Abstract
本发明涉及灵芝酸A在制备治疗CRPC的药物中的应用。本发明通过研究发现,GAA能够显著抑制LNCaP‑AI细胞的增殖、PSA分泌、克隆形成、创面愈合、迁移和侵袭,且GAA可在体内显著抑制去势抵抗性前列腺癌细胞的生长;LNCaP‑AI细胞中AKT、GSK‑3β等的异常激活可以被GAA进行有效抑制;Top‑flash荧光素酶测定确定了活化的β‑catenin也可以被抑制。由此明确了AKT/GSK‑3β/β‑catenin通路参与了GAA对CRPC的作用过程。本发明明确了灵芝酸A对于CRPC的治疗活性,并具体研究了其作用机制,为延缓前列腺癌ADT后转化为CRPC的进程、治疗去势抵抗性前列腺癌提供了新的活性物质,为后续去势抵抗性前列腺癌药物的开发提供了新的治疗靶点和方向。

技术领域
本发明属于生物医学技术领域,涉及灵芝酸A在制备治疗去势抵抗性前列腺癌的药物中的应用。
背景技术
前列腺癌存在雄激素依赖特性,降低雄激素水平或阻断雄激素受体(AndrogenReceptor,AR)可抑制前列腺癌的进展,雄激素剥夺治疗(androgen deprivation therapy,ADT)是局部晚期和转移性前列腺癌的标准治疗方式。虽然初期ADT对大多数前列腺癌患者有效,但是经过18-20个月的缓解期后,几乎所有的前列腺癌患者都将进展为去势抵抗性前列腺癌(castration resistant prostate cancer,CRPC)。
CRPC指经过初次持续雄激素剥夺治疗(ADT)后疾病仍然进展的前列腺癌,患者应同时具备以下条件:(1)检测血清睾酮达到去势水平(<0.5μg/L或<1.7nmol/L);(2)间隔1周检查前列腺特异性抗原(PSA)连续3次上升,升高幅度较基础值>50%。 CRPC目前仍是临床治疗的一个难题。现有技术中明确了抗骨转移治疗、免疫治疗是当前CRPC的主要治疗方法;但是,上述药物仅能在有限的时间内控制肿瘤,并且有不同副作用,患者无法通过这些药物治疗获得更长的生存时间。CRPC患者的生存预后都不理想,患者一旦进展为CRPC,则预后极差,中位生存期通常不超过12个月,最终将死于去势抵抗性前列腺癌。
自从Huggins和Hodges发现前列腺癌存在雄激素依赖特性,降低雄激素水平或阻断雄激素受体可抑制前列腺癌的进展,ADT成为局部晚期和转移性前列腺癌的标准治疗方式。大部分前列腺癌患者接受ADT治疗后将出现前列腺特异性抗原(Prostate SpecialAntigen,PSA)水平下降、肿瘤缩小等明显的治疗效果,不过在2年内大多数患者都终将不可避免的出现PSA水平升高,肿瘤进展,对ADT治疗失去敏感性,成为CRPC。
有研究发现灵芝中的三萜类化合物(例如Ganoderic acid A和Ganoderic acidDM等)能通过能通过Wnt/β-catenin信号通路对乳腺癌、肺癌、肝癌、骨肉瘤、前列腺癌等恶性肿瘤发挥治疗作用。然而,由于前列腺癌与去势抵抗性前列腺癌的发生、发展机制存在明显差异,且大量的研究结果显示常规前列腺癌治疗药物在应用于CRPC的治疗时往往会出现治疗效果大幅度降低甚至无效的情况,同时还会给患者带来严重的不良反应,大大降低患者的生活质量,从而使得常规前列腺癌治疗药物无法继续用于去势抵抗性前列腺癌的正常治疗。因此,针对去势抵抗性前列腺癌的发病机制进行深入研究并探寻能够特异性治疗CRPC的相关活性成分成为了亟待解决的关键问题。
发明内容
本发明的目的在于解决现有技术中所存在的去势抵抗性前列腺癌发病因素复杂、难以进行提前预防、常规前列腺癌治疗药物疗效差的技术问题,从而对去势抵抗性前列腺癌的发病机制进行了深入研究,发现灵芝酸A(Ganoderic acid A,GAA)可通过AKT/GSK-3β/β-catenin信号通路对去势抵抗性前列腺癌产生明显的抑制作用,从而为去势抵抗性前列腺癌的治疗提供了新的活性成分以及相关治疗靶点。
为了解决上述技术问题,本发明是通过如下技术方案得以实现的。

信号通路已经成为抗肿瘤药物的新靶点,针对肿瘤信号通路的调节功能及机制的研究具有重要的临床意义。大量的研究发现,灵芝/灵芝三萜类化合物能够对包括前列腺癌在内的多种恶性肿瘤产生治疗活性,其主要通过Wnt/β-catenin信号通路发挥抗肿瘤作用。而在研究中还发现,常规的前列腺癌治疗药物在长期大量使用后往往会使得患者对药物的敏感性大大降低进而出现耐药,尤其当前列腺癌患者进展为去势抵抗性前列腺癌患者后,先前使用的治疗方案往往不再奏效。这使得本领域研究人员不得不考虑是否因为去势抵抗性前列腺癌的发生发展机制发生了变化而导致原有治疗药物的治疗靶点发生了改变,进而影响其治疗效果。
CRPC的发病机制是目前研究的难点和热点。研究人员对此提出了三种可能的CRPC形成机制:(1)AR相关机制,AR信号通路的激活起着重要作用;(2)干细胞形成机制,干细胞生长不依赖于AR, ADT治疗后可发展为CRPC;(3)神经内分泌转化机制,ADT治疗可诱导前列腺癌细胞神经内分泌分化(NED)。前列腺癌NED细胞不再依赖AR信号通路,通过旁分泌和自分泌作用促进前列腺癌细胞的生长。然而,有关CRPC进展的确切机制目前仍尚不清楚。
对此,本发明对去势抵抗性前列腺癌的发生发展机制进行了深入研究,发现灵芝酸A在体外能够明显降低去势抵抗性前列腺癌细胞中AR、PSA、FKBP5、HK2等表达水平,显著抑制去势抵抗性前列腺癌细胞的增殖、克隆形成、迁移和侵袭等活动和功能,同时在体内抑制去势抵抗性前列腺癌细胞的生长。而作为对照的前列腺癌一线治疗药物恩杂鲁胺则对去势抵抗性前列腺癌细胞的活性明显降低,无法有效降低其中AR、HK2等水平,明确了去势抵抗性前列腺癌往往会对传统前列腺癌治疗药物降低敏感性,即常规的前列腺癌药物无法对去势抵抗性前列腺癌进行正常治疗。通过对作用机制进行分析发现,灵芝酸A可以显著下调去势抵抗性前列腺癌细胞内AKT和GSK-3β的磷酸化水平,同时抑制β-catenin水平;且灵芝酸A能够显著抑制去势抵抗性前列腺癌细胞内c-Myc、Cyclin D1、MMP-2的表达水平,并下调了AR的表达水平。由此可以明确灵芝酸A通过AKT/GSK-3β/β-catenin信号通路以调控去势抵抗性前列腺癌的功能和活动,该作用机制则与前列腺癌的治疗机制同样存在明显的差异。
本发明相对于现有技术具有如下技术效果:
(1)本发明对去势抵抗性前列腺癌的发病、发展机制进行了深入研究,发现灵芝酸A在体外能够显著抑制去势抵抗性前列腺癌细胞的增殖、克隆形成、迁移和侵袭等活动和功能,同时在体内抑制去势抵抗性前列腺癌细胞的生长,为延缓前列腺癌ADT后转化为CRPC的进程、治疗去势抵抗性前列腺癌提供了新的活性成分。
(2)本发明对灵芝酸A在去势抵抗性前列腺癌中的具体作用机制进行了深入分析,发现灵芝酸A可以显著下调去势抵抗性前列腺癌细胞内AKT和GSK-3β的磷酸化水平,同时抑制β-catenin水平;且灵芝酸A能够显著抑制去势抵抗性前列腺癌细胞内c-Myc、Cyclin D1、MMP-2的表达水平,并下调了AR的表达水平。明确了灵芝酸A通过AKT/GSK-3β/β-catenin信号通路以调控去势抵抗性前列腺癌的功能和活动,为去势抵抗性前列腺癌的治疗和后续药物的研发提供了新的作用靶点。

具体实施方式
为使本发明的目的、技术方案及效果更加清楚、明确,以下参照实施例对本发明作进一步详细说明。应当理解,此处所描述的具体实施例仅用以解释本发明,并不用于限定本发明。
在无特别说明的情况下,本发明上下文中所使用的试剂中,均通过市售获得。前列腺癌细胞系LNCap从American Type Culture Collection(ATCC、Manassas、VA、USA)购入并根据ATCC指南进行培养。所有细胞系均通过中国典型培养物保藏中心(武汉)的短串联重复分析鉴定,并使用PCR检测试剂盒(上海Biothrive Sci)验证是否存在支原体污染,同时在液氮中冷冻保存并用于后续实验。本发明所使用的试剂、耗材、设备等均通过市售获得,其中灵芝酸A购于南京中科公司。本发明所使用的实验方法,例如分子生物学实验、细胞实验、生物信息学分析等均为本领域的常规方法和技术。对于动物实验的开展,实验方案均经过暨南大学动物实验伦理委员会批准并遵循其准则。
生物学实验重复中选择具有代表性的结果呈现在上下文附图中,数据按照图示中规定的以mean±SD和mean±SEM展示。所有实验至少重复三次。数据的统计分析由GraphPadPrism 9.0或SPSS 20.0完成。采用t检验、方差分析、秩和检验、卡方检验、Mann-Whitney U检验等常规医学统计学方法比较两组或两组以上的平均值差异。p<0.05被认为是一个显著的差异。
实施例1 去势抵抗性前列腺癌细胞系的构建和验证
首先进行去势抵抗性前列腺癌细胞系的构建,具体包括如下步骤:
(1)将对数生长期的LNCaP细胞置于不含FBS的RPMI-1640培养基中培养48h。
(2)弃去培养基,添加含有经活性炭/葡聚糖处理的FBS的RPMI-1640培养基进行连续培养,以筛选稳定的雄激素非依赖性LNCaP细胞(LNCaP-AI细胞)。
在培养15天后,LNcaP细胞逐渐开始凸出假足;培养2个月后,向外的假足相互连接形成网络,细胞处于自分泌状态;培养4个月后,细胞生长转变为扩散样生长,凋亡减少;培养6个月后,则形成不依赖雄激素的LNCaP细胞(LNCaP-AI细胞)(参见图1)。分别取筛选获得的LNCaP-AI细胞和正常LNCaP细胞,分别置于添加含有经活性炭/葡聚糖处理的FBS的RPMI-1640培养基中进行培养,检测两种细胞的生长速度,结果发现,正常LNCaP细胞生长速度缓慢,而LNCaP-AI细胞的生长速度明显更快(参见图2),进一步证明了筛选获得的细胞为雄激素非依赖性前列腺癌细胞。
为了鉴定所建立LNCaP-AI细胞系的基因谱,采用RT-qPCR检测AR、FKBP5和STEAP1(AR相关信号通路)的mRNA水平,具体步骤如下:
(1)将前列腺癌细胞(LNCaP、LNCaP-AI)分别培养于6孔板中,待细胞生长至对数期时分别收集1×106个前列腺癌细胞。
(2)向细胞液中加入1mL Trizol溶液(Invitrogen,Caarlsbad,CA,USA),吹打混匀使细胞充分裂解,静置5min;加入200μL氯仿,剧烈振荡混匀30s,使水相和有机相充分解除,室温静置2min。
(3)4℃下12000g离心10min,可见分为三层,其中RNA在上层水相中,转移至新的RNase-free EP管中;加入等体积异丙醇,轻柔地充分混匀,于-20℃静置10min。
(4)4℃下12000g离心10min,收集RNA沉淀,取上清,用75%乙醇洗涤两次,超净台风干,加入20-60μL DEPC水溶解沉淀;总RNA的纯度控制OD260/OD280值在1.8-2.2,同时用1%琼脂糖凝胶电泳检测RNA的完整性,RNA置于-80℃保存。
(5)取1μg总RNA作为模板RNA,将RNA逆转录为cDNA,通过GenePharm(Shanghai,China)合成以下引物进行mRNA检测:AR1 (F,5'-CCAGGGACCATGTTTTGCC-3';R,5'-CGAAGACGACAAGATGGACAA-3'),AR2(F,5—GACGACCAGATGGCTGTCATT-3';R,5'-ggggagtagagcaTCCT-3'),STEP1(F,5'-actgggacaatacacgcat-3';R,5'-ggtggtcttcccaaccat-3'),FKBP5(F,5'-CTACACCTGCTGAAGGGACG-3',R,5'-CTCCAGCAAACCCTGGTACA-3')和GAPDH(F,5'-TGAAGGTCGGAGTCAACGG-3';R,5'-cctggagagagggggagggg-3'),以GAPDH为内对照。qRT-PCR反应过程按照Power SYBR GreenPCR Master Mix说明书操作;通过融解曲线分析和电泳确定目的条带,采用2-∆∆Ct法测定mRNA的相对表达量。结果如图3所示。结果显示,LNCap-AI细胞中AR1、AR2和FKBP5的mRNA水平明显高于正常LNCap细胞,而STEAP1则无显著变化。
前列腺特异性抗原(PSA)的表达水平是识别雄激素非依赖性现象的另一关键标志之一,对此,采用Western blot对LNCaP和LNCaP-AI细胞内PSA以及AR表达水平进行检测,具体步骤如下:
(1)分别收集对数生长期的LNCaP和LNCaP-AI细胞,弃掉原培养基,加入PBS清洗2遍。
(2)加入细胞裂解液(含PMSF),于冰上裂解30min。
(3)用细胞刮刮落细胞,转移至灭菌离心管中,于4℃、12000rpm离心10min,取上清,获得细胞裂解液提取细胞总蛋白。采用BCA蛋白定量法测量各组蛋白浓度(Thermo)。使用细胞裂解液将各组蛋白稀释至等浓度,按照体积比4:1加上5×上样缓冲液进行混合,98℃变性5min。
(4)提取总蛋白20μg,10%SDS-PAGE凝胶电泳(100V,90min),转膜以恒流300mA,转移100min至PVDF膜;5%BSA室温摇床封闭2h,孵育一抗(1:1000)4℃过夜(CST),TBST洗涤3次,每次5min,封闭液稀释二抗(1:20000)后,常温下孵育1h;TBST洗涤3次,每次7min,ECL发光试剂盒(Santa Cruz)在曝光仪中进行曝光,以内参GAPDH为对照,采用凝胶图像处理系统(Image-Pro Plus 6.0)分析条带净光密度值。
结果如图4所示。结果显示,LNCaP-AI细胞中PSA和AR表达水平均明显高于正常LNCaP细胞,差异具有统计学意义(**p<0.01)。在细胞培养0d、1d、3d、5d时分别取样重复上述Western blot实验,结果如上述一样,LNCaP-AI细胞中PSA表达水平显著高于正常LNCaP细胞,且该表达水平的差异在一定程度上与培养时间呈现正相关(参见图5),差异具有统计学意义(**p<0.01)。
恩杂鲁胺(Enzalutamide)作为一种雄激素受体抑制剂,临床上用于前列腺癌的治疗。对此,分别于LNCaP和LNCaP-AI细胞中加入恩杂鲁胺,通过ELISA检测细胞中AR和HK2蛋白的表达情况。结果如图6-7所示。结果显示,经恩杂鲁胺处理的正常LNCaP细胞,其中AR和HK2表达水平均得到了明显抑制;而LNCaP-AI细胞则对恩杂鲁胺不敏感,相较于阴性对照组,经恩杂鲁胺处理的LNCaP-AI细胞中无论是AR还是HK2的表达水平均无明显差距。由此可以明确去势抵抗性前列腺癌往往会对传统前列腺癌治疗药物缺乏敏感性,即常规的前列腺癌药物无法对去势抵抗性前列腺癌进行正常治疗。
实施例2 灵芝酸A对LNCaP-AI细胞的影响
首先考察灵芝酸A对LNCaP-AI细胞增殖的影响,具体包括如下步骤:
(1)待LNCaP-AI细胞生长至对数期后,胰酶消化并计数,按各种细胞的倍增时间选择合适的细胞密度,接种到96孔板中,分为2个组,其中实验组细胞给予灵芝酸A处理,浓度为20μM,灵芝酸A的结构式如图8所示;对照组(Control组)给予等体积溶剂处理,每组设置4个重复。
(2)于37℃培养箱中进行培养,并分别于0h、24h、72h以及120h收集细胞,每孔加10μL CCK-8,将培养板在培养箱内孵育1h,测定450nm吸光度,评估细胞的增殖状况。
实验结果如图9所示。结果显示,灵芝酸A能够有效抑制LNCaP-AI细胞的增殖(***p<0.001,vs Control组)。随后通过ELISA实验检测LNCaP-AI细胞中PSA的表达情况,结果发现,灵芝酸A能够显著抑制LNCaP-AI细胞中PSA的表达水平(参见图10)(***p<0.001,vsControl组)。
接着,利用LNCaP-AI细胞进行细胞划痕实验,具体步骤如下:
(1)对处于对数生长期的LNCaP-AI细胞进行消化,细胞计数2×105个/mL种在6孔板中,待细胞密度长到95%以上时进行划痕实验,实验前进行血清饥饿4h。
(2)用10 μL的移液管枪头垂直孔板划线,形成一条狭窄的划痕,用2mL PBS清洗掉的细胞,共3次;将细胞分为2组,其中实验组中加入20μM的灵芝酸A,对照组(Control组)中加入等体积溶剂;继续于CO2培养箱(37℃,5% CO2)中培养24h。
(3)拍照记录0h、48h的细胞迁移情况,用ImageJ软件进行数据处理。
结果如图11所示,结果显示,在无灵芝酸A处理的对照组中,48h内划痕闭合迅速。而在经灵芝酸A处理后,LNCaP-AI细胞划痕闭合速度明显降低,差异具有统计学意义(**p<0.01,vs Control组)。由此可见确定,灵芝酸A能够有效降低去势抵抗性前列腺癌细胞的迁移能力。
随后,通过集落形成实验研究灵芝酸A对去势抵抗性前列腺癌的影响,具体步骤如下:
(1)取对数生长期的LNCaP-AI细胞,分别用0.25%胰蛋白酶消化并吹打成单个细胞,并把细胞悬浮在10%胎牛血清的DMEM培养液中备用。
(2)将细胞悬液后稀释, 按2000/孔的密度接种于含10mL 37℃预温培养液的6孔板中,并轻轻转动,使细胞分散均匀,置37℃ 5% CO2及饱和湿度的细胞培养箱中培养。
(3)培养48h至培养皿中出现肉眼可见的克隆时,终止培养,弃去上清液,用PBS小心浸洗2次,每孔加入含0.5%结晶紫的甲醇1mL,染色30min;弃甲醇,用水清洗残留的甲醇;即可观察到细胞克隆;显微镜下观察,细胞数>50个才计为一个有效克隆。
结果如图12所示。结果显示,与对照组(Control组)相比,灵芝酸A能够显著抑制去势抵抗性前列腺癌细胞的集落形成,差异具有统计学意义(**p<0.01,vs Control组)。
进一步地,采用LNCaP-AI细胞进行Transwell实验,具体步骤如下:
(1)于-20℃取出Matri-gel置于4℃过夜水化。
(2)采用无血清培养液稀释水化后的Matri-gel(Matri-gel:无血清培养基为1:4),混匀后置于冰上备用,用200μL枪头吸取70µl稀释后的Matri-gel,枪头垂直对准侵袭上室中间小心加入Matri-gel,轻轻画圈,使胶铺满小室底部。
(3)将铺好Matri-gel胶的小室轻轻放入37℃培养箱,放置30-40min,使Matri-gel聚合成凝胶。
(4)实验前将LNCaP-AI细胞血清饥饿4-6h;随后取出细胞进行细胞消化计数,用DPBS洗涤细胞一次,用500μL无血清培养基将细胞重悬、调整细胞密度至1×105/mL。
(5)取出铺好胶的Transwell小室,于小室下部加入600μL含10% FBS的培养基;轻轻在胶上加入200μL细胞悬液(即2×104个细胞)。
(6)在细胞培养箱中培养24h后,吸去上室内培养基,用DPBS湿润棉签,用棉签轻轻擦去上室内的细胞,用DPBS洗涤上室,吸取多余细胞。
(7)采用4%多聚甲醛固定细胞20min,0.1%结晶紫染色20min,ddH2O清洗至背景清晰。
(8)镜下随机选取3个视野,进行拍照。
结果如图13所示,结果显示,与无灵芝酸A处理的正常LNCaP-AI细胞对比,在经灵芝酸A处理后,LNCaP-AI细胞迁移和跨膜速度明显降低,差异具有统计学意义(*p<0.05,vsControl组)。由此可见确定,灵芝酸A能够有效降低去势抵抗性前列腺癌细胞的迁移和侵袭能力。
实施例3 灵芝酸A对去势抵抗性前列腺癌作用机制研究
前述实验中证明了灵芝酸A能够有效抑制去势抵抗性前列腺癌细胞的增殖、克隆形成、迁移以及侵袭。对此,本实施例对其作用机制进行了深入探索。首先采用实施例1中方法对经灵芝酸A处理后的LNCaP-AI细胞(GAA组)以及未作任何处理的LNCaP-AI细胞(Control组)进行RNA-seq测序,以对其主成分(PCA)进行分析。分析结果显示,GAA组和对照组的3个样本聚类良好,重复性好(参见图14)。两组中共检测出15759个基因,鉴定出存在824个差异表达基因,其中618个基因表达上调,206个基因表达下调(参见图15)。
随后对上述824个差异表达的基因进行GO分析和KEGG分析,以明确最为相关的信号通路,结果如图16-17所示。结果显示,PI3K-AKT信号通路参与了GAA的作用过程;另一方面,GSK-3β/β-catenin调控与去势抵抗性前列腺癌可能存在相关性。对此,通过对GAA组和Control组细胞进行Western blot来明确AKT/GSK-3β信号是否通过调节β-catenin水平来调节去势抵抗性前列腺癌。结果如图18所示,GAA处理后可以显著下调LNCaP-AI细胞内AKT和GSK-3β的磷酸化水平,同时抑制β-catenin水平。
为了检测典型的β-catenin信号活性,根据试剂盒的说明书,将LNCaP-AI细胞转染pRL-renilla(Promega Corp.,USA)载体或2μg具有野生型TCF结合位点的Topflash荧光素酶载体和具有突变型TCF结合位点Fugene HD(Roche Applied Sciences,USA)的Topflash荧光素酶载体。4小时后,用GAA处理细胞48小时(对照组采用等体积溶剂处理)。裂解细胞后用双荧光素酶测定系统(Promega)测定荧光素酶活性。检测结果如图19所示。结果显示,GAA可以抑制LNCaP-AI细胞中β-catenin活性。
进一步地,通过Western blot对GAA组和Control组细胞内AKT/GSK-3β/β-catenin信号通路的靶基因c-Myc、Cyclin D1、MMP-2表达水平进行检测。结果表明,GAA能够显著抑制LNCaP-AI细胞内c-Myc、Cyclin D1、MMP-2的表达水平,同时下调了AR的表达水平(参见图20),其与上述RNA-seq结果相一致。由此可以明确GAA可通过AKT/GSK-3β/β-catenin信号通路以调控去势抵抗性前列腺癌的功能和活动。
实施例4 灵芝酸A对去势抵抗性前列腺癌的体内作用研究
前述实验在体外验证了灵芝酸A能够有效抑制去势抵抗性前列腺癌细胞增殖、克隆形成、迁移以及侵袭等功能。为了进一步明确灵芝酸A对于去势抵抗性前列腺癌的功能,进一步通过体内实验加以验证,具体步骤如下:
(1)实验前一天,将已经分装好的Matrigel基质胶从-20℃提前放入4℃冰箱过夜,使其从固体状态融化为液体状态。
(2)取4周龄Balb/c-nu小鼠,分别于右背部皮下注射LNCaP-AI细胞;待成瘤后将小鼠随机分为2组,记为组1和组2,每组5只。其中组1给予灵芝酸A处理,20mg/kg,每隔天注射一次;组2给予相同体积的PBS进行处理,每隔天注射一次。
(3)每日观察小鼠生长及精神状况,待成瘤后,定期测量各组小鼠的肿瘤大小,计算肿瘤体积,其中肿瘤体积采用如下公式计算:Volume(mm3)=Length(mm)×Width2(mm2)/2。
(4)在30天后处死小鼠,剥离各组小鼠肿瘤并拍照。
实验结果如图21-22所示。结果显示,与对照组(组2)相比较,在通过注射灵芝酸A(GAA,组1)进行处理后,无论是肿瘤的生长速度还是肿瘤的生长体积明显得到有效的控制(*p<0.05,****p<0.0001,vs Control组);表明灵芝酸A能够有效抑制裸鼠去势抵抗性前列腺癌细胞异种移植瘤生长,当抑制MIR210HG的表达时,则能够显著抑制肿瘤的生长,该结果与前述体外实验结果相一致。
现有技术中已经明确了Wnt/β-catenin通路的异常激活与多种癌症密切相关。大量研究发现,包括肝癌、结直肠癌、前列腺癌等在内的多种恶性肿瘤中Wnt/β-catenin通路会发生异常,使得该通路成为了上述癌症治疗的关键靶点。然而进一步的研究发现,以Wnt/β-catenin信号通路作为靶点进行治疗的一系列前列腺癌药物例如恩杂鲁胺、多西他赛等,在对去势抵抗性前列腺癌治疗时,会出现明显的耐药性导致治疗效果急剧下降甚至无效,在对CRPC患者肿瘤样本进行分析后发现,其肿瘤细胞内Wnt/β-catenin信号通路明显激活。由此可以明确,通过对Wnt/β-catenin信号通路进行抑制来治疗前列腺癌的常规药物已经无法用于去势抵抗性前列腺癌的治疗,其原因在于CRPC的相关发生发展机制可能已经发生了明显改变。
而CRPC的发病机制是目前研究的难点和热点。研究人员对此提出了三种可能的CRPC形成机制:(1)AR相关机制。AR信号通路的激活起着重要作用;(2)干细胞形成机制。干细胞生长不依赖于AR, ADT治疗后可发展为CRPC;(3)神经内分泌转化机制。ADT治疗可诱导前列腺癌细胞神经内分泌分化(NED)。前列腺癌NED细胞不再依赖AR信号通路,通过旁分泌和自分泌作用促进前列腺癌细胞的生长。然而,有关CRPC进展的确切机制目前仍尚不清楚。
在本发明中,通过构建雄激素非依赖性细胞系LNCaP-AI来模拟CRPC,并对LNCaP-AI细胞模型的相关特性进行了验证。后续的研究发现,经GAA处理后,LNCaP-AI细胞的增殖、PSA分泌、克隆形成、创面愈合、迁移和侵袭能力均明显受到抑制,且GAA可在体内显著抑制去势抵抗性前列腺癌细胞的生长。通过RNA-seq测序分析鉴定出824个差异表达基因,发现LNCaP-AI细胞中AKT、GSK-3β等的异常激活可以被GAA进行有效抑制;通过Top-flash荧光素酶测定确定了活化的β-catenin也可以被抑制。由此明确了AKT/GSK-3β/β-catenin通路参与了GAA对去势抵抗性前列腺癌的作用过程。这些发现表明GAA可能通过AKT/GSK-3β/β-catenin通路抑制前列腺癌的CRPC进展。总的来说,本发明明确了灵芝酸A对于去势抵抗性前列腺癌的治疗活性,并具体研究了其相关作用机制,为延缓前列腺癌ADT后向CRPC转化的进程、治疗去势抵抗性前列腺癌提供了新的活性物质,为后续去势抵抗性前列腺癌药物的开发提供了新的治疗靶点和方向。

1.灵芝三萜类化合物在制备治疗去势抵抗性前列腺癌的药物中的应用。
2.根据权利要求1所述的应用,其特征在于,所述灵芝三萜类化合物选自灵芝酸A。
3.灵芝三萜类化合物在制备延缓前列腺癌患者治疗后去势抵抗性前列腺癌出现的药物中的应用。
4.根据权利要求3所述的应用,其特征在于,所述灵芝三萜类化合物选自灵芝酸A。
5.根据权利要求3所述的应用,其特征在于,所述治疗选自雄激素剥夺治疗。
6.一种用于治疗去势抵抗性前列腺癌的药物组合物,其特征在于,包括灵芝三萜类化合物和至少一种抗肿瘤药物。
7.根据权利要求6所述的药物组合物,其特征在于,所述灵芝三萜类化合物选自灵芝酸A。
8.根据权利要求6所述的药物组合物,其特征在于,所述抗肿瘤药物选自多西他赛、卡巴他赛、阿比特龙、恩杂鲁胺中的一种或多种。
9.一种用于治疗去势抵抗性前列腺癌的药物制剂,其特征在于,包括灵芝三萜类化合物、至少一种抗肿瘤药物以及药学上可接受的载体。
10.根据权利要求9所述的药物制剂,其特征在于,所述灵芝三萜类化合物选自灵芝酸A。
