Abstract

技术领域
本发明涉及肿瘤治疗药物相关领域,具体涉及的是一种基于肿瘤细胞和免疫细胞膜融合纳米囊泡的肿瘤疫苗及其制备方法和应用。
背景技术
肿瘤疫苗主要由肿瘤抗原、适当的佐剂、有效的传递载体组成,并辅以有效的给药方式。根据抗原的不同传递形式,目前肿瘤疫苗可以分为三大类,包括可以编码抗原的核酸、肿瘤抗原的蛋白质和能够装载或表达癌症抗原的工程细胞。肿瘤疫苗可以通过不同的方法促进肿瘤抗原的识别,进而增强抗肿瘤免疫,达到抑制肿瘤生长甚至消灭肿瘤的目的。目前肿瘤疫苗主要是靠以下几个方法:设计含有肿瘤相关抗原的核酸并通过电穿孔等转染方法转入细胞中使其表达肿瘤抗原增强肿瘤免疫;基于多肽或者蛋白质的疫苗则是通过筛选或计算机设计与肿瘤免疫相关的,或是可以影响肿瘤生物学特性的蛋白质或多肽,使其进入肿瘤内部发辉作用,从而达到杀伤肿瘤的目的;工程细胞疫苗是在体外对自体或异体免疫细胞进行操作(如通过与抗原共孵育刺激,或抗原负载等),之后将其回输体内进行肿瘤治疗)。
目前肿瘤疫苗的局限性主要有以下几点:
(1)疫苗的免疫原性低:肿瘤抗原表达低下或抗原表位消失是造成疫苗免疫原性低的原因。(2)疫苗输送系统影响肿瘤疫苗的效果(3)研发成本高,合成时间长:其他方法合成路径中设计到的技术复杂,且周期长成本高。(4)安全性参差不齐:一些使用全细胞进行设计的肿瘤疫苗,如果未进行充分的灭活,进入体内会促进肿瘤的发生。相对地,使用细胞膜进行肿瘤疫苗的合成就不用担心这点。
发明内容
本发明要解决的技术问题是针对以上问题和要求,提供一种基于肿瘤细胞和免疫细胞膜融合纳米囊泡的肿瘤疫苗及其制备方法和应用。
为解决以上技术问题,本发明采用以下技术方案:
基于肿瘤细胞和免疫细胞膜融合纳米囊泡的肿瘤疫苗的制备方法,包括以下步骤:
步骤1、对过表达CD40L的肿瘤细胞和树突状细胞分别进行处理获得肿瘤细胞细胞膜和树突状细胞膜;
步骤2、按质量比(4-5):1对肿瘤细胞膜和树突状细胞膜进行混匀,使用装有过滤膜的挤压器进行10次以上挤压循环,获得基于肿瘤细胞和免疫细胞膜融合纳米囊泡的肿瘤疫苗。
进一步的,步骤1具体包括以下内容:
利用细胞膜蛋白与细胞浆蛋白提取试剂盒对过表达CD40L的肿瘤细胞和树突状细胞在冰上进行细胞的裂解,之后在转速800g下离心10分钟,提取上清,将上清于15000g离心30分钟获得细胞膜。
进一步的,步骤2具体包括以下内容:使用BCA法对肿瘤细胞细胞膜和树突状细胞膜进行定量,按肿瘤细胞膜与树突状细胞膜质量比5:1的比例进行混匀,使用装配了5μm,1μm,400nm的聚碳酸酯过滤膜的挤压器进行15次挤压循环,获得基于肿瘤细胞和免疫细胞膜融合纳米囊泡的肿瘤疫苗。
基于肿瘤细胞和免疫细胞膜融合纳米囊泡的肿瘤疫苗,由上述的方法制备得到。
基于肿瘤细胞和免疫细胞膜融合纳米囊泡的肿瘤疫苗在制备肿瘤治疗药物中的应用。
本发明采用以上技术方案后,与现有技术相比,具有以下优点:
(1)采用融合树突状细胞和过表达CD40L的肿瘤细胞膜的纳米囊泡作为肿瘤抗原载体,具有良好的组织相容性及免疫器官靶向性,CD40L可以促进树突状细胞更好的发挥免疫功能。
(2)基于细胞膜形成的纳米囊泡研发成本低,工序简单,提高临床转化应用率。
(3)采用细胞膜为纳米囊泡的材料使得其细胞毒性下降,安全性提高。

具体实施方式
以下结合附图对本发明的原理和特征进行描述,所举实例只用于解释本发明,并非用于限定本发明的范围。
1.CDLVDV的合成和鉴定
CDLVDV的制备方案如图1A所示。构建CD40L过表达的LLC细胞,并通过qPCR和western bolt对其进行表征(图1B)。获得细胞膜碎片后,用DiR和DiO标记CD40L过表达的LLC细胞和DC膜,然后通过挤压和离心制备CDLVDV。用荧光显微镜确认纳米颗粒的杂交情况(图1C)。然后,我们用TEM对其结构进行了确认(图1D)。DLS分析表明,TMPs的粒径分布主要在200nm左右(图1E)。
2.CDLVDV能够体外靶向DC
我们研究了CDLVDV对DC2.4的靶向能力。我们通过荧光成像和流式细胞术研究了DC2.4细胞的吸收效率。用DiI标记的CDLV和CDLVDV刺激DC2.4细胞4小时。荧光显微镜结果显示细胞吸收了纳米颗粒(图1F)。流式细胞仪分析表明,CDLVDV组细胞的荧光信号明显高于CDLV组,这表明DC细胞膜的同源靶向在吸收过程中起了作用,膜杂交也继承了其特性(图1G和H)。
3.CDLVDV在体外刺激DC成熟并间接诱导T细胞活化
DCs是免疫反应中最关键的APCs之一,负责识别和处理抗原。DCs表面的CD40与CD40L之间的相互作用可以增强DCs的抗肿瘤功能。因此,我们首先以BMDC为研究对象,研究CDLVDV的作用。实验的总体设计如图2A所示。我们通过流式细胞术和酶联免疫吸附试验测定了CDLVDV对BMDC的免疫刺激作用。首先,我们用不同比例的DC和LLC细胞制备CDLVDV,以确定成熟DC的最佳比例。确定的比例为1:5(DCs:LLC)(图S2A)。流式细胞术结果表明,CDLVDV刺激的BMDC比其他组表达更高水平的CD86和CD80,表明其成熟BMDC的能力更强(图2B和C)。此外,CDLVDV组的CD80和CD86阳性细胞明显高于CDLV组,这表明杂交也能增强效果。
然后,我们检测了树突状细胞上清液中的IL-12水平。CDLVDV组的IL-12水平高于其他组(图2D)。结果表明,随着CD40L的过表达LLC膜与DC膜的杂交,囊泡的免疫刺激能力被上调,BMDC受到了CDLVDV的影响。然后,为了评估受刺激的DC对T细胞的影响,我们对DC和T细胞进行了共培养。将从脾脏中提取的T细胞与受到不同囊泡刺激的DC共同培养。48小时后,用流式细胞术检测各种T细胞的比例。结果显示,CDLVDV组的CD4+和CD8+T细胞比例最高,高于其他组。同时,混合囊泡组的CD4+和CD8+T细胞比例高于纯囊泡组。这证明CDLVDV可通过与DCs的相互作用后令其激活T细胞。
4.CDLVDV在淋巴结和脾脏中蓄积,并在体内表现出潜在的免疫刺激作用
给C57BL6/J小鼠尾部皮下注射等量的Dil标记CDLVDV和CDLV。24小时后,体内成像及其荧光信号的结果显示CDLVDV在腹股沟淋巴结中明显聚集(图3A和B)。流式细胞术结果显示,CDLVDV组对CD11c+细胞的摄取效率也高于CDLV组(图3C和D)。此外,我们还发现CDLVDV组的淋巴结比其他组大。我们还进行了另一批实验来检测CDLVDV对淋巴结的影响。用PBS、DV、LV、CDLV、LVDV和CDLVDV处理C57BL6/J小鼠。分离腹股沟淋巴结,进一步进行流式细胞术分析。结果表明,淋巴结中CD11c+细胞中CD86+和CD80+的比例增加,表明CDLVDV可刺激体内DCs成熟(图3E和F)。
脾脏是最重要的免疫器官,负责识别和吸收肿瘤抗原。为了检测CDLVDV在脾脏的生物分布,小鼠尾静脉注射了DiR标记的颗粒。进行了体内成像以测量生物分布。结果显示,尾静脉注射CDLVDV 24小时后,CDLVDV主要在肝脏和脾脏积聚。然后,我们评估了CDLVDV对脾脏的免疫刺激能力。脾脏指数显示CDLVDV组最大,表明其潜在的免疫刺激作用(图4G)。此外,CDLVDV组脾脏中CD4+T细胞和CD8+T细胞均增加,明显高于其他组(图3中H到K图)。因此,我们证实了CDLVDV在体内的免疫刺激作用。
5.CDLVDV通过增加T细胞浸润在体内发挥抗肿瘤作用
为了验证CDLVDV的抗肿瘤效应,我们通过尾静脉注射对荷瘤小鼠进行干预。在种瘤7天后进行干预,每隔三天进行一次,于第19天进行取材检测。在观察期间我们记录肿瘤体积,实验结束时测量肿瘤重量。结果显示,CDLVDV组的肿瘤重量和体积在各组中最小,说明CDLVDV能抑制肿瘤的生长(图4A和B,图5)。同时,我们发现各组的肿瘤体积每天都在增加,但CDLVDV组始终保持最低,这证明CDLVDV能在肿瘤早期发挥作用。CDLV组与CDLVDV组的比较表明,CDLV组在一定程度上具有CD40L引起的抗肿瘤作用,但融合囊泡能更好地激活免疫反应。在另一个为期40天的治疗期内,死亡或肿瘤体积超过2000立方毫米的小鼠被排除在外。CDLVDV组的存活率高于其他组,表明CDLVDV可以延长肿瘤小鼠的存活时间(图4C)。肿瘤组织的流式细胞术结果显示,CDLVDV组的CD4+和CD8+T细胞是所有组中最高的。此外,与单组分囊泡相比,融合产物具有更好的效果(图4中D到G图)。
6CDLVDV和PD-L1抗体的协同治疗效果
我们证实了CDLVDV的抗肿瘤免疫通过T细胞浸润增强抗肿瘤免疫。PDL1抗体在肿瘤免疫治疗上已经是一线用药。因此我们想确认我们的产品是否能够一起用,增强PDL1抗肿瘤疗效。
随着ICB药物被应用于临床实践,免疫疗法成为治疗癌症的一种前景广阔的方法。应用PD-1或PD-L1抑制剂可以激活T细胞,增强T细胞的抗肿瘤作用。鉴于CDLVDV可通过与DCs相互作用激活T细胞,我们测试了CDLVDV与PD-L1抗体联合治疗的疗效。图6A所示的体重曲线进一步证实了这些治疗的安全性,体重在治疗期间没有下降。与之前的实验类似,在干预结束时,测量了裸肿瘤的重量,如图6B所示。此外,4组皮下肿瘤体积变化见图6C。很明显,CDLVDV+ICB组对肿瘤生长的抑制效果更好,而单用ICB或CDLVDV对肿瘤的抑制效果较差。这证实了CDLVDV和ICB与PD-L1抗体一样,对肿瘤消退具有协同作用。值得注意的是,流式细胞术还显示,CDLVDV+ICB组肿瘤中CD4+T细胞和CD8+T细胞的百分比显著高于CDLVDV和ICB组,进一步证明了免疫治疗效率的提高(图6中D到G图),图7可以说明CDLVDV联合ICB(PD-L1抗体)的治疗效果较好。
以上所述为本发明最佳实施方式的举例,其中未详细述及的部分均为本领域普通技术人员的公知常识。本发明的保护范围以权利要求的内容为准,任何基于本发明的技术启示而进行的等效变换,也在本发明的保护范围之内。
Claims (5)
1.基于肿瘤细胞和免疫细胞膜融合纳米囊泡的肿瘤疫苗的制备方法,其特征在于,包括以下步骤:
步骤1、对过表达CD40L的肿瘤细胞和树突状细胞分别进行处理获得肿瘤细胞细胞膜和树突状细胞膜;
步骤2、按质量比(4-5):1对肿瘤细胞膜和树突状细胞膜进行混匀,使用装有过滤膜的挤压器进行10次以上挤压循环,获得基于肿瘤细胞和免疫细胞膜融合纳米囊泡的肿瘤疫苗。
2.根据权利要求1所述的基于肿瘤细胞和免疫细胞膜融合纳米囊泡的肿瘤疫苗的制备方法,其特征在于,步骤1具体包括以下内容:
利用细胞膜蛋白与细胞浆蛋白提取试剂盒对过表达CD40L的肿瘤细胞和树突状细胞在冰上进行细胞的裂解,之后在转速800g下离心10分钟,提取上清,将上清于15000g离心30分钟获得细胞膜。
3.根据权利要求1所述的基于肿瘤细胞和免疫细胞膜融合纳米囊泡的肿瘤疫苗的制备方法,其特征在于,步骤2具体包括以下内容:使用BCA法对肿瘤细胞细胞膜和树突状细胞膜进行定量,按肿瘤细胞膜与树突状细胞膜质量比5:1的比例进行混匀,使用装配了5μm,1μm,400nm的聚碳酸酯过滤膜的挤压器进行15次挤压循环,获得基于肿瘤细胞和免疫细胞膜融合纳米囊泡的肿瘤疫苗。
4.基于肿瘤细胞和免疫细胞膜融合纳米囊泡的肿瘤疫苗,其特征在于,由权利要求1-3中任一项所述的方法制备得到。
5.权利要求4所述的基于肿瘤细胞和免疫细胞膜融合纳米囊泡的肿瘤疫苗在制备肿瘤治疗药物中的应用。
