Abstract
本发明公开一种在组织细胞移植抗排异反应中局部注射的趋化因子CCL28免疫调节剂及其应用,所述趋化因子CCL28免疫调节剂包括趋化因子CCL28及趋化因子CCL28的辅料和/或载体。所述辅料和/或载体为水凝胶,所述水凝胶包括甲基丙烯酰化明胶和光引发剂,浓度为甲基丙烯酰化明胶质量/含光引发剂的PBS体积为8‑10%W/V。本发明的趋化因子CCL28免疫调节剂,在组织细胞移植抗排异反应中的移植物局部注射,可以显著改善移植后的排异反应。
Description
技术领域
本发明移植抗排异领域,尤其涉及一种在移植抗排异反应中局部注射的趋化因子CCL28免疫调节剂及其应用。
背景技术
移植是一种广泛应用于临床的有效治疗手段,其术后免疫抑制治疗技术是避免免疫排斥反应必不可少的措施。针对T细胞杀伤的免疫抑制剂持续治疗是抑制移植物排斥反应一直以来的常用策略,但免疫抑制剂的使用可能会引起严重的毒副作用,如肝肾毒性、高血压和感染以及恶性肿瘤等。
近年来,针对移植物保护的Treg免疫疗法发展迅速,可以在不破坏免疫平衡的基础上,形成移植物抗原特异性免疫耐受,以希望最大限度地减少药物毒性反应的基础上,实现移植物的长期保护。但目前该技术仍存在体内Treg细胞数量少,体外扩增困难的技术瓶颈,如何在移植物局部聚集足够数量的调节性T细胞形成移植物保护和诱导免疫耐受等问题。
因此,目前还缺乏一种能够稳定保持移植物局部聚集足够数量的调节性T细胞的技术方案,解决移植抗排异反应的问题。
发明内容
本发明所要解决的技术问题是无法在移植物局部聚集足够数量的调节性T细胞,克服以上背景技术中提到的不足和缺陷,提供一种在组织细胞移植抗排异反应中局部注射的趋化因子CCL28免疫调节剂及其应用。
为解决上述技术问题,本发明提出的技术方案为:
一种在组织细胞移植抗排异反应中局部注射的趋化因子CCL28免疫调节剂,所述趋化因子CCL28免疫调节剂包括趋化因子CCL28。
在移植抗排异反应中的移植物局部使用含趋化因子CCL28的免疫调节剂,能够精准地局部缓释CCL28,在移植物局部募集调节性T细胞。
所述趋化因子CCL28免疫调节剂包括趋化因子CCL28。
优选的,所述趋化因子CCL28免疫调节剂包括趋化因子CCL28的辅料和/或载体。
优选的,所述辅料和/或载体为水凝胶,所述水凝胶包括甲基丙烯酰化明胶和光引发剂,水凝胶浓度为甲基丙烯酰化明胶质量/含光引发剂的PBS体积为8-10%(W/V)。
水凝胶的使用使得CCL28只在局部得到缓释,更好地增加皮肤移植局部的药物浓度,增强对局部的免疫抑制细胞的募集能力,提高对皮肤移植物局部的抗排斥能力。同时,降低了其大范围扩散而对周围正常组织造成免疫抑制,减缓了对正常组织所带来的不利影响。
优选的,所述组织细胞移植抗排异反应为异种皮肤移植,其中的移植物为猪皮肤组织。
本申请的趋化因子CCL28免疫调节剂可用于多种异种动物移植,但利用猪器官作为人类移植器官的来源是解决当前人源器官严重短缺的途径,猪的心脏、肾脏、肺、胰岛、角膜等器官生理组织与人类相似;在猪和人之间交叉传染的疾病相对比较少,大大降低了跨种系感染风险。
在同一个技术构思下,本发明还提供一种在组织细胞移植抗排异反应中局部注射的趋化因子CCL28免疫调节剂的应用,所述趋化因子CCL28免疫调节剂局部注射在组织细胞移植环境的移植物处,参与调节组织细胞移植抗排异反应中的移植物局部免疫微环境。
优选的,所述调节移植物局部免疫微环境为将效应T细胞主导的免疫促进微环境转变为Treg(调节性T细胞)介导的免疫抑制微环境。
优选的,所述趋化因子CCL28免疫调节剂的配置方法包括:
(1)配制引发剂标准溶液;
(2)配制辅料溶液:取GelMA试剂,加入引发剂标准溶液,加热震荡,得到GelMA辅料溶液;
(3)趋化因子与辅料混合物配制:取趋化因子CCL28,采用预热的GelMA辅料溶液重悬,光源固化,得到趋化因子CCL28免疫调节剂。
优选的,步骤(3)所述光源固化时间为12-18s。
与现有技术相比,本发明的有益效果为:
(1)本发明的趋化因子CCL28免疫调节剂,在组织细胞移植抗排异反应中的移植物局部注射,可以显著改善移植后的排异反应。
(2)本发明通过移植物局部注射趋化因子的方法,稳定保持移植物局部聚集足够数量的调节性T细胞;首次将趋化因子CCL28用于皮肤移植,可以增加免疫抑制性细胞Treg,降低局部总CD4+T细胞含量,逆转移植物局部免疫微环境。
(3)本申请将趋化因子与辅料相结合。辅料选取水凝胶,水凝胶的使用使得CCL28只在局部得到缓释,更好地增加皮肤移植局部的药物浓度,增强对局部的免疫抑制细胞的募集能力,提高对皮肤移植物局部的抗排斥能力。同时,降低了其大范围扩散而对周围正常组织造成免疫抑制,减缓了对正常组织所带来的不利影响。
(4)本发明相较于使用免疫抑制剂的方法,局部使用趋化因子对于皮肤移植模型具有较好的效果,能够保证移植物局部聚集足够数量的调节性T细胞的,且具有更低的毒副作用。

除非另有特别说明,本发明中用到的各种原材料、试剂、仪器和设备等均可通过市场购买得到或者可通过现有方法制备得到。
实施例1:
本实施例采用的趋化因子CCL28免疫调节剂包含趋化因子CCL28。
在组织细胞移植抗排异反应中局部注射的趋化因子CCL28免疫调节剂的应用包括以下几个步骤:
第一步,异种皮肤的移植:免疫组化证实免疫缺陷鼠的猪皮异种移植成功
实验步骤如下:
我们利用NSG小鼠为受体,SPF级别新生猪来源的皮肤组织为供体进行皮肤移植。
1、3%戊巴比妥将新生猪(3mL/kg)麻醉后,剃除背部毛发,用温水洗净,酒精消毒后,用心脏取血处死,固定于超净工作台内的小型手术台上,再次用酒精及络合碘消毒背部皮肤;
2、选择合适的取皮机宽度,调整厚度为200μm,从新生猪背部取下数条皮肤移植物保存于预冷的生理盐水;
3、将取下的皮肤用络合碘浸泡30秒,然后用预冷的生理盐水洗涤两次;
4、使用灭菌手术器械将取下的皮肤修剪成1x1cm大小,去除太薄或破损的部分,于预冷的生理盐水中保存;
5、6-8周龄的小鼠用1%巴比妥(6μL/g)麻醉后去除后背部毛发,在超净工作台内固定于小型手术台上,用碘酒消毒3次;
6、用无菌手术器械去除小鼠下背部约1x1cm大小皮肤;
7、将准备好的异种皮肤移植物置于小鼠下背部皮肤缺损处,用美容线缝合四个角后继续用生物胶粘合四边,保证异种皮肤移植物与小鼠皮肤边缘对齐;
8、用棉花和绷带给小鼠背部移植物打上加压绷带固定;
9、术后小鼠置于温垫上至完全苏醒,异种皮肤移植术后小鼠单笼饲养以防止互相破坏皮肤移植物;
10、术后7天每天腹腔注射庆大霉素(800IU/mL,5μL/g)预防感染;术后4-5周评估皮肤移植存活情况判断模型是否建立成功。稳定的异种皮肤移植物呈现较移植时变小的移植面积,异种皮肤移植物外观为黑色,当移植部位皮肤颜色变浅成粉红色、边缘挛缩时被认为是移植物完全丢失。
实验结果如图1所示,其中,对照为未添加CCL28的溶剂PBS对照组,CCL28为添加趋化因子CCL28免疫调节剂实验组。通过免疫组化分析,猪皮移植到NSG小鼠后的生存状况良好,猪皮移植后与周围组织衔接完好,表明猪皮异种移植的免疫缺陷鼠模型构建成功。
第二步,形成排异反应基础:免疫组化证实人免疫重建后皮肤异种移植小鼠局部发生排异反应
经伦理委员会批准,从中南大学湘雅三医院获得正常人外周血样本。所有收集的样本均知情同意。
1、用抗凝管采集正常人外周血样本50mL,于超净工作台内进行后续操作;
2、干细胞保护液配制:取一洁净无菌的50mL离心管,用电子天平称取2.5g BSA粉末,新取一瓶500mL的1×DPBS,在超净工作台中用10mL移液电枪吸取30mL的1×DPBS加入到上述50mL离心管中,剧烈震荡混匀后用再加入2mL 0.5M EDTA,颠倒混匀后使用无菌的50mL注射器和0.22μM滤器将溶液滤回上述1×DPBS瓶中,颠倒混匀之后即为干细胞保护液(Buffer);
3、外周血单个核细胞(PBMC)细胞的分离:取4支50mL的离心管,各用巴氏吸管加入15mL淋巴细胞分离液,然后吸取细胞重悬液缓缓加入至淋巴细胞分离液中,35mL/管,使总体积达50mL左右;无刹车300g离心20min后,用巴氏吸管小心吸取位于离心管中层的白色单个核细胞层至一新的无菌50mL离心管,然后加入buffer至40mL,颠倒混匀后吸取10μl进行细胞计数,300g离心10min后弃上清;
4、利用第一步中皮肤异种移植成功的免疫缺陷小鼠,每只老鼠取107个细胞(体积为100ul)进行尾静脉注射,进行人免疫系统重建;
5、一周后,通过眼眶取血方式取人免疫系统重建的NSG小鼠外周血,进行流式检测分析人和鼠的CD45+细胞,计算人和鼠的CD45+细胞比例,判断免疫重建是否成功;
6、利用免疫重建成功的小鼠,在皮肤移植局部连续10天进行溶剂PBS注射;
7、10天后,取猪皮移植局部及其周围小鼠皮肤组织进行拍照,拍照后立即放入4%多聚甲醛中固定,进行免疫组化染色。
实验结果如图2所示,结果显示,免疫重建的皮肤异种移植小鼠局部注射PBS后,异种移植物局部边缘排斥脱落,血管生成增加,免疫组化结果显示皮肤组织结构出现空泡,表明人免疫重建后皮肤异种移植小鼠局部发生排异反应。
第三步,采用趋化因子CCL28免疫调节剂调节排异反应
经伦理委员会批准,从中南大学湘雅三医院获得正常人外周血样本。所有收集的样本均知情同意。
1、用抗凝管采集正常人外周血样本50mL,于超净工作台内进行后续操作。
2、干细胞保护液配制:取一洁净无菌的50mL离心管,用电子天平称取2.5g BSA粉末,新取一瓶500mL的1×DPBS,在超净工作台中用10mL移液电枪吸取30mL的1×DPBS加入到上述50mL离心管中,剧烈震荡混匀后用再加入2mL 0.5M EDTA,颠倒混匀后使用无菌的50mL注射器和0.22μM滤器将溶液滤回上述1×DPBS瓶中,颠倒混匀之后即为干细胞保护液(Buffer)。
3、外周血单个核细胞(PBMC)细胞的分离:取4支50mL的离心管,各用巴氏吸管加入15mL淋巴细胞分离液,然后吸取细胞重悬液缓缓加入至淋巴细胞分离液中,35mL/管,使总体积达50mL左右;无刹车300g离心20min后,用巴氏吸管小心吸取位于离心管中层的白色单个核细胞层至一新的无菌50mL离心管,然后加入buffer至40mL,颠倒混匀后吸取10μl进行细胞计数,300g离心10min后弃上清。
4、利用第一步中中皮肤异种移植成功的免疫缺陷小鼠,每只老鼠取107个细胞(体积为100ul)进行尾静脉注射,进行人免疫系统重建。
5、一周后,通过眼眶取血方式取人免疫系统重建的NSG小鼠外周血,进行流式检测分析人和鼠的CD45+细胞,计算人和鼠的CD45+细胞比例,判断免疫重建是否成功。
6、利用免疫重建成功的小鼠,在皮肤移植物周围局部连续10天进行趋化因子CCL28免疫调节剂的注射。
7、10天后,取猪皮移植局部及其周围小鼠皮肤组织进行拍照,拍照后立即放入4%多聚甲醛中固定,进行免疫组化染色。
实验结果如图3所示,结果显示,免疫重建的皮肤异种移植小鼠局部注射趋化因子CCL28免疫调节剂后,异种移植物完好,无明显炎症反应,无排异现象,免疫组化结果显示皮肤组织结构正常,表明皮下局部注射趋化因子CCL28免疫调节剂可缓解皮肤异种移植的排异反应。
实施例2:
其余步骤与实施例1相同,在局部注射趋化因子CCL28免疫调节剂时,结合辅料水凝胶提高趋化因子CCL28免疫调节剂对局部抗排异反应的调节程度。
我们采用的水凝胶型号为EFL-GM-60,其浓度为8%(W/V)(甲基丙烯酰化明胶GelMA质量W/含光引发剂的PBS体积V),EFL-LS-1601-405光源固化时间为15s。趋化因子CCL28的使用量为15ng/ul水凝胶,每块移植物注射的水凝胶体积为200ul。
具体配制步骤如下:
1、配制0.25%(w/v)引发剂标准溶液。(1)取20ml PBS,加入装有引发剂LAP的棕色瓶中(内含0.05g LAP);(2)以40-50℃水浴加热溶解15分钟,期间振荡数次。该LAP标准液在4℃避光条件下可保存12个月。
2、配制GelMA溶液(GelMA浓度为8 -10%(w/v))(1)取所需质量的GelMA放入离心管;(2)取引发剂标准溶液加入到上述离心管中,振荡使GelMA充分浸润;(3)以60-70℃水浴避光加热溶解20-30分钟,期间振荡数次;(4)将GelMA溶液立即用0.22μm无菌针头过滤器灭菌(防止低温凝胶化)。
3、趋化因子与水凝胶混合物配制。(1)取3ug趋化因子CCL28,并用200ul 37℃预热的GelMA溶液重悬;(2)用5ml注射器吸入趋化因子和水凝胶混合液,以405nm光源,照射15s使凝胶化,得到含CCL28的水凝胶;
4、立即将凝胶混合物注射到移植物周围,观察并测量水凝胶体积。
采用含水凝胶的趋化因子CCL28免疫调节剂调节排异反应,实验结果如下:
(1)对含CCL28的水凝胶在体内的缓释速度进行测定;
实验结果如图7所示:含CCL28的水凝胶在体内14天完全缓释。
(2)对皮肤组织的生存情况进行记录和统计;
实验结果如图8所示:其中,对照组为实施例1未添加CCL28的水凝胶对照组,CCL28+水凝胶组为实施例2采用含趋化因子CCL28免疫调节剂的水凝胶实验组,皮肤移植小鼠局部注射含CCL28水凝胶后,移植物具有更长的生存期。
(3)采用含CCL28水凝胶移植物周围皮下局部注射治疗,取移植及移植物周围部位皮肤组织,进行Treg细胞的免疫荧光染色;
实验结果如图9所示:其中,对照组为实施例1未添加CCL28的水凝胶对照组,CCL28组+水凝胶组为实施例2采用含趋化因子CCL28免疫调节剂的水凝胶实验组,免疫荧光染色显示皮肤移植小鼠局部注射含CCL28的水凝胶后,移植物组织局部Treg细胞含量升高。相较于单一注射趋化因子CCL28组,结果如图10所示,注射含趋化因子CCL28免疫调节剂的水凝胶组对于调节性T细胞的募集效果得到提升。
对比例1:
其余步骤与实施例1相同,分别保持对照组无免疫调节剂注射,另一组采用CCL28皮下局部注射治疗,进行对照实验。
(1)对皮肤组织的生存情况进行记录和统计;
(2)取移植及移植物周围部位皮肤组织,进行Treg细胞的免疫荧光染色;
实验结果如图4、5所示:其中,对照组为对比例1注射无CCL28的溶剂PBS对照组,CCL28组为CCL28免疫调节剂的皮下局部注射组,皮肤移植物在免疫重建成功后,局部注射CCL28组的皮肤移植物具有更长的生存期,免疫荧光染色显示免疫重建的皮肤异种移植小鼠局部注射CCL28后,异种移植物组织局部Treg细胞含量升高。
(3)取移植及移植物周围部位皮肤组织,进行CD4+T细胞的免疫组化染色;
实验结果如图6所示:其中,对照组为对比例1注射无CCL28的溶剂PBS对照组,CCL28组为CCL28免疫调节剂的皮下局部注射组,免疫组化染色显示免疫重建的皮肤异种移植小鼠局部注射CCL28后,异种移植物组织局部CD4+T细胞含量升高。
Claims (8)
1.一种在组织细胞移植抗排异反应中局部注射的趋化因子CCL28免疫调节剂,其特征在于,所述趋化因子CCL28免疫调节剂包括趋化因子CCL28。
2.如权利要求1所述的趋化因子CCL28免疫调节剂,其特征在于,所述趋化因子CCL28免疫调节剂包括趋化因子CCL28的辅料和/或载体。
3.如权利要求2所述的趋化因子CCL28免疫调节剂,其特征在于,所述辅料和/或载体为水凝胶,所述水凝胶包括甲基丙烯酰化明胶和光引发剂,水凝胶浓度为甲基丙烯酰化明胶质量/含光引发剂的PBS体积为8-10%W/V。
4.如权利要求1所述的趋化因子CCL28免疫调节剂,其特征在于,所述组织细胞移植抗排异反应为异种皮肤移植,其中的移植物为猪皮肤组织。
5.一种在组织细胞移植抗排异反应中局部注射的趋化因子CCL28免疫调节剂的应用,其特征在于,所述趋化因子CCL28免疫调节剂局部注射在组织细胞移植环境的移植物处,参与调节组织细胞移植抗排异反应中的移植物局部免疫微环境。
6.如权利要求5所述的应用,其特征在于,所述调节移植物局部免疫微环境为将效应T细胞主导的免疫促进微环境转变为调节性T细胞介导的免疫抑制微环境。
7.如权利要求5所述的应用,其特征在于,所述趋化因子CCL28免疫调节剂的配置方法包括:
(1)配制引发剂标准溶液;
(2)配制辅料溶液:取辅料试剂,加入引发剂标准溶液,加热震荡,得到辅料溶液;
(3)趋化因子与辅料混合物配制:取趋化因子CCL28,采用预热的辅料溶液重悬,光源固化,得到趋化因子CCL28免疫调节剂。
8.如权利要求7所述的应用,其特征在于,步骤(3)所述光源固化时间为12-18s。
