Description
Technical Field
The invention belongs to the technical field of feed additives, and relates to a feed additive containing plant polysaccharide, and a preparation method and application thereof.
Background
The cultivation mode is divided into traditional cultivation and intensive cultivation, and the intensive cultivation is a main form of cultivation scale due to site limitation and more convenient management. The intensive cultivation can improve the production efficiency of poultry. Through scientific feeding management and environmental control, the growth and the weight growth of the poultry can be optimized, the feed conversion rate is improved, and the waste of resources is reduced, so that the economic benefit of cultivation is improved. However, the intensive animal cultivation is limited in activity, a large amount of nutrient components are contained in the feed, and the excessive nutrient components are converted into fat which is accumulated in the animal body, so that the health is influenced, and the taste is seriously influenced.
Polysaccharides are a class of natural macromolecular polymers linked by glycosidic linkages between aldoses or ketoses. Polysaccharide is widely available in nature, and especially in higher plants, animals, microorganisms and seaweed. Polysaccharides play an extremely important role in cell recognition, control of cell division and differentiation, and the like. The active polysaccharide can directly activate immune cells, enhance phagocytosis and killing effect of animal immune cells, activate complement system, promote antibody production, and synthesize and secrete cytokines.

Further, the preparation method of the compound plant polysaccharide comprises the following steps:
a: removing shells and cores of longan, sun drying, grinding into longan powder, soaking in ethanol solution, and centrifuging to obtain mixed solution a;
b: adding distilled water into the mixed solution a, heating and stirring, standing and filtering to obtain a mixed solution b;
C: removing the core of the red date, adding absolute ethyl alcohol for soaking, and then drying to obtain a red date stock;
d: crushing the red date stock into red date powder, adding a solvent, mixing to form a mixture, carrying out ultrasonic treatment under pressure, and filtering to obtain a mixed solution c;
e: mixing the mixed solution b and the mixed solution c, and then heating, evaporating and concentrating to obtain the compound plant polysaccharide.
Further, in the step A, the mass fraction of the ethanol solution is 80%, the solid-to-liquid ratio of the longan powder to the ethanol solution is 1:5g/mL, the soaking time is 16-24h, and the rotating speed and the time of centrifugal separation are 4000r/min and 5-10min respectively.
Further, in the step B, the volume ratio of the mixed solution a to the distilled water is 1:15-20, the heating means heating to 80 ℃, and the standing time is 2-3h.
Further, in the step C, the solid-to-liquid ratio of the red dates to the absolute ethyl alcohol is 1:10g/mL, the soaking time of the absolute ethyl alcohol is 30-60min, and the drying temperature is 40-50 ℃.
Further, in the step D, the solid-to-liquid ratio of the red date powder to the solvent is 1:5g/mL, the solvent consists of choline chloride and ethylene glycol with the mass ratio of 1:1, the pressurizing pressure is 20-30Mpa, and the ultrasonic time is 20-30min.
Further, in the step E, the volume ratio of the mixed solution b to the mixed solution c is 1:1, and the temperature is raised to 50-60 ℃.
Further, the preparation method of the feed additive containing plant polysaccharide comprises the following steps: all the raw material components are mixed to obtain the feed additive containing plant polysaccharide.
The application of the plant polysaccharide-containing feed additive is that the plant polysaccharide-containing feed additive is applied to fish feed, poultry feed and livestock feed.
The invention has the beneficial effects that:
1. The plant polysaccharide prepared by compounding the longan and the red dates enhances the activity of probiotics, and in addition, the free glycerol is released by inducing lipolysis of fat cells, so that the accumulation of lipid is reduced, and meanwhile, the lipase activity is inhibited, so that the intestinal absorption of the glycerol is obviously reduced. The feed additive containing plant polysaccharide prepared by the invention can effectively reduce the grease content in animals.
2. The soybean isoflavone and the polysaccharide are added for compound use, the expression of related proteins and genes can be influenced through various signal paths, and the synthesis of fatty acid organisms, the synthesis of steroid hormone organisms, the synthesis of unsaturated fatty acid organisms, the elongation of fatty acid, the metabolism of glyceride and the metabolism of glycerophospholipids are influenced, so that the possibility that nutrition is converted into fat is further prevented.
3. Kelp powder and spirulina are rich in iodine, vitamins, amino acids and other substances, promote the synthesis of animal thyroid hormone, make animals more active, reduce fat accumulation through exercise and have better taste to meat quality.

1. Performance testing
The feed additives containing plant polysaccharide prepared in examples 1 to 3 and comparative examples 1 to 5 were added to a poultry feed as a sample in an amount of 10%, the poultry feed consisting of the following raw materials in parts by weight: 2.0 parts of corn, 1.3 parts of sorghum, 1.7 parts of barley, she Kang 2.0.0 parts of bean cake and bean pulp, 1 part of sweet potato powder and 1.5 parts of wheat bran. Selecting 800 healthy chickens which are born for 30 days, dividing the chickens into 8 groups, feeding the chickens in a containment mode and using a sample for feeding, wherein the specific feeding mode is as follows: once in the morning, in the middle and in the evening. In addition, 100 chickens were selected as a control group using feed without additives.
Fat testing: the fat content of the chicken is measured by scanning the abdomen of the chicken with an ultrasonic instrument, and the fat increment is calculated by measuring every 1 month and testing for 3 months respectively. The results of the above tests are shown in Table 1.
Table 1 test results
As can be seen from Table 1, the grease increment per month was smaller in examples 1-3 than in comparative examples 1-5 and the control group, wherein the grease increment per month was the largest in the control group. In comparative example 1, longan is not added for compounding; comparative example 2 was not formulated with red date added; comparative example 3 lacks the addition of soy isoflavones; comparative examples 4 to 5 only one of kelp powder and spirulina powder was added. Thus the absence of either component affects the fat content.
The present invention is not limited to the above embodiments, but is capable of modification and variation in detail, and other modifications and variations can be made by those skilled in the art without departing from the scope of the present invention.
