Abstract
本发明公开了一种新型水果防裂液及其制备方法,属于防裂果技术领域。所述防裂液包括以下原料:壳寡糖肽钙、钨酸钠、氨基酸螯合物、S‑诱抗素、羧甲基壳聚糖和介孔二氧化硅纳米球;所述原料的质量比为:(15‑28)∶(5‑9)∶(5‑9)∶(2‑5)∶(7‑15)∶(16‑20)。采用本发明的方法得到的水果防裂液可应用于多种水果的种植过程中,可以有效降低水果的裂果率,保证水果的质量,进而能够带来较大的经济效益。
Description
技术领域
本发明属于防裂果技术领域,具体涉及一种新型水果防裂液及其制备方法。
背景技术
果实裂果的发生是果树生产上存在的一个普遍的问题,裂果病是一种常见的危害果实的生理性病害,容易发生在果实的转色期。裂果是指在植物生长过程中或成熟后,果实崩裂或开裂射出的现象,是由于果实内部生长应力不断或突然地增加和集中,超过了果皮所能承受的力学强度,便在果皮发育的薄弱处撑破果皮的结果。该现象在许多种类的果实中均普遍存在,比如,瓜果、蔬菜,如番茄、西瓜、甜瓜、苹果、脐橙、芒果、桃、石榴、荔枝、葡萄等在生长过程中经常会出现裂果的现象。果实产生裂痕后,容易引起病原菌入侵,进而导致果实腐烂或被污染,使果实降低或失去商品价值,造成巨大的经济损失。
经研究调查发现,造成裂果的原因有很多,例如:(1)不良的外界条件,如遭遇强光照射、高温干旱或暴雨;(2)不当的栽培技术,如干旱时大水灌溉、氮肥太多或钙肥不足等均容易使果实发生裂果现象。为了降低裂果现象的产生,研究人员对防裂果液进行了大量研究。但是,现有的防裂液中普遍存在营养物质不能被有效吸收,造成补充营养物质的浪费的问题。而且,目前的防裂液对防水性能方面设计考虑不足,防水性能较差也会导致果实开裂。因此,如何研究一种具有较好的防裂效果的新型水果防裂液是本领域技术人员亟需解决的技术问题。
发明内容
为解决现有技术中的上述问题,本发明提供了一种新型水果防裂液及其制备方法。
为实现上述目的,本发明提供了如下技术方案:
本发明提供了一种新型水果防裂液,所述防裂液包括以下原料:壳寡糖肽钙、钨酸钠、氨基酸螯合物、S-诱抗素、羧甲基壳聚糖和介孔二氧化硅纳米球;
所述原料的质量比为:(15-28)∶(5-9)∶(5-9)∶(2-5)∶(7-15)∶(16-20)。
有益效果:本发明将钙和壳寡糖结合,可以更好地预防果实裂果。而且壳寡糖肽钙在喷于果面时,会发生部分成膜,附着于果面上,从而可以阻挡水通过果皮渗透进入果肉,防止果肉细胞改变渗透压吸水,果肉迅速膨胀,导致裂果的问题。壳寡糖是由N-乙酞-D-葡萄糖胺以β-1,4糖苷键结合而成的多糖,与纤维素具有相似的结构,且分子量较小,可以通过果实表皮渗透进果实内部,快速合成纤维素,形成新的细胞壁和果皮,起到较好的预防果实裂果的效果;壳寡糖肽钙用于形成果胶物质分子间的钙桥,钙桥交联成网状结构,它们作为细胞间的中层起粘合作用,进而提高果实对营养物质钙的吸收。
本发明中的钨酸钠也具有一定的防水作用,其和壳寡糖肽钙协同作用,进一步提高了防裂液的防水性能,降低了其裂果率。本发明在防裂果液中加入介孔二氧化硅纳米球,其可以在防裂液的成膜过程中使其形成具有一定柔韧性的透气膜,覆盖在水果表皮,然后结合羧甲基壳聚糖的杀菌性和抗病毒性,进而实现对水果表皮的进一步保护。
进一步地,所述壳寡糖肽钙的制备方法为:
将壳聚糖溶解于酸性溶液中,经β-葡萄糖苷酶酶解后,加热,接着加入氢氧化钠进行水解,最后加入钙的盐溶液混合均匀,即得所述壳寡糖肽钙。
有益效果:本发明利用β-葡萄糖苷酶对破坏壳聚糖之间的β-1,4糖苷键,使壳聚糖酶解,使最终得到的产物更加稳定。
更进一步地,所述酸性溶液由去离子水、顺丁烯二酸和碳酸氢铵按照质量比为4∶2∶1.8混合后,再利用质量浓度为5%的氢氧化钠溶液调pH至5.0制备得到;
所述钙的盐溶液为质量浓度为70-80%的氯化钙溶液。
有益效果:本发明中水溶性不饱和二元有机酸顺丁烯二酸与碳酸氢铵反应生成不饱和酰胺酸,在一定压力和温度下,发生热聚合生成聚酰亚胺,聚酰亚胺水解得到聚氨基酸盐,聚氨基酸盐具有较好的螯合性,与钙离子鳌合,不会生成沉淀,同时,又可以避免钙离子与壳寡糖反应生成沉淀,增加了壳寡糖与肥料的配伍性。
更进一步地,所述壳聚糖、酸性溶液、氢氧化钠和钙的盐溶液的质量比为:(8-15)∶(30-42)∶(12-22)∶(8-12)。
更进一步地,所述酶解的温度为38-45℃,时间为18-24h;
所述加热的压力为0.5-0.8MPa,温度为110-150℃,时间为70-120min;
所述水解的时间为30-50min。
进一步地,所述氨基酸螯合物的制备方法为:
将氯化钾、聚合硼酸钠、表面活性剂、黄原胶和甲酸混合后溶解于水中,然后加入pH缓冲剂,经加热后,加入氨基酸和柠檬酸继续加热,最后经静置、过滤后,即得所述氨基酸螯合物。
有益效果:本发明利用氨基酸将营养物质钾、钠保护起来,形成氨基酸螯合物,使营养物质通过螯合物形式被吸收,增强营养物质的移动性;而且以氨基酸作为螯合体的配体,还具有配合体与微量元素两者的双重肥效,更有利于果实对营养成分的有效吸收。
更进一步地,所述氯化钾、聚合硼酸钠、表面活性剂、黄原胶、甲酸、水、pH缓冲剂、氨基酸和柠檬酸的质量比为:(8-10)∶(8-10)∶(3-5)∶(1-2)∶(1-2)∶(65-90)∶(0.8-1.0)∶(12-16)∶(0.8-1.0)。
更进一步地,所述表面活性剂为OP-10;
所述pH缓冲剂为六次甲基四胺。
更进一步地,所述两次加热的温度均为40-50℃,保持时间为7.5-12h。
本发明还提供了一种新型水果防裂液的制备方法,包括以下步骤:
按所述比例称取各原料,将所述原料混合均匀后,即得所述水果防裂液。
与现有技术相比,本发明具有以下有益效果:
本发明在制备水果防裂液时,将壳寡糖肽钙和钨酸钠联用,其在应用于水果表面时,形成的膜层具有很好的防水性能,进而提高了其防裂效果,降低了果实的裂果率,同时加入S-诱抗素,促进果实对吸收水、肥和协调体内代谢的平衡能力,可有效调控果实的生长,对提高果实的品质具有重要作用。采用本发明的方法得到的水果防裂液可应用于多种水果的种植过程中,可以有效降低水果的裂果率,保证水果的质量,进而能够带来较大的经济效益。

本发明实施例中的β-葡萄糖苷酶购买于沧州夏盛酶生物技术有限公司;
羧甲基壳聚糖购买于西安秋禾生物科技有限公司;
本发明的实施例中其他原料均可以通过市售购买,本发明对其他原料的来源和购买途径不作限制。
实施例1
一种新型水果防裂液的制备方法,包括以下步骤:
(1)将去离子水、顺丁烯二酸和碳酸氢铵按照质量比为4∶2∶1.8混合后,加入质量浓度为5%的氢氧化钠溶液调pH至5.0,得到酸性溶液,然后将壳聚糖加入至得到的酸性溶液中,再经β-葡萄糖苷酶在38℃酶解20h后,在压力0.5MPa,温度150℃下加热120min,接着加入氢氧化钠水解45min,最后加入质量浓度为80%的氯化钙溶液,混合均匀,得到壳寡糖肽钙,其中壳聚糖、酸性溶液、氢氧化钠和钙的盐溶液的质量比为:8∶36∶20∶10。
(2)将氯化钾、聚合硼酸钠、表面活性剂OP-10、黄原胶和甲酸混合后溶解于水中,然后加入pH缓冲剂六次甲基四胺,在温度40℃下,保持12h后,加入氨基酸和柠檬酸继续在温度45℃下,保持10h,最后经静置、过滤后,得到氨基酸螯合物,其中,氯化钾、聚合硼酸钠、表面活性剂OP-10、黄原胶、甲酸、水、六次甲基四胺、氨基酸和柠檬酸的质量比为:9∶8∶3∶2∶1∶80∶0.8∶15∶1。
(3)将20份步骤(1)制备的壳寡糖肽钙、6份钨酸钠、9份步骤(2)制备的氨基酸螯合物、5份S-诱抗素、12份羧甲基壳聚糖和18份介孔二氧化硅纳米球混合均匀,即得到水果防裂液。
实施例2
一种新型水果防裂液的制备方法,包括以下步骤:
(1)将去离子水、顺丁烯二酸和碳酸氢铵按照质量比为4∶2∶1.8混合后,加入质量浓度为5%的氢氧化钠溶液调pH至5.0,得到酸性溶液,然后将壳聚糖加入至得到的酸性溶液中,再经β-葡萄糖苷酶在45℃酶解20h后,在压力0.5MPa,温度130℃下加热110min,接着加入氢氧化钠水解45min,最后加入质量浓度为80%的氯化钙溶液,混合均匀,得到壳寡糖肽钙,其中壳聚糖、酸性溶液、氢氧化钠和钙的盐溶液的质量比为:10∶40∶22∶12。
(2)将氯化钾、聚合硼酸钠、表面活性剂OP-10、黄原胶和甲酸混合后溶解于水中,然后加入pH缓冲剂六次甲基四胺,在温度45℃下,保持10h后,加入氨基酸和柠檬酸继续在温度45℃下,保持10h,最后经静置、过滤后,得到氨基酸螯合物,其中,氯化钾、聚合硼酸钠、表面活性剂OP-10、黄原胶、甲酸、水、六次甲基四胺、氨基酸和柠檬酸的质量比为:9∶10∶4∶2∶2∶85∶1∶16∶1。
(3)将26份步骤(1)制备的壳寡糖肽钙、5份钨酸钠、8份步骤(2)制备的氨基酸螯合物、4份S-诱抗素、7份羧甲基壳聚糖和20份介孔二氧化硅纳米球混合均匀,即得到水果防裂液。
实施例3
一种新型水果防裂液的制备方法,包括以下步骤:
(1)将去离子水、顺丁烯二酸和碳酸氢铵按照质量比为4∶2∶1.8混合后,加入质量浓度为5%的氢氧化钠溶液调pH至5.0,得到酸性溶液,然后将壳聚糖加入至得到的酸性溶液中,再经β-葡萄糖苷酶在38℃酶解24h后,在压力0.6MPa,温度130℃下加热70min,接着加入氢氧化钠水解30min,最后加入质量浓度为70%的氯化钙溶液,混合均匀,得到壳寡糖肽钙,其中壳聚糖、酸性溶液、氢氧化钠和钙的盐溶液的质量比为:8∶30∶12∶8。
(2)将氯化钾、聚合硼酸钠、表面活性剂OP-10、黄原胶和甲酸混合后溶解于水中,然后加入pH缓冲剂六次甲基四胺,在温度45℃下,保持7.5h后,加入氨基酸和柠檬酸继续在温度40℃下,保持12h,最后经静置、过滤后,得到氨基酸螯合物,其中,氯化钾、聚合硼酸钠、表面活性剂OP-10、黄原胶、甲酸、水、六次甲基四胺、氨基酸和柠檬酸的质量比为:8∶9∶4∶1∶2∶65∶0.8∶12∶0.8。
(3)将15份步骤(1)制备的壳寡糖肽钙、9份钨酸钠、9份步骤(2)制备的氨基酸螯合物、2份S-诱抗素、12份羧甲基壳聚糖和16份介孔二氧化硅纳米球混合均匀,即得到水果防裂液。
实施例4
一种新型水果防裂液的制备方法,包括以下步骤:
(1)将去离子水、顺丁烯二酸和碳酸氢铵按照质量比为4∶2∶1.8混合后,加入质量浓度为5%的氢氧化钠溶液调pH至5.0,得到酸性溶液,然后将壳聚糖加入至得到的酸性溶液中,再经β-葡萄糖苷酶在45℃酶解18h后,在压力0.8MPa,温度110℃下加热120min,接着加入氢氧化钠水解50min,最后加入质量浓度为80%的氯化钙溶液,混合均匀,得到壳寡糖肽钙,其中壳聚糖、酸性溶液、氢氧化钠和钙的盐溶液的质量比为:10∶42∶18∶12。
(2)将氯化钾、聚合硼酸钠、表面活性剂OP-10、黄原胶和甲酸混合后溶解于水中,然后加入pH缓冲剂六次甲基四胺,在温度50℃下,保持8h后,加入氨基酸和柠檬酸继续在温度50℃下,保持10h,最后经静置、过滤后,得到氨基酸螯合物,其中,氯化钾、聚合硼酸钠、表面活性剂OP-10、黄原胶、甲酸、水、六次甲基四胺、氨基酸和柠檬酸的质量比为:10∶8∶5∶1∶1∶90∶1∶16∶1。
(3)将28份步骤(1)制备的壳寡糖肽钙、7份钨酸钠、5份步骤(2)制备的氨基酸螯合物、4份S-诱抗素、15份羧甲基壳聚糖和18份介孔二氧化硅纳米球混合均匀,即得到水果防裂液。

一种新型水果防裂液的制备方法,与实施例2的区别在于:不加入钨酸钠;
其余步骤同实施例2。
对比例2
一种新型水果防裂液的制备方法,与实施例2的区别在于:将步骤(1)中的β-葡萄糖苷酶等量替换为溶菌酶;

一种新型水果防裂液的制备方法,与实施例2的区别在于:将8份氨基酸螯合物替换为8份氨基酸、8份氯化钾和8份聚合硼酸钠;
其余步骤同实施例2。
对比例4
一种新型水果防裂液的制备方法,与实施例2的区别在于:将20份壳寡糖肽钙替换为20份壳聚糖和20份氯化钙;
其余步骤同实施例2。
效果验证:
将同一樱桃园中长势相差不大的樱桃树随机分成8个区,每个区内有10棵樱桃树。分别在大雨来临前24h向每个区中的樱桃树喷洒实施例1-4和对比例1-4制备的水果防裂液的稀释液(每1mL水果防裂液中加入500mL水),每颗樱桃树喷洒2L稀释液,期间,对樱桃园采用常规操作进行肥水管理,考察裂果率。喷洒水果防裂液的稀释液的时间为樱桃采摘前1个月。结果如表1所示。

由表1可知,施用本发明所制备的水果防裂液后,可以大大减少番茄裂果现象的发生,降低裂果率。

1.一种新型水果防裂液,其特征在于,所述防裂液包括以下原料:壳寡糖肽钙、钨酸钠、氨基酸螯合物、S-诱抗素、羧甲基壳聚糖和介孔二氧化硅纳米球;
所述原料的质量比为:(15-28)∶(5-9)∶(5-9)∶(2-5)∶(7-15)∶(16-20)。
2.根据权利要求1所述的一种新型水果防裂液,其特征在于,所述壳寡糖肽钙的制备方法为:
将壳聚糖溶解于酸性溶液中,经β-葡萄糖苷酶酶解后,加热,接着加入氢氧化钠进行水解,水解完成后加入钙的盐溶液混合均匀,即得所述壳寡糖肽钙。
3.根据权利要求2所述的一种新型水果防裂液,其特征在于,所述酸性溶液由去离子水、顺丁烯二酸和碳酸氢铵按照质量比为4∶2∶1.8混合后,再利用质量浓度为5%的氢氧化钠溶液调pH至5.0制备得到;
所述钙的盐溶液为质量浓度为70-80%的氯化钙溶液。
4.根据权利要求2所述的一种新型水果防裂液,其特征在于,所述壳聚糖、酸性溶液、氢氧化钠和钙的盐溶液的质量比为:(8-15)∶(30-42)∶(12-22)∶(8-12)。
5.根据权利要求2所述的一种新型水果防裂液,其特征在于,所述酶解的温度为38-45℃,时间为18-24h;
所述加热的条件为压力0.5-0.8MPa,温度110-150℃,时间70-120min;
所述水解的时间为30-50min。
6.根据权利要求1所述的一种新型水果防裂液,其特征在于,所述氨基酸螯合物的制备方法为:
将氯化钾、聚合硼酸钠、表面活性剂、黄原胶和甲酸混合后溶解于水中,然后加入pH缓冲剂,经加热后,加入氨基酸和柠檬酸继续加热,最后经静置、过滤后,即得所述氨基酸螯合物。
7.根据权利要求6所述的一种新型水果防裂液,其特征在于,所述氯化钾、聚合硼酸钠、表面活性剂、黄原胶、甲酸、水、pH缓冲剂、氨基酸和柠檬酸的质量比为:(8-10)∶(8-10)∶(3-5)∶(1-2)∶(1-2)∶(65-90)∶(0.8-1.0)∶(12-16)∶(0.8-1.0)。
8.根据权利要求6所述的一种新型水果防裂液,其特征在于,所述表面活性剂为OP-10;
所述pH缓冲剂为六次甲基四胺。
9.根据权利要求6所述的一种新型水果防裂液,其特征在于,所述两次加热的温度均为40-50℃,保持时间为7.5-12h。
10.一种如权利要求1-9任一项所述的新型水果防裂液的制备方法,其特征在于,包括以下步骤:
按比例称取各原料,将所述原料混合均匀后,即得所述水果防裂液。
