Abstract
本发明公开了增强屏障的敏感皮肤用修护霜及其制备方法,涉及化妆品技术领域。本发明通过选取多种具有保湿、修复、抗氧化、镇静或舒缓功效的天然成分或提取成分,透明质酸、大米发酵滤液和绿豆蛋白水解物内的多糖、多肽、氨基酸成分,协同发挥吸水、保湿、滋润肌肤的功效;乙酰基六肽‑8和棕榈酰五肽‑4作为合成多肽协同提高皮肤的抗炎、修复、再生功能,增强皮肤屏障,适合敏感皮肤修复;椰子油和罗马洋甘菊挥发油作为脂溶性成分,能够更加良好的克服角质层屏障,进一步渗透至真皮层,提升深层次皮层的抗炎、抗菌性能;搭配润肤剂和乳化剂,使得修护霜呈现的性状稳定且更易吸收,尤其适合敏感型皮肤使用者的补水、滋润和修复。

背景技术
皮肤是人体最大的器官,身体表面包在肌肉外面的组织称为皮肤,皮肤由表皮、真皮和皮下组织构成,并含有附属器官以及血管、淋巴管、神经和肌肉等。敏感性皮肤是一种问题性皮肤,具体是指皮肤在生理或病理条件下发生的一种反应,主要发生于面部皮肤,敏感性皮肤分为先天性和后天性,后天性敏感性皮肤的根本原因是皮肤细胞受损而使皮肤的免疫力下降,角质层变薄导致皮肤滋润度不够,最终皮肤屏障功能下降,不能抵抗外界对皮肤的刺激,在受到外界刺激后,皮肤易发生亢奋,从而产生泛红、发热、瘙痒、刺痛等不适的现象。
现有技术(CN112569151A)公开了一种抗敏修护屏障组合物及其制备方法和应用。该抗敏修护屏障组合物包含寡肽、积雪草提取物、凤凰单丛茶茶叶挥发油、反式-4-叔丁基环己醇、神经酰胺2脂质体、低分子量透明质酸钠、高分子量透明质酸钠、尿素和去离子水。该组合物能够达到舒缓皮肤敏感状态,修护皮肤屏障,抗炎等功效。但是研究发现,适用于敏感皮肤的修护霜,没有从提高皮肤抗炎、再生能力并结合深层次渗透,来综合提高补水、滋润、修复功效。

本发明的目的在于提供一种增强屏障的敏感皮肤用修护霜及其制备方法,用于解决现有技术中适用于敏感皮肤的修护霜,没有从提高皮肤抗炎、再生能力并结合深层次渗透,来综合提高补水、滋润、修复功效的技术问题。
为了实现上述目的,本发明采用了如下技术方案:
本发明公开一种增强屏障的敏感皮肤用修护霜,包括以下重量份数的成分:3~8份透明质酸、0.1~0.3份乙酰基六肽-8、0.02~0.05份棕榈酰五肽-4、0.6~2份椰子油、0.3~1份罗马洋甘菊挥发油、0.5~2份大米发酵滤液、1~3份绿豆蛋白水解物、0.05~0.2份pH调节剂、5~12份润肤剂、0.01~0.03份抗菌剂、2~5份乳化剂和60~80份去离子水。
作为本发明进一步优选的方案,所述绿豆蛋白水解物的制备方法如下:
步骤一,绿豆蛋白提取:将绿豆使用破壁机粉碎后,过100~200目筛得到绿豆细粉;向绿豆细粉中加入其重量20~30倍的水,70~80℃下保温提取1~2小时并重复提取三次,合并提取液;提取液减压抽滤除去滤渣,滤液在50~60℃下减压浓缩处理得到绿豆蛋白浓缩液;绿豆蛋白浓缩液通过D3520大孔吸附树脂进行吸附,50~70vt%的乙醇洗脱2~3次,合并洗脱液,洗脱液减压浓缩,80~90℃干燥至恒重,得到纯化绿豆蛋白;
步骤二,绿豆蛋白水解:向纯化绿豆蛋白中加入纯化水,混合均匀,再加入预热至35~40℃的碱性蛋白酶液,滴加5wt%的氢氧化钠溶液至pH=8,50℃的条件下水解2~3小时,沸水浴中放置20min使酶灭活,滴加5vt%的盐酸溶液调节pH至7,4000~6000rpm转速离心分离,上清液使用0.22μm的微孔滤膜过滤,滤液在70~80℃下干燥至含湿量小于5%,再置于-20~-5℃真空冷冻干燥得到绿豆蛋白水解物。
作为本发明进一步优选的方案,,所述纯化绿豆蛋白与纯化水、碱性蛋白酶液的用量比为8~10g:100~120mL:1~1.5mL;碱性蛋白酶液由碱性蛋白酶粉与Tris缓冲液、EDTA、去离子水按照2mg:20mL:5mg:10mL混合而成,碱性蛋白酶粉的酶活力为50000u/g。
作为本发明进一步优选的方案,所述罗马洋甘菊挥发油的制备方法如下:将罗马洋甘菊的新鲜花朵洗净、风干,碾碎后加入超临界高压萃取罐内,通入二氧化碳加压至500~600Psi,升温至40℃,保温超临界萃取70~80min,共提取3~4次,减压抽滤除去杂质,滤液即为黄色油状物的罗马洋甘菊挥发油。
作为本发明进一步优选的方案,所述大米发酵滤液由酵母菌对糙米进行发酵后过滤得到,pH值为5.5;pH调节剂为精氨酸或柠檬酸中的一种或两种;润肤剂为十一烷、十三烷、白池花籽油、向日葵籽油、山嵛醇、硬脂酸、辛酸甘油三酯中的一种或多种的混合物。
作为本发明进一步优选的方案,所述抗菌剂为吡啶硫酮锌、谷甾醇、黄芪黄素、蜂胶中的一种或多种的混合物;乳化剂为氢化卵磷脂、鲸蜡硬脂醇橄榄油酸酯、山梨坦橄榄油酸酯、硬脂酰谷氨酸钠中的一种或多种的混合物。
本发明还提供了上述增强屏障的敏感皮肤用修护霜的制备方法,包括以下步骤:
第一混合相制备:将透明质酸、乙酰基六肽-8、棕榈酰五肽-4、大米发酵滤液、绿豆蛋白水解物和去离子水在400~600rpm转速下搅拌均匀,得到第一混合相;
第二混合相制备:将椰子油、罗马洋甘菊挥发油、pH调节剂和润肤剂混合,升温至60~70℃,600~800rpm转速下搅拌均匀得到第二混合相;
混合乳化:将第一混合相、第二混合相、抗菌剂和乳化剂在60~70℃下混合乳化处理,得到预混霜;
降温均质:待预混霜的温度降低至30~40℃时,进行高速搅拌均质处理,得到修护霜半成品;
检测灌装:修护霜半成品经过理化性质检测合格后灌装至容器内;
包装:将灌装后的修护霜放置于包装盒内得到修护霜产品。
综上所述,由于采用了上述技术方案,本发明的有益效果是:
1、本发明的敏感皮肤用修护霜,选取多种具有保湿、修复、抗氧化、镇静或舒缓功效的天然成分或提取成分,科学合理的复配而成;其中,透明质酸、大米发酵滤液和绿豆蛋白水解物内的多糖、多肽、氨基酸成分,协同发挥吸水、保湿、滋润肌肤的功效;乙酰基六肽-8和棕榈酰五肽-4作为合成多肽协同提高皮肤的抗炎、修复、再生功能,增强皮肤屏障,适合敏感皮肤修复;椰子油和罗马洋甘菊挥发油作为脂溶性成分,能够更加良好的克服角质层屏障,进一步渗透至真皮层,舒缓、滋润皮肤,提升深层次皮层的抗炎、抗菌性能;搭配润肤剂和乳化剂,使得修护霜呈现的性状稳定且更易吸收,尤其适合敏感型皮肤使用者的补水、滋润、修复。
2、本发明绿豆蛋白水解物的制备方法,将绿豆破壁后增大比表面积并促进有效成分的溶出释放,再经水提、浓缩、树脂吸附、干燥的方式,得到纯化绿豆蛋白;纯化绿豆蛋白经碱性蛋白酶水解、膜过滤、真空冷冻干燥的方式得到绿豆蛋白水解物,经双缩脲法测得其中蛋白质含量为45.24%±0.15%,经蒽酮法测得总糖含量为2.97%±0.03%,且多肽的分子量是分布不均的,冻干粉末极易吸潮保湿,对敏感皮肤使用者的皮肤保湿和抗氧化具有显著的提升。
3、本发明罗马洋甘菊挥发油的制备方法,采用超临界二氧化碳萃取的方法,严格控制压力、温度和萃取时间,得到含有α-苯基乙酯、β-苯基乙酯、α-癸烯、氧化樟脑、罗马洋甘菊内酯、辛烯酸、己烯酸、己醛、己酸醋酸酯、香豆素等多种有效成分的天然挥发油,这些成分共同赋予修护霜独特的香气并发挥镇静、舒缓和抗炎的作用。

本申请中,透明质酸是一种天然多糖,在水中具有极强的保水能力,能够吸收和保持皮肤水分,增加皮肤湿润度,改善皮肤弹性和紧致度。
乙酰基六肽-8是一种合成多肽,结构类似于肉毒杆菌素的一部分,但不同于肉毒杆菌素,通常被称为抗皱肽,能够减少肌肉收缩,有助于减少表情纹和细纹的出现,对于抗衰老有一定的作用。
棕榈酰五肽-4是一种合成的脂肪酰化肽,具有良好的稳定性;被认为具有抗炎和抗氧化特性,有助于减少皮肤炎症,促进皮肤修复和再生,同时也有助于抗衰老。
椰子油是从椰子的果肉中提取的天然油脂,含有丰富的脂肪酸和抗氧化物质;具有优秀的保湿性能,能够滋润皮肤,同时也具有抗氧化和抗菌作用,有助于保护皮肤健康。
罗马洋甘菊挥发油是从罗马洋甘菊花朵中提取的天然挥发油,具有特殊的香气;具有镇静和舒缓的作用,有助于缓解紧张和焦虑情绪,同时也对皮肤有一定的舒缓效果。
大米发酵滤液是通过大米发酵产生的液体提取物,富含氨基酸、多肽和抗氧化物质,具有保湿和抗氧化作用,有助于改善肌肤质地,增加皮肤的柔软度和光泽。
绿豆蛋白水解物是从绿豆中提取的蛋白质,经过水解处理得到,具有保湿和滋润作用,有助于改善皮肤的柔软度和光泽,同时也可作为护发成分,增加头发的柔软度和光泽。

本实施例的一种增强屏障的敏感皮肤用修护霜,包括以下重量的成分:3.6kg透明质酸、0.12kg乙酰基六肽-8、0.02kg棕榈酰五肽-4、0.8kg椰子油、0.5kg罗马洋甘菊挥发油、0.8kg大米发酵滤液、1.3kg绿豆蛋白水解物、0.08kg pH调节剂精氨酸、5.8kg润肤剂辛酸甘油三酯、0.01kg抗菌剂谷甾醇、2.6kg乳化剂鲸蜡硬脂醇橄榄油酸酯和62kg去离子水。大米发酵滤液由酵母菌对糙米进行发酵后过滤得到,pH值为5.5。
绿豆蛋白水解物的制备方法如下:

效果验证
抗炎试验和增强细胞屏障能力验证
炎症因子白介素IL-6会引起细胞内炎症产生;而血管内皮生长因子在适当浓度下可促进HaCat细胞的增殖,修护皮肤屏障。配制HaCat细胞悬液浓度为3×105cell/mL,100μL/孔置于10孔板内,于37℃、5%CO2条件下培养至贴壁。将实施例1-6、对比例1-4制备的1mg修护霜分别加水稀释成10mg,向10孔板内分别对应加入3mg稀释后的修护霜,培养箱培养48小时后,取出离心,去除沉淀;通过多重液相蛋白定量技术,采用具有特定荧光信号的微球链接特定的捕获抗体,再用于捕捉溶液中的不同的待检测物,根据荧光信号强度,对待检测物进行定量检测,得到白介素IL-6的浓度。具体结果见表1:

从上表的抗炎试验和增强细胞屏障能力验证结果可以看出,本发明实施例制备的修护霜产生的白介素IL-6的浓度更小,说明细胞炎症反应小,对皮肤细胞的屏障增强能力更强。对比例1-4的浓度均有一定的增加,对比例2由于未添加罗马洋甘菊挥发油,无法通过挥发油内的多种成分赋予修护霜独特的香气并发挥镇静、舒缓和抗炎的作用,白介素IL-6的浓度增幅较大。
保湿效果验证
选取年龄18~22周岁的受试者30名,随机分配为实施例1-6组、对比例1-4组,每组随机分配3名受试者,在温度25±2℃、相对湿度50±2的环境下,擦净受试者脸颊,静坐半小时后,使用电容法皮肤水分测定仪测定脸颊部位的皮肤水分初始值,再按照2g/cm2的量涂抹实施例1-6、对比例1-4的修护霜,测试每组2小时、8小时后皮肤水分值相对于皮肤水分初始值的增长率平均值。具体结果见表2:

从上表的保湿效果验证结果可以看出,本发明实施例的修护霜在2小时、8小时后的皮肤水分值增长率平均值均大于对比例,说明实施例制备的修护霜成分的吸水保湿效果更加持久,适合长期补水保湿;对比例1-4皮肤水分值增长率平均值均有一定程度的下降,尤其是对比例4未添加大比例的润肤剂,使得润滑吸湿效果变差,吸水保湿性能降低明显。
敏感皮肤受试者测试效果验证
选取具有皮肤红肿、过敏、瘙痒、刺痛症状且年龄处于19~30周岁的敏感皮肤受试者30人,随机分为6组,每组5人,分别对应实施例1-6组,早晚使用对应的修护霜一次,并统计关于舒适性、保湿性、质地接受度的感官评价的总人数问卷,舒适性为使用24小时内不刺激、不瘙痒泛红,保湿性为使用24小时内感觉皮肤水润,质地接受度为接受修护霜的触感凉爽、可接受。具体结果见表3:

从上表的敏感皮肤受试者测试效果验证结果可以看出,本发明实施例的修护霜在感官评价的舒适性、保湿性、质地接受度上满意度高,说明低敏感、刺激性小,适合敏感皮肤使用。

步骤一,绿豆蛋白提取:将绿豆使用破壁机粉碎后,过100~200目筛得到绿豆细粉;向绿豆细粉中加入其重量20~30倍的水,70~80℃下保温提取1~2小时并重复提取三次,合并提取液;提取液减压抽滤除去滤渣,滤液在50~60℃下减压浓缩处理得到绿豆蛋白浓缩液;绿豆蛋白浓缩液通过D3520大孔吸附树脂进行吸附,50~70vt%的乙醇洗脱2~3次,合并洗脱液,洗脱液减压浓缩,80~90℃干燥至恒重,得到纯化绿豆蛋白;
步骤二,绿豆蛋白水解:向纯化绿豆蛋白中加入纯化水,混合均匀,再加入预热至35~40℃的碱性蛋白酶液,滴加5wt%的氢氧化钠溶液至pH=8,50℃的条件下水解2~3小时,沸水浴中放置20min使酶灭活,滴加5vt%的盐酸溶液调节pH至7,4000~6000rpm转速离心分离,上清液使用0.22μm的微孔滤膜过滤,滤液在70~80℃下干燥至含湿量小于5%,再置于-20~-5℃真空冷冻干燥得到绿豆蛋白水解物。
3.根据权利要求2所述的增强屏障的敏感皮肤用修护霜,其特征在于,所述纯化绿豆蛋白与纯化水、碱性蛋白酶液的用量比为8~10g:100~120mL:1~1.5mL;碱性蛋白酶液由碱性蛋白酶粉与Tris缓冲液、EDTA、去离子水按照2mg:20mL:5mg:10mL混合而成,碱性蛋白酶粉的酶活力为50000u/g。
4.根据权利要求1所述的增强屏障的敏感皮肤用修护霜,其特征在于,所述罗马洋甘菊挥发油的制备方法如下:将罗马洋甘菊的新鲜花朵洗净、风干,碾碎后加入超临界高压萃取罐内,通入二氧化碳加压至500~600Psi,升温至40℃,保温超临界萃取70~80min,共提取3~4次,减压抽滤除去杂质,滤液即为黄色油状物的罗马洋甘菊挥发油。
5.根据权利要求1所述的增强屏障的敏感皮肤用修护霜,其特征在于,所述大米发酵滤液由酵母菌对糙米进行发酵后过滤得到,pH值为5.5;pH调节剂为精氨酸或柠檬酸中的一种或两种;润肤剂为十一烷、十三烷、白池花籽油、向日葵籽油、山嵛醇、硬脂酸、辛酸甘油三酯中的一种或多种的混合物。
6.根据权利要求1所述的增强屏障的敏感皮肤用修护霜,其特征在于,所述抗菌剂为吡啶硫酮锌、谷甾醇、黄芪黄素、蜂胶中的一种或多种的混合物;乳化剂为氢化卵磷脂、鲸蜡硬脂醇橄榄油酸酯、山梨坦橄榄油酸酯、硬脂酰谷氨酸钠中的一种或多种的混合物。
7.增强屏障的敏感皮肤用修护霜的制备方法,其特征在于,包括以下步骤:
第一混合相制备:将透明质酸、乙酰基六肽-8、棕榈酰五肽-4、大米发酵滤液、绿豆蛋白水解物和去离子水在400~600rpm转速下搅拌均匀,得到第一混合相;
第二混合相制备:将椰子油、罗马洋甘菊挥发油、pH调节剂和润肤剂混合,升温至60~70℃,600~800rpm转速下搅拌均匀得到第二混合相;
混合乳化:将第一混合相、第二混合相、抗菌剂和乳化剂在60~70℃下混合乳化处理,得到预混霜;
降温均质:待预混霜的温度降低至30~40℃时,进行高速搅拌均质处理,得到修护霜半成品;
检测灌装:修护霜半成品经过理化性质检测合格后灌装至容器内;
包装:将灌装后的修护霜放置于包装盒内得到修护霜产品。
