Abstract

一种可用于化学发光检测,以进一步提高灵敏度的化学发光液,由发光剂、氧化剂、增强剂、辅助增强剂以及缓冲液组成。该系统具有检测灵敏度高、成本低、易于配制的特点,在临床诊断、科学研究、环境及卫生检测、法医鉴定等诸多部门具有广泛的应用前景。

Description

含辅助增强剂和增强剂的增敏化学发光液
技术领域
本发明是一种适用于化学发光检测,以进一步提高灵敏度的化学发光液。
背景技术
生物大分子的检测被誉为现代生物技术的重要组成部分,它们不仅带动了整个生物学的迅速发展,而且在生命科学的各个领域得到了越来越广泛的应用。但以往使用的放射性标记和检测方法存在诸多缺点,如:某些放射性同位素由于半衰期短,不利于贮存和运输;同时由于放射性物质的污染对工作人员的健康及环境造成一定危害,因此需要特殊的防护及废物处理设备;同时在某些应用中该方法的灵敏度也显不足,从而大大限制了它们的普及推广。因此,人们一直在寻找更高灵敏度的生物大分子非同位素标记及检测方法。
上世纪末出现了一类称为化学发光(chemiluminescence)标记及检测技术的新型非同位素标记及检测方法,它不仅具有简便、快速、安全、有效期长的特点,而且具有超过同位素标记技术的灵敏度。因此短短十几年中,该技术在免疫学检测、分子杂交等诸方面得到了越来越广泛的应用。其中的一种技术被称为增敏化学发光(ECL:Enhanced chemiluminescence),其突出的特点是使用了一类称为增强剂(Enhancer)的物质,从而大大增强了其发光强度和检测灵敏度。其中效果最好、应用最广泛的增强剂有两大类,一类是6-羟基苯并噻唑及某些其衍生物,另一类是某些其有对位取代基的酚和苯硼酸类。但这两类增强剂之间并不存在相加或协同作用,即二者同时存在时其增强作用仅相当于两者的平均值,因而无法进一步提高其检测灵敏度。
发明内容
我们经过大量的研究和筛选终于发现:在现有增敏化学发光体系中加入1μMol/L至150μMol/L的氨基比林(也可称为:4-二甲氨基安替比林;或者称为:4-二甲氨基-1,5-二甲基-2-苯基-3-吡唑啉酮)可以增强上述两类增强剂的增敏效果2-3倍,从而进一步大大提高其检测灵敏度。而氨基比林单独存在时无增强效果,所以称之为辅助增强剂(Coenhancer)。进一步研究发现,其分子结果中的4-二甲氨基与辅助增强效果有直接关系,该基团的消失和改变将导致其辅助增敏能力丧失。
同时发现:氨基比林的一个衍生物,即4-(4-羟苯甲亚氨基)安替比林,或者称为:4-(4-羟苯甲亚氨基)-1,5-二甲基-2-苯基-3-吡唑啉酮,虽然不象上述氨基比林那样具有明显的辅助增敏能力,但其单独存在时,却对化学发光具有很强的增强作用。实验发现:在其浓度为1μMol/L至150μMol/L时增强作用较明显,最大增强效果可以达到一千倍左右;如果在该体系中再加入氨基比林,同样也可看到上述辅助增敏作用,即发光强度在原有基础上进一步增加2-3倍。
综上所述,我们认为上述辅助增强剂和增强剂均具有效果明显、成本低、易溶于水的特点,特别是辅助增强效应和辅助增强剂为首次发现。故这两种物质可广泛应用于以化学发光为检测手段的化学、生化、免疫及基因分析产品中,从而大大提高其检测灵敏度。

具体实施方式
以下是该发明的一个实施例,该实施例仅用于说明本发明,但本发明的内容不限于以下
实施例。
在10mMol/LTris-HCl(pH8.7)的缓冲液中加入1.25mMol/L鲁米诺和5mMol/LH2O2作为基础发光液,并先后加入20μMol/L 4-(4-羟苯甲亚氨基)安替比林和20μMol/L氨基比林分别作为增强剂和辅助增强剂。然后在该体系内分别加入不同浓度的辣根过氧化物酶(HRP)并测定其发光强度如下:

Claims (8)

1.含辅助增强剂和增强剂的增敏化学发光液,由发光剂、氧化剂、增强剂、辅助增强剂以及缓冲液组成,其中所述辅助增强剂为氨基比林。
2.根据权利要求1的增敏化学发光液,其中所述增强剂是4-(4-羟苯甲亚氨基)安替比林。
3.根据权利要求2的增敏化学发光液,其中4-(4-羟苯甲亚氨基)安替比林的浓度为1μMol/L至150μMol/L。
4.权利要求1-3中任一项的增敏化学发光液,其中氨基比林的浓度为1μMol/L至150μMol/L。
5.权利要求4的增敏化学发光液,其中所述发光剂为鲁米诺,所述氧化剂为H2O2,所述缓冲液为Tris-HCl。
6.含增强剂的增敏化学发光液,由发光剂、氧化剂、增强剂以及缓冲液组成,其中所述增强剂为4-(4-羟苯甲亚氨基)安替比林。
7.权利要求6的增敏化学发光液,其中所述4-(4-羟苯甲亚氨基)安替比林的浓度为1μMol/L至150μMol/L。
8.权利要求7的增敏化学发光液,其中所述发光剂为鲁米诺,所述氧化剂为H2O2,所述缓冲液为Tris-HCl。
