Abstract
本发明公开了一种改善皮肤老化松弛的组合物及其制备方法和应用,所述改善皮肤老化松弛的组合物包括如下重量份组分:交替单胞菌发酵产物提取物20‑30份、茵陈蒿花提取物30‑35份、可溶性蛋白多糖30‑35份;通过复配特定比例的交替单胞菌发酵产物提取物、茵陈蒿花提取物、可溶性蛋白多糖,清除DPPH自由基,从根源处解决皮肤衰老问题,能够有效提升肌肤弹性、肌肤紧致感,减少细纹、淡化细纹,改善肌肤干燥,起到改善皮肤老化松弛的作用。
Description
技术领域
本发明属于日化品领域,尤其涉及一种改善皮肤老化松弛的组合物及其制备方法和应用。
背景技术
衰老是任何生物生命过程中必须经历的阶段,是生命体发育成熟后,随着年龄的增长,受内源性因素(遗传因素)以及外源性因素(辐射污染光损伤光老化等)影响,而产生的渐进的形态结构和生理功能的衰退过程。皮肤作为人体最大的器官,必然会经历衰老过程。
皮肤衰老表现有:皮肤变薄、角质层异常、皱纹增多、加深、皮肤弹性降低松弛下垂、皮肤柔韧度下降、脆性增加、干燥缺水。其中皮肤弹性降低松弛下垂、皱纹增多加深最为普遍。这主要是由于弹性蛋白酶引起的胶原蛋白流失造成的。紧致抗衰老,一要解决及时性二要解决长效性,通过抑制神经递质乙酰胆碱释放,解决即时紧致的问题;通过促进胶原合成、抑制基质金属蛋白酶的活性,解决长效紧致抗衰的问题,通过增加I型、III型、IV型胶原蛋白的含量,让皮肤丰满弹性。
虽然现在皮肤抗衰老方面的研究有了较大的进步,但是抗衰老原料及护肤品的开发还是存在很多缺陷。目前,抗衰老原料及护肤品多存在一些片面性,很多是使用了单一具有抗氧化作用的物质,或使用低含量的植物提取物,或无法清除自由基,从根源处解决皮肤衰老问题。
发明内容
针对上述现有技术中的不足,本发明的目的在于提供一种改善皮肤老化松弛的组合物。通过复配特定比例的交替单胞菌发酵产物提取物、茵陈蒿花提取物、可溶性蛋白多糖,清除DPPH自由基,从根源处解决皮肤衰老问题,能够有效提升肌肤弹性、肌肤紧致感,减少细纹、淡化细纹,改善肌肤干燥,起到改善皮肤老化松弛的作用。
本发明的目的在于提供一种改善皮肤老化松弛的组合物,包括如下重量份组分:交替单胞菌发酵产物提取物20-30份、茵陈蒿花提取物30-35份、可溶性蛋白多糖30-35份。
交替单胞菌发酵产物提取物源自海洋微生物KOPARA产生的胞外多聚糖,由半乳糖、半乳糖醛酸、葡萄糖、葡萄糖醛酸和甘露糖组成,能快速填充皱纹、紧实皮肤;具有显著促进胶原蛋白、弹性蛋白、透明质酸合成的效果,从肌肤内部减少皱纹,对抗衰老,收紧胶原蛋白晶格,能够抵御抗自由基的能力以增强、平抚皱纹、焕发光彩、恢复弹性、肌肤润泽细腻紧致。
茵陈蒿是一种菊科蒿属植物,可以入药,民间过去点燃其干燥茎叶,用于驱蚊虫。茵陈蒿花提取物可以抑制黑素细胞合成的黑色素转移到角质形成细胞,具有美白功效,还可以抑制细菌活性,清除自由基,抑制透明质酸酶、5a-还原酶活性,保护透明质酸免受降解,减少油脂分泌。
可溶性蛋白多糖源自宽叶榆绿木,可通过促进成纤维细胞增殖与I型胶原蛋白产生,从而达到提升胶原蛋白密度、淡化皱纹、改善皮肤纹理及淡化色斑。
更优选地,所述交替单胞菌发酵产物提取物24份、茵陈蒿花提取物32.5份、可溶性蛋白多糖33.5份。
优选地,所述改善皮肤老化松弛的组合物还包括芋螺肽0.002-0.2份。
芋螺肽可增加细胞活性,促进胶原蛋白合成,但皮肤对芋螺肽有屏障作用,导致芋螺肽无法穿过表皮层或无法被更深皮肤层次所吸收,影响芋螺肽抗皱功效的发挥。
更优选地,所述改善皮肤老化松弛的组合物还包括芋螺肽0.01份。
优选地,所述改善皮肤老化松弛的组合物还包括β-烟酰胺单核苷酸0.002-0.5份。
β-烟酰胺单核苷酸是细胞外烟酰胺磷酸核糖转移酶反应的产物和NAD+的前体,可有效提高机体NAD+的水平,间接参与细胞内线粒体的糖类、脂肪代谢、三磷酸循环等多种代谢途径,从而维持线粒体正常的能量代谢效率、维持细胞组织的正常功能、减少ROS对机体的损伤、延缓衰老。但β-烟酰胺单核苷酸稳定性差,对温度、湿度、光和氧等因素敏感。研究发现,β-烟酰胺单核苷酸在液体中稳定性差,在膏霜乳液剂型等化妆品中降解快,将其应用于化妆品中,β-烟酰胺单核苷酸的含量会在短时间内大幅度下降,导致其在液体组合物中无法有效发挥抗衰老作用。
更优选地,所述改善皮肤老化松弛的组合物还包括β-烟酰胺单核苷酸0.01份。
优选地,所述改善皮肤老化松弛的组合物还包括β-丙氨酰羟脯氨酰二氨基丁酸苄胺0.002-0.15份。
β-丙氨酰羟脯氨酰二氨基丁酰苄胺可通过抑制弹性蛋白酶的活性保护肌肤弹性蛋白,激活I型、III型、IV型胶原新增。
更优选地,所述改善皮肤老化松弛的组合物还包括β-丙氨酰羟脯氨酰二氨基丁酸苄胺0.01份。
优选地,所述改善皮肤老化松弛的组合物还包括磷酸氢二钠0.002-0.1份、磷酸二氢钠0.002-0.1份和丁二醇0.57-10份。
更优选地,所述改善皮肤老化松弛的组合物还包括磷酸氢二钠0.05份、磷酸二氢钠0.01份和丁二醇5份。
优选地,所述茵陈蒿花提取物的提取工艺如下:
将干燥并粉碎的茵陈蒿花,过筛,加入萃取液,浸提,超声,过滤,滤液浓缩,得茵陈蒿花提取物。
优选地,所述萃取液包括乙醇、甲醇、丁醇、水中的至少一种。
更优选地,所述萃取液选自乙醇。
优选地,所述茵陈蒿花与萃取液的质量比为1:8~12。
更优选地,所述茵陈蒿花与萃取液的质量比为1:10。
优选地,所述超声的频率为20~40kHz,时间为3~5小时。
更优选地,所述超声的频率为30kHz,时间为4小时。
优选地,所述浸提的温度为60~70℃,浸提的时间为2~3小时。
更优选地,所述浸提的温度为65℃,浸提的时间为2.5小时。
本发明另一目的在于提供一种化妆品,由所述改善皮肤老化松弛的组合物添加化妆品上可接受的辅料,制成化妆品。
优选地,所述化妆品选自面霜、精华液、爽肤水、洁面乳、原液、粉底液、卸妆水、遮瑕霜、粉底霜、口红、胭脂、香水、眼影或腮红中的任意一种。
优选地,以质量百分比计,所述改善皮肤老化松弛的组合物的用量为化妆品的5-20%。
更优选地,以质量百分比计,所述改善皮肤老化松弛的组合物的用量为化妆品的10%。
具体实施方式
为了使本技术领域的人员更好地理解本发明中的技术方案,下面将结合本发明实施例对本发明的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都应当属于本发明保护的范围。
交替单胞菌发酵产物提取物(商品名:Exo-PTM,法国Lucas Meyer);可溶性蛋白多糖购买自日本ICHIMARU PHARCOS公司。
实施例1:茵陈蒿花提取物的提取。
称取干燥并用中药粉碎机粉碎的茵陈蒿花18kg,过80~100目筛备用,取过筛后的茵陈蒿花粉末15kg,加入150kg乙醇,混合均匀后搅拌浸提,65℃浸提2.5h,将提取混合物转入超声萃取设备,30kHz超声萃取4h,过滤除去不溶物,滤液中加入滤液质量3%的活性炭,搅拌,50℃保温30min,过滤除去活性炭,浓缩滤液,得茵陈蒿花提取物。
实施例2:改善皮肤老化松弛的组合物。
交替单胞菌发酵产物提取物24质量份、茵陈蒿花提取物32.5质量份、可溶性蛋白多糖33.5质量份混合均匀,得改善皮肤老化松弛的组合物。
实施例3:改善皮肤老化松弛的组合物。
交替单胞菌发酵产物提取物20质量份、茵陈蒿花提取物35质量份、可溶性蛋白多糖35质量份混合均匀,得改善皮肤老化松弛的组合物。
实施例4:改善皮肤老化松弛的组合物。
交替单胞菌发酵产物提取物30质量份、茵陈蒿花提取物30质量份、可溶性蛋白多糖30质量份混合均匀,得改善皮肤老化松弛的组合物。
DPPH自由基清除能力测定:
称取一定量的DPPH,用无水乙醇配制成0.04mg/mL的DPPH溶液。分别取2g的实施例2-4的改善皮肤老化松弛的组合物加入1000mL无水乙醇,分别得到2mg/mL的实施例2-4稀释样品液,分别取2mL的实施例2-4稀释样品液加入2mL DPPH溶液,混合均匀,室温反应30min后,在517nm处测吸光值Ai。并分别取2g的交替单胞菌发酵产物提取物、2g的茵陈蒿花提取物、2g的可溶性蛋白多糖分别加入1000mL无水乙醇,分别得到2mg/mL的对比例1~3的稀释样品液,分别取2mL的对比例1~3稀释样品液加入2mL DPPH溶液,混合均匀,室温反应30min后,在517nm处测吸光值Ai。同时测定2mL DPPH溶液+2mL无水乙醇的吸光度A0和2mL样品液+2mL无水乙醇混合后的吸光度Aj。重复测定2次。样品对DPPH自由基的清除率用以下公式计算:实验结果见表1。
DPPH自由基清除率=(A0-Aj+Ai)/A0×100%。
DPPH自由基清除率越高,则抗氧化能力越强,反之越弱。

由表1可知,本发明通过复配交替单胞菌发酵产物提取物、茵陈蒿花提取物和可溶性蛋白多糖可协同提高DPPH自由基清除率。
面霜的制备:
原料:
表2.面霜配方。
制备方法:(1)按表2的原料分别称取备用;
(2)将A相中的水、对羟基苯乙酮、山梨醇混合搅拌,再加入聚甘油-3-甲基葡糖二硬脂酸酯、小核菌胶、聚乙烯醇,80℃加热搅拌;
(3)将B相中的原料混合,80℃加热搅拌,溶解均匀,再加入A相中,均质10min,得混合物;
(4)将所述的混合物降温至温度小于45℃,依次加入C相原料,搅拌至溶解均匀,分别得实施例5和对比例4~8面霜。
抗衰老性能测试:
选择年龄30-50岁的皮肤健康的志愿者60名,随机平均分成6组,每组10名,每组志愿者分别使用实施例5、对比例4~8的面霜涂抹于志愿者的面部和眼角,受试者每周使用3次,连续使用4周。通过调查问卷的方式,收集受试志愿者对面霜使用后皮肤状态的评价,以分析面霜的使用的肌肤紧致感、细纹减少效果,通过1-3分的评分方式体现,3分表示肌肤紧致强、细纹大大减少,2分表示肌肤紧致一般、细纹轻微减少,1分表示肌肤紧致感基本不变、细纹基本没有减少,肌肤紧致感和细纹减少效果分别计算分值,计算每组总得分,分值越高说明面霜抗衰老效果越好。

由表3可知,本发明实施例5面霜的抗衰老作用明显,通过对比对比例7~8可知,β-丙氨酰羟脯氨酰二氨基丁酸苄胺可促进芋螺肽穿过表皮层或被更深皮肤层次所吸收,提高芋螺肽的抗皱功效,同时,提高β-烟酰胺单核苷酸在液体中稳定性,使其在膏霜乳液剂型等化妆品中不会快速降解,有效发挥β-烟酰胺单核苷酸的抗衰老作用。
刺激性测试:
选择年龄18-40岁的皮肤健康的志愿者20名。采用48h封闭式斑贴试验方法,取实施例5的面霜产品0.02mL至斑试器中,贴敷于受试者前臂内侧,使用射频美容仪使面霜在受试者前臂内侧分散,24小时后去除受试物,保持斑贴部位干燥,避免激烈运动、挠抓斑试部位、长时间日光照射等,48h后去除试器并做好标记,30min后在充足灯光下进行判定,并分别在72h后进行回访观察。

由表5可知,本发明实施例5的面霜安全无刺激。
最后应当说明的是,以上实施例仅用以说明本发明的技术方案而非对其限制,尽管对照上述实施例对本发明进行了详细的说明,所属领域的普通技术人员应当理解,技术人员阅读本申请说明书后依然可以对本发明的具体实施方式进行修改或者等同替换,但这些修改或变更均未脱离本发明申请待批权利要求保护范围之内。

1.一种改善皮肤老化松弛的组合物,其特征在于,包括如下重量份组分:交替单胞菌发酵产物提取物20-30份、茵陈蒿花提取物30-35份、可溶性蛋白多糖30-35份。
2.如权利要求1所述的改善皮肤老化松弛的组合物,其特征在于,还包括芋螺肽0.002-0.2份。
3.如权利要求1所述的改善皮肤老化松弛的组合物,其特征在于,还包括β-烟酰胺单核苷酸0.002-0.5份。
4.如权利要求1所述的改善皮肤老化松弛的组合物,其特征在于,还包括β-丙氨酰羟脯氨酰二氨基丁酸苄胺0.002-0.15份。
5.如权利要求1所述的改善皮肤老化松弛的组合物,其特征在于,还包括磷酸氢二钠0.002-0.1份、磷酸二氢钠0.002-0.1份和丁二醇0.57-10份。
6.如权利要求1所述的改善皮肤老化松弛的组合物,其特征在于,所述茵陈蒿花提取物的提取工艺如下:
将干燥并粉碎的茵陈蒿花,过筛,加入萃取液,浸提,超声,过滤,滤液浓缩,得茵陈蒿花提取物。
7.如权利要求1所述的改善皮肤老化松弛的组合物,其特征在于,所述萃取液包括乙醇、甲醇、丁醇、水中的至少一种;所述茵陈蒿花与萃取液的质量比为1:8~12;所述浸提的温度为60~70℃,浸提的时间为2~3小时。
8.一种化妆品,其特征在于,由权利要求1~7任一项所述改善皮肤老化松弛的组合物添加化妆品上可接受的辅料,制成化妆品。
9.如权利要求8所述的化妆品,其特征在于,所述化妆品选自面霜、精华液、爽肤水、洁面乳、原液、粉底液、卸妆水、遮瑕霜、粉底霜、口红、胭脂、香水、眼影或腮红中的任意一种。
10.如权利要求8所述的化妆品,其特征在于,以质量百分比计,所述改善皮肤老化松弛的组合物的用量为化妆品的5-20%。
