以及36皮克(pg)/毫升。
细胞毒性T淋巴细胞分析:在肿瘤内注射BM-DCs后7天,从每一群的二只老鼠中,采集脾脏细胞并放在一起。在25国际单位/毫升的rhIL-2的存在下,这些细胞(2×106)在活体外以2×105个辐射的(5000拉德)MCA205再刺激。5天后,使用再刺激的细胞作为效应物(effectors),以用作对抗MCA205、YAC-1以及同基因成纤维细胞的标准4小时-51Cr释放分析。简言之,以100微居里(μCi)的Na2 51CrO4将106的每个标的细胞标示1小时。清洗两次之后,以适当的E/T比例,将这些效应物以及标的细胞放置于96槽孔的圆底培养盘中。在4小时的培养之后,收集上清液(100微升),并用伽码计数器计数放射活性。专一性溶解的百分比以下面的公式计算:%专一性溶解=100×(实验上的释放-自发性的释放)/(最大释放-自发性的释放)。
统计分析:使用非成对的双尾学生氏(Student’s)t-试验,执行统计分析。应用皮耳森氏线性回归以检查相关性。当p值小于0.05时,差异被认为是显著的。
Claims (10)
1.一种治疗具有感染或肿瘤的个体的方法,其包括:
在该个体接近感染或肿瘤的位置注射有效量的基因修饰职业抗原呈递细胞(PAPCs),其中该职业抗原呈递细胞已被基因修饰以增进免疫刺激性细胞因子的表达。
2.根据权利要求1所述的方法,其中该PAPCs是树突状细胞。
3.根据权利要求2所述的方法,其中该树突状细胞是选择自CD34+衍生的树突状细胞、骨髓衍生的树突状细胞、单核细胞衍生的树突状细胞、脾脏细胞衍生的树突状细胞、皮肤衍生的树突状细胞、滤泡衍生的树突状细胞,以及胚种中心衍生的树突状细胞所组成的族群。
4.根据权利要求3所述的方法,其中该树突状细胞是CD34+衍生的树突状细胞,其在至少一个因子的存在下培养,该因子是选择自G-CSF、GM-CSF、TNF-α、IL-4、Flt-3配体以及kit配体所组成的族群。
5.根据权利要求1至4中任一项所述的方法,其中该细胞因子是选择自白介素(例如,IL-1α、IL-1β、IL-2、IL-3、IL-4、IL-6、IL-8、IL-9、IL-10、IL-12、IL-18、IL-19、IL-20)、干扰素(例如,IFN-α、IFN-β、IFN-γ)、肿瘤坏死因子(TNF)、转形生长因子-β(TGF-β)、粒细胞群丛形成促进因子(G-CSF)、巨噬细胞群丛形成促进因子(M-CSF)、粒细胞-巨噬细胞群丛形成促进因子(GM-CSF)、Flt-3配体以及kit配体所组成的族群。
6.根据权利要求1至5中任一项所述的方法,其中该树突状细胞已藉由编码该细胞因子的病毒载体的转导作用而基因修饰。
7.根据权利要求6所述的方法,其中该病毒载体是反转录病毒载体。
8.根据权利要求6所述的方法,其中该病毒载体是选择自腺病毒载体以及腺病毒相关的载体所组成的族群。
9.根据权利要求1至5中任一项所述的方法,其中该树突状细胞已藉由选择自脂质感染(lipofection)以及弹道注射(ballistic injection)所组成的族群中的方法而基因修饰。
10.根据权利要求1至9中任一项所述的方法,其中该个体具有选择自黑色素瘤、肝癌、腺癌、基细胞癌、口腔癌、鼻咽癌、喉癌、膀胱癌、头项部癌、肾细胞癌、胰脏癌、肺癌、子宫颈癌、卵巢癌、食道癌、胃癌、前列腺癌、睪丸癌、以及乳癌所组成的族群中的癌症。
