发明的背景
在许多技术中,从用於流体中存在物质的测试能力来看,把物质,一般是颗粒状物质,从流体中分离出来的能力和设施是一个关键问题。例如,实验室现在有能力用红外光谱测定癌细胞,但是,这一项技术的运用受限于样品制备方面的困难。样品制备中带来的干扰经常使我们难以分析目标细胞,使分析过程不够可靠,或者要花很多的钱排除这种干扰。
类似的情况也发生在许多其它与检测和/或诊断有关的领域:包括环境测试,辐射研究,癌细胞筛选,细胞学测试,微生物测试和有害废物污染等等。
所有这些测试中,制备样品方案存在一个限制因素,即很难从流动载体(如生理、生物和环境等各种流动载体)中准确分离出固态物质,同时,也很难简单、有效地以一种易于实现的电磁辐射照射的方式进行采集和浓缩这些固态物质。例如,有报告说红外技术可以用来区分恶性细胞和正常细胞。细胞具有特征吸收波长,可以用来鉴别细胞的存在、类型以及它的数量。样品制备过程包括艰难地从组织或体液中分离出目标细胞,然后让红外光束通过含有细胞样品的支撑物。一般是采集细胞并涂在支撑物上,如显微镜的承物玻璃片上。采集和涂片需要一些技巧,而且细胞涂片仍可能不适合于用红外技术作分析。
在诊断微生物学,细胞学,特别是临床病理学领域,其基础是细胞的显微镜检查和其它显微镜分析。诊断的精度和最佳采样的制备取决于合适的样品制备方法。
本发明部分是以用红外辐射之类的电磁辐射标志物质的较新的发展为基础。例如,用一红外光束通过在预定位置的某种含有固态物质(如细胞)的支撑物。光束通过固态物质,固态物质吸收其中的特征波长,这种测量和特征吸收谱型可以用耒鉴别样品中的固态物质的类型和数量,以及分子构成或组份。

本发明和一般常用的各种样品制备技术相比有显著的优点。在铸膜法中,样品被溶解在溶剂中,溶液滴在红外窗口材料上(溴化钾或碘化铯),溶液蒸发后,在窗口材料上形成一簿膜。有时在红外辐射照射前,薄膜要从窗口材料上移去放在无活性的固体支撑物上。
热压膜技术:聚合样品被小心地融化到两片红外盐板(溴化钾或碘化铯)之间,小心地把一块盐板压到另一块上,直到薄膜形成。另一种类似的技术,压粘滞流体样品,产生毛细薄膜以形成液体涂片。
溴化钾小球技术:样品磨成大小一微米的粒子,样品和红外级的溴化钾粉末混合(必须小心,保证均匀),然后把粉状混合物用高压制成小球。
低浓度样品用热解法制备,即用液态镏出物干镏。

还有,本发明的装置和方法不需要操作采集点或固态物质支撑物,就可以把采集到的物质置于电磁辐射之下。这和现有的红外光谱仪不同,现有仪器中要移去支撑物如膜层,物质要固定在另一支撑物如显微镜承物玻璃片上,然后支撑物必须正确地放置在支架上。
本发明的装置也可以拆装,放入物质采集介质,可以方便其他需要的测试。例如,在用红外光谱分析后,可以打开装置,取下含有细胞的膜层,细胞可以放在显微镜承物玻璃片上,或作进一步处理,如培养或溶血,处理后可进一步测试细胞。
另一方面,按本发明,物质采集装置也可以包括另外的处理流体组件,可以拆开,也可以组合在一起。例如,流体可以用一物质采集组件处理,结合碎片去除组件,层析组件和分析组件或和其它装置组合。按本发明,这些组件和其它组件或处理方案具有一些必须的特色,可以并入样品制备装置。
新方法,如免疫细胞化学和图象分析,需要用色基标识目标细胞,或需要光吸收或发射探测物,要求制备过程是可重复的,快速的,没有生物损害的,而且是花费不大的。本发明的固态物质采集技术由于在样品制备中分许多步骤,可以标明物质密度的不均匀,分布的不均匀和样品缺失。本发明的制备形成固态物质的平均分布,这样的分布具有优越的形态,改进可观察性,而且容易定位,在光吸收分析时没有必要进一步操作或再制备样品。
例如,这些方法对普通的微生物学和血液学测试有许多优点。这些采集到的细胞处于预定的范围,容易对准辐射光源和吸收波长计。因为细胞都集中在一个单层里,几乎总是可以处于一个焦平面上,这样就消除了或减少了其他粒子的干扰。实际上,减少了技术人员获得正确读数的时间和专门技术需求,本发明的装置甚至允许用自动化装置检测和分析特定密度的任何固态物质。也可用来分析物质化学构成的细节。
用这一处理方法,物质重迭最小,可以确保方便地检测所有物质,很少可能有成为一团或重迭的固态物质或碎片。

本发明的详细说明
本发明包括一个套壳,和在该套壳内有一个物质采集部件,还有一穿过该物质采集部件联系一辐射源该采集点的光学通道。
本发明还包括一个装置,用耒制备让辐射能源照射的样品,它有一个采集点,辐射照射后可以测定采集物质的吸收光谱谱型。
本发明也包括采集流体,如生物学、生理学或环境学的流体,不用离心这些流体采出所要的物质,进而把辐射能照到装置中的物质上进行诊断和测试。
本发明也包括一种用于分析物质的方法,包括在采集部件上采集物质,使采集的物质接受辐射能源的辐照。有利的是,采集步骤和辐射步骤发生在同一套壳内。本发明的方法可以包括检测和/或测定物质吸收辐射能的量,也可以包括用特征吸收谱型检测和/或鉴定物质,而且可以包括测定所采集的样品的组分。
本发明也包括测定流体中某种物质是否存在和/或预定特质的数量多少的自动化方法。
本发明也包括具有分析组件的套件,其包括本发明的物质采集部件,流体样品杯,和一个使流体流过分析组件的泵。
本发明也包括一个固态物质采集部件,它具有一个受支撑的采集点和电磁辐射通过支撑物辐照采集点的通道。
本发明的一个最佳实施例是采集组件在流体中预定的位置上采集并浓缩固体达到预定的厚度。这样,该固体可以方便地,可重复地接受电磁辐射,以鉴别所采集的固态物质并测出数量。
这里所说的流体是指任何流体,只要是用来采集流体的一部分成份,鉴定其是否存在。一般来说,这种成份是固态物质,如颗粒物质。例如,流体可能是气体或空气,或生物流体如尿,可能要求测定是否存在癌细胞,或者测定生物流体中是否有某种蛋白质。另一例子是,在电子工业中用的超纯水,可能要估测杂质,比如分子杂质的性质。其它流体的例子包括但不限制于其它体液如血液,脊椎液,羊膜液;支气管分泌物;痰液;细针吸出物;地下水;工业处理液;电子或医学分解液等,但不限于这些。我们认为本发明不应受所处理的流体类型限制。
这里所说的,固态物质是指在流体中的任何物质,只要这些物质是可以被采集的并用辐射源作估测的。作为例子,物质包括但不限于细胞、细胞碎片、蛋白质、分子、聚合物、橡皮、安定剂、抗氧化剂、加速剂、硅、醇酸树脂、聚硫橡胶、石腊、热塑料、细菌、农药和杀草剂、但并不限于这些。作为化学聚合物的具体例子包括,但不限于聚乙烯、聚丙烯、聚异丁烯、聚丙烯晴、聚乙二醇、聚氯乙烯、聚苯乙烯、多硫化合物、聚甲基丙烯酸甲酯、聚对苯二甲酸乙酯、双酚A(一种常有的环境沾染物)、乙基纤维素、硝化纤维素、聚氨基甲酸乙酯和尼龙。作为生物物质的具体例子包括,但不限于癌细胞(含区分正常及变形的癌细胞)、蛋白质、核酸、抗体等诸如此类的物质。此项发明应不会被所例举的需处理的物质所限制。
这里所说的电磁辐射是指能被固态物质吸收的辐射能,包括,但不限于红外辐射,近红外辐射,可见光谱和近紫外辐射。例如,可用来测定结构,立体化学,添加物类型,退化度,是否存在共聚物,链长,取向,结晶度,碳氢结构强度区,饱和与非饱和碳一氢吸收之区别以及是否存在各别分子的电磁辐射。电磁辐射也可以用来测定样品组分,也就是特殊细胞,蛋白质,分子或聚合物的组分,我们认为本发明包括可以用来鉴别固态物质并测定数量的任何类型能量。
这里所说的流通,是从交通借用来的,或类似描述,是指以任何手段,或方法使流体流过系统,如工艺专家所知的,一个构造流通的有名结构是LUER锁。
本发明的系统装置实际上适合于含有特定物质的流体,此类物质对于辐射估测很灵敏。例如尿液中的癌细胞可以用红外辐射照射采集的细胞测得吸收谱型来鉴别。
按本发明的装置或测试化验组件,包括一个套壳,一个进口,一个出口,确定至少一条通过套壳的流体流动路径,采集部件安置在套壳中流体的通路当中。按本发明,采集部件包括一个采集点。虽然可以用各种各样的结构,如下述细节描述所指出的,这组件也包括一个光学通道,允许辐射能如红外光通过采集点到达吸收率计(Absorbance Meter)。
下面参考各图描述本发明的方法和装置。
图1是一个按本发明设计的典型组件,每一组件有套壳10,入口11,出口12,和采集部件13(见图3)。
如图2-4所示,一个分析组件或物质采集装置包括一个套壳10,该套壳10有入口11和出口12。套壳10确定容室18,入口11和出口12确定了通过套壳10的至少一条流体流径。采集部件13有一个采集点14用于采集物质,放在流体流动路径当中,采集点14与入口11相通。采集部件13在物质采集装置中适应于有两支路的流动路径,第一分支21通过采集点14流出去,第二分支22绕过采集点14(如图5)。
在最佳实施例中,本发明包括一个采集部件13,其具有第一个多孔介质23用于阻止物质流过,第二多孔介质24用于让流体通过。第二多孔介质24有可能从流体取走颗粒物质,也可能不取走,设计时按特殊装置的要求选择。在最佳实施例中,第一多孔介质用于俘获或采集固态物质,以及更优的是俘获或采集固态物质成均匀层或单层。在最佳实施例中,第二多孔介质也用于支撑第一多孔介质。
采集部件13也包括一个光通道15B,它允许电磁辐射照到第一多孔介质23上,而不接触第二多孔介质24。光通道15A,15B和15C是通过组件或套壳的光路,它允许电磁辐射照射到固态物质。如图3所示,光通道15B是一个通道,孔或任何形状的,只要通过第二多孔介质,例如置于中心的环形孔。
第一多孔介质和第二多孔介质可以做成任何形式,只要其功能如这里所描述的即可。熟悉工艺的会知道采集部件可以是各种各样结构与位置的,可根据需要获得特定的结果。例如。第一,第二多孔介质可以分开,中间留一间距;两层介质可以做成薄片迭在一起;第一介质可以和第二多孔介质一体化或可拆卸;或者采集部件可以包括高密度区,起第一多孔介质的作用和低密度区,起第二多孔介质的作用。选择这些不同结构对工艺专家耒说是完全可以做到的。
本发明的最佳实施例中,第一多孔介质是聚合碳化合物膜;而第二多孔介质是一厚的过滤片。
应该注意到,可以用各种类型的第一、第二多孔介质,美国专利5,301,685公开了几种多孔介质,可以用于本发明,因此列出作为参考。虽然聚合碳化合物膜是特别适于本发明的固态物质采集装置应用的,任何不干扰电磁波读出的膜或隔膜都是适用的。例如聚合碳化合物膜和其它多孔膜,赛璐珞或尼龙多孔膜,因为和红外谱波方案相容,都是可以用的。可以例举的,用耒筛选流体的介质,包括PALL公司的PALL生物支撑物分公司生产的一种白细胞置留介质LEUCOSORBTM。PALL公司制造并销售的其他膜是BIODYNE ATM,一种未改性的尼龙,表面化学成份是50%胺,和50%羧基群,它具有pH65的等电离点;BIODYNE BTM,一种经表面修饰的尼龙,其化学特性由表面高密度的正四价阳离子团所决定(pH>10时,Zeta电势为正);BIODYNE CTM,另一种经表面修饰的尼龙,其化学特性由其表面高密度带负电的羧基基团所决定(当pH>3时,Zeta电势为负);LOPRODYNETM,一种低聚蛋白质包捆的尼龙66膜,具有严格的可控的微孔结构。它具有大量的孔可以迅速有效地通过液体,而牢固地吸附液体中的小颗粒,因此被应用于细胞的分离和细菌细胞的免疫检测。在最佳实施例中,第一多孔介质是聚合碳化合物膜,适用于阻止细胞通过。优选的聚合化物膜可以从NUCLEOPORE买到,对于熟悉这项工艺的人耒说它是有名的公司。
采集部件13还可以包括一个厚的过滤片作为第二多孔介质24。第二多孔介质允许流体经第二分支流径22通过介质,同时也作为第一多孔介质的支撑。厚过滤片即第二多孔介质24可以用聚丙烯或高密度聚乙烯POREXR多孔塑料制成,也可用其他任何适用作第一多孔介质支撑的材料制成。
如图1、图2和6所示,第一部分16或入口11包括构成一个连接器的部分,可以和容器之类东西连接,或者以针或套管32构成。第二部分17或出口12包括一个构成连结器的部分,可以和泵30(如注射针之类)连接。
优选多孔膜的孔尺寸大约从0.22微米到8微米,从1微米到6微米更好;最好为2微米,它可以用来俘获大于3微米的细胞。膜层可以让流体流过它而阻挡颗粒物质20通过。第二多孔介质可让流体通过而且能从流体中把颗粒物质移走。第二多孔介质的孔尺寸可以从5微米到60微米,优选尺寸是大约15微米到45微米,最好是35微米。
本发明的另一实施例见图6,包括一个分析组件或物质采集装置10,装在采集杯31上,还有一个泵30用于引导流体流过采集组件。采集杯31可以是样品杯之类东西,和/或泵30可以是注射器或者其他可以引导流体流动的设备。如图6所示,采集杯31或采集组件可能包括一套管32或类似的东西,这样便于流体从采集杯抽入套壳10。在最佳实施例中,套管不同的位置上有穿孔33,便于抽入采集杯31中不同水平面的流体。
分析组件套壳10可以有不同设计,只要允许流体流过采集部件,形成一个整体即可。如图所示,分析组件套壳10最好由两件组成,一个可拆部份16和第二可拆部分17。其他能让流体进入采集部件13的套壳都可以用。
通过系统的流体的运动会受到流体源和流体出口之间压力差的影响。产生压力差的一种方法是对系统套壳入口处(例如采集杯)加压;把系统套壳出口处抽真空(例如用注射器)或者用任何种泵,例如自动玻璃丝过滤器(GENEX公司制造);重力头;或柔性的可折迭容器,例如样品容器,用它可以把流体挤入物质采集装置,通过该装置挤入注射器。本发明的最佳实施例是用一注射器从采集杯抽取流体,流过套壳。
在流体流过套壳10时,流体流过采集点14和采集部件13,如图5所示。技术专家知道按要采集的物质的种类和/或尺寸,调节多孔膜的孔目尺寸和多孔厚过滤片的尺寸,就可以在采集点14采集到所需的物质。在本发明的最佳实施例中,孔目尺寸选择得能在采集点上形成均匀的物质层,最好是单层。
技术专家也知道物质层的厚度可以调节,调到预定的厚度或要求达到的厚度,例如:大约3微米至大约40微米或更大被认为有效,但是,我们认为本发明不限于某一尺寸或厚度范围。
一旦均匀层或单层物质形成,沿着第一分支流动路径21的流体在多孔膜的中心减少,而沿第二分支流动路径22流动的流动的流体向采集部件13边缘增加。不管理论上如何解释,我们认为流动路径22中流体的增加原因是流动路径21中的流体在采集点14被采集的物质所阻挡。在第二分支流动路径22中的物质绕过采集点14,因而保持采集点14上的物质层实质上是均匀层或单层。第二分支流动路径22通过采集部件13的侧面部位,就象排泄通道(对流动的阻力小),可以防止物质的堆积。
以上所描述的物质采集装置或者组件可以用来和其它相适应的过滤器或处理设备相结合。作为例子,处理设备包括其它碎片处理装置,和/或分析装置或可以与套壳10连结的组件。常用的此类附加组件包括一个有入口和出口的套壳,包括过滤,分析或检测单元,放置在套壳内流体流过的路径上。例如,该装置包括一个由入口和出口部分确定流动路径的套壳;一个放在流动路径上的过滤器;和可以移动的层析/分析单元,如基质珠,放在过滤器的出口部位。层析/分析单元可以自由地与流体中的物质混合,俘获物质然后分析其是否存在。相适应的设备涉及已公开的美国专利4,953,561;5224,489;5,016,644;5,139,031;5,301,685;5,042,502和5,137,031,列在这里作参考。
按照本发明的方法,物质在采集部件上被采集,采集部件包括一个使采集物质暴露于电磁辐射的光学通道。在物质被采集后,电磁辐射通过光通道分析这些物质,测定其吸收量和/或吸收类型。
例如,流体从采集杯31中被抽出,通过套壳10,流体中的物质在采集点14被采集成均匀层或单层。最好可以抽取新的流体通过套壳,也可以抽取同样的流体,返回采集杯31,然后再抽,要反复多少次,就可以反复多少次。采集物质后,套壳10被放在固定架之类的东西上,调整位置,使电磁辐射如红外光束通过光学通道。光束沿光学通道15A,15B和15C通过出口12。在光通道15B,光束和采集点14上的采集物质作用。采集物质吸收特定波长的辐射,在光通道15C的路径上放一个吸收计可以测出这一吸收。
按本发明的方法也可以包括以上提到的物质的处理,然后把物质转到另一介质上作进一步分析。例如本发明也可以把细胞转到显微镜承物玻璃片上。和目前所用的方法不同之处在于,用膜过滤方法可以使细胞或其它物质均匀地沉积在显微镜承物玻璃片上,重迭最少。这样,可以清楚地观察并可获得最佳的诊断精度。
本发明也包括从单一病人或源样品产生多种取样。
还有,俘获的微有机物可以在培养介质如标准培养皿中培养。在采集装置套壳10中采集到细胞层后,流体可以通过采集点14通向入口11,这样可以把微有机物转移到标准培养皿。
在细菌测试时,采集点14可以用于用QUALTURE设备(未显示)培养细菌,以便测定是否存在特殊的细菌群体。QUALTURE设备是一个塑料小皿,其中含有过滤膜和四层脱水的,有选择性的介质营养衬垫。
本发明的装置和方法有广泛的应用,主要是因为许多工业,许多处理要从流体中分离出固态物质,然后检测这些固态物质。举例说,这样的工业有食品和饮料,药品,医学和环境(即水、土壤或空气样品),生物,微生物,血液、细胞和病理等。
本发明的装置和方法在这样的测试中特别有用:即用用光谱方法鉴别混合物,分子,细胞,蛋白质等固态物质,无论其是单一的或混合的,测定依据是这些物质在特定波长上吸收辐射能。本发明的装置和方法在以下测试中更有用:物质受由磁波辐照后研究其吸收谱型,电磁波是红外光,波长约为2.5微米到15微米。在血液分析中,一滴血可用耒分析特定细胞是否存在,密度多少,其原理是每一种细胞在红外线之类辐射下有不同的标准图型。例如,可以测定淋巴细胞和白细胞之比,是否存在癌细胞,癌细胞的类型,蛋白质或脂肪的多少等。
在各种工业部门,需要测定空气、水之类流体中的杂质,比如饮用水,或者测定食品和饮料加工厂里的细菌。环境分析中需要测定污染是否存在,类型与含量,如甾醇类化合物,杀虫剂(DDT,高氯化合物,阿杀辛),芳香烃,多氯化联苯。在医学和环境领域,需要测定是否存在断裂产物,如一种降价塑料双酚A,塑料配合剂(Ingredient in plastics)。
本发明的装置与方法在物质用色基(Chromophre),一种光吸收或发射的探针(probe),标志或用其它可见的试剂标志时有用。例如细胞和DNA可以用直接或间接地和感兴趣物质相连的探针来分析,这种探针可以和第一多孔介质结合在一起或混在流体样品中。
应该清楚这一点:本发明的装置与方法可以用在许许多多工业部门,测定任何种类固态物质的存在,数量和组成。
虽然本发明描述的只是最佳实施例,但并不受这些实施例的限制。技术熟练的人员,参照以上描述,可以作出仍在本发明范围之内的结构变更,实例和改进。所以,以下的权利要求包括应该在本发明的概念和范围之内所有变更,实例,改进或相当的结构。具体范围见权利要求。
Claims (18)
1、一种固态物质采集装置,包括一个套壳,和该套壳内装设一个物质采集部件,以及有一采集点,并有一穿过该物质采集部件联系一辐射源与该采集点的光学通道。
2、根据权利要求1所述的固态物质采集装置,其特征在于采集部件适应于有第一和第二分支的流动路径,其中,第一分支通过采集点,第二分支绕过采集点。
3、根据权利要求2所述的固态物质采集装置,其特征在于采集部件包括能阻止固态物质流过的第一个多孔介质,所说的多孔介质有一个位于光学通道上的采集点。
4、根据权利要求3所述的固态物质采集装置,其特征在于多孔介质是一多孔膜。
5、一个采集和分析固态物质的系统,包括一电磁辐射源、一固态物质采集组件和一吸收系数表,该固态物质采集组件包括一套壳和装在该套壳内的采集部件,其特征在于所说的系统包括一个从该电磁辐射源到到该吸收系数表的光学通道,该光学通道通过该采集部件。
6、根据权利要求5所述的采集和分析固态物质的系统,其特征在于电磁辐射源是红外辐射源。
7、一种固态物质采集部件,包括一个具有光学通道的多孔支撑物和一位于支撑物上的采集点,所说的采集点适合于采集在预定位置上预定密度的固态物质,并适合于电磁辐射的照射。
8、根据权利要求7所述的固态物质采集部件,其特征在于所述的固态支撑物是一厚的过滤片。
9、根据权利要求7所述的固态物质采集部件,其特征在于所述的采集点是一种膜。
10、根据权利要求9所述的固态物质采集部件,其特征在于所述的膜的孔目尺寸约从0.3微米到35微米。
11、一种分析在液体中的固态物质的方法,包括从液体中分离固态物质并把固态物质采集到采集点上;把采集点上采集的固态物质置于电磁辐射之下;并分析固态物质置于电磁辐射之下所产生的吸收谱型。
12、根据权利要求11所述的分析在液体中的固态物质的方法,其特征在于从液体中分离固态物质并把固态物质采集到采集点上,包括让流体流过有采集点的采集部件,在采集点上,液体中的固态物质从液体中分离出来并被采集成一均匀层。
13、根据权利要求11所述的分析在液体中的固态物质的方法,其特征在于把采集点上采集的固态物质置于电磁辐射之下,包含把固态物质置于红外辐射之下。
14、根据权利要求11所述的分析在液体中的固态物质的方法,其特征在于分析固态物质置于电磁辐射之下所产生的吸收谱型,包含鉴定采集点上是否有固态物质。
15、根据权利要求14所述的分析在液体中的固态物质的方法,其特征在于鉴定采集点上是否有固态物质,包括确定固态物质的量。
16、一种为红外光谱仪制备固态物质的方法,包括:
a)让含有固态物质的流体流过采集装置,该装置包括有一个适合采集固态物质的采集点的采集部件,和一个用来使采集点上的固态物质接受红外辐射的照射的光学通道;以及
b)在采集点上沉积均匀的固态物质层。
17、根据权利要求1所述的固态物质采集装置,其特征在于还包含附有可见着色剂的物质采集部件。
18、根据权利要求11所述分析在液体中的固态物质的方法,其特征在于分析吸收谱型包括分析可见着色剂(Agent)。
