技术领域
本发明涉及生物医药技术领域,特别是涉及锥丝碱在制备抗疱疹病毒药物中的应用。
背景技术
单纯疱疹病毒(Herpes simplex virus,HSV)属于疱疹病毒科α病毒亚科,其基因组为线性双链DNA,病毒颗粒呈球形,有包膜。根据抗原性的差别,HSV分为HSV-1(单纯疱疹病毒Ⅰ型)和HSV-2(单纯疱疹病毒Ⅱ型单纯疱疹病毒第二型)两种血清型。HSV是一种常见的易引起人类皮肤性疾病的病原体,据报道60%-95%的人至少会感染一次。HSV-1主要感染口腔相关区域(嘴唇、舌头、口腔粘膜),可引起口腔黏膜炎、疱疹性角膜炎和疱疹性脑炎等疾病,而HSV-2主要感染生殖器区域,可引起生殖器疱疹。
目前,针对HSV-1尚无有效的疫苗,临床上治疗单纯疱疹感染主要是通过核苷类抗病毒药物,如阿昔洛韦可干扰病毒DNA合成,膦甲酸钠通过与焦磷酸结合位点结合而抑制病毒聚合酶的活性。虽然这些药物在急性感染中有显著疗效,然而临床上多见不良反应,且长期使用可导致耐药病毒株的出现等缺陷。而植物源性天然产物含有大量具有生物活性的化合物,包括酚类、多酚类、鞣质、萜类和生物碱,且天然产物具有作用时间长、低毒以及病毒不易产生耐药性等优点,是开发抗病毒药物的理想资源。
化合物conessine也称为锥丝碱,是一种从夹竹桃科植物-止泻木的种子中分离出的甾体类生物碱,目前大部分生物碱相关的研究集中于抗菌、肿瘤、心血管系统及神经系统疾病的治疗等,与病毒相关的研究较少。conessine的生理功能有:①抗菌:抑制细菌外排泵;抑制细菌生物膜的形成。②抗疟原虫:其IC50值为1.9μg/mL。③抗癌:可降低基础NF-κB活性,调节自噬,具有抗炎作用,被认为是一种潜在的抗癌药物。④有较强的抑制乙酰胆碱酯酶活性,是进一步开发抗阿尔茨海默病(AD)等神经系统疾病候选药物,以上内容均提示化合物conessine具有较强的药用潜力。
发明内容
基于此,有必要针对目前在临床上治疗单纯疱疹感染的药物不良反应多、长期使用易产生耐药性、生物利用度低等问题,提供一种锥丝碱在制备抗疱疹病毒药物中的应用。

具体实施方式
为了便于理解本发明,下面将参照相关附图对本发明进行更全面的描述。附图中给出了本发明的较佳实施例。但是,本发明可以以许多不同的形式来实现,并不限于本文所描述的实施例。相反地,提供这些实施例的目的是使对本发明的公开内容的理解更加透彻全面。
需要说明的是,当元件被称为“固定于”另一个元件,它可以直接在另一个元件上或者也可以存在居中的元件。当一个元件被认为是“连通”另一个元件,它可以是直接连通到另一个元件或者可能同时存在居中元件。
除非另有定义,本文所使用的所有的技术和科学术语与属于本发明的技术领域的技术人员通常理解的含义相同。本文中在本发明的说明书中所使用的术语只是为了描述具体的实施例的目的,不是旨在于限制本发明。本文所使用的术语“和/或”包括一个或多个相关的所列项目的任意的和所有的组合。
以下实施例所用试剂,如非特别说明,均为市售可得;以下实施例所用方法,如无特别说明,均为常规方法可实现。
以下实施例所使用的conessine(锥丝碱)均购自GlpBio公司,货号GC12749。
实施例1
检测化合物conessine的细胞毒性。
(1)将A549、Huh7.5、BHK21细胞(均为市售)以2×104个/孔的密度分别接种于96孔板中,细胞用含10% FBS(胎牛血清)的高糖型DMEM培养基(加1%双抗)在37℃、5% CO2的培养箱中培养。
(2)待18-24小时后,细胞生长至指数期,加入conessine处理,使conessine的终浓度分别为6.25μmol/L、12.5μmol/L、25μmol/L、50μmol/L、100μmol/L、200μmol/L,并设置空白对照组,每个浓度梯度设3个复孔,在37℃、5% CO2的培养箱中继续培养。
(3)用conessine处理24小时后,每孔加入10μL的CCK-8溶液。
(4)继续培养4小时后,按照常规方法,用多功能酶标仪测量450nm处各孔的吸光值,细胞生存率曲线代表conessine处理组与对照组的吸光值之比。
结果如图1所示,化合物conessine在A549、Huh7.5和BHK21细胞中的半数毒性浓度(CC50)分别为76.11μmol/L、60.81μmol/L、85.76μmol/L,10μmol/L及以下浓度的conessine对A549、Huh7.5和BHK21细胞的生长增殖无明显影响。
实施例2
检测不同浓度的conessine在A549、Huh7.5、BHK21细胞中的抗HSV-1复制的效果。
(1)将A549、Huh7.5、BHK21细胞(均为市售)以1×105个/孔的密度分别接种于24孔板中,细胞用含10% FBS(胎牛血清)的高糖型DMEM培养基(加1%双抗)在37℃、5% CO2的培养箱中培养。
(2)待18-24小时后,细胞生长至指数期,以不含FBS的高糖型DMEM洗两遍,再加入HSV-1病毒液(MOI 0.1,感染体积100μL),每隔15分钟摇一次。1小时后弃液,加入500μL的含10% FBS(胎牛血清)的高糖型DMEM培养基。
(3)在加入病毒的1小时前至实验结束的时间段,全程加入conessine处理,使conessine的终浓度分别为2.5μmol/L、5μmol/L、10μmol/L、20μmol/L、40μmol/L,并设置空白对照组。
(4)加入病毒24小时后,收取细胞及上清,反复冻融3次,以空斑实验的方法检测病毒滴度。
结果如图2所示,化合物conessine在A549、Huh7.5和BHK21细胞中对HSV-1的半数抑制浓度(IC50)分别为3.835μmol/L、4.267μmol/L、4.725μmol/L。在conessine浓度为10μmol/L时,HSV-1滴度下降至10%以下。
结合实施例1的结果可见,化合物conessine在不影响A549、Huh7.5、BHK21细胞生长增殖的浓度下,即可发挥抗HSV-1复制的效应。
实施例3
检测化合物conessine对HSV-1感染的预防作用。
(1)将A549、Huh7.5、BHK21细胞(均为市售)以1×105个/孔的密度接种于24孔板中,细胞用含10% FBS(胎牛血清)的高糖型DMEM培养基(加1%双抗)在37℃、5% CO2的培养箱中培养。
(2)待18-24小时后,细胞生长至指数期,分别加入ethanol(乙醇,对照组)和10μmol/L的conessine,在37℃、5% CO2的培养箱中继续培养。
(3)处理1小时后,弃液,以不含FBS的高糖型DMEM洗两遍,加入HSV-1病毒液(MOI0.1,感染体积100μL),每隔15分钟摇一次。1小时后弃液,加入500μL的含10% FBS(胎牛血清)的高糖型DMEM培养基。
(4)加入病毒24小时后,收取细胞及上清,反复冻融3次,按照常规方法进行空斑实验,检测病毒滴度。
结果如图3所示,相对于对照组,10μmol/L的conessine预处理使HSV-1滴度下降至10%,证明化合物conessine可预防HSV-1感染。
实施例4
检测化合物conessine对HSV-1感染的治疗作用。
(1)将A549、Huh7.5、BHK21细胞(均为市售)以1×105个/孔的密度接种于24孔板中,细胞用含10% FBS(胎牛血清)的高糖型DMEM培养基(加1%双抗)在37℃、5% CO2的培养箱中培养。
(2)待18-24小时后,细胞生长至指数期,以不含FBS的高糖型DMEM洗两遍,加入HSV-1病毒液(MOI 0.1,感染体积100μL),每隔15分钟摇一次。
(3)加入病毒1小时后,弃液,以PBS洗两遍,加入500μL的含10% FBS(胎牛血清)的高糖型DMEM培养基,并分别加入ethanol(对照组)和10μmol/L的conessine,在37℃、5%CO2的培养箱中继续培养。
(4)加入病毒24小时后,收取细胞及上清,反复冻融3次,按照常规方法进行空斑实验,检测病毒滴度。
结果如图4所示,相对于对照组,在HSV-1感染后加入10μmol/L conessine处理使HSV-1滴度下降至30%,证明化合物conessine可治疗HSV-1感染。
实施例5
检测化合物conessine对细胞mRNA表达谱的影响。
(4)将A549细胞(市售)以5×105个/孔的密度接种于6孔板中,细胞用含10% FBS(胎牛血清)的高糖型DMEM培养基(加1%双抗)在37℃、5% CO2的培养箱中培养。
(5)待18-24小时后,细胞生长至指数期,分别加入ethanol(对照组)或10μmol/L的conessine,在37℃、5% CO2的培养箱中继续培养。
(6)处理24小时后,收取细胞RNA,进行转录组测序并进行生物信息学分析。
结果如图5所示,其中,图5-A为火山图(log2 fold change),显示对照组和conessine处理组之间存在300个差异表达基因(DEGs,differentially expressed genes)(倍数变化≥2.0且p值≤0.05),其中167个上调,133个下调;图5-B为热图,有相似生理功能并参与相同代谢过程的基因被分成一组;图5-C为GO分析结果,显示DEGs主要与甾醇、胆固醇和类固醇的生物合成或代谢过程相;图5-D为KEGG分析结果,显示DEGs主要参与类固醇、萜类骨架、脂肪酸和胆固醇的生物合成或代谢;图5-E为基于分子特征数据库(MSigDB)的基因集合富集分析(GSEA),显示DEGs与胆固醇生物合成、代谢或平衡密切相关。
综合上述结果,证明conessine可能对甾醇,特别是胆固醇的生物合成或代谢有影响。
实施例6
检测化合物conessine对细胞胆固醇水平的影响。
(1)将A549细胞(市售)以1×105个/孔的密度接种于24孔板中,细胞用含10% FBS(胎牛血清)的高糖型DMEM培养基(加1%双抗)在37℃、5% CO2的培养箱中培养。
(2)待18-24小时后,细胞生长至指数期,分别加入ethanol(对照组)和10μmol/L的conessine,在37℃、5% CO2的培养箱中继续培养。
(3)处理24小时后,收取细胞,按照常规方法,通过胆固醇检测试剂盒测定细胞胆固醇浓度,利用Filipin III对胆固醇染色并通过激光共聚焦拍照。
结果如图6所示,图6-A为细胞胆固醇浓度,显示conessine处理后,细胞胆固醇浓度升高。图6-B为细胞胆固醇染色结果,显示conessine处理后,细胞胆固醇含量增多。
结合实施例5的结果可见,化合物conessine可上调细胞胆固醇水平。
实施例7
检测化合物conessine是否通过上调胆固醇抑制HSV-1复制。
(1)将A549细胞(市售)以2×105个/孔的密度接种于12孔板中,细胞用含10% FBS(胎牛血清)的高糖型DMEM培养基(加1%双抗)在37℃、5% CO2的培养箱中培养。
(2)待18-24小时后,细胞生长至指数期,加入DMSO(对照组mock)或1μg/mLFilipin III(可螯合细胞内胆固醇),在37℃、5% CO2的培养箱中继续培养。
(3)4小时后,加入HSV-1病毒液(MOI 0.1,感染体积100μL),同时分别加入溶剂对照ethanol或10μmol/L的conessine,在37℃、5% CO2的培养箱中继续培养。
(4)加入病毒24小时后,收取细胞及上清,反复冻融3次,按照常规方法进行空斑实验,检测病毒滴度。
结果如图7所示,利用Filipin III螯合胆固醇后,conessine不再具有抗HSV-1复制的效果,证明conessine是通过上调胆固醇抑制HSV-1复制。
实施例8
检测化合物conessine对NF-κB表达及活化、自噬的影响。
(1)将A549细胞(市售)以2×105个/孔的密度接种于12孔板中,细胞用含10% FBS(胎牛血清)的高糖型DMEM培养基(加1%双抗)在37℃、5% CO2的培养箱中培养。
(2)待18-24小时后,细胞生长至指数期,分别加入ethanol(对照组)和10μmol/L的conessine,在37℃、5% CO2的培养箱中继续培养。
(3)处理24小时后,收取细胞蛋白,按照常规方法,进行western blot分析,GAPDH为内参。
结果如图8所示,10μmol/L conessine处理后,NF-κB、p-NF-κB、p62(自噬标志物之一,如有自噬发生则p62减少)水平无明显变化,证明conessine对NF-κB表达及活化、自噬无明显影响。
以上所述实施例的各技术特征可以进行任意的组合,为使描述简洁,未对上述实施例中的各个技术特征所有可能的组合都进行描述,然而,只要这些技术特征的组合不存在矛盾,都应当认为是本说明书记载的范围。
以上所述实施例仅表达了本发明的几种实施方式,其描述较为具体和详细,但并不能因此而理解为对发明专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进,这些都属于本发明的保护范围。因此,本发明专利的保护范围应以所附权利要求为准。
