FUM诱发的真菌毒素中毒症的预防性治疗目前限于良好的农业实践以减少作物上的真菌毒素产生与食品和饲料商品的控制程序以确保真菌毒素水平保持低于某些限度。

发明内容
为了满足本发明的目的,已经发现偶联的伏马毒素(FUM)适用于保护动物免受FUM诱发的真菌毒素中毒症的方法。发现不需要将FUM转化为类毒素,偶联的毒素似乎对接受治疗的宿主动物是安全的。而且,令人惊讶地发现,在治疗后,针对诸如真菌毒素的小分子诱导的免疫应答强到足以保护动物自身免受摄入真菌毒素后的真菌毒素中毒症。对于任何真菌毒素,通过在动物中诱导针对真菌毒素本身的免疫应答的动物的这种实际保护在本领域中还没有展示。
定义

佐剂是非特异性免疫刺激剂。原则上,能够支持或放大免疫事件级联中的特定过程,最终导致更好的免疫应答(即,对抗原的综合身体应答,特别是由淋巴细胞介导的并且通常涉及特异性抗体或先前致敏的淋巴细胞对抗原的识别的应答)的每种物质可被定义为佐剂。佐剂对于所述特定过程的发生通常是不被需要的,而只是有利于或放大所述过程。佐剂通常可以根据它们诱导的免疫学事件进行分类。第一类,包含i.a.ISCOM(免疫刺激复合物)、皂苷(或其馏分和衍生物如Quil A)、氢氧化铝、脂质体、耳蜗酸盐(cochleate)、聚乳酸/乙醇酸、促进APC(抗原呈递细胞)的抗原摄取、转运和呈递。第二类,包含i.a.油乳液(W/O、O/W、W/O/W或O/W/O)、凝胶、聚合物微球(Carbopol)、非离子嵌段共聚物和最可能还有氢氧化铝,提供储存效应(depot effect)。第三类,包含i.a.富含CpG的基序、单磷酰脂质A、分枝杆菌(胞壁酰二肽)、酵母提取物、霍乱毒素,是基于保守微生物结构的识别,因此被称为病原体相关的微生物模式(PAMP),定义为信号0。第四类,包含i.a.油乳液表面活性剂、氢氧化铝、缺氧,是基于刺激免疫系统在危险和无害(其不必与自身和非自身相同)之间的区分能力。第五类,包含i.a.细胞因子,是基于共刺激分子信号2在APC上的上调。
在本发明的意义上,疫苗是适于施用于动物的组合物,其包含免疫学有效量的一种或多种抗原(即,能够充分刺激靶标动物的免疫系统以至少减少用疾病诱发剂激发的负效应,通常与药学上可接受的载体(即,生物相容性介质,即在施用后不在受试动物中诱导显著不良反应的介质,能够在施用疫苗后将抗原呈递至宿主动物的免疫系统)组合,例如含有水和/或任何其它生物相容性溶剂的液体或固体载体,例如通常用于获得冻干疫苗(基于糖和/或蛋白质)。任选地包含免疫刺激剂(佐剂),其在施用给动物后诱导用于治疗疾病或病症的免疫应答,即辅助预防、改善或治愈疾病或病症。
本发明的进一步实施方式
在本发明的进一步实施方式中,将偶联的FUM全身性施用于动物。尽管局部施用(例如通过胃肠道(口腔或肛门腔)或眼中(例如当免疫鸡时)的粘膜组织),已知是在各种动物中诱导免疫应答的有效途径,但发现全身性施用会引发足够的免疫应答,以保护动物免受FUM诱发的真菌毒素中毒症。特别发现,通过肌内、口服和/或皮内施用可以获得有效的免疫。
尽管优选在动物能够摄取被大量FUM污染的饲料之前进行施用,但施用年龄不是关键的。因此,施用时的优选年龄为6周或更年幼。进一步优选4周龄或更年幼,例如1-3周龄。
在本发明的另一个实施方式中,将偶联的FUM施用于动物至少两次。尽管许多动物(特别是猪、小鸡、反刍动物)通常仅通过一次注射免疫原性组合物就易于免疫,但被认为,为了经济可行的抗FUM的保护,优选两次注射。这是因为实际上动物的免疫系统不会通过自然暴露于FUM而被触发产生抗FUM抗体,这仅仅是因为自然发生的FUM不是免疫原性的。因此,动物的免疫系统完全依赖于偶联的FUM的施用。偶联的FUM的两次注射之间的时间可以是1周和1-2年之间的任何时间。对于幼年动物,被认为初次免疫方案(例如在1-3周龄时),1-4周后,通常1-3周后,如2周后,接着加强施用就足够了。年长的动物可能需要每几个月加强施用(如最后一次施用后4、5、6个月),或在每年或每半年一次的基础上加强施用,如从其它商业应用的动物免疫方案知晓的那样。
在另一个实施方式中,将偶联的FUM在除了偶联的FUM之外还包含佐剂的组合物中使用。如果偶联物自身不能诱导免疫应答以获得预定水平的保护,则可以使用佐剂。尽管已知偶联物分子能够在没有另外的诸如KLH或BSA的佐剂情况下充分刺激免疫系统,但使用另外的佐剂可能是有利的。这可以消除对加强施用的需要或延长其施用间隔。所有这些取决于在特定情况下需要的保护水平。当使用偶联的FUM作为免疫原时显示出能够并诱导针对FUM的良好免疫应答的一类佐剂是水和油的乳液,例如油包水乳液或水包油乳液。前者通常用于家禽,而后者通常用于更容易发生佐剂诱导的部位反应的动物,例如猪和反刍动物。
在另一个实施方式中,偶联的FUM包含与分子量高于10,000Da的蛋白质偶联的FUM。已经发现这些蛋白质,特别是匙孔血蓝蛋白(KLH)和卵清蛋白(OVA),能够在动物中,特别是在猪和鸡中诱导足够的免疫应答。蛋白质的实际上限可以是100MDa。
关于抗真菌毒素中毒症的保护作用,特别发现,被认为使用本发明动物得到了保护免受平均日增重的减少,肺水肿,肝、心和肾损伤,从而免受由FUM诱发的一种或多种这些真菌毒素中毒症体征。
现在将使用以下实施例进一步解释本发明。
本发明的实施例
在第一个系列的实验中(参见实施例1-4),评估了使用偶联的真菌毒素是否可以引起针对真菌毒素的主动免疫应答,如果是可以引起,则能够保护接种的动物在摄取该真菌毒素后免受由该真菌毒素诱发的病症。对于后者,使用了DON激发的猪模型。此后(实施例5),评估了在疫苗中使用偶联的FUM是否可以在接种的动物中诱导抗伏马毒素的抗体。
实施例1:使用偶联的DON的免疫激发实验
目的
本研究的目的是评价偶联的脱氧雪腐镰刀菌烯醇保护动物免受由DON摄取引起的真菌毒素中毒症的功效。为了检测这一点,在用毒性DON激发之前用DON-KLH免疫猪两次。使用了不同的免疫途径来研究施用途径的影响。
研究设计
来自8只母猪的40只1周龄猪被用于本研究,被分成5组。第1-3组的24只仔猪在1和3周龄时免疫两次。第1组在两个年龄时都进行了肌内(IM)免疫。第2组在1周龄时接受了IM注射,在3周龄时接受了口服加强。第3组皮内(ID)免疫两次。从51/2周龄起,用在液体中口服施用的DON在4周期间激发第1-3组。第4组未免疫,但如第1-3组所述仅用DON激发。第5组作为对照,仅从5.5周龄开始接受对照液体,持续4周。
液体制剂中的DON浓度对应于5.4mg/kg饲料的量。这对应于每天平均2.5mgDON的量。激发四周后,对所有动物进行尸检研究,特别注意肝、肾和胃。此外,在研究的第0天、第34天、第41天、第49天、第55天、第64天(安乐死后)进行血液采样,除了第5组仅在第0天、第34天、第49天以及安乐死后直接进行血液采样。
试验品
配制三种不同的免疫原性组合物,即试验品1,其包含在用于IM免疫的注射用水包油乳液(X-solve50,MSD AH,Boxmeer)中50μg/ml的DON-KLH;试验品2,其包含在用于口服免疫的油包水乳液(GNE,MSD AH,Boxmeer)中50μg/ml的DON-KLH以及试验品3,包含在用于ID免疫的注射用水包油乳液(X-solve50)中500μg/ml的DON-KLH。
将激发脱氧雪腐镰刀菌烯醇(获取自Fermentek,Israel)以100mg/ml的终浓度稀释在100%甲醇中并在<-15℃下储存。在使用之前,将DON被进一步稀释并提供在施用的治疗剂中。
入选标准
仅使用健康动物。为了排除不健康的动物,在研究开始之前检查所有动物的一般身体外观和没有临床异常或疾病。每组使用来自不同母猪的仔猪。在日常实践中,所有动物都将被免疫,即使是当通过摄入DON污染的饲料预先暴露于DON时。因为DON本身不引起免疫应答,所以认为在预先暴露于DON的动物和未接触过DON的动物之间没有原则性的区别。
结果
没有动物具有与用DON-KLH免疫相关的负面影响。因此,该组合物似乎是安全的。
在实验开始时,所有猪的抗DON滴度均为血清阴性。在激发期间,肌内免疫组(第1组)和皮内免疫组(第3组)产生了抗DON的抗体应答,这是通过用原生DON-BSA作为包被抗原的ELISA所测定的。表1描述了研究期间4个时间点上的平均IgG值及其SD值。肌内免疫和皮内免疫均诱导了显著的抗DON滴度。

如表2所示,所有免疫动物,包括组2中显示无显著抗DON IgG滴度增加的动物,在第一个15天期间与激发动物相比显示出显著更高的增重。对于被激发的动物,所有动物在研究过程中都增加了更多的重量。

还监测了小肠的状况(由空肠中的绒毛/隐窝比率确定)。在表3中描述了绒毛/隐窝比率。可以看出,第3组中的动物具有与健康对照(第5组)相当的平均绒毛隐窝/隐窝比率,而未免疫的激发组(第4组)具有低得多的(统计学显著的)绒毛隐窝比率。此外,与未免疫的激发对照组相比,第1组和第2组具有显著更好(即更高)的绒毛/隐窝比率。这表明免疫防止了由DON引发的肠损伤。

还监测了其他器官的一般状况,更具体地是肝、肾和胃。观察到所有三个测试组(第1-3组)比未免疫的激发对照组(第4组)的健康状况更好。在表4中描述了一般健康数据的总结。胃溃疡的程度报告为-(没有溃疡形成的证据)至++(多发溃疡)。胃炎症的程度从-(没有炎症的证据)至++/-(胃炎症的开始)报告。

实例2:免疫对DON水平的影响
目的
本研究的目的是评价用DON偶联物免疫对DON摄取的毒代动力学的影响。为了检测这一点,在饲喂毒性DON之前用DON-KLH免疫猪两次。
研究设计
本研究使用10只3周龄的猪,其被分成2组,每组5只。第1组的猪在3周和6周龄时用DON-KLH(试验品1;实施例1)进行了两次IM免疫。第2组作为对照,仅接受对照液。在11周龄时,通过大丸剂以0.05mg/kg的剂量向每只动物施用DON(Fermentek,Israel),其(基于每日饲料摄取)类似于1mg/kg饲料的污染水平。在DON施用前和DON施用后0.25、0.5、0.75、1、1.5、2、3、4、6、8和12小时取猪的血样。
入选标准
仅使用健康动物。
血浆DON分析
在与TQ-SMS仪器(Waters,Zellik,Belgium)连接的/>UPLC系统上使用经过验证额LC-MS/MS方法进行未结合DON的血浆分析。使用该方法的猪血浆中DON的定量下限为0.1ng/ml。
毒代动力学分析
通过非区室分析(Phoenix,Pharsight Corporation,USA)对DON的血浆浓度-时间曲线进行毒代动力学建模。计算以下参数:从时间零点至无穷大的曲线下面积(AUC0→∞)、最大血浆浓度(Cmax)和最大血浆浓度下的时间(tmax)。
结果
毒代动力学结果如下表5所示。可以看出,用DON-KLH免疫降低了所有毒代动力学参数。由于未结合的DON负责发挥毒性作用,因此可以得出结论,用DON-KLH免疫将通过减少动物血液中未结合的DON的量来减少由DON引起的毒性作用。

研究设计
本研究使用18只3周龄的猪,其被分成3组,每组6只。第1组的猪在3和5周龄时用DON-KLH肌内免疫两次(使用实施例1的试验品1)。第2组相应地用DON-OVA免疫。第3组作为阴性对照。在3周龄、5周龄和8周龄时检查所有动物的抗DON IgG应答。
结果
血清学结果在下表中以log2抗体滴度表示。

实例4:鸡的血清学应答
目的
本研究的目的是评价DON-KLH在鸡中的血清学应答。
研究设计
本研究使用了30只4周龄的鸡,其被分成三组,每组10只鸡。用DON-KLH肌内免疫鸡。第1组用作对照,仅接受PBS。第2组接受没有任何佐剂的DON-KLH,第3组接受GNE佐剂配制的DON-KLH(购自MSD Animal Health,Boxmeer)。在第0天用0.5ml疫苗注入右腿进行初次免疫。在第14天,小鸡在左腿接受了类似的加强免疫。
在第0天和第14天以及第35天、第56天、第70天和第84天进行血液取样。分离血清用于测定抗DON的IgY。在第0天和第14天,血样在免疫前被分离。
结果
血清学结果以log2抗体滴度表示在表7中。PBS背景已从数据中减去。

可以看出,偶联的DON还在鸡中诱导抗DON滴度。GNE佐剂显著增加应答,但似乎对于获得这样的净应答不是必需的。
实例5:猪对FUM偶联物的血清学应答
目的
本实验的目的是评价在疫苗中使用偶联的FUM是否可以在接种的动物中诱导抗伏马毒素的抗体。
研究设计
为此,使用了包含与匙孔血蓝蛋白偶联的伏马毒素B1(FUM-KLH)的疫苗。将偶联物与水包油乳液佐剂(XSolve 50,MSD Animal Health,The Netherlands)混合,其终浓度为50μg/ml(用于肌内(IM)施用)或500μg/ml(用于皮内(ID)施用)。
在该实验中,也将如上所述的DON疫苗用作阳性对照。除此之外,配制和使用了具有其他偶联的真菌毒素的疫苗。特别地,将与匙孔血蓝蛋白偶联的玉米赤霉烯酮(ZEA)(ZEA-KLH)和与KLH偶联的T-2真菌毒素(T2-毒素)(T2-KLH)配制成疫苗。将偶联物与如上所述的水包油乳液佐剂(XSolve)混合。针对ZEA-KLH和DON-KLH,其终浓度分别为50μg/ml(用于肌内(IM)施用)或500μg/ml(用于皮内(ID)施用);针对T2-KLH,其终浓度分别为115μg/ml(IM)或1150μg/ml(ID)。
在实验中,6组5只动物(猪)在三周龄时用于接种,第1组皮内接种0.2ml FUM-KLH两次,第2组接受0.2ml ZEA-KLH两次,第3组用2.0ml DON-KLHIM的X-solve 50溶液接种两次,第4组IM接种2.0ml FUM-KLH两次,第5组IM接种2.0ml ZEA-KLH两次,最后第6组IM接种2.0ml T2-KLH两次。对照组有三只仔猪,对照组未接受疫苗接种。所有的初次接种均在3周龄时,加强接种均在5周龄时。研究开始后监测动物14周。
结果
在实验开始时,所有猪抗FUM、ZEA、T2和DON的滴度为血清学阴性,并且所有接种组产生抗体滴度。所得log2滴度示于下表8中。

实例6:在鸡中针对FUM偶联物的血清学应答
目的
本实验的目的是评价在疫苗中使用偶联的FUM是否能在鸡中诱导抗伏马毒素的抗体。
研究设计
为此,根据实施例5使用了包含与匙孔血蓝蛋白偶联的伏马毒素B1(FUM-KLH)的疫苗。使用与实施例5中使用的相同的矿物油将偶联物与油乳液佐剂混合,并且作为替代方案,使用非矿物油的类似乳液,两者的终浓度均为50μg/ml。
研究中使用一组15只鸡。使用了三组,每组5只动物。第1组用作阴性对照并施用PBS溶液,第2组用混合在含矿物油的佐剂中的FUM-KLH接种,第3组用不含矿物油的佐剂接种。在T=8和T=22时,用0.5ml疫苗肌内接种鸡(为了适应环境,在T=0时禽被纳入研究中)。
结果
在实验开始时,所有鸡的抗FUM滴度均为血清阴性,并且所有接种组均产生抗体滴度。所得log2滴度示于下表9中。可以看出,在两组中,针对偶联的伏马毒素,都可以产生高水平的抗体。这支持了共同的理解,即佐剂的类型对于引起足够的免疫应答本身不是必需的。

实例7:猪中针对FUM激发的保护
目的
本实验的目的是评估疫苗中使用偶联的FUM是否能在猪中诱导针对伏马毒素激发的保护。
研究设计
为此,使用了在两种不同佐剂(如实施例6所述,一种基于矿物油,另一种基于非矿物油)中包含与匙孔血蓝蛋偶联的伏马毒素B1(FUM-KLH)的相同疫苗。在本研究中,使用了一组24只猪。第一组8只仔猪用FUM-KLH接种,尽管第一亚组4只动物接受基于含矿物油佐剂的疫苗,第二亚组接受备选疫苗。两种疫苗以2ml的量以50μg/ml的浓度肌内施用。在7-12日龄(T=0)时对动物进行初次免疫接种,在21-26日龄(T=14)时对动物进行加强免疫接种。第2组未接种疫苗,但用伏马毒素B1激发并作为阳性对照。第3组未接种且未激发,用作阴性对照。16只激发的仔猪(第1组和第2组)在约5.5周龄时每天接受13mg/kg FUM饲料,连续4周,相当于5.99mg/天。FUM以液体制剂施用:在第一周,仔猪接受在16ml液体中接受2.41mgFUM/天;在第二周,在32ml液体中接受5.0mg/天;在第三周,在45ml液体中接受7.2mg/天;在第四周,在60ml液体中接受9.3mg FUM/天。随时间监测抗体滴度。在研究结束时,进行肝、肺、肾和肠评估。
结果
在实验开始时,所有仔猪的抗FUM滴度均呈血清学阴性。在激发期间,用FUM-KLH接种的疫苗产生抗FUM的抗体应答,如表10所示,其显示了研究期间6个时间点上的IgG值。

表11描述了在激发期间,较之激发的起始重量,每组动物的增重百分比(%),此外还描述了对特定器官有损伤的动物的增重%。这均显示偶联的伏马毒素可以成功地用于保护动物免受FUM诱发的真菌毒素中毒症的方法中。
