本发明公开了一种代谢指纹图谱可视化的细胞芯片及应用方法,涉及细胞芯片技术领域,包括显微镜圆形玻片、密封储液腔、阵列块细胞芯片和微通道芯片;显微镜圆形玻片置于最底部,密封储液腔为中空圆柱体,设置在显微镜圆形玻片上;阵列块细胞芯片设置于密封储液腔中心,两者之间形成储液腔室,阵列块细胞芯片设置有细胞培养腔;微通道芯片叠置于阵列块细胞芯片上,微通道芯片上设置有若干微通道储液腔、微流道和第一流道,微通道储液腔与细胞培养腔空间位置一一对应,微通道储液腔经由微流道与第一流道相连通。本发明能够实现整个阵列腔室内数种不同代谢物的高通量成像,无需单独培养细胞再逐一图像获取,换液和药物处理批量化,节约时间和成本。

背景技术
细胞芯片技术是一种集成了细胞培养、化学处理、细胞分析和可视化成像等功能的微流控芯片技术,可以对细胞样本进行高通量和高精度的分析。尽管细胞阵列块芯片已在基因序列分析、蛋白组图谱、外显子成像、SNP位点展示、染色质甲基化修饰等研究领域有广泛应用,但目前仍缺乏一种对活细胞实时、多参数、高通量地监测的技术。目前常规使用的细胞成像微孔板(如384孔板等),虽然可以实现高通量和自动化的细胞培养和显微镜成像,但贴壁细胞会受到微孔板材料的影响,导致实验结果产生偏差,且多孔板无法一次性快速完成换液,操作繁琐,加药时间长,增加了细胞在体外空气环境中的暴露时间,导致了与空气中O2/CO2的交换变化,显著影响了敏感类型如神经和免疫细胞的生理活性及其代谢变化。
代谢指纹图谱是描述细胞代谢物谱的方法,通过分析细胞内节点代谢产物的种类和变化,可以评估细胞整体代谢状态和健康状况。该技术在生物医学和药物研发中取得了进展,广泛应用于疾病早期诊断、药物筛选和临床监测等领域。常用的基于原位信息的细胞代谢指纹图谱可视化方法有质谱成像、核磁共振成像和红外成像、受激拉曼散射等。然而,质谱成像技术存在以下不足:首先,质谱成像不具备足够的空间分辨率,不能观察单细胞或亚细胞(如线粒体)中代谢物的变化;其次,质谱成像无法实现活细胞连续动态代谢变化分析;再次,质谱技术在检测单细胞代谢物时样本获取和处理困难且繁琐,检测丰度、通量低,干扰物影响大,标准化和控制样本采集具有挑战性,其数据处理和分析需要专业知识和高水平的技术支持,包括峰识别、质谱图谱对应、代谢物定量和鉴定等;数据标准化和共享存在问题,不同机构使用的分析平台、样本处理方法和数据流程不同,限制了数据比较和验证的可靠性。虽然核磁共振成像和红外成像在活体或活细胞动态可视化代谢指纹图谱方面具有一定的优势,但它们在准确化学组成、空间分辨率成本和设备要求以及特异性等方面仍存在局限。受激拉曼散射是近年新兴的有望实现活细胞动态代谢成像的技术,结合非标或无标技术,利用碳氢震动区域(2800-2050cm-1)或指纹区域(1600-1800cm-1)对固醇或脂肪酸等脂质代谢分子进行分析,但对于特征峰的分析,还存在对目标代谢物的准确化学定义和定量方面显著的不足。光学显微镜成像是研究细胞代谢过程的常用工具。相比于质谱成像、核磁共振成像和红外成像技术,结合使用多种探针的多通道成像,光学显微镜能够提供详细的分子定位信息,可实时观察和记录细胞内的动态过程,因而具有高时空分辨率,并有效提供多种代谢指标的信息。结合新型遗传编码的功能型探针,光学显微镜通过可见光或荧光信号高灵敏度检出,不会造成明显细胞毒性,从而保持其原始状态和代谢活性。此外,光学显微镜的设备操作相对简单,适用于实验室和研究团队使用。然而,常规光学显微镜成像,通常只对活细胞中单一代谢物的检测,成本高且效率低;同时,新型功能荧光探针多为基于FRET原理的比例型探针,无法在同一细胞中同时检测多个代谢物参数(同时使用多种荧光探针标记,存在激发或者发散串扰)。
应用光学显微镜成像与活细胞代谢指纹图谱联用的装置,使得研究人员有望在更高的空间和时间分辨率下观察和分析细胞甚至亚细胞的代谢过程,以更深入理解细胞的代谢机制和功能。虽然近年来基于光学显微镜的细胞代谢分析已有很多研究,但目前还缺乏基于光学显微镜成像和代谢指纹图谱联用的高通量一体化的细胞芯片。
因此,本领域亟需一种可以集成像、快速换液、均匀加药一体化的细胞芯片,可快速、高效、自动化地获得高分辨率、高通量、可视化的细胞代谢指纹图谱。
发明内容
有鉴于此,本发明提供了一种代谢指纹图谱可视化的细胞芯片及应用方法,本发明能够实现整个阵列腔室内细胞代谢物的实时、快速、高效、自动化、高通量成像,无需单独培养细胞再进行图像获取,节约了时间和成本。
为了实现上述目的,本发明采用如下技术方案:
一种代谢指纹图谱可视化的细胞芯片,包括:显微镜圆形玻片、密封储液腔、阵列块细胞芯片和微通道芯片;
所述显微镜圆形玻片置于最底部,所述密封储液腔为中空圆柱体,设置在显微镜圆形玻片的上表面;
所述阵列块细胞芯片设置于密封储液腔中心,位于所述显微镜圆形玻片的上表面,所述密封储液腔和阵列块细胞芯片之间形成储液腔室,用于储液和换液操作,所述阵列块细胞芯片设置有细胞培养腔;
所述微通道芯片叠置于所述阵列块细胞芯片上表面,所述微通道芯片上设置有若干微通道储液腔、微流道和第一流道,所述微通道储液腔与细胞培养腔空间位置一一对应,所述微通道储液腔经由微流道与第一流道相连通,所述第一流道与密封储液腔相连通。
进一步的,所述密封储液腔高度高于所述阵列块细胞芯片与微通道芯片的堆叠高度。
进一步的,所述显微镜圆形玻片的材料为玻璃、聚苯乙烯、聚碳酸酯中的任意一种。
进一步的,所述密封储液腔、阵列块细胞芯片和微通道芯片的材料为聚二甲基硅氧烷、聚苯乙烯、聚碳酸酯中的任意一种。
进一步的,所述细胞培养腔呈阵列设置,阵列的细胞培养腔相互独立,互不相通。
进一步的,所述细胞培养腔内设置有培养基。
进一步的,所述培养腔底部接种带有荧光探针标记的细胞。
一种代谢指纹图谱可视化的细胞芯片的应用方法,包括以下操作步骤:
S01.细胞样品的制备;
S02.将所述密封储液腔、阵列块细胞芯片和微通道芯片进行高压灭菌20分钟,灭菌完成后放入超净台进行风干;
S03.将风干完成的阵列块细胞芯片和密封储液腔依次置于显微镜圆形玻片的上表面,将所述微通道芯片置于阵列块细胞芯片上方,使微通道储液腔与细胞培养腔的空间位置一一对应,按压使其紧密贴合;
S04.在细胞培养腔内加入培养基,用不同种类的荧光探针分别与细胞样品混合均匀后,分别接种到细胞培养腔内;
S05.在储液腔室中注入细胞基础培养基,维持正常的培养环境,防止细胞培养腔内的培养基风干,储液腔室中细胞基础培养基液面高度不高于低于阵列块细胞芯片的高度,防止其流入微通道芯片中;
S06.样品细胞培养若干天后,去除旧的培养基,再将活细胞成像溶液加入到储液腔室内,活细胞成像溶液会通过第一流道进入微通道芯片,依次流入各个细胞培养腔内,一次性快速地完成换液操作;
S07.用镊子拆除微通道芯片,借助光学显微镜进行快速、高效、高通量、自动化成像。
本发明的有益效果为:
(1)本发明的细胞芯片能够一体化成像和快速换液、均匀加药,能够实现整个阵列腔室内细胞多种代谢物的实时、快速、高效、自动化、高通量成像,无需单独培养细胞再逐一地进行图像获取,节约了时间和成本;可实现在相同的微环境下,活细胞多种代谢物参数同时动态监测,这些代谢产物在相同的生理条件下能够更加准确的反应细胞的生理和病理状况,减少了代谢数据的检测波动;
(2)本发明装置尺寸小,操作简便快速,待检测的细胞处于相同的微环境,即培养过程满足同质化条件,消除了环境因素的影响,使实验结果更加可靠和可重复,发明装置可与传统宽场光学显微镜联用成像,设备需求环境简单易行;
(3)本发明由密封储液腔和叠放在一起的阵列块细胞芯片、微通道芯片组成。整个装置可以进行拆卸和装配,在需要对芯片内部细胞进行代谢物检测时,无需破坏整个芯片。检测完成后,密封储液腔、阵列块细胞芯片和微通道芯片可以被重复使用,从而进一步降低实验成本;
(4)本发明可以在单个细胞水平上针对代谢物的动态变化进行高通量筛选(如多种小分子药物靶点筛选)和分析,从而提高药物研发和毒理学研究的效率。通过对细胞芯片上的细胞进行物理刺激或药物处理,然后利用光学显微镜成像技术获取细胞代谢产物的信息,再将这些信息转化为可视化的指纹图谱,帮助研究人员进行数据分析和结果展示,以便更好地理解细胞代谢的变化规律。

下面将结合本发明实施例中的附图,对本发明实施例中的技术方案进行清楚、完整地描述。
在本发明的描述中,需要说明的是,术语“中心”、“上”、“下”、“左”、“右”、“水平”、“内”、“外”、“一侧”等指示的方位或位置关系为基于附图所示的方位或位置关系,仅是为了便于描述本发明和简化描述,而不是指示或暗示所指的装置或元件必须具有特定的方位、以特定的方位构造和操作,因此不能理解为对本发明的限制;术语“第一”、“第二”、“第三”仅用于描述目的,而不能理解为指示或暗示相对重要性,此外,除非另有明确的规定和限定,术语“安装”、“相连”、“连接”应做广义理解,例如,可以是固定连接,也可以是可拆卸连接,或一体地连接;可以是机械连接,也可以是电连接;可以是直接相连,也可以通过中间媒介间接相连,可以是两个元件内部的连通。对于本领域的普通技术人员而言,可以具体情况理解上述术语在本发明中的具体含义。
实施例1
如图1-3所示,本发明提出的一种代谢指纹图谱可视化的细胞芯片,包括:显微镜圆形玻片1、密封储液腔2、阵列块细胞芯片31和微通道芯片41,显微镜圆形玻片1的材料为玻璃、聚苯乙烯、聚碳酸酯中的任意一种,本实施例中显微镜圆形玻片1的材料为玻璃,密封储液腔2、阵列块细胞芯片31和微通道芯片41的材料为聚二甲基硅氧烷、聚苯乙烯、聚碳酸酯等其他生物相容性材料,本实施例中密封储液腔2、阵列块细胞芯片31和微通道芯片41的材料均为聚二甲基硅氧烷。
显微镜圆形玻片1置于细胞芯片的最底部,其在使用前需要进行多聚赖氨酸包被,密封储液腔2为中空圆柱体,设置在显微镜圆形玻片1的上表面,阵列块细胞芯片31和微通道芯片41依次堆叠在密封储液腔2的中心位置,密封储液腔2高度高于阵列块细胞芯片31与微通道芯片41的堆叠高度。
具体的,显微镜圆形玻片1的直径为25mm,厚度为0.17mm,阵列块细胞芯片31高度为2mm,直径为18mm,微通道芯片41的高度为2mm,直径为18mm,密封储液腔2的高度为6mm,内外直径分别为24mm和21mm。
密封储液腔2和阵列块细胞芯片31两者之间形成新的储液腔室5,用于储液和换液操作。阵列块细胞芯片31设置有若干成阵列设置的细胞培养腔32,阵列细胞培养腔32相互独立,互不相通。细胞培养腔32内设置有培养基,培养基的体积不大于细胞培养腔32的容积,培养腔中接种带有荧光探针标记的细胞。
微通道芯片41叠置于阵列块细胞芯片31上表面,微通道芯片41上设置有若干微通道储液腔42、微流道43和第一流道44,微通道储液腔42与细胞培养腔32空间位置一一对应,微通道储液腔42经由微流道43与第一流道44相连通,第一流道44与密封储液腔2相连通,储液腔42直径为2.4mm;微流道43直径为2mm;第一流道44的长为3.7mm,宽为3mm,深度为2mm。
实施例2
本实施例所提供的代谢指纹图谱可视化的细胞芯片在神经科学领域具体应用场景包括神经元代谢状态、神经发育和神经退行性疾病过程中的代谢变化等,下面将本发明所提供的代谢指纹图谱可视化的细胞芯片应用于神经研究神经元代谢状态为例进行举例说明。
如图4-5所示,一种代谢指纹图谱可视化的细胞芯片的应用方法,包含以下操作步骤:
S01.皮层神经元样品的制备,首先,对哺乳动物的胚胎或出生后的神经组织进行解剖,将其剪碎并放入离心管中,经过静置后,丢弃解剖缓冲液;其次,向离心管中添加木瓜蛋白酶溶液,在37℃的培养箱中水平放置40分钟;每隔10分钟,轻轻颠倒离心管使沉淀细胞充分混匀,最后,加入胰蛋白酶抑制剂终止消化;最后,经过细胞过滤筛后,取第二个过滤后的离心管,在200g的转速下离心5分钟;离心后,将上清液丢弃,保留神经元细胞沉淀,并将其重悬于神经元基础培养基中,形成神经元细胞悬浮液;
S02.将显微镜圆形玻片1用无菌过滤后的多聚赖氨酸包被12小时,然后用无菌过滤后的1×PBS清洗3遍,每次15分钟,等待显微镜圆形玻片1风干完成;
S03.将密封储液腔2、阵列块细胞芯片31和微通道芯片41进行高压灭菌20分钟,灭菌完成后放入超净台进行风干;
S04.将上述风干完成的阵列块细胞芯片31和密封储液腔2依次置于显微镜圆形玻片1的上表面,轻轻按压使其贴合,将微通道芯片41按照图2所示的相对位置置于阵列块细胞芯片31的上方,使微通道储液腔42与细胞培养腔32的位置一一对应,轻轻按压至紧密贴合;
S05.在细胞培养腔32内加入培养基,用不同种类的荧光探针分别与皮层神经元混合均匀后,分别接种到细胞培养腔32内,细胞培养腔32的直径为2毫米,皮层神经元细胞用量为2000个/孔,培养基液面高度不高于2毫米;
S06.在腔室5中注入神经元细胞基础培养基,维持正常的培养环境,防止细胞培养腔32内的培养基风干,腔室5中神经元细胞基础培养基液面高度低于2毫米,防止其流入微通道芯片41中;
S07.神经元培养5天后,去除旧的培养基,再将适量活细胞成像溶液加入到腔室5内,活细胞成像溶液会通过第一流道44进入微通道芯片41,依次流入各个细胞培养腔32内,一次性快速地完成换液操作;
S08.用镊子拆除微通道芯片41,借助光学显微镜进行快速、高效、高通量、自动化成像,可视化的4×4阵列细胞芯片中,培养7天神经元代谢物的光学显微镜图片如图4所示,培养第7天神经元中各个代谢物指标如表1所示。神经元在培养7天的发育状态下,其细胞内细胞器亚结构的氧化还原代谢参数如线粒体活性氧(mitoROS)的平均荧光强度(MFI)为211.386,能量代谢参数如线粒体ATP的MFI为448.187。此外,从图5可知,未用荧光探针标记的神经元进行光学显微镜成像时只有显示明场条件下的细胞形态,没有检测到荧光信号。

实施例3
如图6-7所示,当实施例1所提供的代谢指纹图谱可视化的细胞芯片应用于药物毒理学研究时,可将来源于人体的人前列腺癌细胞(PC3)分别与不同种类的荧光探针混合均匀后,分别接种到细胞培养腔32内,PC3细胞培养2天后,先将储液腔室5和细胞培养腔32中的旧培养基取出,再将适量含有待测药物(Erastin)的培养基加入到储液腔室5内,含有待测药物的培养基会通过第一流道44进入微通道芯片41,依次流入各个细胞培养腔32中,一次性快速地完成加药的换液操作。等待药物处理24小时后,取下微通道芯片41,借助光学显微镜对未经待测药物处理和经过待测药物处理的PC3细胞中的代谢物进行多通道、快速、高效、高通量、自动化的成像和统计分析。
在代谢指纹图谱中,20μM剂量的Erastin药物对人前列腺癌细胞处理24小时的毒性作用小,但其改变了人前列腺癌细胞的代谢物,降低了总的ATP的生成(GoAteam)、NADP+/NADPH比值(iNap1)、线粒体自噬(mitoQC)、线粒体数量(mitodendra2)、线粒体ATP的生成(m-GoAteam)、谷胱甘肽(GSH)以及线粒体膜电位(CMTMRos),增加了前列腺癌细胞中乳酸(Laconic)、总的活性氧(H2DCFDA)、线粒体超氧化物(mitoSOX)、脂质过氧化物(LipidROS)。除此之外,Erastin药物对其他的代谢物无显著性改变。
上述虽然结合附图对本发明的具体实施方式进行了描述,但并非对本发明保护范围的限制,所属领域技术人员应该明白,在本发明的技术方案的基础上,本领域技术人员不需要付出创造性劳动即可做出的各种修改或变形仍在本发明的保护范围以内。
Claims (8)
1.一种代谢指纹图谱可视化的细胞芯片,其特征在于,包括:包括显微镜圆形玻片(1)、密封储液腔(2)、阵列块细胞芯片(31)和微通道芯片(41);
所述显微镜圆形玻片(1)置于最底部,所述密封储液腔(2)为中空圆柱体,设置在显微镜圆形玻片(1)的上表面;
所述阵列块细胞芯片(31)设置于密封储液腔(2)中心,位于所述显微镜圆形玻片(1)的上表面,所述密封储液腔(2)和阵列块细胞芯片(31)之间形成储液腔室(5),用于储液和换液操作,所述阵列块细胞芯片(31)设置有细胞培养腔(32);
所述微通道芯片(41)叠置于所述阵列块细胞芯片(31)上表面,所述微通道芯片(41)上设置有若干微通道储液腔(42)、微流道(43)和第一流道(44),所述微通道储液腔(42)与细胞培养腔(32)空间位置一一对应,所述微通道储液腔(42)经由微流道(43)与第一流道(44)相连通,所述第一流道(44)与密封储液腔(2)相连通。
2.根据权利要求1所述的一种代谢指纹图谱可视化的细胞芯片,其特征在于,所述密封储液腔(2)高度高于所述阵列块细胞芯片(31)与微通道芯片(41)的堆叠高度。
3.根据权利要求1所述的一种代谢指纹图谱可视化的细胞芯片,其特征在于,所述显微镜圆形玻片(1)的材料为玻璃、聚苯乙烯、聚碳酸酯中的任意一种。
4.根据权利要求1所述的一种代谢指纹图谱可视化的细胞芯片,其特征在于,所述密封储液腔(2)、阵列块细胞芯片(31)和微通道芯片(41)的材料为聚二甲基硅氧烷、聚苯乙烯、聚碳酸酯中的任意一种。
5.根据权利要求1所述的一种代谢指纹图谱可视化的细胞芯片,其特征在于,所述细胞培养腔(32)呈阵列设置,阵列的细胞培养腔(32)相互独立,互不相通。
6.根据权利要求1所述的一种代谢指纹图谱可视化的细胞芯片,其特征在于,所述细胞培养腔(32)内设置有培养基。
7.根据权利要求6所述的一种代谢指纹图谱可视化的细胞芯片,其特征在于,所述培养腔底部接种带有荧光探针标记的细胞。
8.根据权利要求1-7任意一项所述的一种代谢指纹图谱可视化的细胞芯片的应用方法,其特征在于,包含以下操作步骤:
S01.细胞样品的制备;
S02.将所述密封储液腔(2)、阵列块细胞芯片(31)和微通道芯片(41)进行高压灭菌20分钟,灭菌完成后放入超净台进行风干;
S03.将风干完成的阵列块细胞芯片(31)和密封储液腔(2)依次置于显微镜圆形玻片(1)的上表面,将所述微通道芯片(41)置于阵列块细胞芯片(31)上方,使微通道储液腔(42)与细胞培养腔(32)的空间位置一一对应,按压使其紧密贴合;
S04.在细胞培养腔(32)内加入培养基,用不同种类的荧光探针分别与细胞样品混合均匀后,分别接种到细胞培养腔(32)内;
S05.在储液腔室(5)中注入细胞基础培养基,维持正常的培养环境,防止细胞培养腔(32)内的培养基风干,储液腔室(5)中细胞基础培养基液面高度低于阵列块细胞芯片(31)的高度,防止其流入微通道芯片(41)中;
S06.样品细胞培养若干天后,去除旧的培养基,再将活细胞成像溶液加入到储液腔室(5)内,活细胞成像溶液会通过第一流道(44)进入微通道芯片(41),依次流入各个细胞培养腔(32)内,一次性快速地完成换液操作;
S07.用镊子拆除微通道芯片(41),借助光学显微镜进行快速、高效、高通量、自动化成像。
