Abstract
本发明涉及干细胞生物技术领域,公开了一种诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基及多巴胺能神经前体细胞的制备方法与应用。按照使用时期划分,所述诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基包括Ⅰ期培养基、Ⅱ期培养基、Ⅲ期培养基和IV期培养基;所述多巴胺能神经前体细胞能表达多巴胺能神经前体细胞相关基因,并能分化成能分泌多巴胺的多巴胺能神经元细胞,本发明实现的人多能干细胞向多巴胺能神经前体细胞分化的2D分化,方法操作简单快速,诱导周期短,可在21天内获得多巴胺能神经前体细胞,为帕金森模型构建以及药物筛选提供稳定的研究平台。

技术领域
本发明涉及干细胞生物技术领域,公开了一种诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基及多巴胺能神经前体细胞的制备方法与应用。
背景技术
神经退行性疾病,是一种大脑和脊髓的细胞神经元丧失的疾病状态,随着时间的推移而恶化,以导致功能障碍。帕金森病(Parkinson’s disease,PD)是一种常见的神经系统变性疾病,老年人多见,平均发病年龄为60岁左右,40岁以下起病的青年帕金森病较少见。我国65岁以上人群PD的患病率大约是1.7%。帕金森病最主要的病理改变是中脑黑质多巴胺(dopamine,DA)能神经元的变性死亡,由此而引起纹状体DA含量显著性减少而致病。帕金森病的确切病因仍不清楚,遗传因素、环境因素、年龄老化、氧化应激等均可能参与PD多巴胺能神经元的变性死亡过程。帕金森临床上主要表现为肌强直、运动迟缓、静止性震颤、姿势步态障碍、语言能力以及其他功能。药物治疗是帕金森病最主要的治疗手段,左旋多巴制剂是最有效的药物。手术治疗是药物治疗的一种有效补充。目前治疗手段主要是改善症状,但尚不能阻止病情的进展。
人诱导性多能干细胞(iPSC)和人胚胎干细胞(ESC)均具有多向分化和强大自我复制的潜能,适用于大规模培养,批次间特性相对稳定。相对于ESC,iPSC可以来源于自身,伦理争议较小,是细胞药物最好的种子细胞来源,可以大规模体外生产多巴胺能神经前体细胞,满足临床移植的需求。
目前,帕金森病的细胞治疗开始应用于临床试验,受到医学领域、干细胞生物学领域与病人的热切关注。将多能干细胞诱导分化成多巴胺能神经前体细胞的方法有多种,大多数方法是在滋养层细胞的基础上,利用一些细胞因子和小分子进行诱导,诱导过程周期较长、效果不稳定、效率低,不利于后续临床级细胞制剂的生产等问题。
发明内容
本发明提供了诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基及多巴胺能神经前体细胞的制备方法与应用,解决了现有技术,诱导过程周期较长、效果不稳定、效率低,不利于后续临床级细胞制剂的生产的技术问题。
为实现上述目的,本发明公开了一种诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基,按照使用时期划分为Ⅰ期培养基、Ⅱ期培养基、Ⅲ期培养基和IV期培养基;
所述I期培养基包括N-2、B27、Glutmax、KnockOut血清替代物、hLIF、CHIR99021、SB431542、DAPT小分子,其中KnockOut血清替代物浓度为5%-15%,hLIF浓度为7.5-17.5ng/mL,CHIR99021浓度为465-1395ng/mL,SB431542浓度为768-1536ng/mL,DAPT浓度为2162-4324ng/mL;
所述II期培养基包括N-2、B27、Glutmax、KnockOut血清替代物、CHIR99021、SB431542、SHH、Purmorphamine,其中KnockOut血清替代物浓度为5%-15%,CHIR99021浓度为465-1395ng/mL,SB431542浓度为384-1536ng/mL,SHH浓度为100-200ng/mL、Purmorphamine浓度为521-1563ng/mL;
所述的III期培养基包括DMEM/F12、N-2、B27、cAMP、Vitamin C、FGF8b,其中cAMP浓度为250-400ng/mL,Vitamin C浓度为17.6-35.2μg/mL,FGF8b浓度为50-200ng/mL;
所述IV期培养基包括DMEM/F12、N-2、B27、cAMP、Vitamin C、BDNF、GDNF、IGF-1、TGF-β3、db-cAMP,其中cAMP浓度为250-400ng/mL,Vitamin C浓度为17.6-35.2μg/mL,BDNF浓度为5-20ng/mL,GDNF浓度为5-20ng/mL,IGF-1浓度为5-20ng/ml,TGF-β3浓度为0.5-2ng/mL,db-cAMP浓度为123-492μg/mL。
进一步的,所述诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基将诱导人多能干细胞分化为多巴胺能神经前体细胞,后再诱导分化为成熟的多巴胺能神经元细胞。
进一步的,所述CHIR99021的浓度为930ng/mL。
进一步的,所述多巴胺能神经前体细胞表达FOXA2+、OTX2+、LMXIA+和CORIN+,另外,FOXG1、PTX3和SOX10表达低于5.6%。
进一步的,所述多巴胺能神经元细胞表达MAP2+、Nurr1+、TH+、PTX3+。
本发明的另一个方面,提供了一种所述的诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基制备多巴胺能神经前体细胞的方法,包括以下步骤,
步骤一:Day-1,使用单细胞传代方式将人诱导多能干细胞以2x 105/cm2的接种密度接种于Laminin521包被的2D容器中,添加Rock抑制剂Y27632的多能干细胞的培养基培养过夜;
步骤二:Day0至Day6,弃去细胞培养上清液,使用不添加Rock抑制剂Y27632的I期培养基,维持步骤一中的人诱导多能干细胞的定向分化,每天换液;
步骤三:Day7,弃去细胞培养上清液,使用DPBS洗涤贴壁细胞,弃去DPBS,使用Accutase将步骤二中的分化细胞消化解离成单细胞,使用添加有Rock抑制剂Y27632的I期培养基进行终止,300x g,离心5min,弃上清,获得细胞沉淀,再使用含有Rock抑制剂Y27632的II期分化培养基重悬细胞沉淀制备成单细胞悬液,接种于Laminin521包被的2D容器中进行过夜培养;
步骤四:Day8至Day13,弃去细胞培养上清液,使用不含有Rock抑制剂Y27632的II期分化培养基培养分化细胞,每天换液;
步骤五:Day14,弃去细胞培养上清液,使用DPBS洗涤贴壁细胞,弃去DPBS,使用Accutase将步骤四中的分化细胞消化解离成单细胞,使用添加Rock抑制剂Y27632的II期培养基进行终止,300x g,离心5min,弃上清,获得细胞沉,再使用含有Rock抑制剂Y27632的III期分化培养基重悬细胞沉淀制备成单细胞悬液,接种于Laminin和Poly-L-ornithine包被的2D容器中进行过夜培养;
步骤六:Day15至Day20,弃去细胞培养上清液,使用含有Rock抑制剂Y27632的III期分化培养基培养分化细胞,每天换液,进行定向分化培养;
步骤七:Day21,弃去细胞培养上清液,使用DPBS洗涤贴壁细胞,弃去DPBS,使用Accutase将步骤六中的多巴胺能神经前体细胞消化解离成单细胞,使用添加Rock抑制剂Y27632的III期培养基进行终止,300x g,离心5min,弃上清,获得细胞沉淀,得到多巴胺能神经前体细胞。
进一步的,在iPSC阶段、NPC阶段及DAP阶段,Y27632的浓度是10μM。
本发明还提供了,一种验证多巴胺能神经前体细胞具有定向分化成多巴胺神经元能力的方法,包括以下步骤:
步骤一:使用含有Y27632的IV期分化培养基,重悬多巴胺能神经前体细胞沉淀,制备成单细胞悬液,接种于Laminin和Poly-L-ornithine包被的2D容器中进行过夜培养24h;
步骤二:弃去细胞培养上清液,使用不含有Rock抑制剂Y27632的IV期分化培养基培养多巴胺能神经前体细胞,每天换液,进行定向分化成熟培养2周,获得多巴胺能神经元细胞;
步骤三:弃去细胞培养上清液,使用DPBS洗涤贴壁细胞,弃去DPBS,将步骤二中的多巴胺能神经元细胞用KCL溶液刺激,用ELISA检测多巴胺的分泌,同时通过qPCR、免疫荧光及流式检测多巴胺能神经元细胞。
一种包含多巴胺能神经前体细胞用于治疗帕金森病的细胞药物。
本发明相比现有技术的有益效果是:
本发明公开的诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基及多巴胺能神经前体细胞的制备方法与应用,适用于ESC细胞系与其他不同来源的iPSC细胞系;将iPSC诱导成多巴胺能神经前提的周期为21天,诱导周期相对较短,有效降低时间成本;利用本发明对不同细胞株进行了多轮诱导分化,均发现分化效率较高,分化体系稳定;本发明所述的培养基中不含有动物血清或动物血清白蛋白,分化方法没有使用滋养层细胞,避免了带有动物源性病原体的风险,可应用于科研及临床实验,且不存在安全性问题。

本发明提供了一种诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基及多巴胺能神经前体细胞的制备方法与应用。本发明的诱导体系涉及LDN193189、SB431542、SHH、Purmorphamine、FGF8、CHIR99021、BDNF、GDNF、dbcAMP、TGF-β3、DAPT等小分子以及细胞因子,在不同时期以固定比例添加到基础培养基中,用以诱导分化人多能干细胞(hPSC)向多巴胺能神经前体细胞分化。所述多巴胺能神经前体细胞能表达多巴胺能神经前体细胞相关基因,其中FOXA2+和OTX2+表达量高于90%、CORIN+表达量高于95%、LMXIA+表达量高于85%,并能发育为成熟的多巴胺能神经元(mDAN),并分泌多巴胺。本发明实现人多能干细胞(hPSC)向多巴胺能神经前体细胞分化的2D分化,方法操作简单快速,可在21天内获得多巴胺能神经前体细胞,为帕金森模型构建以及药物筛选提供稳定的研究平台。

N-2是一种化学成分确定的无血清添加剂,用于支持源自外周神经系统和中枢神经系统的原代神经母细胞瘤及有丝分裂后神经元的生长和表达。
B-27是一种优化的无血清添加剂,用于支持胚胎、出生后和成年海马神经元和其他中枢神经系统神经元的生长和活性维持。
Glutmax是一独立的L-丙氨酰-L-谷氨酰胺二肽,在水溶液中更稳定,不会自发降解。从而实现高效的能量代谢和高生长产量,而不会产生对细胞造成不利影响的过量氨。
KnockOut血清替代物是一种无血清添加剂,支持在成纤维细胞饲养层上培养的多能干细胞的生长,可通过添加细胞所需生长因子进行定向分化。
hLIF(Human Leukemia Inhibitory Factor)是人白血病抑制因子,是一种多效性细胞因子,能高效抑制干细胞的自发分化,促进干细胞增殖分裂。同时具有多样性的细胞学作用,可调节机体许多组织细胞的功能,包括血细胞、胚胎细胞、肝细胞、神经细胞、骨细胞以及肿瘤细胞。
CHIR99021是一种糖原合酶激酶3(GSK3)抑制剂,可促进体细胞重编程为干细胞,也可以减少神经元中Tau蛋白磷酸化。
SB431542是TGF-β受体激酶抑制剂,可诱导多能干细胞高效分化为神经元。
DAPT是一种强效的γ分泌酶复合体抑制剂,可以促进神经细胞的分化。
Shh(Sonic hedgehog)是音猬因子,主要参与调节腹侧细胞的分化,在胚胎早期发育对细胞的分化方向和空间分布起重要调控作用。
Purmorphamine又称嘌吗啡胺,是平滑受体激动剂,是一种嘌呤衍生物,是第一个针对Smoothened蛋白开发的小分子激动剂,通过直接结合激活Smoothened,活化Hedgehog通路,和Shh一起控制神经干细胞的命运。
FGF8b是一种成纤维细胞生长因子,促进细胞增殖和分化,加速组织修复和再生过程。
cAMP(环磷酸腺苷)是一种胞内信使分子,负责包括神经元在内的许多细胞的功能,促进轴突的生长,维持神经元间的通信。
Vitamin C又叫L-抗坏血酸,是一种水溶性维生素,是一种抗氧化剂,减少细胞凋亡。
GDNF(胶质细胞源性神经营养因子)是一种神经营养因子,在中枢神经系统和外周神经系统发育的不同阶段均能促进不同神经元亚群的存活。是一种能促进运动神经元、中脑多巴胺能神经元、浦肯野细胞和交感神经元存活的分泌性蛋白。
BDNF(脑源性神经营养因子)属于一个被称为神经营养因子的蛋白质家族,对胚胎神经元的生长、发育、诱导分化及突触连接具有调节功能,在神经网络的形成中发挥着重要作用,它也参与了活性依赖的神经元可塑性。GDNF与BDNF一起调控神经元的生长、发育、分化和存活。
IGF-1(胰岛素样生长因子)是一种在肝脏中产生的内分泌激素,可作用于神经元、星形胶质细胞和少突胶质细胞,通过减少细胞凋亡和促进细胞分化成熟在中枢神经系统发育过程中发挥着重要作用。
TGF-β3(transforming growth factor-β3)是转化生长因子-β3,属于一组调节细胞生长和分化的TGF-β超家族,在促进神经元分化、生长、存活和轴突再生中发挥重要作用。
db-cAMP(Dibutyryl-cAMP)是一种细胞可渗透性PKA激活剂,通过模拟内源性cAMP发挥作用,可以诱导神经突起的生长。
本发明公开了一种培养基,该种培养基按照使用阶段分为4组,分别称为Ⅰ期培养基(表1),Ⅱ期培养基(表2),Ⅲ期培养基(表3)和IV期培养基(表4)。

实施例1
一种诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基,按照使用时期划分为Ⅰ期培养基、Ⅱ期培养基、Ⅲ期培养基和IV期培养基;
I期培养基包括有N-2、B27、Glutmax、KnockOut血清替代物、hLIF、CHIR99021、SB431542、DAPT小分子,且在hPSC的最适工作浓度如下:其中KnockOut血清替代物浓度为5%,hLIF浓度为7.5ng/mL,CHIR99021浓度为465ng/mL,SB431542浓度为768ng/mL,DAPT浓度为2162ng/mL。
II期培养基包括有N-2、B27、Glutmax、KnockOut血清替代物、CHIR99021、SB431542、SHH、Purmorphamine,并且对神经祖细胞的最适工作浓度如下:其中KnockOut血清替代物浓度为5%,CHIR99021浓度为465ng/mL,SB431542浓度为384ng/mL,SHH浓度为100ng/mL、Purmorphamine浓度为521ng/mL。
III期培养基包括有DMEM/F12、N-2、B27、cAMP、Vitamin C、FGF8b,并且对神经祖细胞的最适工作浓度如下:其中cAMP浓度为250ng/mL,Vitamin C浓度为17.6μg/mL,FGF8b浓度为50ng/mL。
IV期培养基包括有DMEM/F12、N-2、B27、cAMP、Vitamin C、BDNF、GDNF、IGF-1、TGF-β3、db-cAMP,其中cAMP浓度为250ng/mL,Vitamin C浓度为17.6μg/mL,BDNF浓度为5ng/mL,GDNF浓度为5ng/mL,IGF-1浓度为5ng/ml、TGF-β3浓度为0.5ng/mL,db-cAMP浓度为123μg/mL。
实施例2
一种诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基,按照使用时期划分为Ⅰ期培养基、Ⅱ期培养基、Ⅲ期培养基和IV期培养基;
I期培养基包括有N-2、B27、Glutmax、KnockOut血清替代物、hLIF、CHIR99021、SB431542、DAPT小分子,且在hPSC的最适工作浓度如下:其中KnockOut血清替代物浓度为10%,hLIF浓度为12.5ng/mL,CHIR99021浓度为930ng/mL,SB431542浓度为1052ng/mL,DAPT浓度为3243ng/mL。
II期培养基包括有N-2、B27、Glutmax、KnockOut血清替代物、CHIR99021、SB431542、SHH、Purmorphamine,并且对神经祖细胞的最适工作浓度如下:其中KnockOut血清替代物浓度为10%,CHIR99021浓度为930ng/mL,SB431542浓度为768ng/mL,SHH浓度为150ng/mL、Purmorphamine浓度为1042ng/mL。
III期培养基包括有DMEM/F12、N-2、B27、cAMP、Vitamin C、FGF8b,并且对神经祖细胞的最适工作浓度如下:其中cAMP浓度为325ng/mL,Vitamin C浓度为26.4μg/mL,FGF8b浓度为125ng/mL。
IV期培养基包括有DMEM/F12、N-2、B27、cAMP、Vitamin C、BDNF、GDNF、IGF-1、TGF-β3、db-cAMP,其中cAMP浓度为325ng/mL,Vitamin C浓度为0.2mM,BDNF浓度为10ng/mL,GDNF浓度为10ng/mL,IGF-1浓度为10ng/ml,TGF-β3浓度为1.25ng/mL,db-cAMP浓度为332μg/mL。
实施例3
一种诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基,按照使用时期划分为Ⅰ期培养基、Ⅱ期培养基、Ⅲ期培养基和IV期培养基;
I期培养基包括有N-2、B27、Glutmax、KnockOut血清替代物、hLIF、CHIR99021、SB431542、DAPT小分子,且在hPSC的最适工作浓度如下:其中KnockOut血清替代物浓度为15%,hLIF浓度为17.5ng/mL,CHIR99021浓度为1395ng/mL,SB431542浓度为1536ng/mL,DAPT浓度为4324ng/mL。
II期培养基包括有N-2、B27、Glutmax、KnockOut血清替代物、CHIR99021、SB431542、SHH、Purmorphamine,并且对神经祖细胞的最适工作浓度如下:其中KnockOut血清替代物浓度为15%,CHIR99021浓度为1395ng/mL,SB431542浓度为1536ng/mL,SHH浓度为200ng/mL、Purmorphamine浓度为1563ng/mL。
III期培养基包括有DMEM/F12、N-2、B27、cAMP、Vitamin C、FGF8b,并且对神经祖细胞的最适工作浓度如下:其中cAMP浓度为400ng/mL,Vitamin C浓度为35.2μg/mL,FGF8b浓度为200ng/mL。
IV期培养基包括有DMEM/F12、N-2、B27、cAMP、Vitamin C、BDNF、GDNF、IGF-1、TGF-β3、db-cAMP,其中cAMP浓度为400ng/mL,Vitamin C浓度为35.2μg/mL,BDNF浓度为20ng/mL,GDNF浓度为20ng/mL,IGF-1浓度为20ng/ml,TGF-β3浓度为2ng/mL,db-cAMP浓度为491μg/mL。
本发明优化了CHIR99021的使用浓度,在使用本发明提供的方法及培养基组分的基础上,添加不同浓度的CHIR99021,例如浓度为0μM、0.7μM、1μM、2μM、3μM,对人iPSC进行诱导分化,得到多巴胺能神经前体细胞,使用Accutase消化解离成单细胞,经过终止消化、离心,获得细胞沉淀样本,通过qPCR检测样本的分子标志物表达,通过对比,筛选出了CHIR99021最佳使用浓度是2μM,能够获得分化效率最好的多巴胺能神经前体细胞,此时的多巴胺能神经前体细胞标志物FOXA2+、OTX2+、CORIN+、LMXIA+高表达,其中所述多巴胺能神经前体细胞群中FOXG1、PTX3和SOX10表达低于5.6%,图1为不同CHIR99021浓度下iPSC分化成DAP的分子标志物表达。
本发明提供了Rock抑制剂Y27632在不同阶段的最佳使用浓度,使用本发明提供的方法及培养基组分,使用最佳浓度2μM的CHIR99021,在不同阶段细胞培养的第一天,添加不同浓度的Y27632,例如浓度为0μM、5μM、10μM、20μM、40μM,但不限于此。经过比较不同浓度下不同细胞阶段的细胞活率,筛选出不同阶段的最有浓度如下:在iPSC阶段Y27632的最佳使用浓度是10μM(图2A),在NPC阶段及DAP诱导至成熟的多巴胺神经元阶段,Y27632的最佳使用浓度是5μM(图2B-2D)。综合得到一个浓度组合,早期iPSC细胞培养阶段,使用最佳浓度10μM的Y27632提高细胞存活率,后续的神经相关细胞培养阶段,使用最佳浓度5μM的Y27632提高细胞存活率。
图2中,图2A为:使用CCK-8细胞计数试剂盒评估Y-27632处理24h的分化的人iPSC的存活率(n=3)。
图2B为:使用CCK-8细胞计数试剂盒评估Y-27632处理24h的分化的人iPSC来源的NPC的存活率(n=3)。
图2C为:使用不同浓度的Y27632将DAP诱导为mDAN后MAP2的荧光强度图。
图2D为:使用不同浓度的Y27632将DAP诱导为mDAN后MAP2的流式阳性细胞比率。
图2E为:免疫荧光检测通过使用不同浓度的Y27632将DAP诱导为mDAN后MAP2的表达。
实施例4
使用上述培养基(表1-表3),添加上述本实施例中最佳使用浓度的CHIR99021,使用上述本实施例中的一组Y27632的最佳浓度组合,公开了一种诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基及多巴胺能神经前体细胞的制备方法,具体流程图如图3所示,包括以下步骤:
步骤一:Day-1,使用单细胞传代方式将人诱导多能干细胞以2x 105/cm2的接种密度接种于Laminin521包被的2D容器中,添加Rock抑制剂Y27632的多能干细胞的培养基培养过夜;
步骤二:Day0至Day6,弃去细胞培养上清液,使用不添加Rock抑制剂Y27632的I期培养基,维持步骤一中的人诱导多能干细胞的定向分化,每天换液;
步骤三:Day7,弃去细胞培养上清液,使用DPBS洗涤贴壁细胞,弃去DPBS,使用Accutase将步骤二中的分化细胞消化解离成单细胞,使用添加有Rock抑制剂Y27632的I期培养基进行终止,300x g,离心5min,弃上清,获得细胞沉淀,再使用含有Rock抑制剂Y27632的II期分化培养基重悬细胞沉淀制备成单细胞悬液,接种于Laminin521包被的2D容器中进行过夜培养;
步骤四:Day8至Day13,弃去细胞培养上清液,使用不含有Rock抑制剂Y27632的II期分化培养基培养分化细胞,每天换液;
步骤五:Day14,弃去细胞培养上清液,使用DPBS洗涤贴壁细胞,弃去DPBS,使用Accutase将步骤四中的分化细胞消化解离成单细胞,使用添加Rock抑制剂Y27632的II期培养基进行终止,300x g,离心5min,弃上清,获得细胞沉,再使用有Rock抑制剂Y27632的III期分化培养基重悬细胞沉淀制备成单细胞悬液,接种于Laminin和Poly-L-ornithine包被的2D容器中进行过夜培养;
步骤六:Day15至Day20,弃去细胞培养上清液,使用不含有Rock抑制剂Y27632的III期分化培养基培养分化细胞,每天换液,进行定向分化培养;
步骤七:Day21,弃去细胞培养上清液,使用DPBS洗涤贴壁细胞,弃去DPBS,使用Accutase将步骤六中的多巴胺能神经前体细胞消化解离成单细胞,使用添加Rock抑制剂Y27632的III期培养基进行终止,300x g,离心5min,弃上清,获得细胞沉淀,获得大量多巴胺能神经前体细胞,用于质检及后续分化为成熟多巴胺神经元的验证实验。
实施例5
本发明使用上述IV期培养基(表4),按照图3的流程,提供了一种验证多巴胺能神经前体细胞具有定向发育为成熟多巴胺神经元能力的方法,具体步骤如下:
步骤一:使用含有Rock抑制剂Y27632的本发明所述的IV期分化培养基,将获得的多巴胺能神经前体细胞沉淀,制备成单细胞悬液,接种于Laminin和Poly-L-ornithine包被的2D容器中进行过夜培养24h。
步骤二:弃去细胞培养上清液,使用不含有Rock抑制剂Y27632的IV期分化培养基培养多巴胺能神经前体细胞,每天换液,进行定向分化成熟培养2周,获得多巴胺能神经元细胞;
步骤三:弃去细胞培养上清液,使用DPBS洗涤贴壁细胞,弃去DPBS,将步骤二中的多巴胺能神经元细胞用KCL溶液刺激,用ELISA检测多巴胺的分泌。同时,通过qPCR、免疫荧光及流式检测多巴胺能神经元细胞。
图3中,iPSC为诱导多能干细胞;NPC P0为iPSC直接分化成的神经祖细胞;NPC P1为第一次传代培养后的NPC;DAP为多巴胺能神经前体细胞;mDAN为成熟多巴胺神经元。
实施例6
添加上述本实施例中最佳使用浓度的CHIR99021,使用实施例4的方法,将人iPSC分化成多巴胺能神经前体细胞,并使用实施例5中的方法验证多巴胺能神经前提细胞(DAP)可发育成成熟的多巴胺神经元(mDAN),经过KCL刺激后,能分泌多巴胺,整个应用过程中,在不同的培养阶段观察细胞的形态结构(图4),同时对不同阶段的细胞进行免疫荧光和流式检测(图5-图6),或qPCR检测(图7)其分子标志物,并对成熟的多巴胺神经元(mDAN)进行免疫荧光、流式检测,还同时进行了多巴胺分泌的ELISA检测(图8)。
在整个细胞培养过程中,使用不同阶段的培养基,获得预期内的不同阶段的细胞类型,形态结构正常,细胞活率较高(图4),对起始种子细胞iPSC进行了免疫荧光和流式检测(图5),发现iPSC维持较高的干性标志物表达,对获得的多巴胺能神经前体细胞进行了免疫荧光、流式检测(图6,其中,图6A为免疫荧光检测DAP的分子标志物示意图,图6B为流式检测DAP的分子标志物示意图),同时,对不同阶段的细胞类型进行qPCR检测(图7),结果均显示制备的多巴胺能神经前体细胞群,其表达FOXA2+、OTX2+、LMXIA+和CORIN+,其中FOXA2+和OTX2+表达量高于90%、CORIN+表达量高于95%、LMXIA+表达量高于85%;另外,FOXG1、PTX3、SOX10表达低于5.6%。证实制备的DAP纯度较高,同时iPSC残留较少,致瘤风险较低,安全性较高。
通过免疫荧光(图8A)、流式(图8B)检测mDAN阶段细胞的分子标志物表达,显示通过本发明制备的DAP能发育为成熟的多巴胺能神经元细胞,其表达MAP2+、Nurr1+、TH+、PTX3+,通过ELISA检测mDAN的多巴胺分泌,发现通过本发明制备的多巴胺能神经元细胞能够分泌较高的多巴胺(图8C),图8A为免疫荧光检测mDAN的分子标志物示意图,图8B为流式检测mDAN的分子标志物示意图,图8C为ELISA检测mDAN的多巴胺分泌示意图,证实本发明可以制备能分化成多巴胺神经元并分泌多巴胺的多巴胺能神经前提细胞,为后续帕金森的细胞治疗准备种子细胞。
以上所有实施例均用于帮助理解本发明所公开的培养基的配制、小分子浓度的优化、培养方法及应用。所有实施例证实使用本发明所述的培养基和本发明提出的培养方法,对不同iPSC细胞株进行了多轮诱导分化,均发现分化效率较高,分化稳定,均可制备出能分化成多巴胺神经元并分泌多巴胺的多巴胺能神经前提细胞,为后续帕金森的细胞治疗准备种子细胞,具有广泛的应用前景。由于不同的细胞系具有不同的特性,分化效率略有差别,本领域的研发人员,可以在本发明框架的前提下,进行一定浓度范围内的优化,均可达到制备DAP的目的,这些优化也属于本发明权利要求的保护范围。
Claims (9)
1.一种诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基,其特征在于:按照使用时期划分为Ⅰ期培养基、Ⅱ期培养基、Ⅲ期培养基和IV期培养基;
所述I期培养基包括N-2、B27、Glutmax、KnockOut血清替代物、hLIF、CHIR99021、SB431542、DAPT小分子,其中KnockOut血清替代物浓度为5%-15%,hLIF浓度为7.5-17.5ng/mL,CHIR99021浓度为465-1395ng/mL,SB431542浓度为768-1536ng/mL,DAPT浓度为2162-4324ng/mL;
所述II期培养基包括N-2、B27、Glutmax、KnockOut血清替代物、CHIR99021、SB431542、SHH、Purmorphamine,其中KnockOut血清替代物浓度为5%-15%,CHIR99021浓度为465-1395ng/mL,SB431542浓度为384-1536ng/mL,SHH浓度为100-200ng/mL、Purmorphamine浓度为521-1563ng/mL;
所述的III期培养基包括DMEM/F12、N-2、B27、cAMP、Vitamin C、FGF8b,其中cAMP浓度为250-400ng/mL,Vitamin C浓度为17.6-35.2μg/mL,FGF8b浓度为50-200ng/mL;
所述IV期培养基包括DMEM/F12、N-2、B27、cAMP、Vitamin C、BDNF、GDNF、IGF-1、TGF-β3、db-cAMP,其中cAMP浓度为250-400ng/mL,Vitamin C浓度为17.6-35.2μg/mL,BDNF浓度为5-20ng/mL,GDNF浓度为5-20ng/mL,IGF-1浓度为5-20ng/ml,TGF-β3浓度为0.5-2ng/mL,db-cAMP浓度为123-492μg/mL。
2.根据权利要求1所述的诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基,其特征在于:所述诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基将诱导人多能干细胞分化为多巴胺能神经前体细胞,然后再被诱导分化为成熟的多巴胺能神经元细胞。
3.根据权利要求1所述的诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基,其特征在于:所述CHIR99021的浓度为930ng/mL。
4.根据权利要求2所述的诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基,其特征在于:所述多巴胺能神经前体细胞表达FOXA2+、OTX2+、LMXIA+和CORIN+,另外,FOXG1、PTX3和SOX10的表达低于5.6%。
5.根据权利要求2所述的诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基,其特征在于:所述多巴胺能神经元细胞表达MAP2+、Nurr1+、TH+、PTX3+。
6.利用权利要求1-5中任一所述的诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基制备多巴胺能神经前体细胞的方法,其特征在于:包括以下步骤,
步骤一:Day-1,使用单细胞传代方式将人诱导多能干细胞以2x 105/cm2的接种密度接种于Laminin521包被的2D容器中,使用添加Rock抑制剂Y27632的多能干细胞培养基培养过夜;
步骤二:Day0至Day6,弃去细胞培养上清液,使用不添加Rock抑制剂Y27632的I期培养基,维持步骤一中的人诱导多能干细胞的定向分化,每天换液;
步骤三:Day7,弃去细胞培养上清液,使用DPBS洗涤贴壁细胞,弃去DPBS,使用Accutase将步骤二中的分化细胞消化解离成单细胞,使用添加有Rock抑制剂Y27632的I期培养基进行终止,300x g,离心5min,弃上清,获得细胞沉淀,再使用含有Rock抑制剂Y27632的II期分化培养基重悬细胞沉淀制备成单细胞悬液,接种于Laminin521包被的2D容器中进行过夜培养;
步骤四:Day8至Day13,弃去细胞培养上清液,使用不含有Rock抑制剂Y27632的II期分化培养基培养分化细胞,每天换液;
步骤五:Day14,弃去细胞培养上清液,使用DPBS洗涤贴壁细胞,弃去DPBS,使用Accutase将步骤四中的分化细胞消化解离成单细胞,使用添加Rock抑制剂Y27632的II期培养基进行终止,300x g,离心5min,弃上清,获得细胞沉淀,再使用含有Rock抑制剂Y27632的III期分化培养基重悬细胞沉淀制备成单细胞悬液,接种于Laminin和Poly-L-ornithine包被的2D容器中进行过夜培养;
步骤六:Day15至Day20,弃去细胞培养上清液,使用含有Rock抑制剂Y27632的III期分化培养基培养分化细胞,每天换液,进行定向分化培养;
步骤七:Day21,弃去细胞培养上清液,使用DPBS洗涤贴壁细胞,弃去DPBS,使用Accutase将步骤六中的多巴胺能神经前体细胞消化解离成单细胞,使用添加Rock抑制剂Y27632的III期培养基进行终止,300x g,离心5min,弃上清,获得细胞沉淀,得到多巴胺能神经前体细胞。
7.根据权利要求6所述的诱导人多能干细胞分化为多巴胺能神经前体细胞的培养基制备多巴胺能神经前体细胞的方法,其特征在于:在iPSC阶段、NPC阶段及DAP阶段,Y27632的浓度是10μM。
8.一种验证多巴胺能神经前体细胞具有定向分化成多巴胺神经元能力的方法,其特征在于,包括以下步骤:
步骤一:使用含有Y27632的IV期分化培养基,重悬多巴胺能神经前体细胞沉淀,制备成单细胞悬液,接种于Laminin和Poly-L-ornithine包被的2D容器中进行过夜培养24h;
步骤二:弃去细胞培养上清液,使用不含有Rock抑制剂Y27632的IV期分化培养基培养多巴胺能神经前体细胞,每天换液,进行定向分化成熟培养2周,获得多巴胺能神经元细胞;
步骤三:弃去细胞培养上清液,使用DPBS洗涤贴壁细胞,弃去DPBS,将步骤二中的多巴胺能神经元细胞用KCL溶液刺激,用ELISA检测多巴胺的分泌,同时通过qPCR、免疫荧光及流式检测多巴胺能神经元细胞。
9.一种用于治疗帕金森病的细胞药物,其特征在于:所述药物包含权利要求2、4、5中任一所述多巴胺能神经前体细胞。
