背景技术
多功能纳米颗粒在广泛的应用,例如生物传感器、诊断纳米探针和靶向药物递送系统中受到关注。这些努力在很大程度上是由于需要通过在各种生理系统中精确、时空控制药剂递送来提高生物特异性并减少诊断和疗法中的副作用。为了实现这一目标,人们致力于开发刺激响应性纳米平台。已被用于确定递送效率的环境刺激包括pH、温度、酶促表达、氧化还原反应和光诱导。在这些激活信号中,pH触发是基于两种pH差异的最广泛研究的刺激之一:(a)病理(例如肿瘤)与正常组织和(b)酸性细胞内隔室。
例如,由于肿瘤细胞外微环境的异常酸度(pH~6.5),已经报道了几种pH响应性纳米系统来增加肿瘤成像的灵敏度或疗法的功效。然而,对于在酸性环境中通过水解释放药物的聚合物胶束组合物,可能需要数天才能释放药物。在那段时间内,身体可以排泄或分解胶束。
为了靶向酸性内体/溶酶体隔室,已经研究了具有pH可裂解连接子的纳米载体以提高有效负载的生物可用性。此外,已经设计了几种具有pH诱导电荷转换的智能纳米载体来提高药物疗效。内吞系统由一系列隔室构成,所述隔室在内化货物的分类、加工和降解中具有独特的作用。由于纳米颗粒停留时间短(<分钟)以及这些隔室中的pH差异小(例如,早期内体与溶酶体之间<1pH单位),因此通过pH敏感性纳米颗粒选择性靶向不同的内吞隔室特别具有挑战性。
免疫疗法已成为癌症治疗的有力策略。诸如白介素-2(IL-2)等免疫调节剂可诱导抗肿瘤免疫反应,但它们的临床应用受到不利的药物动力学特性的限制,所述特性会引起严重的剂量限制性毒性(例如血管渗漏综合征)。
需要改进的用于治疗应用的pH响应性胶束组合物,特别是具有增加的药物有效负载、延长的血液循环时间、在目标部位快速递送药物以及在特定窄pH范围内的响应性(例如用于靶向肿瘤或特定细胞器)的组合物。
发明内容
本文所述的嵌段共聚物为适用于治疗原发性和转移性肿瘤组织(包括淋巴结)的治疗剂。本文呈现的嵌段共聚物和胶束组合物利用癌性组织与正常组织之间的这一普遍存在的pH差异并且在由细胞吸收之后提供高度敏感和特定的响应,因此允许将治疗性有效负载部署到肿瘤组织。

图4显示具有rhIL-2(未修饰)和经修饰IL-2(800CW)两者的生物活性的包封调配物。与IL-2(黑色倒三角形)相比,胶束在IL-2(800CW)(黑色正方形)的中性状态下具有更好的保护作用,而两种调配物在酸化时表现出相似的生物活性。胶束以pH响应性方式包封和释放IL-2和IL-2(800CW)两者。
图5显示与PEG-PDBA相比,PEG5K-PLA16K在IL-2(修饰或未修饰)包封方面效率低下。800CW修饰必须与本文所述的pH敏感性胶束配对以增加IL-2的负载。即使用800CW修饰IL-2,如PEG-PLA的非pH敏感性胶束也不能有效包封IL-2。
图6显示来自用IL-2或IL-2(800CW)制备的非pH敏感性胶束调配物的胶束包封的IL-2峰和游离IL-2峰的考马斯染色。在包封IL-2(800CW)时PDBA>>PEG5K-PLA15K由未包封的游离IL-2池中大比例的IL-2(800CW)证明。通过查看在高离子强度条件下执行PDBA IL-2(800CW)调配物是否会降低包封效率,这证实了静电相互作用的重要性。未标记和800CW标记的IL-2均以约20%EE、2%DL负载至PEG-PLA纳米颗粒中,相比之下,PDBA为约80%EE、8%DL。

II.使用方法
称为瓦尔堡效应(Warburg effect)的好氧糖酵解发生在所有实体癌中,其中癌细胞优先摄取葡萄糖并将其转化为乳酸或其他酸。由于单羧酸转运蛋白或其他转运蛋白,乳酸或其它酸优先积聚在细胞外空间。由此产生的细胞外空间酸化促进了细胞外基质的重塑,以进一步促进肿瘤侵袭和转移。
本文所提供的一些实施例描述在生理pH(7.35-7.45)下形成胶束的化合物。在一些实施例中,本文所述的化合物与治疗剂共价或非共价结合。在一些实施例中,胶束的分子量大于2×107道尔顿。在一些实施例中,胶束的分子量为约2.7×107道尔顿。在一些实施例中,治疗剂在生理pH(7.35-7.45)下(例如,在血液循环期间)被隔离在胶束核心内。在一些实施例中,当胶束遇到酸性环境(例如肿瘤组织)时,胶束解离成平均分子量为约3.7×104道尔顿的单个化合物,从而允许释放治疗剂。在一些实施例中,胶束在pH低于pH转变点的pH(例如肿瘤微环境的酸性状态)下解离。

应了解,提及药学上可接受的盐包括溶剂加成形式。在一些实施例中,溶剂合物含有化学计量或非化学计量的溶剂,且在与药学上可接受的溶剂(如水、乙醇等)结晶期间形成。当溶剂是水时形成水合物,或当溶剂是醇时形成醇化物。本文所描述的化合物的溶剂合物在本文所描述的方法中方便地制备或形成。另外,本文提供的化合物以非溶剂化以及溶剂化形式存在。

除非另有说明,否则采用质谱法、NMR、HPLC、蛋白质化学、生物化学、重组DNA技术和药理学常规方法。嵌段共聚物使用标准有机化学技术,例如《马奇高级有机化学(March'sAdvanced Organic Chemistry)》,第6版,约翰·威利父子公司(John Wiley and Sons,Inc)中所描述的技术制备。

合适的PEG聚合物可购买(例如,购自西格玛阿尔德里奇(Sigma Aldrich))或可根据所属领域中已知的方法合成。在一些实施例中,亲水性聚合物可用作疏水性单体聚合以形成嵌段共聚物的引发剂。例如,MPC聚合物(例如窄分布的MPC聚合物)可通过与市售小分子引发剂,例如2-溴-2-甲基丙酸乙酯(西格玛阿尔德里奇)的原子转移自由基聚合(ATRP)制备。这些所得MPC聚合物可用作大分子ATRP引发剂以进一步与其它单体共聚合以形成嵌段聚合物,可使用原子转移自由基聚合(ATRP)或可逆加成-断裂链转移(RAFT)方法合成。
在一些实施例中,合适的嵌段共聚物和胶束可使用标准合成技术或使用所属领域中已知的方法与专利公开案第WO 2012039741号和第WO 2015188157号中描述的方法组合来合成,所述专利公开案的全部内容以引用的方式并入本文中。
实例2.胶束形成
一般方法
将甲醇添加至玻璃圆底烧瓶中的嵌段共聚物中并且借助于超声处理浴溶解。在溶解之后,将所得溶液定量地转移至含有搅拌棒的HDPE瓶中,并且用冰浴冷却至0℃。在搅拌时,使用蠕动泵将水逐滴添加至HDPE瓶中的甲醇聚合物溶液中。将含有聚合物溶液的HDPE瓶维持于冰浴中,从而形成胶束。使用5个循环的切向流过滤(TFF),通过100k2微型超滤模块从胶束溶液中去除甲醇。
通过简单混合制备的PEG-PDBA-IL-2调配物

通过FPLC纯化PDBA-IL-2调配物

通过斑点印迹测定调配物的IL-2含量和胶束含量。用0.2μm硝酸纤维素膜组装斑点印迹设备。每个孔在真空下用200μL 1×PBS洗涤,然后用100μL PBS再水合。添加样品和标准品(10-100μL),并对膜施加真空。膜用PBS洗涤2×。

实例3.体内肿瘤小鼠模型的一般程序

实例4.体外IL-2生物活性分析的一般程序

实例5.调配物SDS-PAGE分析的一般程序
通过SDS-PAGE评估胶束-IL-2调配物以确认IL-2负载至胶束中和IL-2完整性。制备样品以靶向每条泳道负载的100-200ng蛋白质。为了通过FPLC表征IL-2负载调配物纯化,负载样品构成未经任何纯化的粗调配物,旋转负载样品构成通过高速离心纯化以清除聚集体和大颗粒后的调配物,胶束池由组合含有胶束的级分制备且游离IL-2样品含有含未包封蛋白质的级分。根据还原要求,将调配物样品在含有或不含β-巯基乙醇的4×勒姆利(Laemmli)缓冲液(伯乐)中稀释,并在65℃下变性5分钟。通过在50V下堆叠30分钟,然后在100V下分离90分钟,将样品负载于任何kDTM或4-20%SDS-PAGE梯度Mini-Protean凝胶(伯乐)中。IL-2的检测通过Simply Blue Stain(茵维特罗根)进行。在转移至0.2μm硝化纤维素膜后,还通过蛋白质印迹如下确定IL-2:通过用抗IL-2Ab克隆(赛信通技术公司(CellSignaling Technology),克隆D7A5,1:4000稀释)探测,然后用HRP结合抗兔二抗(LI-COR,1:2000稀释)探测,并通过ECL试剂(皮尔斯(Pierce))进行检测,并使用ChemiDoc MP成像仪(伯乐)捕获化学发光。使用ImageLab(伯乐)进行图像处理和光密度测定法分析。如果需要,通过拟合IL-2标准曲线来进行IL-2的定量。

43.根据权利要求37或38所述的方法,其中所述治疗剂为抗体或其片段。
44.根据权利要求43所述的方法,其中所述抗体或其片段为双特异性抗体或其片段。
45.根据权利要求44所述的方法,其中所述双特异性抗体或其片段为融合蛋白。
46.根据权利要求45所述的方法,其中所述融合蛋白为双特异性T细胞衔接子(BiTE)。
47.根据权利要求37至46中任一权利要求所述的方法,其中所述荧光染料的激发光谱为约400nm至约900nm。
48.根据权利要求47所述的方法,其中所述荧光染料的激发光谱为约:400、450、500、550、600、650、700、750、800或850nm。
49.根据权利要求47或48所述的方法,其中所述荧光染料为香豆素、罗丹明、花青、呫吨、荧光素或其磺化或带负电荷的形式,或来自图22的化合物。
