本发明属于纳米材料和生物医药技术领域,涉及一种无载体自组装药物纳米粒子及其制备方法和应用。本发明提供的无载体自组装药物纳米粒子由焦亡促进剂、光敏剂、IDO‑1抑制剂三种组分自组装而成;其中,焦亡促进剂为辛伐他汀,光敏剂为二氢卟吩e6,IDO‑1抑制剂为NLG919。本发明还提供了制备这种纳米粒子的方法,以及包含这种纳米粒子的药物及纳米粒子在制备肿瘤治疗药物中的应用。本发明提供的纳米粒子具有载药率高、制备简单、无载体的特点,体外细胞实验和动物实验证实,该纳米粒子具有显著的肿瘤治疗效果。

本发明属于纳米材料和生物医药技术领域,涉及一种无载体自组装药物纳米粒子及其制备方法和应用,具体涉及一种由三种药物经自组装形成的无载体自组装药物纳米粒子及其制备方法和应用。
背景技术
光动力治疗(Photodynamic therapy,PDT)是一种药物-器械结合的治疗技术,通过对局部给与光敏剂后,再给予特定波长的光源照射,处于基态的光敏剂吸收光能后转化成高能量的单重态,单重态光敏剂通过发射荧光释放能量回到基态,或者经系间跨越转化成稍稳定的三重态,三重态的光敏剂再释放能量回到基态,而释放出的能量可使周边的分子氧转换成单态氧(II型光动力学治疗);又或者通过氢离子、电子转移生成超氧化物、过氧化物等氧自由基(I型光动力学治疗)。单态氧和氧自由基都是活性氧,具有氧毒性,可氧化损伤周边的细胞器和生物分子,进而激活细胞坏死、凋亡、自噬、血管损伤、免疫激活等涉及活性氧的信号通路。光动力学已经被应用于损伤肿瘤、病毒感染细胞及其他过度增殖细胞、激活抗肿瘤抗病毒免疫、损伤血管,杀伤细菌、真菌、病毒,消除炎症等过程中。其中,光敏剂指的是能够吸收一定波长光源后激活一系列光化学、光物理反应,生成荧光或能杀伤细胞的氧活性物质的一类物质。理想的光敏剂应具备选择性高,在病变/正常组织中具有较高的分布比,在靶组织中分布均匀,照光后光动力反应效率高等特点。
作为一种光动力治疗的光敏剂,二氢卟吩e6的化学结构式如下。
焦亡是一种程序性细胞死亡。生理条件下焦亡通常在先天免疫系统的细胞(例如单核细胞、巨噬细胞和树突状细胞)中被诱导。发生焦亡的细胞显示出诸如细胞肿胀、膜起泡、DNA断裂和最终细胞裂解等特征。但是,细胞核通常保持完整,这与凋亡和坏死性凋亡中观察到的细胞核破坏现象不同。近年来,焦亡机制在肿瘤治疗中应用的研究取得进展。综述文章《Emerging mechanisms of pyroptosis and its therapeutic strategy incancer》(Cell Death Discovery,(2022)8:338)指出,焦亡和免疫反应之间存在正反馈循环,这个过程可能被利用于治疗肿瘤;具体地,焦亡肿瘤细胞释放大量炎性因子,这些炎性因子能够反过来召集免疫细胞并增强系统性免疫应答。该综述文章还列举了多种诱导肿瘤细胞焦亡的因素,包括细菌、病毒、毒素、活性氧和化疗药物如顺铂、姜黄素、5-氟尿嘧啶、辛伐他汀等。
其中,辛伐他汀的化学结构式如下。
吲哚胺-2,3-双加氧酶(IDO)是一种含有血红素的酶,该酶是哺乳动物色氨酸代谢途径中的第一个酶并且是限速酶。IDO能催化必需氨基酸中的色氨酸转化为N-甲酰犬尿氨酸,并且负责清理人体中的色氨酸。而N-甲酰犬尿氨酸进一步代谢则会生成犬尿氨酸。IDO通过降解色氨酸,造成体内色氨酸缺失的微环境,进而导致了癌症、病毒感染、抑郁、器官移植排斥或自身免疫疾病等多种与色氨酸缺失密切相关的疾病的发生。特别地,在肿瘤微环境中,色氨酸耗竭会抑制癌细胞中主要代谢调节因子mTOR和蛋白激酶C,从而分别促进自噬和Treg细胞的形成;而犬尿氨酸会激活芳香烃受体AhR,AhR是一种配体激活的转录因子,对免疫细胞有显著影响,并参与诱导Treg分化,有利于逆转免疫抑制性肿瘤微环境。因此,寻找基于IDO靶点的高效抑制剂已成为近年来肿瘤免疫治疗药物开发的新赛道。综述文章
《IDO1:An important immunotherapy target in cancer treatment》(International Immunopharmacology 47(2017)70-77)指出,针对IDO-1(IDO家族成员之一)的高效抑制剂,如NLG919(1-环己基-2-(5H-咪唑并[5,1-A]异吲哚-5-基)乙醇,cas号为1402836-58-1)等,具有作为肿瘤化疗药物治疗实体瘤和转移实体瘤的前景。
其中,NLG919的化学结构式如下。
迄今为止,大量不同的纳米载体被开发用于将疏水性药物递送至肿瘤部位。但是,大多数载体都面临制备过程繁琐,载体安全性研究不充分,代谢途径不清楚等问题。因此,由纯药物分子通过自组装技术形成的无载体纳米粒子成为纳米材料用于肿瘤治疗领域技术的发展趋势。中国专利申请202110644226.5公开了一种协同化疗/声-光动力治疗的自组装纳米药物,将齐墩果酸和二氢卟吩e6通过π-π堆积和疏水相互作用自组装成无载体纳米药物,可以同时发挥化疗和声-光动力治疗的协同效果。中国专利申请202110913922.1公开了一种协同光动力-免疫联合治疗的无载体自组装纳米药物,该纳米自组装药物由光敏剂以及咪唑并喹啉类Toll样受体激动剂两种药物分子构成,该纳米自组装药物制备方法简单,同时解决了两个疏水性药物分子水溶性差,生物利用度低的问题。
然而,现有技术中所涉及无载体自组装纳米体系,均无法实现光动力-促进肿瘤细胞焦亡-抑制IDO-1提升免疫治疗的协同肿瘤治疗的目的。
发明内容
有鉴于此,针对现有技术中,无载体自组装纳米粒子无法实现光动力-促进肿瘤细胞焦亡-基于IDO-1抑制剂敏化的免疫治疗的协同肿瘤治疗的问题,本发明的目的是提出一种无载体自组装药物纳米粒子,以及这种纳米粒子的制备方法和应用。此纳米粒子集光动力学治疗、促进肿瘤细胞焦亡的肿瘤治疗、抑制IDO-1提升免疫治疗的功能于一身,具有制备方法简单无需载体、纳米粒子尺寸可控、在溶液中稳定性好等优点,克服了单药水溶性低带来的问题;体外细胞实验和动物实验检测表明,此纳米粒子杀灭肿瘤细胞的效果显著优于未经自组装的三种药物同时使用的效果,且转移瘤动物实验表明,该纳米颗粒能够显著抑制实体转移肿瘤。
为了实现本发明目的,采用的技术方案如下:
本发明提供一种无载体自组装纳米粒子,由以下三种组分自组装而成:
焦亡促进剂,光敏剂,IDO-1抑制剂;所述凋亡促进剂为辛伐他汀、所述光敏剂为二氢卟吩e6、所述IDO抑制剂为NLG919。
其中,所述自组装过程的驱动力,包括疏水相互作用,静电相互作用中的至少一种。
优选地,所述自组装过程的驱动力为疏水相互作用和静电相互作用。
优选地,所述辛伐他汀、二氢卟吩e6、NLG919的摩尔比为1-10:1-10:1-10。
再优选地,所述辛伐他汀、二氢卟吩e6、NLG919的摩尔比为1-6.75:1-2.65:1-10。
更优选地,且作为本发明的一个具体实施例,所述辛伐他汀、二氢卟吩e6、NLG919的摩尔比为1.43:1.41:1。
另一方面,本发明还提供上述纳米粒子的制备方法,包括以下步骤:
S1:取焦亡促进剂,光敏剂,IDO-1抑制剂分别溶于溶剂中,制成A溶液、B溶液和C溶液;
S2:取步骤S1制得的A溶液加水超声,得混合液1;
S3:取步骤S1制得的B溶液加入步骤S2制得的混合液1中,加水超声吹打,得混合液2;
S4:取步骤S1制得的C溶液加入步骤S3制得的混合液2中超声,得混合液3;
S5:将步骤S4中制得的混合液3透析,得纳米粒子。
其中,步骤S1中所述溶剂为有机溶剂,包括但不限于DMSO、DMF、THF、离子液体,等。
优选地,所述溶剂为DMSO。
其中,步骤S1中所述A溶液、B溶液、C溶液浓度为1-50mg/mL。
更优选地,所述A溶液、B溶液、C溶液的浓度均为10mg/mL。
更优选地,所述步骤S1具体如下:
取辛伐他汀、二氢卟吩e6、NLG919分别溶于DMSO,形成浓度为8-12mg/mL的A溶液、B溶液、C溶液。
进一步优选地,且作为本发明的一个具体实施例,所述步骤S1具体如下:
取辛伐他汀、二氢卟吩e6、NLG919分别溶于DMSO,形成浓度为10mg/mL的A溶液、B溶液、C溶液。
更优选地,步骤S2具体为:取步骤S1制得的45-55μL的A溶液,加入0.8-1.1mL水,超声1-3min,得混合液1。
再优选地,且作为本发明的一个具体实施例,步骤S2具体为:取步骤S1制得的50μL的A溶液,加入1mL水,超声2min,得混合液1。
更优选地,步骤S3具体为:取步骤S2制得的20-40μL的B溶液加入步骤S2制得的混合液1中,超声1-3min,再加入0.8-1.2mL水,超声吹打1.5-3min,得混合液2。
再优选地,且作为本发明的一个具体实施例,步骤S3具体为:取步骤S2制得的30μL的B溶液加入步骤S2制得的混合液1中,超声2min,再加入1mL水,超声吹打2min,得混合液2。
更优选地,步骤S4具体为:取步骤S3制得的40-60μL的C溶液加入步骤S2制得的混合液2中,超声6-8min,得混合液3。
再优选地,且作为本发明的一个具体实施例,步骤S4具体为:取步骤S3制得的50μL的C溶液加入步骤S2制得的混合液2中,超声7min,得混合液3。
更优选地,步骤S5中所述透析,使用的透析袋孔径为800-1500Da,透析时间为1-5h。
再优选地,且作为本发明的一个具体实施例,步骤S5中所述透析,使用的透析袋孔径为1000Da,透析时间为2h。
上述制备方法中,使用的水包括但不限于蒸馏水、去离子水、超纯水、净化水、纯化水、无菌水等。
再一方面,本发明还提供了一种肿瘤治疗药物,有效成分包括上述纳米粒子和/或上述制备方法制得的纳米粒子。
所述药物可制备成粉剂、栓剂、注射剂、乳剂、贴剂、喷雾剂、气溶胶剂等给药剂型,针对不同的剂型可选择本领域合适的药物载体。
所使用的药物载体可以是固体、液体或气体。固体载体示例包括乳糖、白陶土、蔗糖、滑石粉、明胶、琼脂、果胶、阿拉伯胶、硬脂酸镁和硬脂酸。液体载体的例子包括糖浆、花生油、橄榄油和水。气体载体的例子包括二氧化碳和氮气。
在制备口服剂型的药物时,可以使用任何方便的药物介质。例如,水、乙醇、油、醇、调味剂、防腐剂、着色剂等可用于形成口服液体制剂,如混悬剂、剂和溶液;而载体,如淀粉、糖类、微晶纤维素、稀释剂、制粒剂、乳化剂、润滑剂、粘合剂、崩解剂可用于形成口服固体制剂,如粉剂、胶囊剂和片剂。由于其易于给药,片剂和胶囊是使用固体药物载体的优选口服剂量单位。可选择使用标准水性或非水性技术对片剂进行包衣。
含有本发明纳米粒子的片剂可以通过压片或模塑制备,可选择使用一种或多种辅助成分或佐剂。可通过在适当的机器中以自由流动的形式(如粉末或颗粒)压片活性成分,可选择与粘合剂、润滑剂、惰性稀释剂、表面活性物质或分散剂混合来制备压片。模制片剂可在适当的机器中模制,即用惰性液体稀释剂润湿的粉末状纳米粒子。每片优选含有约0.05mg至约5g纳米粒子,每个小袋或胶囊优选含有约0.05mg至约5g纳米粒子。例如,拟用于人体口服给药的制剂可能含有约0.5mg至约5g纳米粒子,与适量且方便的载体材料混合,其可能约占总组成的5%至95%。单位剂型通常含有约1mg至约2g纳米粒子,通常为25mg、50mg、100mg、200mg、300mg、400mg、500mg、600mg、800mg或1000mg。
本发明中可制备为包括纳米粒子的水溶液或混悬液,以便胃肠外给药。可以包括适当的表面活性剂,例如羟丙基纤维素。也可以在甘油、液体聚乙二醇和其油混合物中制备分散体。此外,可以加入防腐剂以防止微生物的有害生长。
本发明中适用于注射使用的药物包括无菌水溶液或分散体。此外,该药物可以无菌粉末的形式,用于临时制备此类无菌注射液或分散体。在所有情况下,最终注射形式必须是无菌的,并且必须是有效的液体,以便于可注射性药物成分必须在生产和储存条件下保持稳定;因此,最好应保存,以防止微生物(如细菌和真菌)的污染作用。载体可以是溶剂或分散介质,例如含有水、乙醇、多元醇(例如甘油、丙二醇和液体聚乙二醇)、植物油及其合适的混合物。
本发明的药物可以是适合局部使用的形式,例如气雾剂、乳膏、软膏、洗剂、粉剂或类似物。此外,纳米粒子可以是适当的形式用于透皮给药装置。可以使用本发明的纳米粒子,通过常规处理方法制备这些处方。例如,通过混合亲水性材料和水,以及约5wt%至约10wt%的纳米粒子,制备具有所需稠度的乳膏或软膏。
本发明的药物可以是适用于直肠给药的形式,其中载体是固体。最好将混合物制成单位剂量栓剂。合适的载体包括可可脂和其他本领域中常用的材料。栓剂可通过首先形成混合含有软化或熔化载体的组合物,随后在模具中冷却和塑形。
除上述载体成分外,上述药物制剂可能包括(如适用)一种或多种额外的载体成分,如稀释剂、缓冲液、矫味剂、粘合剂、表面活性剂、增稠剂、润滑剂、防腐剂(包括抗氧化剂)等。此外,可加入其它辅料,例如乳糖、淀粉、纤维素衍生物、硬脂酸镁、硬脂酸等、着色剂和矫味剂等。使制剂与预期受体的血液等渗。还可以粉末或浓缩液形式制备含有本发明纳米粒子的组分。
另一方面,本发明还提供了上述纳米粒子、上述制备方法制备的纳米粒子在制备肿瘤治疗药物中的应用。
其中,所述肿瘤治疗包括但不限于光动力治疗、免疫治疗、细胞焦亡治疗、代谢调节治疗,等。
优选地,所述肿瘤治疗包括光动力治疗、免疫治疗、细胞焦亡治疗、代谢调节治疗中的至少一种。
再优选地,所述肿瘤治疗为光动力治疗、免疫治疗和细胞焦亡治疗。
与现有技术相比,本发明的有益效果:
(1)实现了焦亡促进剂、光敏剂和IDO-1抑制剂三种物质自组装形成纳米粒子,该纳米粒子克服了单药水溶性低的缺点,具有载药率高、制备简单、无载体的特点,且实现了光动力-促进肿瘤细胞焦亡-抑制IDO-1提升免疫治疗的协同肿瘤治疗的目的。
(2)荧光成像试验证实,本发明所得纳米粒子能够有效在肿瘤区域富集;体外细胞试验和动物试验证实,该纳米粒子对肿瘤细胞具有很强的杀灭作用,能够有效抑制肿瘤生长。
(3)转移性肿瘤治疗的动物试验证实,本发明所得纳米粒子能够实现焦亡诱导的肿瘤治疗与抑制IDO-1的增强免疫治疗的结合,实现对转移性肿瘤的有效治疗。
(4)本发明所得纳米粒子无显著暗毒性,具有良好的应用前景。

实施例1
制备一种无载体自组装纳米粒子,此纳米粒子由二氢卟吩e6(Ce6)、辛伐他汀(Simvastatin)、NLG919组成,下述将由这三种药物自组装而成的纳米粒子称为CNS纳米粒子。合成的具体过程如下。
取一定量的二氢卟吩e6、辛伐他汀、NLG919分别溶于DMSO中,制得10mg/mL二氢卟吩e6的DMSO溶液、10mg/mL辛伐他汀的DMSO溶液、10mg/mL的NLG919的DMSO溶液。取50μL上述辛伐他汀溶液置于塑料离心管中,加入1mL超纯水,超声2min;接着加入30μL上述二氢卟吩e6溶液,超声2min,再加入1mL超纯水,超声吹打2min;接着加入50μL上述NLG919溶液,超声7min。上述溶液放入透析袋中,透析袋的孔径为1000Da,透析2小时,得到材料CNS纳米粒子。
该纳米粒子中,辛伐他汀、二氢卟吩e6、NLG919的摩尔比为1.43:1.41:1。
采用透射电子显微镜对实施例1制备的无载体自组装纳米粒子进行表征,结果为图1。观察可知,上述方法成功合成了粒径均一度高的球状纳米粒子。
对比例1
制备一种无载体自组装纳米粒子,具体过程与实施例1中的制备过程相比,使用辛伐他汀和NLG919的体积改为30μL,其余皆相同。
对比例2
制备一种无载体自组装纳米粒子,具体过程与实施例1中的制备过程相比,使用辛伐他汀的体积改为30μL,其余皆相同。
对比例3
制备一种无载体自组装纳米粒子,具体过程与实施例1中的制备过程相比,使用NLG919的体积改为30μL,其余皆相同。
采用动态光散射对实施例1、对比例1-3中合成的纳米粒子的水合粒径大小进行了表征,结果为图2。其中,纳米粒子的平均粒径±标准差(nm)、多分散指数(PDI)和是否单分散纳米粒子的结果汇总于下表。

观察图2可知,对比例1-3采用的不同的三种药物用量配比,合成的纳米粒子产物中出现部分粒径过大的团聚物,多分散指数变大且非单分散纳米颗粒。而利用实施例1中采用的三种药物用量配比进行合成,可以得到单分散纳米颗粒,平均粒径253.6nm。
进一步地,对实施例1中制备的CNS纳米粒子的水溶液稳定性进行测试,测定了在水溶液中,实施例1中制备的CNS纳米粒子在7天内的平均粒径和多分散指数,结果如图3。观察图3可见,实施例1中制备的CNS纳米粒子在水溶液中7天内平均粒径和多分散指数没有发生大幅提升,证明实施例1制得的CNS纳米粒子具备良好的水溶液稳定性。
为了证实CNS纳米粒子的分子组装机制,进行如下测试。
检测了二氢卟吩e6、CNS纳米粒子的水溶液在300-800nm范围内的紫外可见吸收光谱。由图4左图可知,二氢卟吩e6和CNS纳米粒子的水溶液的吸收峰位置存在差异,说明CNS纳米粒子中疏水相互作用参与了自组装过程。进一步地,检测了0.2%十二烷基磺酸钠水溶液(0.2%SDS)、CNS纳米粒子水溶液、CNS纳米粒子和0.2%十二烷基磺酸钠混合水溶液(CNS+0.2%SDS)的在300-800nm范围内的紫外可见吸收光谱,结果为图4中图。SDS存在下,表征峰发生明显差异,说明CNS的纳米结构中存在疏水相互作用。更进一步地,将CNS纳米粒子溶于水和梯度浓度(0.5M、1M、2M)的氯化钠溶液中,分别测定溶液在300-800nm范围内的紫外可见吸收光谱,其结果为图4右图。当氯化钠浓度为2M时,光谱发生了明显的变化,说明静电作用是CNS纳米粒子自组装的主要驱动力之一。综上所述,可以推断出CNS纳米粒子是由二氢卟吩e6、NLG919和辛伐他汀通过分子间的疏水相互作用和静电相互作用自组装而成的稳定、均匀的无载体纳米粒子。
试验例1
纳米粒子用于杀灭体外肿瘤细胞的试验,具体过程如下。
1.细胞培养
4T1细胞(购自中国科学院细胞库),培养条件依据为美国模式培养物保藏所(ATCC)提供的4T1细胞培养手册,具体如下。
细胞培养箱:温度为37℃,气氛为95%空气、5%二氧化碳。培养基:RPMI-1640培养基,含10%浓度的胎牛血清。传代密度:贴壁生长的4T1细胞,生长密度达到80%时进行传代。传代比例:1比6至1比8。传代周期:每2-3日传代一次。
2.MTT试验
将4T1细胞消化、离心、分散、种植至96孔板上,每孔100μL溶液、内含约5000个4T1细胞,培养24小时。移除孔板中培养基,添加含有梯度浓度的各组药物RPMI-1640溶液各100μL,与各组药物共培养6小时(Ce6、CNS组以二氢卟吩e6的质量浓度计;NLG和Sim以各自的质量浓度计;C+N+S组以Ce6浓度计,且其中二氢卟吩e6、NLG919、辛伐他汀的摩尔比例与CNS组相同)。采用标准MTT比色法进行处理,对570nm波长处吸收进行检测。
将实施例1对应的纳米颗粒采用上述方法进行检测,结果为图5中A。该实验结果表明,在体外CNS纳米粒子对4T1肿瘤细胞几乎没有暗毒性。
进一步地,为了证实CNS纳米粒子具有优秀的光动力治疗效果,将4T1细胞消化、离心、分散、种植至96孔板上,每孔100μL溶液、内含约5000个4T1细胞,培养24小时。移除孔板中培养基,添加含有梯度浓度的各组药物的RPMI-1640溶液各100μL,与各组药物共培养6小时后光照,波长为630nm,光源能量密度为29.8mW·cm-2,光照时长为5分钟。共培养24小时后,采用标准MTT比色法进行处理。对570nm波长处吸收进行检测,结果为图5中的B。进一步地,重复上述试验3次,对Ce6(+)、C+N+S(+)、CNS(+)组别的IC50进行计算,结果为图5中的C。该结果表明,CNS纳米粒子在光照下对体外肿瘤细胞的杀灭效果,显著优于等量三种药物叠加下光照对体外肿瘤细胞的杀灭效果。
3.活/死双染试验
将1000μL、每1μL中含细胞量10个的处于对数期生长的4T1细胞种植在12.56cm2的培养皿上,培养24小时,分别加入RPMI-1640培养基(Blank组)、浓度为2mg/L的二氢卟吩e6(Ce6组、Ce6(+)组)、浓度为1.8mg/L的NLG919(NLG组)、浓度为1.3mg/L的辛伐他汀(Sim组)、浓度为5.1mg/L的CNS纳米粒子(CNS组、CNS(+)组)、三种药物浓度分别为2mg/L,1.8mg/L,1.3mg/L的二氢卟吩e6+NLG919+辛伐他汀组(C+N+S组、C+N+S(+)组),给药体积为1mL每组。培养6h后,对Ce6(+)组、CNS(+)组、C+N+S(+)组进行光照,光照波长为630nm,光源能量密度为29.8mW·cm-2,光照时长为15分钟。移除各组中含有药物的培养基,使用RPMI-1640培养基清洗细胞。使用Calcein-AM/PI活/死双染试剂盒进行染色,使用激光共聚焦扫描显微镜观察。试验结果为图5中的D。进一步地,统计红色荧光PI强度,总结为图5中的E。显著性差异:***表示p<0.001,计算显著性差异所使用的统计方法为采用双尾Student t检验分析两组间差异的统计学方法。上述试验结果表明,与单纯添加二氢卟吩同时光照和等量三种药物叠加同时光照的组别相比,CNS纳米粒子同时光照组产生了更强的红色荧光,这同样说明了CNS纳米粒子对体外培养肿瘤细胞具有显著增强的光动力治疗效果。
试验例2
CNS纳米粒子用于引发体外肿瘤细胞焦亡的试验,具体过程如下。
1.细胞培养
与试验例1中细胞培养部分相同。
2.Cas-1和c-Cas-1检测试验
试验方法
4T1细胞在培养皿中培养24h。随后,将细胞与Ce6、NLG、Sim、C+N+S、CNS材料孵育10小时,光照组光照5min,其他组的细胞在黑暗中培养。实验分组为Blank组、Ce6暗处理组、NLG组、Sim组、Ce6光照组、C+N+S暗处理组、C+N+S光照组、CNS暗处理组和CNS光照组。等效浓度为:Ce6:4.5μg/mL;NLG:4.125μg/mL;Sim:3μg/mL。然后,将所有组别的细胞再培养4h,用PBS洗涤细胞随后,用4%多聚甲醛固定细胞15min,用0.1%Triton-100通透细胞20min。用山羊血清封闭后,用抗cas-1和c-Cas-1抗体孵育细胞在4℃下过夜。PBS洗涤后,分别用山羊抗兔IgG H&L和DAPI染色1h和15min。最后,采用激光共聚焦扫描显微镜进行细胞免疫荧光观察和分析。
试验结果为图6中A。
通过免疫荧光实验检测细胞中焦亡相关蛋白Cas-1和c-Cas-1的激活情况,在激光共聚焦扫描显微镜(激光共聚焦扫描显微镜)成像中可观察到Sim,C+N+S,CNS,Ce6(+),C+N+S(+)和CNS(+)组有明显的绿色荧光,说明PDT和Sim能够上调Cas-1和c-Cas-1的表达,此外,分别对绿色荧光进行定量分析,结果为图6中B和C。显著性差异:*表示P<0.05,**表示P<0.01,***表示P<0.001,计算显著性差异所使用的统计方法为采用双尾Student-t检验分析两组间差异的统计学方法。试验结果显示,CNS(+)组别的绿色荧光最强,这也说明了形成纳米药物之后促进了细胞内吞从而增强了PDT和Sim诱导的焦亡。
3.GSDMD-N、c-Cas-1的免疫印记分析(Western blot)
试验方法
将4T1细胞接种于6孔板中培养24h。随后,将细胞与Ce6、NLG、Sim、C+N+S、CNS材料孵育12小时,光照组光照5min,其他组的细胞在黑暗中培养。实验分组为Blank组、Ce6暗处理组、NLG组、Sim组、Ce6光照组、C+N+S光照组、C+N+S暗处理组、CNS光照组和CNS暗处理组。等效浓度为Ce6(4.5μg/mL)、NLG(4.125μg/mL)、Sim(3μg/mL)。然后,将所有组别的细胞再培养4h,用PBS洗涤细胞,用RIPA裂解缓冲液处理细胞,提取蛋白用于免疫印迹检测,分析c-Cas-1和GSDMD-N的表达。
免疫印记分析结果为图7中的A,各组GSDMD-N、c-Cas-1的表达量灰度值分析总结为图7中的B和C。显著性差异:*表示P<0.05,计算显著性差异所使用的统计方法为采用双尾Student-t检验分析两组间差异的统计学方法。可见,在CNS(+)组中,观察到GSDMD-N和c-Cas-1的表达增强,这些实验结果表明,CNS纳米粒子通过光动力和辛伐他汀的双重作用,有效诱导了体外培养的肿瘤细胞的焦亡过程。
上述试验说明:光动力学治疗产生的ROS和药物Sim可进一步激活核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)炎症小体和caspase-1蛋白,从而导致GSDMD的裂解,释放GSDMD-N结构域。GSDME-N易位到细胞膜上并形成膜孔,从而驱动细胞肿胀、膜破裂和DAMPs的释放,从而导致焦亡诱发的免疫反应。
试验例3
CNS纳米粒子引发体外肿瘤细胞发生免疫原性细胞死亡(ICD)的试验,具体过程如下。
1.细胞培养
与试验例1中细胞培养部分相同。
2.HMGB1和CRT检测试验
试验方法
HMGB1:4T1细胞在培养皿中培养24h。随后,将细胞与Ce6、NLG、Sim、C+N+S、CNS材料孵育6小时,光照组光照5min,其他组的细胞在黑暗中培养。实验分组为Blank组、Ce6光照组、NLG组、Sim组、Ce6暗处理组、C+N+S光照组、C+N+S暗处理组、CNS光照组和CNS暗处理组。等效浓度为Ce6浓度为:5μg/mL;NLG:4.6μg/mL;Sim:
3.3μg/mL。然后,将所有组别的细胞再培养4h,用PBS洗涤细胞随后,用4%多聚甲醛固定细胞15min,用0.1%Triton-100通透细胞10min。用山羊血清封闭后,用抗HMGB1抗体孵育细胞在4℃下过夜。PBS洗涤后,分别用山羊抗兔IgG H&L和DAPI染色1h和15min。最后,采用激光共聚焦扫描显微镜进行细胞免疫荧光观察和分析。
CRT:4T1细胞在培养皿中培养24h。随后,将细胞与Ce6、NLG、Sim、C+N+S、CNS材料孵育6小时,光照组光照8min,其他组的细胞在黑暗中培养。实验分组为Blank组、Ce6暗处理组、NLG组、Sim组、Ce6光照组、C+N+S光照组、C+N+S暗处理组、CNS光照组和CNS暗处理组。等效浓度为:Ce6:5μg/mL;NLG:4.6μg/mL;Sim:3.3μg/mL。然后,将所有组别的细胞再培养4h,用PBS洗涤细胞随后,用4%多聚甲醛固定细胞15min。用山羊血清封闭后,用抗CRT抗体孵育细胞在4℃下过夜。PBS洗涤后,分别用山羊抗兔IgG H&L和DAPI染色1h和15min。最后,采用激光共聚焦扫描显微镜进行细胞免疫荧光观察和分析。
通过免疫荧光实验检测CRT和HMGB1的表达,使用激光共聚焦扫描显微镜检测CRT和HMGB1的亚细胞定位。我们发现经过CNS(+)处理后,CRT暴露于死亡肿瘤细胞的细胞膜中,HMGB1从细胞核中释放到细胞外基质,从而增强抗肿瘤免疫反应,检测结果为图8中的A,CRT和HMGB1绿色荧光强度定量分析结果分别为图8中的B、C。显著性差异:*表示P<0.05,**表示P<0.01,***表示P<0.001,计算显著性差异所使用的统计方法为采用双尾Student t检验分析两组间差异的统计学方法。
PDT是基于在光敏剂的光激活下产生细胞毒性ROS来诱导细胞焦亡和组织破坏的原理。焦亡细胞焦亡是一种新识别的ICD形式,由具有n端结构域和c端抑制结构域的gasdermin家族蛋白介导,与质膜孔形成、细胞肿胀和炎症反应诱导的多种DAMPs释放有关。焦亡细胞促进DAMPs释放,如内质网应激后CRT易位到死亡细胞表面,可以促进巨噬细胞和未成熟的树突状细胞被吞噬到死亡的肿瘤细胞及其碎片中,以及HMGB1向细胞外释放,以赋予肿瘤细胞高免疫原性。
3.HMGB1、CRT的免疫印记分析(Western blot)
试验方法
将4T1细胞接种6孔板中培养24h。随后,将细胞与Ce6、NLG、Sim、C+N+S、CNS材料孵育6小时,光照组光照7min,其他组的细胞在黑暗中培养。实验分组为Blank组、Ce6暗处理组、NLG组、Sim组、Ce6光照组、C+N+S光照组、C+N+S暗处理组、CNS暗处理组和CNS光照组。等效浓度为:Ce6(5μg/mL)、NLG(4.6μg/mL),Sim(3.3μg/mL)。然后,将所有组别的细胞再培养4h,用PBS洗涤细胞,用RIPA裂解缓冲液处理细胞,提取蛋白用于免疫印迹检测,分析CRT和HMGB1的表达。
免疫印记分析结果为图9中的A,各组CRT、HMGB1的表达量灰度值分析总结为图9中的B、C。显著性差异:*表示P<0.05,**表示P<0.01,***表示P<0.001,计算显著性差异所使用的统计方法为采用双尾Student t检验分析两组间差异的统计学方法。可见,CNS纳米粒子能够有效降低细胞表面HMGB1的量,升高CRT的表达。
上述试验表明:CNS纳米粒子能够通过诱导细胞焦亡引起强烈的ICD,有利于进一步启动免疫应答。
试验例4
评估CNS纳米粒子在活体动物乳腺癌肿瘤模型中的抗肿瘤效果,具体过程如下。
1.动物模型建立和分组
周龄6-7周的Balb/c雌性小鼠,购自北京斯贝福。向小鼠的背部皮下注射106个4T1乳腺癌细胞,同时尾静脉注射1×106个4T1细胞,构建乳腺癌的肺转移瘤模型。当皮下种植的异质瘤体积约为100mm3时,建模完成。
将所有建模完成的小鼠随机分组,每组小鼠数目为5只。7组小鼠分别命名为空白对照组(Blank),二氢卟吩e6组(Ce6),辛伐他汀组(Sim),NLG919组(NLG),CNS纳米粒子组(CNS),二氢卟吩e6光动力治疗组(Ce6(+)),CNS纳米粒子光动力治疗组(CNS(+))。
2.治疗试验过程
向分组后小鼠尾静脉中注射溶液,不同组注射溶液不同,具体如下。
Blank:500μL生理盐水。
Ce6、Ce6(+):500μL浓度为1.5mg/kg的二氢卟吩e6生理盐水溶液。
Sim:500μL浓度为1mg/kg的辛伐他汀生理盐水溶液。
NLG:500μL浓度为1.375mg/kg的NLG919生理盐水溶液。
CNS、CNS(+):500μL浓度为3.875mg/kg(以二氢卟吩e6计)的CNS纳米粒子生理盐水溶液。
光动力治疗组(包括Ce6(+)组和CNS(+)组)照光过程为:每次给药8h后,对光照组以638nm、0.68Wcm-2的功率照射10分钟。
分别在尾静脉注射药物后的第1、3、5、7、9、11、13、15、17、19、21、23天对小鼠异质瘤体积和小鼠体重进行测量。在尾静脉注射药物后的第23天,处死各组试验小鼠,取小鼠的各主要器官、肿瘤组织和血液进行检测分析。
3.试验结果
3.1针对异质瘤模型的抑瘤试验
尾静脉注射药物后,第1-23天隔日测量异质瘤体积,结果汇总为图10中的A。纳米粒子组尾静脉注射药物后,第1-23天隔日测量小鼠体重,结果汇总为图10中的D。该结果说明,CNS纳米粒子光动力治疗组(CNS(+)组)小鼠体重既未因纳米粒子的毒性明显减轻,又未因肿瘤治疗效果不好导致的恶病质明显减轻。治疗23天后,处死各组小鼠,分别取小鼠异质瘤组织和脾脏进行分析。小鼠异质瘤组织照片为图10中的B,异质瘤组织重量汇总为图10中的C,可以看出,CNS(+)组小鼠相较二氢卟吩e6光动力治疗组(Ce6(+)组)治疗效果明显更好。小鼠脾脏照片为图10中的E,脾脏重量汇总为图10中的F,可以看出在其他组中脾脏肥大现象,这表明脾脏随着肿瘤的发展呈负的免疫调节。肿瘤可通过分泌神经营养因子诱导脾组织红细胞大量积累,促进肿瘤进展。而CNS光照组介导的免疫治疗被发现可使脾脏大小正常化,这可能是由于脾脏组织中红细胞的聚集减少所致。有利于免疫激活,从而根除肿瘤。
进一步地,将空白对照(Blank)、在黑暗状态下二氢卟吩e6(Ce6)、NLG919(NLG)、辛伐他汀(Sim)、CNS纳米粒子(CNS)、光照条件下二氢卟吩e6(Ce6(+))、光照条件下CNS纳米粒子(CNS(+))组,治疗23天后,取肿瘤区域进行切片和组织染色(H&E染色、Ki67染色和TUNEL染色),结果汇总为图11。由该结果可以看出,经CNS光动力免疫治疗后,肿瘤组织严重受损,其Ki67最弱而Tunel信号最强,说明细胞增殖最小,凋亡最大。综上所述,自传递CNS可以通过PDT和免疫治疗选择性地积累在肿瘤部位,消除肿瘤细胞。
3.2免疫细胞分析
为了研究体内免疫效应,我们将空白对照(Blank)、在黑暗状态下二氢卟吩e6(Ce6)、NLG919(NLG)、辛伐他汀(Sim)、CNS纳米粒子(CNS)、光照条件下二氢卟吩e6(Ce6(+))、光照条件下CNS纳米粒子(CNS(+))组中,经过不同药物治疗23天后的小鼠处死后,提取肿瘤细胞和脾脏细胞,并采用流式细胞术进行免疫细胞分析,结果汇总于图12。通过观察可以发现,Sim,CNS,Ce6(+)和CNS(+)组脾细胞中辅助性T淋巴细胞(CD3+CD4+)(图12中的A)和细胞毒性T细胞(CD3+CD8+)(图12中的B)与Blank,Ce6和NLG组相比明显增加,CNS(+)增加最显著。CNS(+)组治疗后,肿瘤细胞中的细胞毒性T细胞(CD3+CD8+)显著扩增(图12中的C),这说明引起了强大的免疫应答。此外,由于NLG相关的U-1抑制,CNS(+)也大大降低了Treg的百分比(图12中的D)。但游离NLG的作用不大,这可能是由于其生物利用度较差所致。我们进一步检测了肿瘤组织中的CD11c+CD80+(图12中的E)和CD11c+CD86+树突状细胞(图12中的F)。结果CNS(+)大大增加了树突状细胞。可以推测CNS通过诱导焦亡激活了ICD级联反应,并释放了肿瘤相关抗原,激活了抗肿瘤免疫,联合IDO抑制改善免疫抑制肿瘤微环境,促进抗肿瘤免疫治疗。
3.3针对乳腺癌转移模型的抑瘤试验
对空白对照(Blank)、在黑暗状态下二氢卟吩e6(Ce6)、NLG919(NLG)、辛伐他汀(Sim)、CNS纳米粒子(CNS)、光照条件下二氢卟吩e6(Ce6(+))、光照条件下CNS纳米粒子(CNS(+))组中,针对小鼠肺转移模型治疗23天后,取小鼠肺脏观察转移瘤发展情况并拍照,结果如图13中的A。观察发现,空白组肺部存在大量转移瘤病灶,各治疗组转移情况相对轻,经光动力治疗的CNS纳米粒子组转移灶最少,显示出相当大的抗转移作用。
取小鼠心脏、肝脏、肾脏、脾脏、肺脏组织进行切片染色。
其中,小鼠脾脏、肺脏组织切片的H&E染色图如图13中的B所示。该结果显示,空白组、Ce6,NLG,Sim,CNS以及Ce6(+)组脾脏组织红髓增加,白髓减少,红白髓界限清晰。经光照后,CNS处理的小鼠红髓与白髓的比例表现出比Ce6处理的小鼠高得多,显示了诱导焦亡和增强免疫治疗组合的绝对优势。
其中,小鼠心脏、肝脏、肾脏组织切片的H&E染色图如图14中的A所示。经观察可以发现:Ce6,NLG,Sim,CNS以及Ce6的光照均不能阻止肿瘤向肝脏转移,相比之下,光照射下的CNS通过光动力免疫治疗成功地抑制了肝转移。这些结果表明诱导焦亡,可有效激活抗肿瘤免疫,与IDO-1抑制剂联用抑制肿瘤转移。
3.4生物安全性分析
空白对照(Blank)、在黑暗状态下二氢卟吩e6(Ce6)、NLG919(NLG)、辛伐他汀(Sim)、CNS纳米粒子(CNS)、光照条件下二氢卟吩e6(Ce6(+))、光照条件下CNS纳米粒子(CNS(+))组中,针对小鼠肺转移模型治疗23天后,提取各组中小鼠血样进行生化分析,结果汇总于图14中的B、C、D、E,可知,各组间均无显著性差异,生化指标基本保持在正常范围内。这些结果表明,静脉注射后的治疗药物的系统毒性可以忽略不计。
实验结果小结
通过考察无载体纳米药物CNS在体内的治疗疗效,建立了原位瘤和肺转移模型对纳米粒CNS进行了疗效评估。各组的肿瘤生长抑制及肿瘤组织切片H&E染色及Tunel、ki67荧光切片结果显示,CNS光照组作为一种有效的联合方式实现了最好的协同抗肿瘤效应。在整个周期小鼠体重没有明显的变化验证了纳米药物CNS的初步安全性。治疗结束,主要器官病理切片和生化检测提示CNS是没有明显毒性的。因此制备的纯药物无载体自主装纳米药物CNS被证明是安全有效的,能够以协同的方式治疗乳腺癌。为了研究体内免疫效应,提取肿瘤细胞和脾脏细胞,并采用流式细胞术进行免疫细胞分析。我们发现,CNS光照组脾细胞中辅助性T淋巴细胞(CD3+CD4+)和细胞毒性T细胞(CD3+CD8+)增加显著。并且肿瘤细胞中的细胞毒性T细胞(CD3+CD8+)显著扩增,这说明引起了强大的免疫应答。此外,由于NLG相关的IDO抑制,CNS光照组也大大降低了Treg的百分比。但游离NLG的作用不大,这可能是由于其生物利用度较差所致。我们进一步检测了肿瘤组织中的CD11c+CD80+和CD11c+CD86+树突状细胞,结果显示CNS光照组大大增加了树突状细胞。可以推测,CNS通过诱导焦亡激活了ICD级联反应,并释放了肿瘤相关抗原,唤起了抗肿瘤免疫,联合IDO抑制改善免疫抑制肿瘤微环境,促进抗肿瘤免疫治疗。而肺组织照片和H&E切片结果,CNS的光照组小鼠具有最少的肺转移结节,表明CNS激活的免疫疗法有效的抑制肿瘤细胞的转移。在免疫细胞分析以及肿瘤组织中c-Cas-1免疫荧光和CD3/CD8免疫荧光切片的结果也验证了CNS光照组诱发焦亡和免疫的效果最佳,实现了抗肿瘤疗效的提升。

Claims (12)
1.一种无载体自组装纳米粒子,其特征在于,由以下三种组分自组装而成:
焦亡促进剂,光敏剂,IDO-1抑制剂;其中,所述焦亡促进剂为辛伐他汀,所述光敏剂为二氢卟吩e6,所述IDO-1抑制剂为NLG919;
所述辛伐他汀、二氢卟吩e6、NLG919的摩尔比为1.43:1.41:1;
所述纳米粒子的制备方法包括以下步骤:
S1:取焦亡促进剂,光敏剂,IDO-1抑制剂分别溶于溶剂中,制成A溶液、B溶液和C溶液;
S2:取步骤S1制得的A溶液加水超声,得混合液1;
S3:取步骤S1制得的B溶液加入步骤S2制得的混合液1中,加水超声吹打,得混合液2;
S4:取步骤S1制得的C溶液加入步骤S3制得的混合液2中超声,得混合液3;
S5:将步骤S4中制得的混合液3透析,得纳米粒子。
2.根据权利要求1所述的纳米粒子,其特征在于,所述自组装的驱动力,包括疏水相互作用、静电相互作用中的至少一种。
3.根据权利要求2所述的纳米粒子,其特征在于,所述自组装的驱动力为疏水相互作用和静电相互作用。
4.权利要求1-3中任意一项所述纳米粒子的制备方法,其特征在于,包括以下步骤:
S1:取焦亡促进剂,光敏剂,IDO-1抑制剂分别溶于溶剂中,制成A溶液、B溶液和C溶液;
S2:取步骤S1制得的A溶液加水超声,得混合液1;
S3:取步骤S1制得的B溶液加入步骤S2制得的混合液1中,加水超声吹打,得混合液2;
S4:取步骤S1制得的C溶液加入步骤S3制得的混合液2中超声,得混合液3;
S5:将步骤S4中制得的混合液3透析,得纳米粒子。
5.根据权利要求4所述的制备方法,其特征在于,所述步骤S1具体为:取辛伐他汀、二氢卟吩e6、NLG919分别溶于DMSO,形成浓度为8-12mg/mL的A溶液、B溶液、C溶液。
6.根据权利要求4所述的制备方法,其特征在于,所述步骤S2具体为:取步骤S1制得的45-55μL的A溶液,加入0.8-1.1mL水,超声1-3min,得混合液1。
7.根据权利要求4所述的制备方法,其特征在于,所述步骤S3具体为:取步骤S1制得的20-40μL的B溶液加入步骤S2制得的混合液1中,超声1-3min,再加入0.8-1.2mL水,超声吹打1.5-3min,得混合液2。
8.根据权利要求4所述的制备方法,其特征在于,所述步骤S4具体为:取步骤S1制得的40-60μL的C溶液加入步骤S3制得的混合液2中,超声6-8min,得混合液3。
9.根据权利要求4所述的制备方法,其特征在于:步骤S5中所述透析,使用的透析袋孔径为800-1500Da,透析时间为1-5h。
10.一种肿瘤治疗药物,其特征在于,有效成分包括权利要求1-3中任意一项所述的纳米粒子或者权利要求4-9任意一项所述制备方法制备的纳米粒子。
11.权利要求1-3中任意一项所述的纳米粒子或者权利要求4-9任意一项所述制备方法制备的纳米粒子在制备肿瘤治疗药物中的应用。
12.根据权利要求11所述的应用,其特征在于,所述肿瘤治疗包括光动力治疗、免疫治疗、细胞焦亡治疗、代谢调节治疗中的至少一种。
