Abstract
本发明提供了一种两亲性聚多肽,具有式A‑D任一所示结构。本发明提供的两亲性聚多肽可以有效原位捕获功能性蛋白且在病理微环境响应以释放药物/探针。其中,式C或式D结构的两亲性聚多肽,亲水链端含有功能基团R5,自组装成胶束纳米粒子并经静脉注射后,胶束纳米粒子表面的功能基团R5原位捕获内源性血清蛋白,主动调节胶束纳米粒子上的蛋白质冠。胶束纳米粒子表面主动捕获功能性蛋白并对其表面蛋白质冠调节,极大的增加了血液循环时间,促进了肿瘤的积累,同时减少了肝脏和脾脏的隔离,并提高了肿瘤细胞的选择性摄取。进而实现调节纳米粒子表面蛋白质冠和病理微环境响应以释放药物/探针的功能。

技术领域
本发明涉及材料技术领域,尤其涉及一种两亲性聚多肽及其制备方法和应用。
背景技术
近年来,纳米材料作为纳米药物被广泛应用于体内治疗和诊断应用。由于具有较高的表面自由能,这些纳米材料在静脉注射给药后与血液成分接触时,会自发地吸附生物分子。例如血清蛋白与纳米粒子表面结合形成生物分子涂层,称为蛋白质冠。蛋白质冠的形成显著改变了由此产生的纳米药物的生物物理化学性质,对细胞摄取、组织靶向、免疫反应以及单核吞噬细胞系统(MPS)隔离等方面产生了深远的影响。在提高药物传递效率和最大限度地提高治疗效果的长期追求中,了解蛋白质冠形成机制以及通过调控纳米生物界面来利用蛋白质冠组分已成为研究热点。除了对纳米载体的物理化学参数进行工程设计,如化学成分、大小、形状和zeta电位,人们还广泛致力于用防污材料(包括聚乙二醇(PEG)、两性离子聚合物、低声压蛋白等)对纳米药物进行表面功能化。由此产生的具有防污涂层的纳米药物增强了胶体稳定性,延长了血液循环时间,减少了MPS清除率,从而优化了治疗效果,减少了副作用。尽管取得了很大的成就,但人们普遍认为这些防污聚合物并不能完全消除蛋白质冠的形成,这为通过减少血清蛋白吸附来调节纳米药物上的蛋白质冠提供了一种可行的手段。然而,是否可以通过主动调控吸附血清蛋白种类及含量来提高治疗性能仍然是一个悬而未决的问题,这为在纳米药物上设计蛋白质冠提供了一种独特的方法。
血清白蛋白是血液中含量最高的血浆蛋白(如人血清白蛋白为35-50g/L),参与许多关键的病理过程,如营养物质的运输,这一过程由其多个配体结合位点和细胞受体参与促进。血清白蛋白也被证明在许多类型的肿瘤中积累,常见的肿瘤过表达白蛋白结合受体如糖蛋白60(gp60)、分泌的酸性蛋白和半胱氨酸(SPARC)。事实上,血清白蛋白已成功用于递送成像或治疗药物,如紫杉醇伊文思蓝(Evans Blue)、多肽和siRNA,并显示出独特的优势,包括增加生物稳定性、扩大循环、靶向肿瘤等。阿霉素(DOX)的马来酰亚胺基团功能化衍生物aldoxorubicin在静脉注射后立即与血清白蛋白上的半胱氨酸34(Cys34)的巯基结合,与DOX相比,显示出更好的治疗效果和较低的心脏毒性。我们推测血清白蛋白也可以用于纳米药物表面的功能化,以原位调节蛋白冠的组成,这可能会利用内源性白蛋白的长血液循环和肿瘤靶向能力提高治疗效果。
在用于诊断治疗的各种纳米药物中,多肽和多肽/合成聚合物复合物因其可规模化生产、易于功能化、可递送不同治疗药物、良好的生物相容性和独特的生物降解性而受到越来越多的关注。为了使基于多肽的纳米药物能够特异性响应病理微环境,人们引入了大量的刺激响应键,使得合成的纳米药物具有良好的血液循环稳定性,并在活性氧(ROS)、酸性pH和酶过表达等病理微环境下触发药物释放。在这种情况下,肿瘤酸性已被认为是重要的癌症特征之一,pH响应性化学键如腙和缩氨基脲已通过利用细胞外肿瘤组织和细胞内酸性细胞器(例如,内涵体和溶酶体)中的酸性pH得到广泛研究。
发明内容
有鉴于此,本发明要解决的技术问题在于提供一种两亲性聚多肽及其制备方法和应用,制备的两亲性聚多肽可实现与含醛/酮衍生物高效动态可逆的共价结合。
本发明提供了一种两亲性聚多肽,具有式A-D任一所示结构:

S2)脱去上述中间体d1的保护基R1,得到中间体f1;或脱去上述中间体e1的保护基R1,得到中间体g1;
S3)将中间体f1或中间体g1与含醛/酮基团的F衍生物进行反应,得到式A或式B所示的两亲性聚多肽;

本发明提供了一种直接经单体聚合并经动态可逆共价结合含醛酮功能性小分子的方法,解决了之前含酰肼功能基元单体无法直接经α-氨基酸-N-羧酸酐(NCA)开环聚合问题,实现了含醛酮药物、探针等小分子的高效可逆共价结合至聚多肽上。
具体地,用甲氧基/功能基团R5封端的PEG大分子引发剂直接聚合NPCA单体,合成了侧链上含有氨基脲或羟胺基团的聚多肽。通过含醛酮功能性小分子形成酸性微环境响应脲酰腙或肟键得到聚多肽-药物缀合物。
在本发明的一些具体实施例中,上述含醛酮功能性小分子为含酮喜树碱(ketone-CPT)或含醛基乏氧探针(Ir(btpna)(bpy)2)。
本发明还提供了上述制备方法制备的纳米粒子。
本发明还提供了上述纳米粒子在制备可控释放的负载型药物中的应用。
本发明还提供了上述纳米粒子在制备可控释放的抗肿瘤药物、抗炎药物中的应用。
本发明还提供了上述纳米粒子在肿瘤区域病灶诊断成像中的应用。
本发明提供了一系列具有式A-D结构的主链为聚多肽侧链悬挂功能性小分子F的两亲性聚合物,可以在酸性条件下释放功能性小分子F。肿瘤组织呈弱酸性且胞内存在溶酶体等酸性区域,在该酸性区域内,纳米粒子疏水区动态可逆共价结合键式I或式II结构断裂,释放功能性小分子F药物或探针,释放药物进行化疗作用,探针则实现肿瘤区域成像。随着功能性小分子F释放,纳米粒子逐渐解组装最后形成亲水单链形式的嵌段聚合物,且主链为聚多肽生物相容性好。上述聚合物基于这一设计策略,实现了聚多肽侧链高效动态可逆的接枝含醛/酮衍生物,同时其纳米粒子中键合功能性小分子F可控释放。
与现有技术相比,本发明提供了一种两亲性聚多肽,具有式A-D任一所示结构。本发明提供的两亲性聚多肽可以有效原位捕获功能性蛋白且在病理微环境响应以释放药物/探针。其中,式C或式D结构的两亲性聚多肽,亲水链端含有功能基团R5,自组装成胶束纳米粒子并经静脉注射后,胶束纳米粒子表面的功能基团R5原位捕获内源性血清蛋白,主动调节胶束纳米粒子上的蛋白质冠。胶束纳米粒子表面主动捕获功能性蛋白并对其表面蛋白质冠调节,极大的增加了血液循环时间,促进了肿瘤的积累,同时减少了肝脏和脾脏的隔离,并提高了肿瘤细胞的选择性摄取。进而实现调节纳米粒子表面蛋白质冠和病理微环境响应以释放药物/探针的功能。

具体实施方式
为了进一步说明本发明,下面结合实施例对本发明提供的两亲性聚多肽及其制备方法和应用进行详细描述。
以下实施例中所采用的原料均为按照现有技术制备得到或市售得到。
以下实施例中,通过控制引发剂的加入量控制化合物的聚合度,聚合度n=单体物质的量/引发剂物质的量。
实施例1
合成总路线图如下:

合成化合物S-(苯并[d]噻唑-2-基)O-苯基碳硫酸酯,具体步骤如下:
将苯并[d]噻唑-2-硫醇(5.00g,29.897mmol)溶于THF,向其中添加Na2CO3,并在室温下搅拌10分钟;将氯甲酸苯酯(5.15g,32.893mmol)溶解在THF中,并缓慢滴入系统中;在室温下搅拌3小时;过滤,除去溶剂,用PE和甲醇重结晶,得到黄色针状固体,产率87.1%。
合成NPCA单体4a分为三步,具体步骤如下:
合成化合物2a:将肼基甲酸叔丁酯(8.486g,64.212mmol)溶解于28mL DMF中,超低温反应器-15℃;将N,N-羰基二咪唑(CDI,10.412g,64.212mmol)溶于70mL DMF缓慢加入体系中,搅拌2h,待投下一步;将N-Cbz-Lys-OH(16.3636g,58.375mmol)溶于水中,加入2MNaOH调节pH至10,逐滴加入体系内;室温搅拌16h;旋除溶剂,加入适量水,调节pH至2,EA萃取;柱层析纯化PE:EA=4:1→1:2,旋除溶剂,得到无色油状物,产率77.8%。
合成化合物3a:将化合物2a(4.000g,9.122mmol)溶解于40mL CH3OH中,向其中加入400mg Pd/C;抽真空通N2循环3次,H2循环3次;室温搅拌6h;硅藻土助滤,旋除溶剂,得白色固体;产率98.5%。
合成化合物4a:将化合物3a(2.0g,6.571mmol)溶解于28mL水中,加入696.5mgNa2CO3,45℃搅拌预热;将化合物c(2.077g,7.229mmol)溶于100mL THF中,搅拌均匀缓慢滴入体系;45℃搅拌1.5h;向其中加入30mL 20% NaHCO3,过滤,EA洗涤,0℃下调节pH至2,EA萃取,干燥,旋除溶剂;柱层析PE:EA=1:3,旋干溶剂,得白色固体,产率79.2%。
合成NPCA单体4b分为四步,具体步骤如下:
合成化合物1b:在20mL DCM中溶解(Boc-氨基氧基)乙酸(Boc-Aoa,2.00g,10.5mmol),在冰浴中冷却至0℃,加入EDCI(2.41g,12.6mmol)逐渐溶解,然后溶解五氟苯酚(PFp-OH,2.13g,11.6mmol)在5mL DCM溶液中,缓慢滴入系统中,然后移去冰浴,在室温下搅拌5h,然后过滤,获得白色固体产物1b。在真空烘箱中,得到白色固体(产率90.1%)。
合成化合物2b:在冰浴下将化合物1b(3.31g,9.27mmol)溶解在DMF中,加入N-Cbz-Lys-OH(2.86g,10.19mmol),DIPEA(3.59g,27.80mmol),然后返回室温,在室温下搅拌3h,除去DMF,用1M HCl调节pH到2,用EA萃取,用无水硫酸钠干燥,然后通过柱色谱分离。PE:EA=4:1→1:1,溶剂蒸发,得到无色油状物(产率87.0%)。
合成化合物3b:将化合物2b(4.00g,8.820mmol)溶解于40mL CH3OH中,向其中加入400mg Pd/C;抽真空通N2循环3次,H2循环3次;室温搅拌6h;硅藻土助滤,旋除溶剂,得白色固体(产率98.6%);
合成化合物4b:将化合物3b(2.00g,6.263mmol)溶解于28mL水中,加入663.8mgNa2CO3,45℃搅拌预热;将化合物c(1.979g,6.889mmol)溶于100mL THF中,搅拌均匀缓慢滴入体系;45℃搅拌1.5h;向其中加入30mL 20% NaHCO3,过滤,EA洗涤,0℃下调节pH至2,EA萃取,干燥,旋除溶剂;柱层析DCM:CH3OH=20:1,旋除溶剂得白色固体(产率78.5%)。
NPCA单体聚合合成两亲性聚合物5a,5b(即式(Ⅳ)或式(Ⅴ))分为两步,具体步骤如下:
合成化合物5a,5b(聚合度n=单体物质的量/引发剂物质的量):将单体加入甲苯中,共沸除去水3次,然后转移到手套箱中。在手套箱中称出300mg4a或4b单体;加入相应的mPEG-NH2引发剂,加入2mL DMAc溶液;溶解后,转移至60℃,在手套箱中搅拌反应2天;终止反应,并将系统溶于乙醚/甲醇(20:1),得到浅黄色固体(产率87.5%-89.1%)。
合成两亲性聚合物键合药物CPT 7a,7b(即式(Ⅵ)或式(Ⅶ))分为两步,具体步骤如下:
合成化合物6a,6b:将300.0mg化合物5a或5b溶于3mL TFA;在室温下搅拌8小时;加入1.5mL甲醇终止反应;通过减压蒸馏除去TFA(产率98.3%-99.5%)。
合成化合物7a,7b:在10mL圆底烧瓶中的DMSO中溶解6a或6b,和ketone-CPT,然后加入500μL TFA母液(在10mL DMSO中为4mg TFA);在40℃下搅拌5h。在2h,3h和4h的时间内,添加约0.5mL的甲苯以共沸除去水。除去水后,加入等量的TFA继续反应;用100mL甲醇透析(1000Da)12小时(每2小时更换一次溶剂)(产率85.3%-86.8%)。
NPCA单体聚合合成两亲性聚合物9a,9b(即式(Ⅳ)或式(Ⅴ),R5为MI)分为三步,具体步骤如下:
合成化合物8a,8b:将单体加入甲苯中,共沸除去水3次,然后转移到手套箱中。在手套箱中称出300mg 4a或4b单体;加入相应的Fu-MI-PEG-NH2引发剂,加入2mL DMAc溶液;溶解后,转移至60℃,在手套箱中搅拌反应2天;终止反应,并将系统溶于乙醚/甲醇(20:1),得到浅黄色固体(产率82.5%-89.9%)。
合成化合物8a1,8b1:在双颈圆底烧瓶中加入8a或8b(200.0mg,0.048mmol)和甲苯(30mL),通过共沸蒸馏去除甲苯。然后,在N2下加入DMSO(2mL)和异氰酸苄酯(7.0mg,0.053mmol)。然后在室温下将反应混合物搅拌12h。将溶液混合物沉淀到过量乙醚中。最终产物在室温下真空干燥过夜,得到化合物8a1或8b1白色固体(产率91.2%-92.2%)。
合成化合物9a,9b:将8a1或8b1(200.0mg,0.047mol)分散于甲苯(12mL)中,在110℃下在N2气氛下搅拌。通过旋转蒸发去除所有溶剂后,将残留物溶解于TFA(10mL)中,在室温下搅拌7h。将溶液混合物沉淀到过量的冷乙醚中,在真空烘箱中干燥,得到目标物9a或9b(产率99.1%-99.5%)。
合成两亲性聚合物键合药物CPT 10a,10b(即式(Ⅵ)或式(Ⅶ))具体步骤如下:
合成化合物10a,10b:在10mL圆底烧瓶中的DMSO中溶解9a或9b,和ketone-CPT,然后加入500μL TFA母液(0.4mg/mL的DMSO溶液);在40℃下搅拌5h。在2h,3h和4h的时间内,添加约0.5mL的甲苯以共沸除去水。除去水后,加入等量的TFA继续反应;用100mL甲醇透析(1000Da)12小时(每2小时更换一次溶剂)(产率83.1%-92.9%)。
结构表征如图1-图16所示。
图9中DP表示n值。
应用例1共价键合ketone-CPT纳米粒子的制备
以制备0.1mg/mL纳米粒子为例,将1mg两亲性聚合物式7a(n=6),7b(n=8),10a(n=6,R5为MI),10b(n=8,R5为MI)溶解在1mL二甲基亚砜中,在25℃下,采用闪沉法将溶解聚合物二甲基亚砜体系闪沉至7mL去离子水中,采用分子量为14000的透析袋经过去离子水透析除去有机溶剂,获得的溶液称重计算后加入适量去离子水制备得到0.1mg/mL纳米粒子。粒径66nm。
应用例2共价键合ketone-CPT纳米粒子的药物可控释放
共价键合ketone-CPT的纳米粒子(后称纳米粒子),在37℃用20mM不同pH(4.8、6.0、6.5、7.4)的磷酸缓冲液透析纳米粒子,并用稳态荧光光谱测试荧光强度定量得出随着透析时间推移药物ketone-CPT释放曲线,图17所示对应两亲性聚合物(7a)结构,图18所示对应两亲性聚合物(7b)结构。结果表明:共价结合在纳米粒子中的疏水药物随着透析时间延长逐渐释放。
应用例3功能性纳米粒子主动捕获内源性白蛋白调控蛋白质冠
人血浆中蛋白质含量丰富,血清白蛋白含量最高(35-50mg/mL)。两亲性聚合物式(7a,n=6)和两亲性聚合物式(10a)(10a中R5以马来酰亚胺MI为例,n=6)制备纳米粒子与血浆孵育1小时,然后通过蔗糖垫离心。聚丙烯酰胺凝胶电泳分析的蛋白质含量表明,两亲性聚合物式(7a)纳米粒子蛋白质冠中的蛋白质总量是两亲性聚合物式(10a)纳米粒子蛋白质冠的蛋白质总量的1.47倍,主要蛋白质属于分子量为66kDa的白蛋白(图19中a图)。
通过无标记定量液相色谱-质谱联用(LC-MS)分析两亲性聚合物式(7a)和两亲性聚合物式(10a)纳米粒子的蛋白质冠状结构,共检测到171种蛋白质。根据它们的功能对识别的蛋白质进行分组,表明两种胶束主要被白蛋白和免疫球蛋白覆盖。有趣的是,与两亲性聚合物式(7a)纳米粒子相比,两亲性聚合物式(10a)纳米粒子的白蛋白含量增加,免疫球蛋白比例下降(图19和图20)。在已鉴定的蛋白质中,20个含量最高的蛋白质(含量最少大于1wt%)用于绘制热图,并按照丰度降低从上到下依次对齐(图19中c图)。两亲性聚合物式(10a)纳米粒子蛋白质冠中具有更高含量的白蛋白归因于通过巯基-马来酰亚胺点击反应形成共价结合物。因此,本发明提供的两亲性聚多肽引入蛋白反应活性基团增强胶束纳米粒子与内源性白蛋白结合,进一步提高蛋白质冠中白蛋白含量,蛋白质冠组成含量的变化可能会影响所得到的纳米粒子的生物学行为。
与含有具有抗吸附特性的甲氧基PEG相比,胶束纳米粒子表面主动捕获功能性蛋白并对其表面蛋白质冠调节,极大的增加了血液循环时间,促进了肿瘤的积累,同时减少了肝脏和脾脏的隔离,并提高了肿瘤细胞的选择性摄取。这些独特优点的协同作用有助于改善小鼠肿瘤模型的诊断和化疗效果。
以上实施例的说明只是用于帮助理解本发明的方法及其核心思想。应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以对本发明进行若干改进和修饰,这些改进和修饰也落入本发明权利要求的保护范围内。
Claims (10)
1.一种两亲性聚多肽,其特征在于,具有式A-D任一所示结构:
其中,n独立的选自1-3的整数;m独立的选自4-400的整数;x独立的选自5-100的整数;
R5选自以下任一结构:
R6为H、C1~C5的烷基或卤素;
F为键合的功能分子。
2.根据权利要求1所述的两亲性聚多肽,其特征在于,所述F具有以下任一结构:
R为H或C1~C5的烷基。
3.根据权利要求1所述的两亲性聚多肽,其特征在于,所述两亲性聚多肽的数均分子量为1000~500000。
4.权利要求1~3任一项所述的式A-B所示两亲性聚多肽的制备方法,包括以下步骤:
S1)将式a所示单体或式b所示单体与式c1所示的链引发剂在溶剂中反应,得到中间体d1或中间体e1;
S2)脱去上述中间体d1的保护基R1,得到中间体f1;或脱去上述中间体e1的保护基R1,得到中间体g1;
S3)将中间体f1或中间体g1与含醛/酮基团的F衍生物进行反应,得到式A或式B所示的两亲性聚多肽;
R1为叔丁氧羰基或苄氧羰基。
5.权利要求1~3任一项所述的式C-D所示两亲性聚多肽的制备方法,包括以下步骤:
S11)将式a所示单体或式b所示单体与式c2所示的链引发剂在溶剂中反应,然后与苄基异氰酸酯反应,得到中间体d2或中间体e2;
S22)将上述中间体d2脱除保护基,得到中间体f2;或将上述中间体e2脱除保护基,得到中间体g2;
S33)将上述中间体f2或中间体g2和含醛/酮基团的F衍生物进行反应,得到式C或式D所示的两亲性聚多肽;
R1为叔丁氧羰基或苄氧羰基。
6.一种纳米粒子,由权利要求1~3任一项所述的两亲性聚多肽或权利要求4~5任一项所述的制备方法制备的两亲性聚多肽经自组装形成。
7.权利要求6所述的纳米粒子的制备方法,包括以下步骤:
将权利要求1~3任一项所述的两亲性聚多肽或权利要求4~5任一项所述的制备方法制备的两亲性聚多肽溶解在有机溶剂中,采用共溶剂-加水法或闪沉法制备,得到纳米粒子。
8.权利要求6所述的纳米粒子或权利要求7所述的制备方法制备的纳米粒子在制备可控释放的负载型药物中的应用。
9.权利要求6所述的纳米粒子或权利要求7所述的制备方法制备的纳米粒子在制备可控释放的抗肿瘤药物、抗炎药物中的应用。
10.权利要求6所述的纳米粒子或权利要求7所述的制备方法制备的纳米粒子在肿瘤区域病灶诊断成像中的应用。
