Experimental example 2
Animal experiment:
The model preparation is prepared by orally and intragastrically administering a model preparation compound diphenoxylate, establishing a constipation model of the mice, and measuring the first-grain black stool defecation time, the defecation grain number and the defecation weight of the mice within 5 or 6 hours to reflect the defecation condition of the model mice.
Adult male mice were selected and weighing 18-22g, and were divided into 5 groups of 10 animals each. Surgical scissors, ophthalmic forceps, ruler, syringe, balance, activated carbon powder, gum arabic and compound diphenoxylate.
Preparing ink: accurately weighing 100g of gum arabic, adding 800mL of water, boiling until the solution is transparent, weighing 50g of activated carbon (powder) and adding into the solution, boiling for three times, cooling the solution, adding water to 1000mL, keeping the volume at 4 ℃ in a refrigerator, and shaking uniformly before use.
Preparing a compound diphenoxylate suspension: compound diphenoxylate tablet, each tablet contains 2.5mg of compound diphenoxylate. Taking a corresponding amount of compound diphenoxylate tablets, grinding the tablets into powder by a mortar, adding water to 100mL, and preparing the tablets before use.
Dose grouping and time of sample administration: model mice were randomly divided into three high, medium and low dose groups, a blank control group and a model control group. The traditional Chinese medicine composition prepared in the example 1 is given to 2.5 times of human clinical dose in a low dose group, the traditional Chinese medicine composition prepared in the example 1 is given to 5 times of human clinical dose in a medium dose group, and the traditional Chinese medicine composition prepared in the example 1 is given to 10 times of human clinical dose in a high dose group. The administration was continued for 7 days.
The experimental method comprises the following steps: animals in each group were given a 7 day period of time without water withdrawal and no water withdrawal, and the model control group and three dose groups were given a suspension of diphenoxylate (7.5 mg/kg BW) by gavage, and the blank control group was given water gavage. After 0.5 hours, each of the dosing groups was given an ink (5% active carbon powder, 10% gum arabic) with the corresponding test sample, respectively, and the negative and model control groups were given an ink lavage. The black discharge time, number of black discharge particles in 5 or 6 hours and weight of each animal were recorded from the start of ink filling.
Experimental results: as shown in table 2, compared with the model group, the first granule defecation time of each dosage group and the positive control group of the traditional Chinese medicine composition prepared by the invention is obviously shortened, the defecation number is obviously increased, and the gastrointestinal condition can be effectively improved.

The treatment course is 14 days, and the curative effect and the safety are evaluated after 1 treatment course.
1. Efficacy test:
The effect is shown: the symptoms and signs are obviously improved within 14 days of administration. The integral value of syndrome is reduced by more than or equal to 70 percent.
The method is effective: symptoms and signs are all improved within 14 days of administration. The integral value of syndrome is reduced by more than or equal to 30 percent.
Invalidation: symptoms and signs were not significantly improved or even aggravated within 14 days of administration. The integral reduction value of syndrome is less than 30%.
Note that: the calculation formula (nimodipine method) is: [ (pre-treatment integral-post-treatment integral)/(pre-treatment integral ] ×100%
Evaluation of syndrome curative effect:
the effect is shown: symptoms or signs decreased by 2-grade (3 to 1-grade) after treatment compared to before treatment.
The method is effective: symptoms or signs decreased 1-way (3 to 2 points, 2 to 1 point) after treatment compared to before treatment.
Invalidation: the symptoms or signs are not obviously improved after treatment compared with those before treatment, and the standard is not met.

2. Safety evaluation:
Stage 1: safety and no adverse reaction.
2 Stages: is safer, has adverse reaction, and can be continuously administered without any treatment.
3 Stages: has safety problem and moderate adverse reaction, and can be continuously administered after treatment.
4 Stages: the test was discontinued due to adverse reactions.
Experimental results: in the experimental process, no adverse reaction is found, and the safety is good.
