Furthermore, the invention also creatively adds modified hydrolyzed collagen into the shampoo formula. The modified hydrolyzed collagen is the product of the reaction of 4-dodecyloxybenzoyl chloride and hydrolyzed collagen. The collagen is a peptide chain composed of amino acids, and then hydroxyproline is interacted to form a hydrogen bond, so that three peptide chains are wound to form a spiral structure. Amino acids in collagen include, but are not limited to, glycine, proline, hydroxyproline and glutamic acid, all of which contain amino groups in the structure, have activity of reacting with acyl chloride, and have mild reaction, and the formed amide bond has stable structure. After collagen is hydrolyzed, active groups are exposed, and the reaction capacity is stronger. In view of this, the present invention employs 4-dodecyloxybenzoyl chloride to react with hydrolyzed collagen to prepare modified hydrolyzed collagen. The structure of the prepared modified hydrolyzed collagen comprises three parts, namely a collagen part, a long-chain alkyl part and a benzamide part between the two parts.
The preparation process of the modified hydrolyzed collagen comprises the following steps: dissolving hydrolyzed collagen in water, adding 4-dodecyloxybenzoyl chloride, continuously using sodium hydroxide solution to adjust the system to be alkaline, stirring at constant temperature for reacting for a certain time, precipitating the product with anhydrous diethyl ether, filtering, washing with ethanol, and drying in vacuum to obtain the modified hydrolyzed collagen.
Preferably, the hydrolyzed collagen has a molecular weight of 1000 to 2000 daltons. Further preferably, the raw materials are 20-30 parts of hydrolyzed collagen, 3-6 parts of 4-dodecyloxybenzoyl chloride and the balance of water according to 100 parts by weight.
In the preparation process of the modified hydrolyzed collagen, because acyl chloride reacts with amino groups to generate HCl, alkali liquor is needed to be used for neutralization, and the alkaline environment of the system is maintained so as to promote the forward reaction. Preferably, the concentration of the sodium hydroxide solution is 5-10wt%, and the pH value of the system is adjusted between 8-9.
Preferably, the temperature of the reaction is 10-30 ℃ and the time is 2-3h.
The modified hydrolyzed collagen of the invention has the following functions:
First, it is known that the surface of the hair flake of healthy hair is hydrophobic, and the surface of the damaged hair flake becomes hydrophilic, so that when the modified hydrolyzed collagen comes into contact with the damaged hair, the protein portion thereof is liable to come into contact with the hair flake due to the hydrophilic group (derived from amino acid), and the corresponding long chain alkyl portion extends to the outside. On one hand, the protein part can repair damaged hair scales, smooth the hair scales and supplement nutrients for hair; on the other hand, the long chain alkyl moiety forms a hydrophobic layer on the outermost layer of the hair, preventing loss of hair moisture and nutrients. Under the combined action of the two aspects, the hair restoration agent can play a good role in restoring damaged hair, so that the glossiness and the flexibility of the hair are increased, and the phenomena of hair dryness, hair bifurcation, hair breakage and the like can be obviously improved after long-term use.
Secondly, it is known that adding amino acids to the shampoo helps to maintain the balance of the grease, and also supplements the nutrition of the hair to make the hair healthier. The amino acid and the modified hydrolyzed collagen are added simultaneously, and the amino acid and the modified hydrolyzed collagen can play a synergistic promotion role in repairing hair, because: compared with the modified hydrolyzed collagen, the modified hydrolyzed collagen has larger molecular weight, and the existence of the long chain of the hydrophobic alkyl plays a certain role in preventing the modified hydrolyzed collagen from entering the hair to repair, and the molecular weight of the amino acid is obviously smaller and is easier to permeate into the hair, so that the amino acid can assist in repairing the parts which cannot be reached by the modified hydrolyzed collagen and provide deeper nutrition for the hair; the alkyl long-chain hydrophobic layer formed by the modified hydrolyzed collagen can prevent the migration and loss of small molecular amino acid, and promote the amino acid to play a durable role in balancing grease and repairing.
Thirdly, the modified hydrolyzed collagen is obtained by reacting alkoxy benzoyl chloride with amino, and a benzamide structure generated by the reaction is positioned between a protein part and a long-chain alkyl part, and benzoyl is known to have good ultraviolet absorption capacity, so that the use of the modified hydrolyzed collagen can prevent ultraviolet rays from damaging hair and reduce phenomena of dryness, roughness, breakage and the like of the hair.
Fourthly, salicylic acid is adopted to endow shampoo with good anti-dandruff capability, however, the salicylic acid is known to be used on the skin to avoid sunburn, because the salicylic acid can cause the skin to be stimulated by ultraviolet rays to generate adverse reaction, the shampoo can contact the skin in the use process, and a small amount of salicylic acid can still remain on the scalp after washing.
In addition, the invention also provides a preparation method of the collagen shampoo with the sun-proof, anti-dandruff and repairing functions, which comprises the following preparation steps:
(1) Adding salicylic acid, hexamidine bis (isethionate), sodium C14-16 alkenyl sulfonate, sodium laurylsulfate, cocamidopropyl PG-dimethyl ammonium chloride phosphate, cocamidopropyl MEA, sodium chloride, sodium lauroyl sarcosinate, lauryl glucoside, cocamidopropyl betaine, hydroxypropyl guar hydroxypropyl trimethyl ammonium chloride, acrylamide propyl trimethyl ammonium chloride/acrylamide copolymer, sodium lactate, citric acid, potassium citrate, p-hydroxyacetophenone and 2/3 water into a container, heating to 55-65 ℃, and uniformly stirring;
(2) Adding butanediol, allantoin, glycerol and 1, 2-hexanediol into a container, stirring uniformly, and cooling to 35-40deg.C;
(3) Mixing amino acid, modified hydrolyzed collagen, plant extract, and 1/3 water, adding into container, and stirring;
(4) Adding PEG-7 glycerol cocoate and PEG-120 methyl glucose dioleate into a container, stirring uniformly, cooling to room temperature, adding essence, phenoxyethanol, sodium benzoate and dipotassium glycyrrhizinate, stirring uniformly, and discharging.
From the above, the technical scheme of the invention has the following beneficial effects: the invention prepares the modified hydrolyzed collagen by the reaction of 4-dodecyloxybenzoyl chloride and the amino group of the hydrolyzed collagen, and adds the modified hydrolyzed collagen into the shampoo formula, and simultaneously adds the components such as amino acid, salicylic acid and the like, thereby not only playing an obvious role in repairing damaged hair, improving the glossiness and flexibility of the hair, improving the phenomena such as dryness, bifurcation and breakage of the hair, but also preventing the damage of ultraviolet rays to the hair, and simultaneously improving the use safety while endowing the shampoo with good anti-dandruff capability, namely the multifunctional shampoo with sun-screening, anti-dandruff and repairing functions can be prepared by the method.
Drawings
FIG. 1 is a schematic representation of the reaction of hydrolyzed collagen with 4-dodecyloxybenzoyl chloride.
Detailed Description
The present invention will be described in further detail by the following detailed description, but it should not be construed that the scope of the invention is limited to the following examples. Various substitutions and alterations are also within the scope of this disclosure, as will be apparent to those of ordinary skill in the art and by routine experimentation, without departing from the spirit and scope of the invention as defined by the foregoing description.
Example 1
First, a modified hydrolyzed collagen was prepared by the following method: dissolving hydrolyzed collagen in water, adding 4-dodecyloxybenzoyl chloride, continuously using 10wt% sodium hydroxide solution to adjust the pH value of the system to be maintained between 8 and 9, stirring at a constant temperature of 20 ℃ for 2.5 hours, precipitating the product with anhydrous diethyl ether, filtering, washing with ethanol, and drying in vacuum to obtain the modified hydrolyzed collagen.
Wherein, the weight portions of the hydrolyzed collagen are 25 portions, the 4-dodecyloxybenzoyl chloride is 5 portions and the water is 70 portions. Hydrolyzed collagen is available from Jiangsu Yihao added Biotechnology Inc. and has been tested to have a molecular weight of 1250 daltons.
The isoelectric points of the unmodified hydrolyzed collagen and the hydrolyzed collagen after modification are tested by adopting a potentiometric-conductivity titration method, the testing instrument is a full-automatic titration instrument, the sample concentration is 1g/L, and the testing condition is room temperature. The isoelectric point of the unmodified hydrolyzed collagen is about 6.5, and the isoelectric point of the modified hydrolyzed collagen prepared by the method is about 4.5. The decrease in isoelectric point indicates that the amino groups react during the modification process, so that the amino groups of the modified collagen, which are positively charged at the isoelectric point, decrease, requiring more positive charge to reach the electrically neutral state.

Test 1: the method comprises the steps of selecting uniform in-vitro damaged hair bundles as a sample, measuring a brightness value L1 by using a spectrocolorimeter, and then equally dividing into 5 parts, and cleaning by using shampoo in examples 1-4 and shampoo in comparative example 1 respectively, wherein the cleaning method comprises the following steps: wetting hair bundles with tap water at 37deg.C, uniformly applying shampoo on hair bundles, massaging for 2min, standing for 5min, and uniformly washing with tap water at 37deg.C for 1min; after washing, the mixture was dried, and the luminance value L2 was measured, and the degree of increase DeltaL= (L2-L1) ×100%/L1 was calculated. As a result of the test, the ΔL of examples 1 to 4 and comparative example 1 was 34.2%, 31.3%, 26.1%, 18.5% and 6.2%, respectively.
Test 2: selecting uniform in-vitro damaged hair bundles as a sample, measuring combing work W1 by using a hair combing property tester, and then equally dividing into 5 parts, and cleaning by using the shampoos of examples 1-4 and comparative example 1 respectively, wherein the cleaning method is the same as that of test 1; after washing, air-dried, carding work W2 was measured, and the degree of decrease DeltaW= (W1-W2) ×100%/W1 of carding work was calculated. As a result of the test, deltaW was 61.4%, 54.2%, 43.9%, 29.1% and 11.7% in examples 1 to 4 and comparative example 1, respectively.
It can be seen from test 1 and test 2 that the addition of the modified hydrolyzed collagen significantly improves the glossiness (brightness improvement) and the softness (combing work reduction) of the damaged hair, and that this effect is more remarkable with the increase of the addition amount of the modified hydrolyzed collagen. This is because the modified hydrolyzed collagen plays a role in repairing damaged hair scales and smoothing the hair scales.
Test 3: selecting 210 healthy female volunteers 18-30 years old, dividing into 7 groups, wherein each group of 30 people has the hair length of 10-20cm, scalp follicular orifice has skin-lifting symptoms and other symptoms, hair has different damaged states, and all volunteers do not participate in other tests or do hair dyeing and perming in the period of 6 months; the shampoo of examples 1-4 and comparative examples 1-3 was applied to 7 groups of volunteers at a frequency of once every 2 days, using the method: after the hair is wetted, the shampoo is uniformly smeared on the hair, the hair is massaged for 5min, and the hair is rinsed for 2min, and then the hair is dried. The cap is worn in outdoor activities during the use period, and a questionnaire is conducted after 28 days of use, so that subjective evaluation of hair dryness, bifurcation and breakage phenomena of a user is obtained. The results of the questionnaire are shown in table 1.
Table 1:
Test 3 shows that the addition of the modified hydrolyzed collagen can obviously repair hair quality, improve phenomena of hair dryness, bifurcation, breakage and the like, and the effect is more obvious along with the increase of the addition amount of the modified hydrolyzed collagen; the addition of the amino acid can also supplement hair nutrition, so that hair quality is repaired, but the action effect of the amino acid is inferior to that of the modified hydrolyzed collagen when the amino acid is added singly; the modified hydrolyzed collagen and amino acid are added simultaneously, so that the synergistic effect on repairing the hair can be achieved.
Test 4: selecting 60 healthy female volunteers 18-30 years old, dividing into 2 groups of 30 people each, requiring hair length of 10-20cm, with obvious dandruff, and not taking part in other tests or hair dyeing or hair waving for about 6 months; the shampoo of example 1 and comparative example 4 was applied to the group 2 volunteers once every 2 days, using the same method as in test 3. A questionnaire was conducted 28 days after use to obtain a subjective assessment of the user's severe condition of dandruff. The results of the questionnaire are shown in table 2.
Table 2:
Test 4 shows that the addition of salicylic acid and hexamidine bis (isethionic acid) salt to the shampoo can significantly improve the anti-dandruff effect.
Test 5: 4 groups of volunteers were selected, 30 persons each, with the same requirement as test 3; the shampoo of example 1 and the shampoo of comparative example 5 were used by group 2 volunteers, and the hair dryness, bifurcation and breakage phenomena were subjectively evaluated after 28 days of use according to the exactly same method of test 3; the shampoo of the example 1 and the shampoo of the comparative example 5 are respectively used by 2 groups of volunteers, and the use method is the same as that of the test 3, and the difference is that no cap is worn in outdoor activities, the outdoor activities are not less than 2 hours every day, and the phenomena of hair dryness, hair bifurcation and hair breakage are subjectively evaluated after 28 days of use; the results of the questionnaire are shown in table 3.
Table 3:
Test 5 shows that the shampoo of the embodiment 1 has better effect of preventing hair from drying out, bifurcation and fracture when wearing the cap than the shampoo of the comparative embodiment 5, because the embodiment 1 modifies the hydrolyzed collagen, and the hydrophobic layer formed by the long-chain alkyl part can prevent the loss of hair moisture and nutrient substances, thereby having better repairing effect; the shampoo of example 1 showed no significant decrease in the effect of preventing hair from drying out, bifurcation and breakage without the cap, while the shampoo of comparative example 5 showed a significant decrease in the effect of preventing hair from drying out, bifurcation and breakage, because example 1 modified hydrolyzed collagen, a benzamide structure having ultraviolet absorption ability was formed between the protein part and the long chain alkyl part, and damage of ultraviolet rays to hair was prevented, whereas unmodified hydrolyzed collagen did not have such structure and efficacy.
