Background
The medical name of the acne is acne, which is an inflammatory disease caused by sebaceous gland grease secretion and microorganism breeding. Currently, the technologies for acne removal are divided into surgical treatment, physical treatment and drug treatment.
Surgical treatment prevents the infection and further deterioration of vaccinia by thoroughly squeezing out microorganisms, microorganism secretions and lipid metabolites in hair follicles by means of an instrument through a sterile manipulation technique. The operation treatment speed is high, but the pain is strong, the recurrence is easy, and if pathogenic bacteria enter blood vessels in the process, the serious result is caused, so the risk is high.
The physical therapy kills microorganisms such as propionibacterium acnes and the like which induce acnes by utilizing the specific wavelength of laser, has low risk of physical laser therapy and can induce regeneration of collagen cells, and is popular for many people at present, but the physical laser therapy only plays a role in sterilization and cannot solve more problems, so that the problems are easy to relapse, and in addition, the phenomena such as pain and swelling of skin, pigmentation and the like can be caused by laser acne removal. Mishandling of laser acne treatment can also cause a series of injuries to the skin.
The medicine treatment is divided into antibiotics, anti-inflammatory medicines, medicines for inhibiting oil secretion and the like according to different action ways, and the main purpose of the medicine treatment is to inhibit oil secretion and microbial growth of skin, and a plurality of acne-removing products such as acne-removing ointment, acne-removing essential oil, acne-removing essence, acne-removing face cream and the like are available on the market. The drug treatment needs to be selected according to different situations of individuals, if the single drug can not achieve the effect or the effect is not long, a certain professional foundation is needed depending on the combination drug, and the acne removal effect is not achieved as same as the single drug when the combination drug is selected.

5 alpha reductase inhibition test
Precisely weighing 36mg testosterone, and fixing the volume to 25mL with absolute ethyl alcohol, wherein the concentration is 5mmol/L; NADPH solution: precisely weighing 42.5mg of NADPH tetrasodium salt, and fixing the volume to 25mL with water, wherein the concentration is 2mmol/L; enzyme extraction buffer solution: 0.32mol/L sucrose, 0.1mmol/L DTT,20mmol/L sodium phosphate, pH6.5. The reaction solution: 1mmol/L DTT,20mmo/L sodium phosphate.
The reaction components were sequentially (155. Mu.L of reaction solution, constant temperature at 37 ℃ C.; 10. Mu.L of testosterone; 10. Mu.L of NADPH; the compositions of the sample examples and comparative examples; 20. Mu.L of 5α -reductase) and the change of absorbance of the reaction system at 340nm within 4min was continuously monitored. According to the calculation formulas of the NADPH standard curve, the inhibitor activity unit and the inhibition percentage, the inhibition rate of the 5 alpha-reductase can be calculated.
Inhibitor activity unit definition: in a reaction system at 37 ℃, one enzyme activity unit is deactivated to one inhibitor activity unit. Percent inhibition = (inhibitor activity/enzyme activity) ×100%. The test results are shown in Table 6.
Test example 5
Tyrosinase inhibition rate assay
The compositions of examples and comparative examples were diluted to 0.2mg/mL in Phosphate Buffer (PBS) with pH=6.8 at 0.2mol/L as a solvent.
0.9mL of the solution prepared from the above sample and having a concentration of 0.2mg/mL was taken, 1mL of L-tyrosine (0.3 mol) and 1mL of Phosphate Buffer (PBS) having pH=6.8 were added, the mixture was preheated in a constant temperature water tank at 37℃for 10 minutes, 0.1mL of an aqueous tyrosinase solution (1.0 mg/mL, mcllvans buffer) was added thereto, the mixture was stirred well, and after a reaction at 37℃for 15 minutes, absorbance (D1) at 475nm was measured. The same reaction was performed using the phosphate buffer instead of the sample, and the absorbance (D2) was measured. For control, absorbance (D3) was measured without the addition of sample solution and tyrosinase. Tyrosinase activity inhibition was calculated using the following formula. Tyrosinase activity inhibition ratio (%) = [ (D2-D1)/(D2-D3) ]x100%. The test results are shown in Table 6.
