Abstract
为克服现有技术中防脱产品普遍存在机理不明确、靶点单一、副作用大,无法有效改善脱发的问题,提供一种防脱发洗发液,包括发用调理剂和防脱发剂,所述发用调理剂包括以下组分:泛醇、鳄鱼多肽、水解蚕丝以及吡罗克酮乙醇胺盐;所述防脱发剂包括以下组分:二氨基嘧啶氧化物、植物提取物以及三肽‑1铜。同时,本发明还公开了上述防脱发洗发液的制备方法。本发明提供的防脱发洗发液,将不同作用机理的防脱有效活性成分进行组合,有效抑制5α‑还原酶活性、抑制油脂过度分泌及马拉色菌的滋生、改善头皮炎症、改善微循环、强化毛囊,达到防脱固发效果。

技术领域
本发明属于洗护日用品技术领域,具体涉及一种防脱发洗发液及其制备方法。
背景技术
随着生活节奏的加快,生活压力的加大,越来越多的人出现脱发问题,且脱发正呈现年轻化的趋势;目前关于脱发的机理研究尚不十分清楚,但已发现脱发与遗传、内分泌、环境恶化以及精神心理等多种因素有关。
雄激素造成的脱发是常见的一种脱发类型,二氢睾酮(DHT)是由睾丸分泌的一种类固醇激素,是人体内主要的雄激素,5α-还原酶使睾酮转换成强的雄激素双氢睾酮(DHT)与头皮激素受体蛋白(ARP)结合后,导致毛囊萎缩、毛囊生长期缩短及休止期毛囊数增加,即产生脱发;油脂分泌旺盛也是脱发的另一主要原因,如雄激素脱发的症状为患者头皮脂肪过量溢出,常伴有头屑增多,头皮油腻,瘙痒明显,多发生于皮脂腺分泌旺盛的青壮年,患者一般头发细软,有的还伴有头皮脂溢性皮炎症状;头皮菌群失衡也会导致脱发,例如真菌群中的马拉色菌,主要寄生于人体头皮等部位,以头皮分泌的皮脂为营养,过量繁殖会使头皮产生过多头皮屑,致使患者头皮角质层变薄,破坏了头皮正常的防御屏障,进而会造成头皮瘙痒、头皮发红等症状,进而导致头发大量脱落;此外,头部血液循环不良时,头发营养就无法送达至毛囊,休止期毛囊数增加,从而加速了头发的脱落。
现有的脱发治疗药物中,治疗雄激素性脱发的化学生发剂有米诺地尔、非那雄胺等,米诺地尔治疗脱发会产生药物依赖性,甚至会导致头皮瘙痒、发干、红斑等皮炎的症状,副作用明显;非那雄胺则具有男性荷尔蒙活性抑制作用,不能够从根本上解决脱发问题。
由于脱发原因的复杂性,单一的治疗成分及治疗方式,均不能达到有效防脱发的效果,且现有防脱产品普遍存在机理不明确、靶点单一、副作用大等问题,无法达到有效改善脱发的作用。
发明内容
针对现有技术中防脱产品普遍存在机理不明确、靶点单一、副作用大,无法有效改善脱发的问题,提供一种防脱发洗发液及其制备方法。
本发明解决上述技术问题所采用的技术方案如下:

另一方面,本发明提供一种防脱发洗发液的制备方法,包括以下步骤:
将聚季铵盐-10、瓜儿胶羟丙基三甲基氯化铵、柠檬酸加到去离子水中,加热至80~85℃,混合搅拌均匀,保温5~10分钟,得到混合液A备用;

具体的,在优选实施例中所述有机溶剂选自丁二醇,所述丁二醇在防脱发剂中可以发挥保湿和抑菌作用;由于丁二醇的分子粒较大,不易蒸发水分,反而能吸收更多的水分,因此具有保湿功效;另外,由于丁二醇属于多元醇类别,而这种类别具备抗微生物繁殖的能力,能抑制细菌的滋生,并且对直接接触的皮肤无刺激和损伤。
在一些实施例中,所述植物提取物包括以下重量组分:
侧柏叶提取物0.01~0.5份、当归根提取物0.01~0.15份、人参根提取物0.01~0.15份、旱金莲提取物0.01~0.15份、姜根提取物0.005~0.15份。
所述侧柏叶有凉血止血,化痰止咳,生发乌发的功效,本申请中防脱发洗发液添加所述侧柏叶提取物,有利于缓解血热造成的脱发,起到防脱育发的作用;
所述旱金莲提取物具有补肾益阴、乌须发的作用,改善抗氧化特性,防止毛囊因氧化应激导致的萎缩;
所述当归根提取物可以补血活血,帮助扩张头皮及皮肤的毛细血管、促进血液循环;
所述姜根提取物中的姜辣素、姜烯油等成分,可使头部皮肤血液循环正常化,促进头皮新陈代谢,激活毛囊;

本发明的另一实施例提供了一种防脱发洗发液的制备方法,包括以下步骤:
将聚季铵盐-10、瓜儿胶羟丙基三甲基氯化铵、柠檬酸加到去离子水中,加热至80~85℃,混合搅拌均匀,保温5~10分钟,得到混合液A备用;
将月桂醇聚醚硫酸酯铵、月桂醇硫酸酯铵、月桂酰胺丙基甜菜碱、氯化钠以及乙二醇二硬脂酸酯加入混合液A中,均质5~10分钟,搅拌混匀,得到混合液B;
降温至45~50℃时,混合液B中加入泛醇、侧柏叶提取物、水解蚕丝、吡罗克酮乙醇胺盐、二氨基嘧啶氧化物、丁二醇、苯氧乙醇、旱金莲提取物、苯氧乙醇、人参根提取物、水解胶原、姜根提取物、三肽-1铜以及芳香剂,搅拌均匀,均质5~10分钟,搅拌混匀后,加入防腐剂并混匀,得到混合液C;

在一些实施例中,所述混合液C的搅拌时间为10~15min。
具体的,所述混合液C的搅拌时间时间不宜过长,避免有效成分失活。以下通过实施例对本发明进行进一步的说明。
实施例1~3
本实施例用于说明本发明公开的防脱发洗发液及其制备方法,包括以下操作步骤:
将聚季铵盐-10、瓜儿胶羟丙基三甲基氯化铵、柠檬酸加到去离子水中,加热至80℃,混合搅拌均匀,保温10分钟,得到混合液A备用;
将月桂醇聚醚硫酸酯铵、月桂醇硫酸酯铵、月桂酰胺丙基甜菜碱、氯化钠以及乙二醇二硬脂酸酯加入混合液A中,均质10分钟,搅拌混匀,得到混合液B;
降温至45℃时,混合液B中加入泛醇、侧柏叶提取物、水解蚕丝、吡罗克酮乙醇胺盐、二氨基嘧啶氧化物、丁二醇、苯氧乙醇、旱金莲提取物、苯氧乙醇、人参根提取物、水解胶原、姜根提取物、三肽-1铜以及芳香剂,搅拌均匀,均质10分钟,搅拌混匀后,加入防腐剂并混匀,得到混合液C;
待温度35℃,抽真空,出料罐装,得到防脱发洗发液。

1、人体安全性实验
实验物:实施例1产品
受试者:共30名志愿者,男性9人,女性21人,实验人员年龄在29~59岁,平均年龄40.9±9.2岁,符合受试者志愿入选标准;
实验方法:按照化妆品产品标签注明的使用特点和方法让受试者直接使用供试物,每周1次观察或电话随访受试者皮肤反应,按《化妆品安全技术规范》(2015年版)中人体试用试验皮肤不良反应分级标准记录结果,试用时间为4周,将30例受试者不同时间段的皮肤反应情况填入表5,测试中产生的人体不良反应填入表6。

2、产品功效测试:
5α-还原酶抑制试验
实验方法:
利用“一步夹心法”检测供试物对5α-还原酶的抑制率,抑制因子(可能存在于样品中的物质)作用于吸附在抗体(抗体已固着在酶标板凹孔)上5α-还原酶,使其活性受到抑制,处理后微孔上仍然存留的5α-还原酶与HRP标记的免疫酶连接,催化底物显色,终止反应后测试吸光值,通过分析数据,评估供试物5α-还原酶的抑制效果,并以此数据科学合理的评价样品的控油功效。
实验指标
5α-还原酶的活性比较(单位:U/ml)
实验步骤
①将样品用0.1mol/L Tris-HCl缓冲液稀释N倍作为下步试验用样品,从室温平衡20min后的铝袋中取出所需板条,剩余板条用自封袋密封放回4℃,注意:本样品浓度不适宜过高,过高导致样品与还原酶浓度失衡,最适合浓度需要进行预实验。
②设置标准品系列孔、阳性对照孔、样品孔、空白孔;标准品孔各加不同浓度的标准品50μl,样品孔与阳性对照孔均根据预实验的结果加入合适的量。
③标准品系列孔(6个)、阳性标准品孔、样品孔中每孔加入辣根过氧化物酶(HRP)标记的检测抗体100μl。用封板膜封住反应孔,37℃水浴锅或恒温箱温育60min。
④弃去液体,吸水纸上拍干,洗板5次
⑤每孔加入底物A、B各50μl,37℃避光孵育15min
⑥每孔加入终止液50μl,15min内,在450nm波长处测定各孔的OD值,通过标准品系列的吸光值建立曲线得出阳性标准品孔、样品孔中5α-还原酶的活性,并计算样品的抑制率,实验结果填至表7。

由表7数据可知,实施例1~3的5α-还原酶抑制率均在50%以上,对比例1的5α-还原酶抑制率较低,由此说明按照本申请配方制备的防脱发洗发液通过抑制5α-还原酶的活性,从而减少睾酮转换DHT,阻止毛囊周围胶原形成,活化毛囊,刺激毛囊再生,进而达到防脱育发生发效果。
抑菌实验测试
糠秕马拉色菌抑菌抑制试验
实验方法:
取实施例1~3以及对比例1供试产品,进行样品处理,确定作用浓度和作用和时间,加入糠秕马拉色菌菌悬液与样品充分混合,待作用时间结束,选取适宜的稀释度进行活菌菌落计数,30℃培养4天,同时做对照,试验重复3次,求平均值,菌落计数,记录数据,计算抑菌率。
本试验通过抑菌测试,判断待测样品对糠马拉色菌抑菌作用,大量研究表明马拉色菌与头皮屑产生相关,马拉色菌属于具有脂肪酶活性的真菌,当头皮分泌过多皮脂,头皮上的马拉色菌通过分泌脂肪酶将皮脂中的甘油三醋和饱和脂肪酸分解成不饱和脂肪酸,而不饱和脂肪酸具有促炎性,可以渗透进入头皮的角质层,引起头皮的猛痒感,而且能够让钞离子流入头皮细胞,这会进一步导致头皮角质细胞过度角化,从而使得头皮角质异常以及头皮屑加速产生。
按照OB/T 2738-2012《日化产品抗菌抑菌效果的评价方法》供试产品评价抑菌效果,当抑菌率≥50%时表征为产品具有抑菌效果,从而说明供试物具有去屑功效。

由表8测试结果可知,实施例1~3对糠秕马拉色菌的抑菌率均大于50%,即实施例1~3制备的产品抑菌效果好,供试物具有去屑功效。
防脱发功效测试:
材料和方法:
试验产品:实施例1产品。
对照产品:对比例1产品,不含防脱发功效成分的相应试验产品基质配方产品,与试验产品平行测试。
洗脱期产品:同对照产品,洗脱期内试验组和对照组均使用对照产品。
受试者:完成测试有效受试者共65人,年龄21~59岁,其中试验组33人,男性6人,女性27人,平均年龄40±10岁,对照组32人,男性5人,女性27人,平均年龄39±9岁。两组性别和年龄均无统计学差异(P>0.05),符合受试者志愿入选和排除标准。

试验方法:按照《化妆品安全技术规范》(2015年版)化妆品防脱发功效测试方法的具体要求进行检验。入组的合格受试者首先使用洗脱期产品2周,洗脱期结束后将符合要求的受试者根据随机表分为试验组和对照组,分别发放试验产品和对照产品,按照产品使用说明正确使用产品12周,试验期间要求受试者记录使用日记并严格遵守限制要求。分别在产品使用前和使用后4周、8周、12周访视,每次访视进行脱发计数、毛发密度评估测试和图像拍摄。

综合表9~12的防脱发功效测试,试验产品使用4周、8周、12周后,脱发计数显著减少(P<0.05),且使用12周后,前后脱发计数差值显著低于对照组(P<0.05);使用8周、12周后,前后局部毛发密度差值显著高于对照组(P<0.05),认定试验产品有防脱发、促进毛发生长的作用功效;即本申请提供的防脱发洗发液通过添加多种固发、生发营养成分,将不同作用机理的防脱有效活性成分进行组合,有效抑制5α-还原酶活性、抑制油脂过度分泌及马拉色菌的滋生、改善相关炎症、改善微循环、改善毛囊微小化、强化毛囊,达到防脱发、生发固发效果,进一步提升防脱发产品的有效性及安全使用性能。

降温至45~50℃时,混合液B中加入泛醇、侧柏叶提取物、水解蚕丝、吡罗克酮乙醇胺盐、二氨基嘧啶氧化物、丁二醇、苯氧乙醇、旱金莲提取物、苯氧乙醇、人参根提取物、水解胶原、姜根提取物、三肽-1铜以及芳香剂,搅拌均匀,均质5~10分钟,搅拌混匀后,加入防腐剂并混匀,得到混合液C;待温度降至30~35℃,抽真空,出料罐装,得到防脱发洗发液。
