Abstract

背景技术
人白介素4受体α(人IL-4Rα)是IL-4/IL-13的共同受体,分两种类型:I型受体由IL-4Rα和γc亚基组成,只与IL-4结合。II型受体由IL-4Rα和IL-13Rα1亚基组成,可与IL-4或IL-13结合。IL-4和IL-13共享同一受体信号传导系统,功能部分重叠。IL-4能促进嗜酸性粒细胞和肥大细胞的活化及向炎症位点的趋化;IL-4/IL-13能促进B细胞活化和IgE的分泌,上调B细胞、肥大细胞等表达IgE受体;IL-13能促进粘液分泌、细胞增生、胶原沉积。IL-4R介导的信号转导在2型炎症相关疾病的发生、发展中发挥重要作用。
相比单独抑制IL-4或IL-13,IL-4R单抗通过特异性结合IL-4Rα,可同时抑制IL-4/IL-13通路的信号传导,治疗由Th2介导的2型炎症性疾病,疗效更好。研究表明80-90%哮喘、特异性皮炎、慢性鼻窦炎伴鼻息肉病人患有2型炎症疾病。目前,单抗度普利尤单抗已获批治疗5个适应症,分别为中重度特异性皮炎,中重度哮喘,慢性鼻窦炎伴鼻息肉,嗜酸性食管炎,结节性痒疹。然而,面对患者对于疾病治疗的需求,亟待开发新型、高效、特异的IL-4R抗体,改善患有2型炎症性疾病人群的生活质量。
发明内容
针对上述诸问题,本公开提供了抗体、其制备方法、组合物等。本公开提供的益处广泛地适用于抗体治疗和诊断领域,并且能够与各种靶标反应的抗体联合使用。本发明提供了能够特异性结合IL-4R抗原的抗体,优选嵌合单克隆抗体或人源化单克隆抗体。
一方面,本发明提供一种分离的抗体或其抗原结合片段,其中所述抗体或其抗原结合片段特异性结合IL-4R,且包含重链可变区(VH)和轻链可变区(VL),
所述重链可变区包含:
(i)HCDR1,其中包含与SEQ ID NO:1具有至少80%、至少85%、至少95%、或100%序列同一性的序列或SEQ ID NO:1组成;

定义
为了更好地理解本发明,相关术语的定义和解释提供如下。
术语“抗体”或“Ab”通常是指包含通过共价二硫键和非共价相互作用保持在一起的两条重链(H)和两条轻链(L)多肽链的Y形四聚体蛋白。抗体的轻链可以分为κ或λ轻链。重链可分为μ、δ、γ、α或ε,它们分别将抗体的同种型定义为IgM、IgD、IgG、IgA或IgE。在轻链和重链中,可变区通过约12个或更多个氨基酸的“J”区与恒定区连接,并且重链还包含约3个或更多个氨基酸的“D”区。每条重链由重链可变区(VH)和重链恒定区(CH)组成。重链恒定区由3个结构域(CH1,CH2和CH3)组成。每条轻链由轻链可变区(VL)和轻链恒定区(CL)组成。VH和VL区可以进一步分为由相对保守的区域(称为框架区,简称FR)间隔开的高变区(称为互补决定区,简称CDR)。每个VH和VL由以下顺序的3个CDR和4个FR组成:从N端到C端的FR1、CDR1、FR2、CDR2、FR3、CDR3和FR4。VH上的CDR为HCDR1,HCDR2,HCDR3;VL上的CDR为LCDR1,LCDR2,LCDR3。每个重链/轻链对的可变区(VH和VL)分别形成抗原结合位点/部分。氨基酸在各种区域或结构域中的分布遵循Kabat、IMGT或Chothia等常见系统中的编号定义,在本公开的具体实施例中,CDR序列的确定使用的是Kabat系统中的编号定义。
本公开中抗体也包括抗原结合部分(与术语“抗原结合片段”可互换使用)。抗原结合部分是指包含完整抗体的片段的多肽,其保留了与全长或者完整抗体特异性结合的抗原特异性结合的能力,和/或其与全长抗体竞争结合相同的抗原。在一些条件下,抗原结合部分包括Fab、Fab'、F(ab')2、Fd、Fv、dAb和互补决定区(CDR)片段,单链抗体(例如scFv),嵌合抗体,双抗体和包含足以赋予多肽特异性抗原结合能力的至少一部分抗体。抗体的抗原结合部分可通过本领域技术人员已知的常规技术(例如,重组DNA技术或酶促或化学切割方法)从给定抗体获得,并且可以与完整抗体相同的方式筛选特异性。
术语“同种型”是指由重链恒定区基因编码的抗体类别(例如,IgM或IgG1)。
术语“单克隆抗体”或“mAb”是指单分子组成的抗体分子/制剂。单克隆抗体显示对特定表位的单一结合特异性和亲和力。本发明的抗体可源自不同的物种,包括但不限于小鼠、大鼠、兔、豚鼠和人。
如本文所用的术语“嵌合抗体”是指其可变区序列来自一个物种并且恒定区序列来自另一物种的抗体,例如其中可变区序列源自小鼠抗体和恒定区序列来源于人抗体的抗体。
术语“人源化抗体”旨在指其中来源于另一种哺乳动物物种如小鼠的种系的CDR序列/抗原结合部分或位点已被移植到人框架序列上的抗体。此外,还可以在人框架序列内进行额外的框架区修饰。
术语“KD值”是抗体与其抗原之间的平衡解离常数,即koff/kon或kd/ka(通过SPR技术测定)比值。因此KD值越低,抗体的亲和力越高。因此“KD值”可以用于衡量抗体与其抗原之间的结合亲和力。
术语“IL-4R”和“IL-4R抗原”在本文中可互换使用,包括由细胞天然表达的或者在用IL-4R基因转染的细胞上表达的人IL-4R的任何变体、同等型和物种同系物。术语“抗IL-4R抗体”或“IL-4R抗体”是指如本文所定义的能够与IL-4R抗原结合或与表达IL-4R的细胞结合的抗体。
术语“特异性结合”是指,两分子间的非随机的结合反应,如抗体和其所针对的抗原之间的反应。
术语“分离的”是指通过人工方式从天然状态获得的状态。如果某种“分离的”物质或组分天然存在,则可能是因为其天然环境发生变化,或者物质与天然环境分离,或者两者兼而有之。例如,某种未分离的多核苷酸或多肽天然存在于某个活动物体内,从该天然状态分离的高纯度的相同多核苷酸或多肽被称为分离的多核苷酸或多肽。术语“分离的”既不排除混合的人造或合成物质,也不排除不影响分离的物质的活性的其他不纯物质。例如,分离的抗体可以基本上不含其他细胞材料和/或化学物质。
术语“载体”是指可以在其中插入多核苷酸的核酸媒介物。当载体允许插入其中的多核苷酸编码的蛋白质的表达时,该载体称为表达载体。该载体可以通过转化、转导或转染入宿主细胞而使携带的遗传物质元件在宿主细胞中表达。载体是本领域技术人员所熟知的,包括但不限于质粒,噬菌体,粘粒,人工染色体如酵母人工染色体(YAC),细菌人工染色体(BAC)或P1衍生人工染色体(PAC);噬菌体如λ噬菌体或M13噬菌体和动物病毒。可用作载体的动物病毒包括但不限于逆转录病毒(包括慢病毒)、腺病毒、腺伴随病毒、疱疹病毒(如单纯疱疹病毒)、痘病毒、杆状病毒、乳头瘤病毒、乳多空病毒(如SV40)。载体可以包含用于控制表达的多个元件,包括但不限于启动子序列、转录起始序列、增强子序列、选择元件和报道基因。另外,载体可以包含复制起点。对于表达抗体的载体,可以使用其中抗体重链和轻链存在于不同的载体中的载体类型或者其中重链和轻链存在于同一载体中的载体类型。
术语“宿主细胞”是指可被工程化以产生目标蛋白质、蛋白质片段或肽的细胞系统。宿主细胞包括但不限于培养细胞,例如来源于啮齿类动物(大鼠、小鼠、豚鼠或仓鼠)的哺乳动物培养细胞,如CHO、BHK、NSO、SP2/0、YB2/0;或人体组织或杂交瘤细胞、酵母细胞和昆虫细胞,以及包含在转基因动物或培养组织内的细胞。该术语不仅涵盖特定的受试细胞,而且还涵盖这种细胞的后代。因为某些修饰可由于突变或环境影响而在后续世代中发生,这样的后代可能与亲本细胞不同,但仍包括在术语“宿主细胞”的范围内。

抗IL-4R抗体
在一些方面,本发明包括分离的抗体或其抗原结合片段。
在本申请的上下文中,“抗体”可以包括多克隆抗体、单克隆抗体、嵌合抗体、人源化和灵长类动物化抗体、CDR移植抗体、人抗体、重组产生的抗体、胞内抗体、双功能抗体、多特异性抗体、双特异性抗体、单价抗体、多价抗体、抗独特型抗体、合成抗体、包括其突变蛋白及变体、改进的抗体;及其衍生物(包括Fc融合蛋白和其他修饰),以及任何其他免疫反应性分子,只要其表现出与IL-4R蛋白的优先缔合或结合。此外,除非上下文另外规定,否则该术语还包括所有类别的抗体(即IgA、IgD、IgE、IgG和IgM)和所有亚类(即IgG1、IgG2、IgG3、IgG4、IgA1和IgA2)。在一个优选的实施方案中,抗体是单克隆抗体。在更优选的实施方案中,抗体是嵌合单克隆抗体或人源化单克隆抗体或改进的嵌合单克隆抗体。
无论抗体如何产生,测试抗体与抗原(例如IL-4R)结合的能力的方法在本领域中是已知的,并且包括任何抗体-抗原结合测定,例如放射免疫测定(RIA)、ELISA、蛋白质印迹、免疫沉淀、SPR和竞争性抑制测定(参见例如Janeway等人、下文和美国专利申请公开号2002/0197266以及有关竞争测定的以上章节)。

在一些实施方案中,所述的分离的抗体或其抗原结合片段包含IgG的恒定区。所述IgG的恒定区优选选自IgG1、IgG2、IgG3、或IgG4恒定区。所述IgG的恒定区更优选选自IgG1的恒定区。
在本领域中,采用各种手段进行不改变抗体所期望的性质改造是多样化的,例如本公开中采用的将抗体的轻链和重链重新组合的方式、进行氨基酸的替换等。例如,可以将本发明中的序列,包括嵌合抗体序列或人源化抗体序列,进行保守氨基酸替换。
嵌合抗体是指不同部分来自不同动物物种的抗体,例如,具有来自鼠抗体的可变区和人免疫球蛋白恒定区的抗体。将鼠抗体重链和轻链的可变区与人重链和轻链的恒定区连接在一起从而获得抗体的嵌合(例如,如Kraus等,in Methods in Molecular Biologyseries,Recombinant antibodies for cancer therapy ISBN-0-89603-918-8所述)。在一个优选实施方案中,通过连接人κ轻链恒定区至鼠轻链可变区产生嵌合抗体。在另一优选实施方案中,可通过连接人λ轻链恒定区至鼠轻链可变区产生嵌合抗体。
而人源化抗体是指将其中来源于另一种哺乳动物物种如鼠的种系的CDR序列/抗原结合部分或位点移植到人框架序列上的抗体。
为了降低抗体对人的免疫原性,使用本公开的IL-4R抗体的序列生产人源化抗IL-4R抗体,利用兔源的抗IL-4R抗体的CDR区与人源的框架区(例如,人免疫球蛋白)组合,形成本公开的人源化抗IL-4R抗体,人源化抗体被期望保留与人IL-4R结合的功能以及与猴IL-4R结合的功能。
抗体的制备或产生

本发明的抗体还可以在宿主细胞转染瘤中使用例如本领域众所周知的重组DNA技术和基因转染方法的组合(例如Morrison,S.(1985)Science 229:1202)来产生。在一些实施方案中,将通过标准分子生物学技术获得的编码部分或全长轻链和重链的DNA插入一个或多个表达载体中,使得所述基因可操作地连接于转录和翻译调节序列。在此上下文下,术语“可操作地连接”意在表示将抗体基因连接到载体中,使得载体内的转录和翻译控制序列发挥它们调节抗体基因转录和翻译的预期功能。
可以将抗体轻链基因和抗体重链基因插入相同或不同的表达载体中。在一些实施方案中,可变区用于通过将其插入到已经编码所需同种型的重链恒定区和轻链恒定区的表达载体中来产生任何抗体同种型的全长抗体基因,使得VH区段可操作地连接至载体内的CH区段和VL区段可操作地连接至载体内的CL区段。另外或可选地,重组表达载体可以编码促进抗体链从宿主细胞分泌的信号肽。可以将抗体链基因克隆到载体中,使得信号肽连接到抗体链基因的氨基末端。信号肽可以是免疫球蛋白信号肽或异源信号肽(即来自非免疫球蛋白蛋白质的信号肽)。
为了表达轻链和重链,通过标准技术将编码重链和轻链的表达载体转染到宿主细胞中。术语“转染”的各种形式旨在涵盖通常用于将外源DNA引入原核或真核宿主细胞中的各种技术,例如电穿孔,磷酸钙沉淀,DEAE-葡聚糖转染等。可以在原核或真核宿主细胞例如哺乳动物宿主细胞(其可以装配和分泌适当折叠和免疫活性的抗体)中表达本发明的抗体。
用于表达本发明重组抗体的哺乳动物宿主细胞包括与DHFR选择标记(例如,如R.J.Kaufman and P.A.Sharp(1982)J.MoI.Biol.159:601-621中所述的)一起使用的中国仓鼠卵巢细胞(CHO细胞)(包括在Urlaub和Chasin,(1980)Proc.Natl.Acad.Sci.USA 77:4216-4220中描述的dhfr CHO细胞),NSO骨髓瘤细胞,COS细胞和SP2细胞。特别地,为了与NSO骨髓瘤一起使用,另一种表达系统是WO 87/04462、WO 89/01036和EP 338,841中公开的GS基因表达系统。当将编码抗体基因的重组表达载体导入哺乳动物宿主细胞时,通过培养宿主细胞足以允许抗体在宿主细胞中表达的时间段或将抗体分泌到宿主细胞生长的培养基中来产生抗体。使用标准蛋白质纯化方法可从培养基中回收抗体。
用于生成单克隆抗体的另一个策略是从所定义策略的产生抗体的淋巴细胞中直接分离编码抗体的基因,例如参见Babcock et al.,1996;A novel strategy forgenerating monoclonal antibodies from single,isolated lymphocytes producingantibodies of defined strategy。对于重组抗体工程的细节还参见Welschof and Krau,Recombinant antibodies for cancer therapy ISBN-0-89603-918-8and Benny K.C.LoAntibody Engineering ISBN 1-58829-092-1。

缀合物
一方面,本公开提供了一种缀合物,其包含与至少一个可检测的标记物偶联的如前所述的抗体或其片段。可检测的标记物包括但不限于:(i)提供可检测信号;(ii)与第二标记物相互作用以修饰所述第一或第二标记物所提供的可检测信号,例如FRET(荧光共振能力转移,Fluorescence Resonance Energy Transfer);(iii)通过电荷、疏水性、形状或其他物理参数影响迁移率(例如,电泳迁移率),或者(iv)提供捕获部分,例如亲和力、抗体/抗原或离子络合。
适合作为标记物的结构例如荧光标记物、发光标记物、发色团标记物、放射性同位素标记物、同位素标记物,优选稳定的同位素标记物、同量异位素标记物(isobariclabel)、酶标记物(例如辣根过氧化物酶、碱性磷酸酶、β-半乳糖苷酶、脲酶、葡萄糖氧化酶等)、颗粒标记物(尤其是金属颗粒标记物、磁性颗粒标记物、聚合物颗粒标记物)、有机小分子(例如,生物素、受体的配体或结合分子(例如,细胞黏附蛋白或卵磷脂)、可通过使用结合剂检测包含核酸和/或氨基酸残基的标记物序列等。标记物非限制性地包括硫酸钡、碘西他酸、碘番酸、胺碘苯丙酸钙、泛影酸钠、泛影葡胺、甲泛葡胺、酪泮酸钠和放射诊断剂(包括正电子发射体,(例如氟-18和碳-11)、γ发射体(例如,碘-123、碘125、锝-99m、碘-131和铟-111)、核磁共振核素(例如,氟和钆))、发光物质(例如异鲁米诺和吖啶酯)、荧光物质(例如荧光素和罗丹明)、有色物质(例如乳胶颗粒和胶体金)。

抗体药物缀合物/免疫缀合物

多特异性分子
本发明的抗体或其抗原结合片段可用于形成多特异性分子(例如双特异性分子)。本发明的抗体或其抗原结合片段可以是多特异性分子(例如双特异性分子)的一部分,所述双特异性或多特异性分子包含具有不同于本发明的抗体或其抗原结合片段的结合特异性的第二功能模块(例如第二抗体)或第三功能模块(例如第三抗体),从而能够结合至少两个不同的结合位点和/或靶分子。例如,本发明的抗体或其抗原结合片段可连接至能够特异性结合可用作联合治疗的潜在靶标的任何蛋白质的第二抗体或其抗原结合片段。为产生所述双特异性或多特异性分子,可以将本发明的抗体或其抗原结合片段连接(例如通过化学偶联、基因融合、非共价缔合或其他方式)至一个或多个其他结合分子(例如另外的抗体、抗体片段、肽或结合模拟物)。

在某些优选的实施方案中,所述多特异性分子特异性结合IL-4R(例如人IL-4R或猴IL-4R),并且特异性结合一个或多个其他靶标。

药物组合物

医药用途
本发明的抗体、抗体组合物和方法具有许多体外和体内用途,包括例如IL-4R的检测或免疫应答的增强。例如,可以将这些分子体外或离体施用于培养细胞,或例如体内施用于人受试者。
优选的受试者包括哺乳动物,例如人/患者。本发明上下文中的哺乳动物是人类、非人类灵长类动物、驯养动物如狗、猫、绵羊、牛、山羊、猪、马等,实验室动物如小鼠、大鼠、兔、几内亚猪等以及圈养的动物,如动物园的动物。
诊断
本发明提供了用于检测、诊断或监测增殖性病症的体外和体内方法以及筛选来自患者的细胞以鉴定肿瘤细胞包括致瘤细胞的方法。这样的方法包括鉴定用于治疗的患有癌症的个体或监测癌症的进展,包括将患者或从患者获得的样品(体内或体外)与本文所述的抗体接触,并检测样品中与结合的或游离的靶分子的结合的抗体的存在或不存在或结合水平。在一些实施方案中,抗体将包含如本文所述的可检测标记或已报道分子。

通过参考以下实施例将更容易地理解本文一般地描述的本发明,这些实施例是以举例说明的方式提供的,并且不旨在限制本发明。另外,下述实施例中的实验方法如无特殊说明,均为常规方法。下述实施例中所用的原料、试剂材料等,如无特殊说明,均为市售购买产品。
实施例1:人IL-4R抗体的免疫及筛选
选择Balb/c小鼠(购自北京维通利华实验动物技术有限公司,雌性,8-10周)或SJL小鼠(购自上海斯莱克实验动物有限责任公司,雌性,7-9周)用人IL 4R(Human IL4RProtein,His Tag,购自Acro Inc,货号:ILR-H5221)进行蛋白免疫。首次免疫使用完全弗氏佐剂按1:1比例与抗原蛋白(0.1mg)进行混合乳化,皮下多点注射。随后每隔2周用TiterMax(购自Sigma Inc,货号T2684)与抗原蛋白(0.05mg)按1:1比例进行乳化,皮下多点注射,共免疫4次。为了获得最佳免疫效果,分别抽取0,1,2,3,4次免疫7天后血液分离抗血清,以评估抗抗原反应的免疫效果。
当小鼠达到最佳免疫反应时,安乐死小鼠并分离脾脏,收获表达抗体的B细胞。通过流式分选(购自BD Inc,型号,FACS ARIA III)技术,分选出单个B细胞,调取抗体重、轻链基因并克隆至表达载体,瞬转293细胞获得抗体表达上清,通过检测抗原结合活性以及阻断人IL-4和人IL-4R结合的活性,选择了具有所需功能的单克隆抗体。
通过前述方法获得的1种人IL-4R抗体,被命名为65G5。65G5抗体以及它的生物特性将在以下实施例中更为详细地叙述。
实施例2:人IL-4R轻重链表达质粒的构建及抗体制备
将65G5(分别含轻、重链可变区PCR产物)分别克隆至含人IgG1重链恒定区的载体和含人Kappa轻链恒定区的载体上进行测序,然后制备质粒后转染CHO-S细胞,培养7~10天后收取培养上清,通过Protein A亲和层析纯化获得抗体,并通过超滤置换至1xPBS。
实施例3:65G5与人IL-4R过表达细胞的结合活性
通过FACS检测本公开中的抗人IL-4R抗体与表达于HEK293-IL-4R细胞表面的人IL-4R的结合活性。简而言之,从细胞培养瓶中收获HEK293-IL-4R细胞,洗涤两次,并重悬于FACS缓冲液中(含4%FBS的PBS缓冲液)中,然后,在含有2X105个细胞/孔的96孔板中加入100ul/孔于FACS缓冲液中梯度稀释(以10ug/ml为起始浓度,3倍梯度稀释)的抗IL-4R抗体或对照品,冰浴30min。FACS缓冲液洗涤两次后,加入100ul/孔FITC Anti-human IgG(Merk,货号:F9512,在FACS缓冲液中以1:400的比例稀释),于4℃避光孵育30min后,将细胞洗涤3次,重悬于FACS缓冲液中,使用BD FACS Lyric检测其MFI值。分析数据计算得到EC50值。结果显示(图1),65G5的EC50值为0.334μg/ml。
实施例4:65G5阻断IL4/IL-4R过表达细胞的结合
通过流式细胞术(FACS)检测本公开中的抗人IL-4R抗体阻断人IL-4蛋白与细胞表面IL-4R的结合活性。简而言之,从细胞培养瓶中收获HEK293-IL-4R细胞(内部构建),洗涤两次,并重悬于FACS缓冲液中(含4%FBS的PBS缓冲液)中,然后,在含有2X105个细胞/孔的96孔板中加入50ul/孔于FACS缓冲液中梯度稀释(以20ug/ml为起始浓度,3倍梯度稀释)的抗IL-4R抗体或对照品,然后加入50ul/孔的生物素标记的人IL-4(Acro,货号:IL4-H82E0),于4℃避光孵育30min。FACS缓冲液洗涤两次后,加入100ul/孔PE Streptavidin(BD,货号405204,在FACS缓冲液中以1:400的比例稀释),于4℃避光孵育30min后,将细胞洗涤3次,重悬于FACS缓冲液中,使用BD FACS Lyric检测其MFI值。通过数据分析计算得到IC50值。结果显示(图2),65G5的IC50值为0.075μg/ml。
实施例5:65G5与IL-4R蛋白结合亲和力测定
通过Octet RH96测定抗体与IL-4R蛋白结合亲和力。使用AHC2探针(sartorius,18-5142)捕获候选抗体;与多浓度IL-4R蛋白(Acrobiosystems,IL-H5221)结合300s,解离600s,使用Octet Analysis Studio软件选择1:1模型拟合数据,计算抗原结合亲和力。结果显示(图3),65G5的KD值为3.41E-10M。
实施例6:抗体抑制IL-4诱导的HEK293-hSTAT6/5×STAT6-Luc细胞中STAT6磷酸化活性
将对数期的HEK293-hSTAT6/5×STAT6-Luc(购自中检院)细胞重悬于培养基(DMEM+10%FBS)中,以60μL/孔接种到96孔板中,每孔含4×104个细胞,置于37℃,5%CO2培养箱过夜。然后,每孔加入30μL梯度稀释的抗IL-4Rα抗体或对照品(内部制备的dupilumab)(以400nM为起始浓度,5倍梯度稀释)。每孔加入30μL IL4蛋白(40ng/mL,Sino biological,目录号:11846-HNAE),置于37℃,5%二氧化碳培养箱孵育6小时。加入ONE-GLO检测试剂(Promega,目录号:E6120),室温避光静置3~5分钟,使用SpectraMax i3x读取发光值。通过数据分析计算得到IC50值。结果显示(图4),65G5的IC50值为92.67nM。
实施例7:抗体抑制IL-13诱导的HEK293-hSTAT6/5×STAT6-Luc细胞中STAT6磷酸化活性
将对数期的HEK293-hSTAT6/5×STAT6-Luc细胞重悬于培养基(DMEM+10%FBS)中,以60μL/孔接种到96孔板中,每孔含4×104个细胞,置于37℃,5%CO2培养箱过夜。然后,每孔加入30μL梯度稀释的抗IL-4Rα抗体或对照品(内部制备的dupilumab)(以400nM为起始浓度,5倍梯度稀释)。每孔加入30μL IL-13蛋白(10ng/mL,Sino biological),置于37℃,5%二氧化碳培养箱孵育6小时。加入ONE-GLO检测试剂(Promega,目录号:E6120),室温避光静置3~5分钟,使用SpectraMax i3x读取发光值。通过数据分析计算得到IC50值。结果显示(图5),65G5的IC50值为5.596nM。

1.一种分离的抗体或其抗原结合片段,其中所述抗体或其抗原结合片段特异性结合IL-4R,且包含重链可变区(VH)和轻链可变区(VL),
所述重链可变区包含:
(i)HCDR1,其中包含与SEQ ID NO:1具有至少80%、至少85%、至少95%、或100%序列同一性的序列或SEQ ID NO:1组成;
(ii)HCDR2,其中包含与SEQ ID NO:2具有至少80%、至少85%、至少95%、或100%序列同一性的序列或由SEQ ID NO:2组成;和
(iii)HCDR3,其中包含与SEQ ID NO:3具有至少80%、至少85%、至少95%、或100%序列同一性的序列或由SEQ ID NO:3组成,
所述轻链可变区包含:
(i)LCDR1,其中包含与SEQ ID NO:4具有至少80%、至少85%、至少95%、或100%序列同一性的序列或由SEQ ID NO:4组成;
(ii)LCDR2,其中包含与SEQ ID NO:5具有至少80%、至少85%、至少95%、或100%序列同一性的序列或由SEQ ID NO:5组成;和
(iii)LCDR3,其中包含与SEQ ID NO:6具有至少80%、至少85%、至少95%、或100%序列同一性的序列或由SEQ ID NO:6组成。
2.如上述权利要求1所述的抗体或片段,其中所述轻链可变区包含与SEQ ID NO:8具有至少80%、至少85%、至少90%、至少91%、至少92%、至少93%、至少94%、至少95%、至少96%、至少97%、至少98%、至少99%或100%序列同一性的序列或者由SEQ ID NO:8组成。
3.如上述权利要求中任一项所述的抗体或片段,其中所述重链可变区包含与SEQ IDNO:7具有至少80%、至少85%、至少90%、至少91%、至少92%、至少93%、至少94%、至少95%、至少96%、至少97%、至少98%、至少99%或100%序列同一性的序列或者由SEQ IDNO:7组成。
4.如上述权利要求中任一项所述的抗体或片段,其中还包含重链恒定区和轻链恒定区,该重链恒定区包含与SEQ ID NO:9具有至少80%、至少85%、至少90%、至少91%、至少92%、至少93%、至少94%、至少95%、至少96%、至少97%、至少98%或至少99%或100%序列同一性的序列或者由SEQ ID NO:9组成;
该轻链恒定区包含与SEQ ID NO:10具有至少80%、至少85%、至少90%、至少91%、至少92%、至少93%、至少94%、至少95%、至少96%、至少97%、至少98%或至少99%或100%序列同一性的序列或者由SEQ ID NO:10组成。
5.如上述权利要求中任一项所述的抗体或片段,其中包含:
与SEQ ID NO:12具有至少95%、至少96%、至少97%、至少98%、至少99%或100%序列同一性的轻链或者由SEQ ID NO:12组成的轻链;以及与SEQ ID NO:11具有至少95%、至少96%、至少97%、至少98%、至少99%或100%序列同一性的重链或者由11组成的重链。
6.如上述权利要求中任一项所述的抗体或片段,其中所述抗体为单克隆抗体。
7.如上述权利要求中任一项所述的抗体或片段,其中所述抗体为嵌合抗体或人源化抗体。
8.一种分离的核酸分子,其中包含编码如权利要求1-7的任一项中所述的抗体或片段的核酸序列。
9.一种载体,其中包含如权利要求8中的核酸分子。
10.一种多特异性分子,其中包含如权利要求1-7的任一项中所述的抗体或抗原结合片段,所述多特异性分子还包含至少一种具有针对第二靶标的结合特异性的分子。
