Abstract

技术领域
本公开属于生物化学领域;更具体地,本公开涉及一种肿瘤微环境激活的细胞因子及其应用。
背景技术
细胞因子是分子信使,它允许免疫系统的细胞相互交流,以产生对目标抗原的协调、稳健但自限性的反应。虽然免疫系统的许多形式的交流是通过直接的细胞间相互作用发生的,但细胞因子的分泌使免疫信号以多方面和有效的方式快速传播。
近年来,许多细胞因子,包括GM-CSF、IL-7、IL-12、IL-15、IL-18和IL-21,进入晚期癌症患者的临床试验。基于在动物模型中发现的几种促炎细胞因子的强效抗肿瘤活性,重组干扰素-α和白细胞介素-2用于治疗多种恶性肿瘤临床研究被批准,即使疗效还亟待提高。
干扰素根据它们与称为I型、II型和最近描述的III型IFN受体的特定受体结合的能力进行分类。虽然在向黑色素瘤患者提供辅助IFNα时,大剂量IFNα是最常用的方案,但非常需要具有更好治疗指数的疗法,目前正在研究多种这种情况下的各种其他方案。使用IFN的经验已经形成了识别和管理毒性和副作用的既定指南。IFNα的毒性特征通常与剂量有关,大多数副作用可以在不停止治疗的情况下控制,全身症状包括发热、疲乏、头痛、胃肠道症状和肌痛是很常见的,并且可能会发生在80%或更多的患者身上。IFNα也使一些患者的肝酶升高,特别是在高剂量静脉注射期间,此时应经常监测患者,并在治疗期间对肝酶升高的患者保持治疗或减少剂量。因此,降低IFNα的使用剂量、提高IFNα的给药效果,是这一领域亟待解决的问题。
发明内容
本公开的目的在于提一种供肿瘤微环境激活的IFNα细胞因子及其用途。
在本公开的第一方面,提供一种IFNα细胞因子生物分子偶联物,有以下结构:

Cys代表包含在IFNα表面单点突变成的半胱氨酸残基;

本发明人经过深入的研究,揭示了一种优化改造的INF-α,以其搭配肿瘤高表达的可以识别切割的底物和掩蔽体(R),可形成生物分子偶联物,该生物分子偶联物在正常组织中INF-α活性被阻碍、不造成对正常组织的毒性,在肿瘤环境中INF-α活性被激活、进行有效的肿瘤抑制。本公开的偶联物,可使得INF-α的抑制活性高度聚焦于肿瘤环境中,从而实现非常理想的抑瘤效应。
术语
如本文所用,除非另外说明,“IFNα突变体”、“突变型IFNα”可互换使用,是指对野生型的IFNα进行突变后的产物;较佳地,是指对应于野生型IFNα(如SEQ ID NO:1),发生以下位置的氨基酸残基突变后所构成的蛋白:Arg149、Ala145、His57、Gln61、Phe64或Ser68、Phe27、Ser152、Arg162”;较佳地,所述Arg149、Ala145、His57、Gln61、Phe64或Ser68的残基突变形成的Cys。

INF-α突变体
本公开中,提供一种INF-α突变体,其氨基酸序列的Arg149、Ala145、His57、Gln61、Phe64或Ser68、Phe27、Ser152、Arg162残基突变为Cys;优选地Arg149残基突变为Cys。
本公开的INF-α突变体可以是化学合成的产物,或使用重组技术从原核或真核宿主(例如,细菌、酵母、高等植物、昆虫和哺乳动物细胞)中产生。

通过常规的重组DNA技术,可利用本公开的多聚核苷酸序列来表达或生产重组的INF-α突变体。一般来说有以下步骤:(1).用本公开的编码INF-α突变体的多核苷酸(或变异体),或用含有该多核苷酸的重组表达载体转化或转导合适的宿主细胞;(2).在合适的培养基中培养的宿主细胞;(3).从培养基或细胞中分离、纯化蛋白质。
本公开中,INF-α突变体多核苷酸序列可插入到重组表达载体中。只要能在宿主体内复制和稳定,任何质粒和载体都可以用。表达载体的一个重要特征是通常含有复制起点、启动子、标记基因和翻译控制元件。本领域的技术人员熟知的方法能用于构建含INF-α突变体编码DNA序列和合适的转录/翻译控制信号的表达载体。所述的DNA序列可有效连接到表达载体中的适当启动子上,以指导mRNA合成。表达载体还包括翻译起始用的核糖体结合位点和转录终止子。表达载体优选地包含一个或多个选择性标记基因,以提供用于选择转化的宿主细胞的表型性状。
包含上述的适当DNA序列以及适当启动子或者控制序列的载体,可以用于转化适当的宿主细胞,以使其能够表达蛋白质。
本公开中,所述的宿主细胞可以是原核细胞,如细菌细胞,优选地如大肠杆菌;或是低等真核细胞,如霉菌细胞,酵母细胞;或是高等真核细胞,如植物细胞。代表性例子有:大肠杆菌、枯草杆菌、链霉菌、农杆菌;真核细胞如酵母、植物细胞等。
本领域一般技术人员清楚如何选择适当的载体、启动子、增强子和宿主细胞。本公开建立的重组细胞(宿主细胞)可以用常规方法培养,表达本公开的基因所编码的多肽。根据所用的宿主细胞,培养中所用的培养基可选自各种常规培养基。在适于宿主细胞生长的条件下进行培养。当宿主细胞生长到适当的细胞密度后,用合适的方法(如温度转换或化学诱导)诱导选择的启动子,将细胞再培养一段时间。
在表达时,本公开的INF-α突变体可在细胞内、或在细胞膜上表达、或分泌到细胞外。如果需要,可利用其物理的、化学的和其它特性通过各种分离方法分离和纯化重组的蛋白。这些方法是本领域技术人员所熟知的。这些方法的例子包括但并不限于:常规的复性处理、用蛋白沉淀剂处理(盐析方法)、离心、渗透破菌、超声处理、高速离心、分子筛层析(凝胶过滤)、吸附层析、离子交换层析、高效液相层析(HPLC)和其它各种液相层析技术及这些方法的结合。
本公开的优化的INF-α突变体有多方面的与INF-α特性相关的用途,包括用于作为抑制肿瘤的活性分子。应理解,表达该突变体的宿主细胞或其表达产物(如裂解产物或分泌产物)也具有该用途。

组合物
本公开还包括含有所述生物分子偶联物的组合物(如药物组合物),其包含如本文所述的偶联物。药物组合物可进一步包含药学上可接受的载体。载体可以是任何药学上可接受的载体或赋形剂,其可以根据剂型和给药方式而变化。药学上可接受的载体通常是安全且无毒的,并且可包含在制药工业中配制药物组合物中使用的任何已知物质,包括填料,稀释剂,凝结剂,粘合剂,润滑剂,助流剂,稳定剂,着色剂,润湿剂和崩解剂等。药学上可接受的合适的载体包括糖,如乳糖或蔗糖,甘露醇或山梨糖醇;纤维素制剂和/或磷酸钙,如磷酸三钙或磷酸氢钙;淀粉,包括玉米淀粉,小麦淀粉,大米淀粉,马铃薯淀粉,明胶,黄蓍胶,甲基纤维素,羟丙基甲基纤维素,羧甲基纤维素钠和/或聚乙烯吡咯烷酮;二氧化硅,滑石粉,硬脂酸或其盐,如硬脂酸镁或硬脂酸钙;和/或聚乙二醇等等。当选择药学上可接受的载体时,主要考虑的是药物剂型的给药方式。这在本领域中是众所周知的。

在肿瘤患者中,携带肿瘤抗原的肿瘤细胞或抗原呈递细胞(APC)通过与T细胞结合而部分或完全抑制宿主对肿瘤的免疫杀伤。在一些实施方式中,本公开的偶联物通过蛋白水解酶,尤其是天冬酰胺内肽酶或粒酶,或在酸性条件下,在病理微环境中被活化和释放。
因此,本公开的偶联物可以有效地突破个体的免疫屏障,到达病理微环境,然后在病理微环境中被激活和释放。结果,它可以选择性地促进肿瘤微环境中T细胞等的增殖或杀伤作用,从而实现低自身免疫性和高效力。
本公开开的生物分子偶联物可以用于治疗肿瘤,或者可以用作制备用于治疗肿瘤的药物的活性成分。本文所述的肿瘤可以是已知由本文所述的IFNα治疗的任何肿瘤,包括但不限于前文中所列举的。
本公开还包括用于治疗或预防肿瘤的方法,其包括向有此需要的受试者施用治疗或预防有效量的如本文所述的偶联物或其药物组合物。该方法可以与任何已知的放射疗法或免疫疗法组合使用。
因此,本公开构建了一种细胞因子前药,其通过化学连接子将掩蔽体(R)与突变的细胞因子进行化学偶联,从而使细胞因子在正常组织中的活性丧失,降低该细胞因子的毒性,同时也可以进一步将该细胞因子前药与抗体偶联,进一步提高药物性能(对靶向性、半衰期、以及表达量进行进一步的优化),在靶向组织附近,经酶切激活或者酸性条件激活后,恢复至原来或者更好活性发挥其药效。
生物偶联物与抗体的联用
本公开提供了一种联合用药的方法,包括利用所述生物分子偶联物联合抗肿瘤抗体的用药方法。

作为本公开的优选方式,所述的抗肿瘤抗体为抗PD-1抗体,在发明的具体实施例中,其与本公开的生物分子偶联物联合应用后呈现了非常显著的增效。

本公开的药物组合物或混合物可以通过常规方法制成任何常规的制剂形式。剂型可以是多种多样的,只要是能够使活性成分有效地到达哺乳动物体内的剂型都是可以的。比如可选自:注射剂,输液剂,片剂,胶囊剂,丸剂。其中活性组分可以存在于适宜的固体或液体的载体或稀释液中。适用的药学上可接受的载体同前述。
本公开的生物分子偶联物和抗肿瘤抗体的混合物或药物组合物也可储存在适宜于注射或滴注的消毒器具中。通常,在本公开的药物组合物中,所述生物分子偶联物和抗肿瘤抗体作为活性成分可以占药物组合物总重量的0.0001-20%,其余可以为药学上可接受的载体。
所用的所述生物分子偶联物和抗肿瘤抗体的有效剂量可随给药的模式和待治疗的疾病的严重程度而变化。必要的时候,所述生物分子偶联物和还可与其它活性成分或药物联合给药。
当将所述生物分子偶联物和抗肿瘤抗体联用时,本公开还提供了一种用于治疗肿瘤的药盒,所述的药盒中,含有:容器1,以及置于容器1中的所述生物分子偶联物;以及容器2以及置于容器2中的抗肿瘤抗体。
所述的药盒中,也可含有所述生物分子偶联物和抗肿瘤抗体的混合物,其中所述生物分子偶联物和抗肿瘤抗体的含量如前述。

本公开实验中,选择了一些链接子R1-AAN-PABC-R3进行研究。合成好的连接子化合物统一溶解,并用水稀释10倍到1毫克/毫升。在37℃、2小时条件下在100微克legumain中加入1毫克/毫升的样品化合物,酶能够剪切连接子导致释放出R3,通过HPLC能够检测链接子的减少和R3的增加而比较酶对链接子的激活效率(指被酶剪切而释放出来的R3占原化合物的比例)。如表2,R1=peg5000时,链接子及其激活效率如表2所示。

上述结果说明:从稳定性和激活效率综合评价下,链接子1,链接子3均能呈现非常高的激活效率,链接子10的激活效率也很高。
实施例1、INF-α蛋白表面的氨基酸突变后与R1-AAN-PABC-R3化合物偶联的情况
采用链接子R1-AAN-PABC-R3,R1为peg5000,该链接子为表2中链接子3。
INF-α蛋白表面的任一氨基酸突变为Cys后都可以与R1-AAN-PABC-R3化合物偶联,但是偶联效率各有不同。对INF-α蛋白的氨基酸序列进行第2-165位的单一位点的氨基酸突变,根据含突变位点的INF-α的氨基酸序列及其DNA序列,构建可在哺乳细胞中表达出His*6-INF-α的质粒(金唯智)。表达宿主是HEK293细胞(Life Technologies)。转染之前,HEK293细胞在含有10%FBS完全培养基(Gibico)中于37℃,5%CO2中培养。转染前一天,将细胞以合适的密度接种在15cm的培养盘中,培养基更换为低IgG的FBS。在转染6个小时后或者2天后,培养基更换为Freestyle293(Gibico)。转染当天,当细胞达到一定汇合度时,使用脂质体2000(Life Technologies)和PEI(Sigma)将表达目标蛋白的质粒共转染到293T细胞中。在转染后的第4天和第6天分别更换培养上清。检测蛋白的表达与活性,然后纯化蛋白。
将纯化的突变体以0.3mg/mL的浓度在含有5mM EDTA的20mM磷酸盐缓冲液(pH7.4)中温育。将TCEP溶液以100:1的摩尔比添加到突变体中将所得混合物在4℃温育4小时,同时轻轻搅拌。然后将混合物用含有150mM NaCl的20mM磷酸盐缓冲液(pH7.4)在4℃透析2小时。然后,立即将R1-AAN-PABC-R3以20:1的摩尔比加入混合物中,并将所得混合物在25℃温和搅拌16小时。停止反应并除去残留的R1-AAN-PABC-R3化合物。取等体积的样品,然后进行SDS-PAGE检测,通过扫描条带灰度得到偶联效率(扫描灰度的比例即为偶联效率:偶联后蛋白条带的灰度/(未偶联蛋白条带的灰度+偶联后蛋白条带的灰度))。INF-α蛋白表面的单一位点氨基酸突变为Cys后与R1-AAN-PABC-R3化合物偶联效率列表如表4。

因此对于抑制INFR1结合的突变位点而言,将His57,Gln61,Phe64,Ser68分别突变为Cys后,与部分R1-AAN-PABC-R3进行偶联,其结合INFR1的能力均下降至野生型的10%以下。对于抑制INFR2结合的突变位点而言,将Ala145,Arg149分别突变为Cys后,与一些R1-AAN-PABC-R3进行偶联,其结合INFR2的能力均下降至野生型的1%以下,甚至小于0.1%。第Phe27、Ser152、Arg162也有不同程度的下降。
实施例5、突变的INF-α与R1-AAN-PABC-R3偶联并激活后的结合活性变化情况
在活化试验中,将10μg用于活化的Legumain酶(可在AAN位置切割)加入到1mg/ml的偶联后突变体中,在37℃下反应1小时或者1mg/ml的偶联后突变体放置在pH=6的酸性环境中在37℃下反应4h。通过ELISA检测激活后的结合活性情况。
突变的INF-α与R1-AAN-PABC-R3偶联并激活后的结合活性变化情况如表8。

结果说明,对于抑制INFR1结合的变位点而言,除了侧链与R3结构相似的Phe64,Ser68外,与R3结构差异比较大的His57和侧链带电的Gln61相对应的突变体偶联后再激活,其结合INFR1的能力大部分可以恢复至野生型的200%以上。由于INF-α与INFR2结合活性很高,所以INF-α上的任意变动都可能引起其与INFR2结合活性的降低,因此针对INFR2的突变位点的筛选的关键在于偶联并激活后结合活性的恢复情况。对于抑制INFR2结合的突变位点而言,除了侧链与R3结构相似的Phe27,Ala145,Ser152外,Arg149(R149,也即第149位由Arg突变为Cys)相对应的突变体偶联后活性降至原来的0.27%,再激活后其结合INFR2的能力可以恢复至接近野生型的100%。
实施例6、突变的INF-α与R1-AAN-PABC-R3偶联后以及激活后对细胞信号传导的影响情况
准备280000cells/ml HEK-Blue IFN-a/b细胞悬浮液在含10%热灭活的FBS的培养基中。在96孔细胞板中分别加入20ul不同梯度的INF-α,突变的INF-α+R1-AAN-PABC-R3(其中R1为peg20000),Active form,然后每个孔加入180ul细胞悬浮液。将96孔细胞板放在37℃的二氧化碳培养箱中培养20-24小时。
按照产品说明书准备QUANTI-BlueTM,并在新的96孔细胞板中每个孔中加入180ulQUANTI-BlueTM。然后再加入20ul上述的HEK-Blue IFN-a/b细胞上清液,在37℃条件下孵育1h。在620-055nm波长下检测SEAP水平。
突变的INF-α与R1-AAN-PABC-R3偶联后以及激活后对细胞信号传导的影响情况如表9。

表6的结果表示,INF-αR149+R1-AAN-PABC-R3偶联后的样品和激活后的样品对细胞信号传导的影响效果最理想。
INF-αR149偶联后以及激活后对细胞信号传导的影响情况如图1。结果表明,多种突变形式的INF-α与R1-AAN-PABC-R3偶联后,细胞信号传导减至极弱或没有、而激活后呈现显著的恢复。其中,INF-αR149+R1-AAN-PABC-R3偶联后的样品可以完全抑制相应的细胞信号传导,但激活后的激活形式可以恢复至与野生型的INF-α一样的细胞信号传导水平。
实施例7、在肿瘤微环境中活化的INF-α抑制MC38肿瘤的生长情况
将MC38细胞皮下接种到C57BL/6小鼠(上海模拟生物研究中心),每只小鼠2×106个细胞。一周后,将植入MC38肿瘤的小鼠随机分组。第1组G1注射溶剂对照,每周给药1次,其余每组分别注射100ug突变的INF-α+R1-AAN-PABC-R3,每周给药1次。给药周期为3周。每周记录3次小鼠肿瘤体积。不同的INF-α+R1-AAN-PABC-R3给药后的TGI(肿瘤生长抑制,TumorGrowth Inhibition)情况如表10。

该表的结果表明,多种突变形式的INF-α与R1-AAN-PABC-R3偶联后,具有抑制肿瘤生长的效果。其中,INF-αR149+R1-AAN-PABC-R3抑制肿瘤生长的效果最明显。
实施例8、INF-α的偶联物在肿瘤微环境中活化的INF-α的抑瘤效果
将MC38细胞皮下接种到C57BL/6小鼠(上海模拟生物研究中心),每只小鼠2×106个细胞。一周后,将植入MC38肿瘤的小鼠随机分成4组。
分组如下:
第1组:G1注射100ug INF-α,每周给药3次;
第2组:G2注射300ug INF-αR149+R1-AAN-PABC-R3,每周给药1次;
第3组:G3注射100ug INF-αR149+R1-AAN-PABC-R3,每周给药1次;
第4组:G4注射溶剂对照,每周给药1次。
给药周期为3周。每周记录3次小鼠肿瘤体积。给药结束后的肿瘤体积结果显示在图2中。
其中,R1-AAN-PABC-R3具体为表3中链接子10。
结果表明,300ug INF-αR149+R1-AAN-PABC-R3可有效抑制MC38肿瘤的生长,一只小鼠被治愈,五只小鼠具有显著的抑制作用,其为80%或更高。一周给药一次100ug INF-αR149+R1-AAN-PABC-R3比一周给药三次100ug INF-α的抑制肿瘤效果更好。
该结果表明,INF-α的偶联物可以增强效应T细胞的活性和INF-α在肿瘤微环境中的富集,因此偶联物表现出比原始INF-α更高的功效。
实施例9、INF-α的偶联物与PD-1抗体联合应用的抑瘤效果
将CT26细胞皮下接种到Balb/c小鼠(上海模拟生物研究中心),每只小鼠2×106个细胞。一周后,将植入CT26肿瘤的小鼠随机分成4组:
第1组(G1):注射100ug INF-αR149+R1-AAN-PABC-R3,每周给药1次;
第2组(G2):注射100ug抗PD-1的鼠抗(Sino Biological;Cat:50124-RP02),每周给药2次;
第3组(G3):注射100ug INF-αR149+R1-AAN-PABC-R3,每周给药1次;同时,注射100ug抗PD-1的鼠抗,每周给药2次;
第4组(G4):注射溶剂对照,每周给药1次。
其中,R1-AAN-PABC-R具体为表3中链接子10。
给药周期为3周。每周记录3次小鼠肿瘤体积。给药结束后的肿瘤体积结果显示在图3中。结果表明,100ug INF-αR149+R1-AAN-PABC-R3与抗PD-1抗体的联合可有效抑制CT26肿瘤的生长,四只小鼠被治愈,全部小鼠具有显著的抑制作用。肿瘤体积的降低极为显著,可见两者的联合应用实现了协同增效。
因此,在肿瘤微环境中活化的INF-αR149与PD-1抗体联合表现出更高的功效和治愈率。
以上所述实施例仅表达了本公开的几种实施方式,其描述较为具体和详细,但并不能因此而理解为对本公开专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本公开构思的前提下,还可以做出若干变形和改进,这些都属于本公开的保护范围。因此,本公开专利的保护范围应以所附权利要求为准。同时,在本公开提及的所有文献都在本申请中引用作为参考,就如同每一篇文献被单独引用作为参考那样。
Claims (16)
1.一种IFNα细胞因子生物分子偶联物,有以下结构:
R1-AAN-PABC-R3-S-Cys-IFNα
其中,
Cys代表包含在IFNα表面单点突变成的半胱氨酸残基;
S代表半胱氨酸残基上的硫原子;
IFNα具有SEQ ID NO:1所示的氨基酸序列;
R1是一个阻止IFNα突变体与它的配体或受体结合的基团;
AAN代表三肽,A为丙氨酸,N为天冬氨酸,PABC为对-氨基苄基氨甲酰基;
R3是一个带有EMC基团与IFNα的单点突变的半胱氨酸的硫原子共价偶联形成的连接臂,其作用是当AAN断裂后,R3结构仍偶联在细胞因子上,R3-S-Cys-IFNα部分恢复,完全恢复,甚至提高与IFNα受体的亲合力。
2.如权利要求1所述的生物分子偶联物,其特征在于,其中R1选自分子量44至132000的聚乙二醇,例如1000至50000、3000至80000或10000至60000的聚乙二醇;或
所述IFNα突变体为INF-α进行单点突变后获得的突变体,其氨基酸序列根据SEQ IDNO:1,Arg149、Ala145、His57、Gln61、Phe64或Ser68、Phe27、Ser152、Arg162残基突变为Cys。

8.一种用于抑制肿瘤的组合物或药盒,其特征在于,所述组合物或药盒中含有权利要求1-5任一所述的生物分子偶联物;较佳地,所述组合物还含有药学上可接受的载体。
9.如权利要求8所述的组合物或药盒,其特征在于,所述组合物中还含有抗肿瘤抗体;或
所述药盒中还含有:抗肿瘤抗体或含有抗肿瘤抗体的组合物。
10.如权利要求9所述的组合物或药盒,其特征在于,所述的抗肿瘤抗体是抗PD-1抗体。
11.如权利要求8所述的组合物或药盒,其特征在于,所述的生物分子偶联物与所述抗PD-1抗体的按照质量比为1:(0.05~50);较佳地为1:(0.2~8);更佳地为1:(0.4~5)。
12.一种制备肿瘤微环境中活化的INF-α的方法,包括:将INF-α进行单点突变,使其氨基酸序列的特定残基突变为Cys。
13.一种INF-α突变体,其为INF-α进行单点突变后获得的突变体,其氨基酸序列根据SEQ ID NO:1,Arg149、Ala145、His57、Gln61、Phe64或Ser68、Phe27、Ser152、Arg162残基突变为Cys。
14.分离的多核苷酸,所述的多核苷酸编码权利要求13所述的INF-α突变体。
15.权利要求13所述的INF-α突变体的用途,用于与R1-AAN-PABC-R3偶联形成抑制肿瘤的生物分子偶联物;
其中,
R1是一个阻止IFNα突变体与它的配体或受体结合的基团;
AAN代表三肽,A为丙氨酸,N为天冬氨酸,PABC为对-氨基苄基氨甲酰基;
R3是一个带有EMC基团与IFNα的单点突变的半胱氨酸的硫原子共价偶联形成的连接臂,其作用是当AAN断裂后,R3结构仍偶联在细胞因子上,R3-S-Cys-IFNα部分恢复,完全恢复,甚至提高与IFNα受体的亲合力。
16.如权利要求1-15任一所述,其特征在于,所述的肿瘤包括:黑色素瘤,NSCLC,头颈鳞状细胞癌,尿路上皮癌,经典霍奇金淋巴癌,胃癌,食管胃交界腺癌,宫颈癌,B细胞淋巴癌,肝细胞癌,默克尔细胞癌,肾细胞癌,原发性肝癌,小细胞肺癌。
