Abstract
本发明提供一种含有红茶发酵产物的美白组合物。在该组合物中,本发明使用红茶发酵产物、金铃花提取物、红千层提取物作为原料,实验结果表明,本发明提供的美白活性组合物在抑制酪氨酸活性方面具有优异的效果,可以用于美白化妆产品。
Description
技术领域
本发明涉及化妆品领域,具体涉及一种含有红茶发酵产物的美白组合物。
背景技术
黑色素是决定皮肤颜色深浅的重要影响因素之一,抑制黑色素的产生可以产生美白效果。黑色素是一种氨基酸衍生物,是酪氨酸或3,4-二羟基苯丙氨酸经过一连串化学反应而形成的,其常以聚合的方式存在。黑色素由人表皮基底层中的黑色素细胞合成,当紫外线照射到皮肤上,肌肤就会处于“自我防护”的状态,激活酪氨酸酶的活性,催化酪氨酸或3,4-二羟基苯丙酸氧化生成黑色素,来保护我们的皮肤细胞。
黑色素合成过程主要参与的酶有酪氨酸酶、多巴色素互变酶、二羟基吲哚羧酸氧化酶,其中酪氨酸酶属于氧化还原酶,是黑色素合成的主要限速酶。在黑色素合成过程中,酪氨酸酶能够催化L-酪氨酸羟基化为左旋多巴,然后再将左旋多巴氧化为多巴醌,再经过各种复杂的非酶促反应之后,最终产生黑色素。因此,通过抑制酪氨酸酶的活性,可以减少黑色素的产生,从而达到美白的效果。
现有技术中,专利文献CN114788796A公开了含福鼎白茶提取物的美白抗菌化妆品组合物,使用福鼎白茶提取物、川芎提取物、沙棘多糖提取物和花椒抗菌肽作为原料,福鼎白茶提取物中的黄酮具有抗氧化和抗辐射的功效,川芎中的有机酚酸类、生物碱类、内酯类等成分具有美白的功效。
金铃花(Abutilon striatum),又名灯笼花,富含多酚、多糖等多种抗氧化活性成分。红千层,温中散寒,理气止痛的功效,其鲜叶桉叶素含量较高,提取精油用于化妆品,但现有技术未公开使用提取物制作美白化妆品的方案。本发明人在开发美白化妆品的过程中,经过大量的原料选择发现,将红茶发酵产物、金铃花提取物和红千层花提取物作为原料用于美白化妆品时,在抑制酪氨酸活性方面取得了突出的效果,由此完成了本发明。
发明内容
本发明要解决的技术问题是提供一种美白组合物,使用红茶发酵产物、金铃花提取物、红千层提取物作为原料,用以解决美白的技术问题。
为了解决上述问题,本发明提供一种含有红茶发酵产物的美白组合物,按质量百分比计,包括:

本发明提供了一种含有红茶发酵产物的美白组合物,在该组合物中,本发明创造性的使用了红茶发酵产物、金铃花提取物、红千层提取物作为原料,实验结果表明,本发明提供的美白活性组合物在抑制酪氨酸活性方面具有突出的效果,是良好的美白化妆产品。

本发明提供的含有红茶发酵产物的美白组合物,按质量百分比计,包括17-21%的月桂醇磺基琥珀酸酯二钠、4.5-5.5%的月桂醇聚醚硫酸酯钠、3.9-4.2%的氯化钠、0.1-0.2%的亚硫酸钠、2.9-3.2%的椰油酸胺丙基甜菜碱、0.1-0.4%的PEG-150二硬脂酸酯、0.45-0.58%的1,2-己二醇、0.46-0.59%的对羟基苯乙酮、0.15-0.25%的红茶发酵产物、0.2-0.5%的红千层提取物、0.15%-0.35%的金铃花提取物、0.1-0.2%的亚硫酸钠、0.01-0.03%的甘油、0.07-0.1%的苯甲酸钠。
按照本发明,月桂醇磺基琥珀酸酯二钠的含量优选为17.5%-20.5%,更优选为17.9%-19.5%,更优选为18.1%-19.1%,更优选为18.2%-18.8%;月桂醇聚醚硫酸酯钠的含量优选为4.6-5.3%,更优选为4.7-5.1%,更优选为4.85-4.96%;椰油酸胺丙基甜菜碱的含量为2.96-3.1%,更优选为2.98-3.05%;1,2-己二醇的含量为优选为0.48-0.55%,更优选为0.49-0.52%;羟基苯乙酮的含量优选为0.47-0.55%,更优选为0.49-0.52%;红茶发酵产物的含量优选为0.16-0.26%,更优选为0.17-0.25%,更优选为0.18-0.24%;红千层提取物的含量优选为0.25-0.45%,更优选为0.28-0.38%,更优选为0.29-0.35%;金铃花提取物的含量优选为0.25-0.3%。
在本发明提供的化妆品组合物中,月桂醇磺基琥珀酸酯二钠起到清洁剂和增泡剂的作用;月桂醇聚醚硫酸酯钠有清洁乳化作用;氯化钠起到增稠的作用;椰油酸胺丙基甜菜碱作为泡沫促进剂;PEG-150二硬脂酸酯起到表面活性剂的作用;乳化剂以及黏度控制剂的作用;亚硫酸钠起到抗油氧化剂和防腐剂的作用;甘油起到润泽作用,增强对皮肤的渗透力;苯甲酸钠作为防腐剂;1,2-己二醇起到保湿的作用;对羟基苯乙酮起到抗氧化剂的作用;上述组分均符合行业要求的安全剂量;红茶发酵产物、红千层提取物、金铃花提取物作为美白活性成分。
金铃花提取物的获得方法,包括:取金铃花为原料,洗净并干燥后用打粉机打粉至100目以下,得到粉末;将所述粉末溶于乙醇得到溶液,再利用100体积%乙醇和pH值为2的酸液活化C18固相萃取柱,将所述溶液上样至所述固相萃取柱中,利用pH值为2的酸液淋洗所述固相萃取柱,再利用100体积%甲醇洗脱所述固相萃取柱,收集洗脱液;氮吹所述洗脱液,收集浓缩液进行冷冻干燥处理,得到金铃花酚提取物。
红千层提取物的制备,包括:1)以红千层花为原料,洗净并干燥后用打粉机打粉、过筛、混匀,得到粉末,投入乙醇溶剂中进行回流,合并提取液;2)将提取液减压浓缩得到浸膏,使用石油醚脱脂处理,并用甲醇回流提取处理残渣,进行减压浓缩;3)将减压浓缩产物使用水分散,作为大孔吸附树脂的上样液;4)将上样液用梯度甲醇溶液洗脱,然后收集70%甲醇溶液洗脱液浓缩,得到红千层提取物。
红茶发酵物采用如下工艺制备:
1)按质量份取2~4份的红茶、2~4份的蔗糖和40~80份的水混合煮沸10-20min,冷却,获得红茶糖液;
2)第一次发酵、获得发酵滤液,按质量比往步骤1)所得冷却物中加入1-3%、密度为108~109cfu/mL的酵母菌菌悬液,混合均匀静置发酵,发酵成熟后过滤得到发酵滤液;
3)第二次发酵、获得红茶发酵物制剂,往发酵滤液中加入密度为106~108CFU/mL葡糖酸醋杆菌菌悬液、密度为106~108CFU/mL植物乳杆菌菌悬液和密度为105~107CFU/mL的用来从葡萄糖直接生产葡糖醛酸的微生物悬液进行第二次发酵,发酵成熟后灭活、浓缩得到红茶发酵物制剂;第二次发酵时,葡糖酸醋杆菌菌悬液、植物乳杆菌菌悬液和巴氏醋杆菌菌悬液加入量分别为:1-3v/v%、0.5-2v/v%、0.5-2v/v%;浓缩后的体积为浓缩前的1/2以下。
其中,酵母菌菌悬液由保藏号为CGMCC2.1543的酿酒酵母(Saccharomycescerevisiae)培养所得,葡糖酸醋杆菌菌悬液由保藏编号为CCTCC M 2018745的葡糖酸醋杆菌(Gluconacetobacter xylinum)C2培养所得,所述植物乳杆菌菌悬液由保藏号为CGMCC1.511的植物乳杆菌(Lactobacillus plantarum)培养所得,微生物悬液由保藏号为FERM BP-10820的Pseudogluconobacter saccharok etogenes Rh47-3株培养所得。
所述第一次发酵的工艺条件为:将装有待发酵液的容器置于湿度为30%~40%,温度为25~30℃的环境下静置发酵5-15天;所述第二次发酵的工艺条件为:将装有待发酵液的容器置于湿度为30%~40%,温度为25~30℃的环境下静置发酵发酵3-10天,待形成菌膜即为发酵成熟。
实施例1制备金铃花提取物
1)、取金铃花为原料,洗净并干燥后用打粉机打粉至100目以下、得到粉末;
2)将粉末与乙醇按照1:10配置,得到溶液;
3)利用100体积%乙醇和pH值为2的酸化水活化C18固相萃取柱,将溶液上样至固相萃取柱中,利用pH值为2的酸化水淋洗固相萃取柱,再利用100体积%甲醇洗脱固相萃取柱,收集洗脱液;
4)氮吹洗脱液,收集浓缩液进行冷冻干燥处理,得到金铃花提取物。
实施例2制备红千层提取物
1)以红千层花为原料,洗净并干燥后用打粉机打碎,过90目筛,混匀,得到50kg粉末,投入1t 95%的乙醇中进行回流三次,每次三小时,合并提取液;
2)将提取液减压浓缩得到浸膏11kg,使用30L石油醚脱脂处理三次,并用10L甲醇回流残渣三次,再进行减压浓缩;
3)将减压浓缩产物使用10L水分撒,作为AB-8型大孔吸附树脂的上样液;
4)将上样液分别用上水、20%甲醇溶液、70%甲醇溶液洗脱,收集70%甲醇溶液洗脱,浓缩至干,得到红千层提取物320g。
实施例3制备红茶发酵物
1)按质量份取3份的红茶、3份的蔗糖和60份的水混合煮沸15min,冷却,获得红茶糖液。
2)第一次发酵、获得发酵滤液,按质量比向步骤1)所得冷却物中加入2%、密度为108~109cfu/mL的酵母菌菌悬液,混合均匀静置发酵,发酵成熟后过滤得到发酵滤液。静置发酵方法为,将装有待发酵液的容器置于湿度为30%~40%,温度为25~30℃的环境中静置发酵10天。
3)第二次发酵,获得红茶发酵物制剂,向发酵滤液中加入密度为106~108CFU/mL葡糖酸醋杆菌菌悬液、密度为106~108CFU/mL植物乳杆菌菌悬液和密度为105~107CFU/mL的用来从葡萄糖直接生产葡糖醛酸的微生物悬液进行第二次发酵,发酵成熟或灭活、浓缩得到红茶发酵物制剂;第二次发酵时,葡糖酸醋杆菌菌悬液、植物乳杆菌菌悬液和巴氏醋杆菌菌悬液加入量分别为:2v/v%、1v/v%、1v/v%;浓缩后的体积为浓缩前的1/2。
第二次发酵的工艺条件为:将装有待发酵液的容器置于湿度为30%~40%,温度为25~30℃的环境下静置发酵7天,待形成菌膜即为发酵成熟。
上述酵母菌菌悬液保藏号为CGMCC 2.1543的酿酒酵母(Saccharomycescerevisiae)培养所得,所述葡糖酸醋杆菌菌悬液由保藏编号为CCTCC M 2018745的葡糖酸醋杆菌(Gluconacetobacter xylinum)C2培养所得,所述植物乳杆菌菌悬液由保藏号为CGMCC 1.511的植物乳杆菌(Lactobacillus plantarum)培养所得,所述微生物悬液由保藏号为FERM BP-10820的Pseudogluconobacter saccharoketogenes Rh47-3株培养所得。
实施例4配置美白组合物,获得待测样品1
按质量百分比计取用原料:月桂醇磺基琥珀酸酯二钠为18%,月桂醇聚醚硫酸酯钠5.5%,红茶发酵产物为0.18%,金铃花提取物为0.19%,红千层提取物为0.3%,氯化钠为4.1%,椰油酸胺丙基甜菜碱为3.1%,1,2-己二醇为0.47%,亚硫酸钠为0.15%,甘油为0.03%,苯甲酸钠为0.08%,PEG-150二硬脂酸酯为0.3%,对羟基苯乙酮为0.49%,余量水。
先将基料月桂醇磺基琥珀酸酯二钠、月桂醇聚醚硫酸酯钠、红茶发酵产物、金铃花提取物、红千层提取物与少量去离子水混合搅拌均匀,然后加入剩余原料及去离子水混合搅拌至乳化,获得待测样品1。
实施例5配置美白组合物,获得待测样品2
按质量百分比计取用原料:月桂醇磺基琥珀酸酯二钠为21%,月桂醇聚醚硫酸酯钠4.5%,红茶发酵产物为0.15%,金铃花提取物为0.35%,红千层提取物为0.5%,氯化钠为3.9%,椰油酸胺丙基甜菜碱为3.2%,1,2-己二醇为0.45%,亚硫酸钠为0.15%,甘油为0.03%,苯甲酸钠为0.08%,PEG-150二硬脂酸酯为0.3%,对羟基苯乙酮为0.49%,余量水。
其余工艺同实施例4,获得待测样品2。

先将基料月桂醇磺基琥珀酸酯二钠、月桂醇聚醚硫酸酯钠、红茶发酵产物、与少量去离子水混合搅拌均匀,然后加入剩余原料及去离子水混合搅拌至乳化,获得对比例1样品。
对比例2配置美白组合物
按质量百分比计取用原料:月桂醇磺基琥珀酸酯二钠为19%,月桂醇聚醚硫酸酯钠4.9%,红千层提取物为0.32%,氯化钠为4.1%,椰油酸胺丙基甜菜碱为3.1%,1,2-己二醇为0.58%,亚硫酸钠为0.2%,甘油为0.01%,苯甲酸钠为0.1%,PEG-150二硬脂酸酯为0.4%,对羟基苯乙酮为0.49%,余量水。
先将基料月桂醇磺基琥珀酸酯二钠、月桂醇聚醚硫酸酯钠、红千层提取物、与少量去离子水混合搅拌均匀,然后加入剩余原料及去离子水混合搅拌至乳化,获得对比例2样品。
DPPH自由基清除实验:
取3mL浓度为0.2mmol/L的DPPH-无水乙醇溶液于比色皿中,再加入1mL测试样品,混匀,室温避光30min,在517nm波长处测吸光度,吸光值记为A1,加入3mL无水乙醇溶液和1mL测试样品溶液的吸光度值记为A2,加3mLDPPH-无水乙醇溶液和1mL无水乙醇溶液的吸光值记为A3。按下式计算DPPH自由基清除率:DPPH自由基清除率(%)=[1-(A1-A2)/A3]×100%,取三次测量的平均值,实验结果见表1。
羟自由基清除实验:
取0.5mL浓度为0.75mmol/L邻二氮菲的无水乙醇溶液,分别加入浓度为0.2mol/L的pH7.4磷酸盐缓冲液2mL和1mL测试样品,充分混匀后加入0.5mL浓度为0.75mmol/L的FeSO4·7H2O溶液,再次混匀后加入0.5mL0.01%(v/v)的H2O2,于37℃恒温水浴60min后,在536nm下分别测量各组混合溶液的吸光度值,记为As,以蒸馏水代替样品溶液作为空白组测吸光度值,记为Ab,以蒸馏水替代H2O2作为损伤组,测其吸光度值An,测试样品羟自由基清除率按以下公式计算:羟自由基清除率(%)=[(As-An)/(Ab-An)]×100%,取三次测量的平均值。测试结果如表1所示。

表1的测试结果表明,本发明提供的美白组合物中,使用红茶发酵产物、金铃花提取物和红千层提取物作为活性成分在清除羟自由基清及DPPH方面具有明显的效果,可以用于美白。
酪氨酸酶活性抑制率测试
酪氨酸酶是一种多酚氧化酶,是皮肤黑色素形成过程中的关键酶,其活性与黑色素合成量呈正相关。如果酶活性过高,人们就会患上色素沉积症。能降低酪氨酸酶活性的物质就是酪氨酸酶抑制剂(Tyrosinase Inhibitor,TI)。酪氨酸酶抑制剂通过抑制酪氨酸酶活性,阻止黑色素形成,可用来预防和治疗色素沉着等疾病。本方法通过测定吸光度来判断色素物质的生成量,从而判断酪氨酸酶受抑制的程度。
测试样品:以0.2mol/L pH=6.8的磷酸缓冲液(PBS)为溶剂,分别将测试样品1-4以及对比例1、2中的组合物或天然物质配置成浓度为0.1mg/mL的溶液,即得测试样品。
测试方法:分别取上述0.1mg/mL的测试样品,加入1mL L-酪氨酸(0.3mol)和1mLpH=6.8的磷酸缓冲液(PBS),在37℃恒温水槽中预热10min,再加入0.1mL酪氨酸酶水溶液(1.0mg/mL,mcllvains缓冲液),充分搅拌,在37℃反应15分钟后,测定475nm吸光度(A1)。另外,用上述磷酸缓冲液代替上述实施例1-4和对比组1-5所制得的组合物配制的浓度为0.1mg/mL的溶液进行同样的反应,测得吸光度(A2)。为了对照,在不加试样溶液和酪氨酸酶的条件下,测得吸光度(A3)。用下式计算出酪氨酸酶活性抑制率:
酪氨酸酶活性抑制率(%)=(A2-A1)/(A2-A1)x100%;
测试结果如表2所示。

表1的测试结果表明,本发明提供的美白组合物中,使用红茶发酵产物、金铃花提取物和红千层提取物作为活性成分在抑制酪氨酸酶活性方面具有明显的效果,这样可以抑制黑色素的产生,起到美白的作用。
人体皮肤斑贴实验
空白组:去离子水
样品组:将上述实施例1-6制得产品配置成体积百分比为1%的待测液。
空白对照的斑试器上不加任何物质,阴性对照组的斑试器上添加50μl去离子水,实验组的斑试器上添加50μL上述制得的待测液;将加有受试物的斑试器敷于受试者的前臂曲侧,用手掌轻压使之均匀地贴敷于皮肤上,持续24h。24h后去除斑试器,去除后30min(待压痕消失后)按表3标准观察皮肤反应,并记录观察结果见表4。

根据表4本发明提供的组合物对人体皮肤无刺激作用。
Lab色度系统测量皮肤颜色变化
方法:参照团体标准《化妆品美白祛斑功效测试方法》(T/AHCA 001-2018)测试美白功效,在人脸上分别使用实施例1-4、对比例1-4制得的样品,每10个人使用一个样品,采用CK-PMA6CL400皮肤测试仪测试皮肤的L(亮泽度)、b(黑色素)、ITA(个体肤色度数),分别测试使用前、使用第6天、使用第13天、使用第20天、使用第27天的L、b、ITA值,测得的个样品的L、b、ITA值的平均数如表5

根据表5本发明提供的组合物用于人体皮肤美白时,具有预料不到的美白效果。
以上对本发明所提供的含有红茶发酵产物的美白活性组合物进行了详细介绍。本文中应用了具体个例对本发明的原理及实施方式进行了阐述,以上实施例的说明只是用于帮助了解本发明的方法及其核心思想。应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以对本发明进行若干改进和修饰,这些改进和修饰也落入本发明权利要求的保护范围内。
Claims (10)
1.一种含有红茶发酵产物的美白组合物,其特征在于,按质量百分比计,包括:
余量水。
2.根据权利要求1所述的美白组合物,其特征在于,按质量百分比计,所述月桂醇磺基琥珀酸酯二钠的含量为17.5%-20.5%。
3.根据权利要求2所述的美白组合物,其特征在于,按质量百分比计,所述月桂醇聚醚硫酸酯钠的含量为4.6-5.3%。
4.根据权利要求3所述的美白组合物,其特征在于,按质量百分比计,所述椰油酸胺丙基甜菜碱的含量为2.96-3.1%。
5.根据权利要求4所述的美白组合物,其特征在于,按质量百分比计,所述1,2-己二醇的含量为0.48-0.55%。
6.根据权利要求5所述的美白组合物,其特征在于,按质量百分比计,所述对羟基苯乙酮的含量为0.47-0.55%。
7.根据权利要求6所述的美白组合物,其特征在于,按质量百分比计,所述红茶发酵产物的含量为0.16-0.26%。
8.根据权利要求7所述的美白组合物,其特征在于,按质量百分比,所述红茶发酵产物的含量为0.17-0.25%。
9.根据权利要求6所述的美白组合物,其特征在于,按质量百分比计,所述金铃花提取物的含量为0.25-0.3%。
10.根据权利要求9所述的美白组合物,其特征在于,按质量百分比计,所述红千层提取物的含量为0.28-0.38%。
