背景技术
古树是指树龄达到百年以上的大树,是承载历史文化的重要文脉,对于研究城市建设、社会历史进程及园林发展等方面都具有很高的价值。近年来,随着我国城市地区的规模迅速扩张,人口快速集聚,以致地下水污染、土壤污染、生态系统退化等“城市病”接踵而来,古树面对急剧失衡的城市生态环境,其生长条件受到严重影响和干扰,生存现状令人堪忧。宋路有等(2016)对上海闵行区松柏类古树及周围环境进行了调查,发现土壤矿质养分匮乏及配比失衡是造成部分古树生长衰弱的主要原因,因此创造良好的根际养分环境是保护古树和实施复壮措施的先决条件。
微生物是土壤中最活跃的组分,是评价土壤健康的重要指标。而丛枝菌根真菌是陆地生态系统中分布最为广泛、能够与约80%的高等植物(包括多数高大乔木)形成互利共生系统的菌根真菌。在这一系统中,AM真菌通过菌丝延伸到根系无法进入的土壤缝隙中吸收矿质养分,从而传递给宿主植物以换取自身所需碳源。AM真菌不仅在增强植物养分吸收、促进根系生长、提高植株抗逆性等方面兼具重要作用,还能对土壤产生诸多有益影响,如物理结构改善、有机碳固持、养分活化、空隙率保持等。在古树复壮过程中,添加菌根菌不仅可以降低根际土壤的pH、EC值,而且能促进树木对土壤中的速效氮、有效磷及微量矿质元素的吸收,起到较好的复壮效果。AM真菌具有作为生物肥料的极大潜力,可为促进古树生长提供一种新的经济、有效的生物学选择,但目前国内外关于AM真菌对古树生长、复壮方面的研究及应用鲜有报道。
土壤是影响古树生长的重要环境因子之一。目前古树根际土壤的研究主要集中在探讨土壤理化性质和养分元素与古树生长之间的关系、如何改善理化性质和养分含量来促进古树复壮等相关方面。汤珧华和范善华(2001)在对上海市古树名木的研究中发现,土壤通气性良好对于古树生长至关重要,当土壤容重在1.35g·cm-3以下、土壤有效孔隙度在10%以上时,才有利于古树根系生长。林大仪(2011)对古树根际土壤酸碱度的研究表明,土壤酸碱度直接影响到古树根际土壤中微生物和养分元素的分布和转移。蒲小鹏等(2011)发现在维持古树活力方面,可从增加土壤有机质含量,激活土壤磷酸酶活性和降低土壤pH等方面进行。
菌根在促进古树生长、提高抗病能力方面具有明显效果。金陈斌(2018)将菌根菌应用在白皮松和桧柏古树的复壮处理上,发现接种菌根菌对古树地上部和地下部的生长均有促进效果,尤其是根系活力得到了明显提升;并且接种菌根菌后,不仅可以增加土壤有机质含量,降低pH值,而且对N、P等营养元素也可以起到较好的调节作用。此外,美味牛肝菌(Boletue edulis)可以改善土壤微环境,促进古树根系生长,对古树的复壮产生明显效果。吴小芹等(2006)对黑松的复壮进行研究也表明,美味牛肝菌是黑松的优良外生菌根真菌,对黑松苗具有显著的促生作用。
银杏(Ginkgo biloba)是银杏科银杏属的唯一种,俗称白果、公孙树,在食用、药用和生态方面等均具有较大价值。目前,针对银杏根系是否存在AM真菌以及多样性如何仅有少量报道。经过前人对银杏根系的解剖、镜检,已确定银杏为AM真菌宿主植物。且在自然生长的银杏树和生产种植的实生银杏苗、嫁接银杏苗根系上都发现了丛枝菌根结构,这表明AM真菌侵染银杏是一种普遍现象。在研究中,Fontana还发现被侵染的银杏根系中存在的菌丝大多为胞内菌丝,而胞外菌丝非常稀少。此外,我国学者陈连庆和韩宁林(1999)对浙江地区银杏根际共生真菌资源进行调查,共发现4属9种AM真菌存在于银杏根际土壤中,且以球囊霉属(Glomus)最多,巨孢囊霉属(Gigaspora)次之。在对银杏应用AM真菌方面,吴强盛等发现自然田间条件下与人工接种Glomus epigaeum菌种的形态特征相似,且丛枝菌根在嫁接银杏苗根际的发育比实生苗明显更好。也有研究认为,银杏与AM真菌的共生关系较差,在银杏造林中应用丛枝菌根真菌的潜力不高,但银杏根际土著AM真菌的研究与应用未见报道。

本发明所要解决的技术问题为:提供一种促进银杏树生长的菌剂及其应用。

进一步地,以孢子数计,所述近明球囊霉(Claroideoglomus claroideum)Cc-01和摩西管柄囊霉(Funneliformis mosseae)Fm-01的配比为1∶1。

本发明利用古银杏根际土中筛选、培育的两株土著优势AM真菌近明球囊霉(Claroideoglomus claroideum)Cc-01和摩西管柄囊霉(Funneliformis mosseae)Fm-01,对银杏实生苗采用单接、双接处理,与不接菌处理进行比较,发现接种AM真菌对银杏的生长、光合、抗氧化系统、代谢具有显著影响。并通过隶属模糊函数综合法,比较发现不同处理对银杏实生苗的促生效应以双接处理效果最佳,具有协同效果,以此为选择促进古银杏复壮的接种方式提供一定试验依据。
与现有技术相比,本发明具有以下有益效果:

实施例1菌株的分离和鉴定
1、试验地点概况
试验地点位于上海市嘉定区、静安区,地处31°13′19′N~31°23′3″N,121°14′6″E~121°27′5″E之间,属北亚热带季风性气候,雨热同季,日照充足。该地区年均气温17.7℃,年均日照1809.2h,年均无霜期259d,全年降雨量为1388.8mm,且每年约80%的雨量集中在4~10月的汛期。
2、样品的采集与处理
本试验样品选自上海市嘉定区、静安区二级保护古银杏树(100~299年)和幼龄银杏(50年左右)共13株,测量株高、胸径、地径、冠幅等基本生长指标,记录地理位置及生境,采集细根和根际土壤作为试验样品。具体采样位置及信息见表1。
表1银杏生境调查和土壤采样点基本概况
采集土壤样品时,在目标树木滴水线附近按照东西南北4个方向各选取1个取样点,去除地表凋落物和植被后,用75%酒精消毒后的小铁铲挖至30cm深度(银杏吸收根主要分布深度),采集土壤和根系混合样品500g左右。将每棵树采集的四个土、根样混合,去除凋落物和石块后作为一个样品,进行编号标记并封口。
样品处理时将每个样品中的根系挑出洗净,与土壤分开保存,分别标记为:古银杏根(GYXG)、古银杏根际土(GYXT)、对照根(XYXG)、对照根际土(XYXT)。将土壤分为两份,一份自然风干后过2mm、0.149mm筛后用于土壤常规养分、理化性质测定,另一份暂存于-20℃冰柜保存用于AM真菌分离鉴定和诱导培养。
3、AM真菌鉴定
使用改良的湿筛倾析法分离土壤AM真菌孢子。在体视显微镜(奥特光学SZ680)下从湿筛分离物中挑选健康的AM真菌孢子,于光学显微镜(奥特光学SMART e500)下重点观察孢子的形态、大小、纹饰、连孢菌丝等明显特征。参考《中国丛枝菌根真菌资源与种质资源》、国际AM真菌保藏中心(http://fungi.invam.wvu.edu/)的鉴定方法以及孢子形态描述,对比最新分类系统,对获取的AM真菌进行分类与鉴定。
筛选出两株真菌菌株Cc-01和Fm-01。
菌株Cc-01的形态如下:
孢子果:未见。孢子:在土壤中单生,淡黄色,球形、近球形,表面常附有碎屑,直径50~110μm。内含物:丰富油状物。孢子壁:2层(L1和L2)。L1透明,易脱落,在Melzer's试剂中被染成粉红色;L2层状壁,淡黄色,厚4~6μm,在Melzer's试剂中无反应。连孢菌丝:单根,浅黄色,弯曲,基部宽7~13μm,菌丝壁与孢子壁呈连续结构,连孔小漏斗形。
对其18S rDNA AML1-AML2区域测序结果如SEQ ID No.1所示。
形态学结合18S rDNA分子测序结果,将菌株Cc-01鉴定为近明球囊霉(Claroideoglomus claroideum)。
2019年10月14日将近明球囊霉(Claroideoglomus claroideum)Cc-01保藏在北京市朝阳区北辰西路1号院3号的中国微生物菌种保藏管理委员会普通微生物中心,保藏编号CGMCC No.18593。
菌株Fm-01的形态如下:
孢子果:未见。孢子:在土壤中单生,幼时淡黄色,成熟时黄色,大多数球形,少数近球形,表面光滑,透亮可见内含物,直径100~210μm。孢子壁:3层(L1、L2和L3)。L1无色,易逝,厚1~1.5μm,在Melzer's试剂中被染成粉红色;L2无色透明,厚0.5~1μm;L3层积壁,淡黄色或黄色,厚2~3.5μm。内含物:丰富油状物。连孢菌丝:单根,弯曲,浅黄色,长8~20μm,厚约2μm,连孔漏斗形,菌丝内有隔。
对其18S rDNA AML1-AML2区域测序结果如SEQ ID No.2所示。
形态学结合18S rDNA分子测序结果,将菌株Fm-01鉴定为摩西管柄囊霉(Funneliformis mosseae)。
2023年05月17日,将摩西管柄囊霉(Funneliformis mosseae)Fm-01保藏于北京市朝阳区北辰西路1号院3号的中国微生物菌种保藏管理委员会普通微生物中心,保藏编号CGMCC No.40631。
实施例2真菌菌株接种银杏实验
1、AM真菌扩繁培养
参考王幼珊等(2012)的培养方法,通过单孢培养、扩繁的方式以获得大量有活力的AM真菌孢子和可作实际应用的纯化菌剂。本发明共采用2株供试AM菌种:近明球囊霉(Claroideoglomus claroideum)Cc-01(孢子密度60个/10g干土)、摩西管柄囊霉(Funneliformis mosseae)Fm-01(孢子密度15个/10g干土)。经高粱与沸石∶沙子=1∶1基质单孢培养扩繁,成功制出的2种含有孢子、菌丝体、侵染根段的菌剂。
2、培养基质
沸石∶河沙∶蛭石=1∶1∶1,用121℃高压灭菌30min去除土壤中的微生物。土壤基础理化性质为pH 7.5,EC值91.8,全氮(TN)0.28g/kg,全磷(TP)0.47g/kg,全钾(TK)11.46g/kg。
3、试验设计:
从银杏古树上选取健康的种子,在灭菌河沙内培养出实生苗。挑选长势相同的银杏种苗进行以下处理:
(1)CK:灭菌基质+银杏
(2)处理1(CC):灭菌基质+近明球囊霉+银杏
(3)处理2(FM):灭菌基质+摩西管柄囊霉+银杏
(4)处理3(CC+FM):灭菌基质+近明球囊霉+摩西管柄囊霉+银杏
根据不同处理方式,将银杏种苗栽种到27cm×18cm(内径×高)的塑料花盆中,种苗数量为每盆1株,5个重复,共20盆。每盆按照菌剂孢子密度进行换算接种AM真菌孢子600个,其中对照处理加入等量灭菌基质,处理3加入CC和FM孢子各300个。
4、研究结果
银杏实生苗地上部生长分析
由图1可知,银杏实生苗四个处理的株高、茎粗和叶面积在试验过程中均有增长,120d时CC+FM处理的茎粗已显著高于CK(p<0.05),而叶片数只有接菌处理CC、FM和CC+FM在30~60d时开始增长,CK叶片数在120d内未发生任何变化,与CC+FM呈极显著差异(p<0.01)。
其中,生长120d时株高总增长量最多的处理为CC,增长了1.12cm,茎粗、叶片数和叶面积总增长量最大的均为CC+FM,分别增长了1.04mm、4.4片和4.96cm2。而四个指标总增长量最少的均为CK处理,分别只增长了0.42cm、0.704mm、0片和1.16cm2,株高总增长量仅占CC总增长量的37.5%,茎粗、叶片数和叶面积生长总增长量占CC+FM总增长量的67.7%、0和23.4%。从单阶段增长量来看,CC株高在7~30d间增长最快,为0.51cm,茎粗单阶段增长量最多的则为CK处理7~30d的0.34mm,叶片数和叶面积单阶段增长最快的均为CC+FM,分别为60~90d间增长的1.8片和30~60d间增长的2.05cm2。而株高单阶段增长量最少的为CC和FM,均在90~120d间只增长了0.1cm,仅占CC单阶段增长量的19.6%。茎粗、叶片数和叶面积单阶段增长量最少的均为CK处理,分别只增长了60~90d间的0.11mm、7~120d间的0片和7~30d间的0.01cm2。
从地上部各项指标来看,接种AM真菌对银杏实生苗地上部生长的促进效果较明显,且以CC+FM的处理最优。
银杏实生苗相对叶绿素含量分析
相对叶绿素含量SPAD在一定意义上可以表示植物叶片的“绿色程度”。从不同处理各时期银杏实生苗相对叶绿素含量的动态变化图(图2)中,可见接种AM真菌对银杏实生苗的叶绿素SPAD有显著影响(p<0.05)。随着接菌时间的增长,未接菌银杏实生苗CK的相对叶绿素含量呈整体下降趋势,而接种AM真菌的三个处理CC、FM和CC+FM呈持续增长趋势,在生长60d时分别高于CK处理14.24、19.42和21.77,出现显著性差异(p<0.05),在生长120d时分别高于CK处理21.34、28.32、29.31,呈极显著差异(p<0.01)。由此可知,接种AM真菌提高了银杏实生苗叶片的相对叶绿素含量SPAD,促进了其光合速率。
银杏实生苗叶绿素荧光相关参数分析
接种AM真菌能显著影响银杏实生苗叶片的叶绿素荧光参数(p<0.05)。由图3可知,银杏实生苗最大光能转化效率(Fv/Fm)与潜在活性(Fv/Fo)的变化一致,接种AM真菌的处理均表现为整体上升趋势,而CK处理的Fv/Fm、Fv/Fo在接菌30d后显著低于三个接菌处理(p<0.05),在60d时与CC+FM出现最大差值0.088、1.787。银杏实生苗PSⅡ实际光合效率Y(Ⅱ)从30d后均表现为FM>CC+FM>CC>CK,三个接菌处理显著高于CK(p<0.05),其中在60d时FM与CK出现最大差值0.0968。此外,银杏实生苗的非光化学淬灭NPQ在生长过程中整体呈现下降趋势,其中,在30d时CK与FM出现最大差值0.6795,在60d后三个接菌处理间的NPQ差异逐渐减小,在90d后接菌处理均显著低于CK(p<0.05)。
银杏实生苗根系形态分析
如图4所示,接种AM真菌会对银杏实生苗的根系形态产生一定的影响。在接菌处理120d后,各处理银杏实生苗根系的总根长、根表面积和根体积均呈现FM>CC+FM>CC>CK的结果,但各处理间并无显著性差异。而接种AM真菌对实生苗的根尖数具有显著影响(p<0.05),以CC+FM(746.0个)最多,显著高于CK(367.0个)和CC(478.2个),以FM(624.8)次之,显著高于CK(p<0.05)。但银杏实生苗的根平均直径则与其余四项指标结果相反,CK的根平均直径最大,为1.064cm,显著高于FM的0.929cm和CC+FM的0.932cm(p<0.05),CC的根平均直径为1.003cm,与另外两个接菌处理FM、CC+FM间没有显著性差异。
银杏实生苗生物量分析
植物生物量的测定与分析是研究植物生长情况及物质积累的重要方法,通常可通过测定植物的干重,分别分析地上部分、地下部分和全株的生长状况(表1)。其中,银杏实生苗地上部分和全株干重以CC+FM最大,分别为1.160g和2.504g,地下部分干重则以FM最大,为1.489g,地上、地下部分和全株干重最小的均为CK,仅占最大值的41.90%、62.01%和52.16%,差异显著(p<0.05),而单接处理CC与其余三个处理间均没有显著性差异。可见接种AM真菌对银杏实生苗的生物量积累具有促进作用。

银杏实生苗氮、磷含量及积累量分析
氮是植物生长的必需养分,对植株的生长发育有重要影响。由图5可知,各处理地上部分氮含量均高于地下部分,三个接菌处理银杏实生苗地上、地下部分的氮含量均显著高于CK(p<0.05),但三个处理间无显著差异。其中,地上部分氮含量以FM处理的15.936g/kg最高,高出CK处理4.006g/kg,地下部分以CC处理的13.020g/kg最高,高出CK处理4.390g/kg。此外,全株氮积累量最高的为CC+FM处理(0.066g/株),与FM处理(0.058g/株)均显著高于CK处理(0.033g/株),但CC处理(0.048g/株)与其余三个处理之间差异不显著。
磷对植物生长发育具有重要作用。由图6可知,磷含量与氮含量结果整体相似,各处理地上部分磷含量均高于地下部分,三个接菌处理银杏实生苗地上、地下部分磷含量和全株磷积累量均显著高于CK(p<0.05),但三个处理间无显著差异。地上、地下部分磷含量均以CC处理的1.860g/kg和1.530g/kg最高,分别高出CK处理0.962g/kg和0.906g/kg。此外,全株磷积累量最高的为CC+FM处理(0.007g/株),与FM处理(0.006g/株)均极显著高于CK处理(0.002g/株)。
银杏实生苗抗氧化酶活性分析
接种AM真菌对银杏植株体内三种主要抗氧化酶SOD、CAT、POD的活性有显著影响(图7)。
从SOD活性来看,四个处理银杏实生苗的SOD活性呈现显著差异(p<0.05)。其中,CK的SOD活性939.61U/g,显著高于三个接菌处理。而不同接菌处理银杏实生苗之间的SOD活性则以CC+FM较高,为667.10U/g;FM次之,为629.93U/g;CC最低,为526.33U/g。
从POD活性来看,不同处理的银杏实生苗中POD活性有显著差异(p<0.05)。四种处理中,CC的POD活性最高,为50.25U/g;FM的POD活性最低,为21.19U/g;CK与CC+FM的POD活性相似,分别为29.15U/g和30.41U/g。
从CAT活性来看,不同处理间差异显著(p<0.05)。其中,CK的CAT活性最高,为209.98U/g。三种接菌处理中,FM的CAT活性最高,为154.96U/g;CC的CAT活性次之,为136.34U/g;CC+FM处理的银杏叶中CAT活性最低,为69.80U/g。
银杏实生苗代谢分析
可溶性蛋白是植物体内一类重要的营养物质,能够在一定程度上反映植物体的总代谢水平,并且具有强亲水性和渗透调节能力,可以提高植物的保水能力、保持植物细胞的渗透平衡,因此常用作筛选抗性的指标之一。由图8可知,不同处理的银杏实生苗叶片中可溶性蛋白浓度有显著差异(p<0.05)。接菌处理后,叶片中可溶性蛋白浓度均显著高于CK。其中,CK叶片中的可溶性蛋白浓度仅为7.35mg/g。FM、CC处理后银杏叶片的可溶性蛋白浓度最高,分别为12.57mg/g和12.10mg/g,是不接菌处理的1.7倍左右。
可溶性糖是植物生长发育的关键物质和重要能量来源,在植物体新陈代谢过程中发挥重要作用。经过不同处理的银杏叶中可溶性糖浓度与SOD活性的结果整体相似(图9),不同处理间的可溶性糖浓度有显著差异(p<0.05)。以CK的可溶性糖浓度最高,为35.40mg/g。CC+FM处理的银杏叶片中可溶性糖浓度与CK相近,为34.80mg/g。CC处理的银杏叶可溶性糖浓度最低,为28.10mg/g。
黄酮类化合物(类黄酮)是银杏次生代谢物中的主要成份之一,由图10可知,不同处理间银杏叶中植物类黄酮的浓度有显著差异(p<0.05)。对照组银杏实生苗的植物类黄酮浓度低于接菌处理,为2.35mg/g。接菌处理中,FM处理的银杏实生苗植物类黄酮浓度最高,为3.48mg/g。CC处理的最低,为2.75mg/g。
银杏实生苗综合评价
对不同处理银杏实生苗生长情况的综合评估,采用隶属模糊函数综合法进行。计算本试验测定的紧密度、开展度、投影面积等33个生长、生理指标所得隶属函数值(表2)可知,不同处理对银杏实生苗的促生效果由高到低依次为:CC+FM>FM>CC>CK。
