技术领域
本发明涉及一种人类白细胞主抗原HLA-A和HLA-B共敲除的多血系分化潜能永生化细胞及其制造方法,属于生物医药技术领域。
背景技术
人类白细胞抗原(Human leukocyte antigen,HLA),是人类主要组织相容性复合体的主要组成部分,包含多类抗原成员,它们由一系列紧密连锁的基因座编码,位于6号染色体(6p21 .31)。HLA与人类的免疫系统功能密切相关,这些抗原在人类不同个体之间各有特征。这类抗原之所以被称为白细胞抗原的原因,在于首先发现白细胞膜上高表达这些抗原,但后来发现全身大多数的组织细胞也表达这一类的抗原。供血者和受体的人类白细胞抗原差别越大,免疫排异反应就越大,严重排异可导致输血受体死亡。HLA包括有I、II、III类抗原。HLA的I类抗原是异体间发生免疫排异最主要的抗原,其中HLA-A和HLA-B又是HLA的I类抗原中的主要成分,对异体排异起主导作用。
因此,HLA-A和HLA-B抗原引起的异体免疫排斥,是目前异体血细胞输注(也包括干细胞移植)中存在的最大安全风险因素,临床上需要通过严格的HLA配型以降低输血以及干细胞移植的风险。但是,由于配型成功的机率低,严重限制了输血救治的可及性,并且在多次输血后,易发生抗原抗体反应,从而引起免疫排斥和发热反应。由于基于基因编辑技术的体外造血技术无伦理限制,可以获得无免疫排斥的通用型人类血液或血细胞,所以未来有可能对血液病治疗带来革命性影响,也可能从根本上解决医疗用血困境,以及解决未来养生保健的输血供应。所以,通过体外基因改造,获得与体内完全相同或更优化的人类血液或血细胞,将颠覆现有的医疗范式和健康方式。
体外造血的主要研发方向,是运用基因编辑技术对人血系细胞在体外进行精准编辑,以期获得异体输注后不再有免疫排斥的通用型血液或血细胞。有些血细胞(包括红细胞、血小板)因没有细胞核不能直接进行基因编辑。而且,成熟的血细胞由于不能增殖传代,到达其自然寿命后即发生凋亡,所以有核的成熟血细胞即使能够实施基因编辑,也没有实际应用价值。真正有应用前景的是制造能够可持续或批量地产生成熟血细胞的多血系分化潜能的永生化的细胞。
目前,涉及血细胞抗原基因编辑的细胞包括T细胞、巨核细胞或胚胎干细胞,如专利1(公开号:CN107630006A;发明名称:一种制备TCR与HLA双基因敲除的T细胞的方法),专利2(公开号:CN114929866A;发明名称:基因改造巨核细胞、改造血小板和它们的制造方法)和专利3(公开号:CN107921148A;发明名称:通用供体干细胞和相关方法),都运用了先进的CRISPR/Cas9技术进行基因组精准编辑。但是,专利1涉及T细胞,是制备无免疫排斥的人T细胞,由于这类血细胞已是终末分化的细胞,不再具有长久的分化或增殖能力,而且该专利涉及敲除β2M基因, 制备的T细胞易被自然杀伤细胞杀死,不能正常存活,故该类改造后的T细胞应用产业制备不可行;专利2涉及人巨核细胞,其改造后的巨核细胞只能用于诱导无免疫排斥的血小板这一种类型的血细胞,故产业应用面比较局限;专利3涉及人胚胎干细胞,其公开了不表达一种或几种人类白血病抗原的胚胎干细胞的制备技术,该专利也涉及敲除β2M基因, 制备的胚胎细胞易被自然杀伤细胞杀死,不能正常存活,而且,从胚胎细胞定向诱导成为血液细胞的技术障碍多、难度大。特别是,专利3由于涉及编辑或敲除的基因数量多,其中被编辑或敲除的有些基因是造血过程必需的,对其编辑会影响造血发生。而且,该专利发明没有证明其制备的胚胎干细胞能够定向分化成血细胞。因此该发明制备的胚胎细胞中尽管多个抗原基因被敲除,未来以此制备没有免疫排斥或降低免疫排斥的血细胞没有应用前景。
迄今,尚无一种既能够实现没有抗原免疫排斥,又可以向多种功能血细胞方向分化的用于持续、批量生产成熟血细胞的通用型细胞。
发明内容
为解决上述技术问题,本发明运用精准的基因组敲除技术(CRISPR/Cas9),在具有多血系分化潜能的永生化细胞中同时敲除HLA-A和HLA-B抗原基因,获得没有HLA-A和HLA-B抗原免疫排斥的、可以向多种成熟血细胞方向分化的多血系分化潜能永生化细胞,用以诱导生成无HLA-A和HLA-B免疫排斥的红细胞、血小板、巨噬细胞等。
本发明具体是利用CRISPR/Cas9的精准基因编辑技术,通过设计靶向人类白细胞两个主抗原基因HLA-A/HLA-B的sgRNA序列,构建了lentiCas9-Blast-HLA-A/HLA-B双基因共敲除的表达质粒,进而通过稳定转染方式导入到具有多血系分化潜能的永生化细胞中,从而建立了没有HLA-A和HLA-B免疫排斥而具有红系、巨核系和单核巨噬系多向分化潜能的永生化细胞的制造方法。该多血系分化潜能的永生化细胞,通过定向诱导,可以分别生成红系各级细胞(包括红细胞)、巨核系各级细胞(包括血小板)和单核巨噬系(包括巨噬细胞)各级细胞,产业上可用以生成成熟红细胞、血小板、巨噬细胞,也可作为细胞模型研究体外造血技术。
本发明的第一个目的是提供一种人类白细胞主抗原基因HLA-A和HLA-B共敲除的多血系分化潜能永生化细胞,所述细胞是在多向分化潜能细胞中,稳定转染了一种包含靶向敲除人源HLA-A基因和HLA-B基因的sgRNA的表达载体。
进一步地,所述HLA-A和HLA-B共敲除的多血系分化潜能永生化细胞能够被诱导分化成红系、巨核系或单核巨噬系的祖细胞或成熟血细胞。
进一步地,所述多向分化潜能细胞包括永生化的造血干细胞、永生化的造血祖细胞、永生化的髓系祖细胞、永生化的巨核-红系祖细胞、永生化的红系祖细胞、永生化的巨核系祖细胞、永生化的淋系祖细胞和永生化的人工诱导多功能干细胞中的任何一种。
进一步地,靶向敲除人源HLA-A基因的sgRNA序列如SEQ ID NO.1所示。
进一步地,靶向敲除人源HLA-B基因的sgRNA序列如SEQ ID NO.2所示。
本发明的第二个目的是提供所述多血系分化潜能永生化细胞的制造方法,包括如下步骤:
S1、将靶向敲除人源HLA-A基因和HLA-B基因的sgRNA构建到整合了CRISPR/Cas9的载体上得到表达载体,将表达载体和包装质粒混合液加入病毒包装细胞中进行培养,培养后收集细胞上清液,过滤得到含有表达载体的慢病毒;
S2、采用慢病毒对处于对数生长期的多向分化潜能细胞进行侵染,侵染后洗病毒,并筛选得到所述多血系分化潜能永生化细胞。
进一步地,所述制备方法,具体包括如下步骤:
S1、慢病毒制备:培养病毒包装细胞至70-80%汇合度时进行慢病毒质粒制备,将含有靶向敲除人源HLA-A基因和HLA-B基因的sgRNA的表达载体和包装质粒混合液加入病毒包装细胞中进行培养,20-30 h后更换新鲜的培养基,45-60 h收集细胞上清,过滤膜进行过滤,获得含有表达载体的慢病毒;
S2、细胞转染:取对数生长期的多向分化潜能细胞,按照2-3×105/ mL接种,并按照800 μL/mL添加慢病毒进行侵染,6-10 h后洗病毒,并添加新鲜培养基,按照1-3 μg/mL添加嘌呤霉素进行药筛处理。
进一步地,所述病毒包装细胞为293T细胞、293T/17细胞、293T/17 SF细胞、293H细胞、293-6E细胞、293F细胞、293FT细胞或MEF细胞。
进一步地,所述载体为pLentiCas9-Blast、pLentiCas9-BFP、pLentiCas9-GFP、pLentiCas9-T2A-GFP、dCas9 plasmid、pLenti_dCas9-2xAM、lenti-dCas9-VP64-BFP、pLentiV2-dCas9-VP64、LentiV_Cas9_puro、lenti-SpCas9 puro或lenti-SpCas9 hygro。
本发明的第三个目的是提供所述多血系分化潜能永生化细胞在制造血细胞产品中的应用。
本发明的有益效果是:
本发明利用多血系分化潜能的细胞在体外可诱导分化为红系/巨核系/单核巨噬细胞系的多血系分化特性,运用CIRPR/Cas9技术制备了靶向敲除HLA-A和HLA-B双基因的多血系分化潜能细胞,为未来应用工程化多血系潜能细胞在体外生产成熟红细胞、血小板、巨噬细胞提供了技术方法。
此外,本发明使用更具稳定性和长期性的稳转方式进行基因编辑,可避免电击打孔对细胞造成的损伤。电穿孔方式对细胞易造成较大损伤,尽管可达到将质粒导入细胞的目的,但易导致细胞状态差,功能下降。且利用电穿孔或脂质体转染的方式为瞬转方式,无法维持长期稳定的敲除效果。电穿孔导入方法还会损伤细胞DNA,有致突变风险。

具体实施方式
下面结合具体实施例对本发明作进一步说明,以使本领域的技术人员可以更好地理解本发明并能予以实施,但所举实施例不作为对本发明的限定。
下述实施例中所使用的实验方法如无特殊说明,均为常规方法。
下述实施例中所用的材料、试剂等,如无特殊说明,均可从商业途径得到。
实施例1:利用CRISPR/Cas9技术靶向敲除HLA-A/HLA-B双基因表达载体的构建
1、靶序列确定
利用人源HLA-A/HLA-B基因外显子信息,设计sgRNA序列(如SEQ ID NO:1、2所示),其中这些sgRNA序列如下表1所示:

在其5’端加上KpnI酶切位点GGTACC,在其3’端加上EcoRI酶切位点GAATTC,合成目的序列(如SEQ ID NO:3所示)。其中该目的序列如下表2所示:

2、CRISPR/Cas9整合至载体lentiCas9-Blast,利用KpnI和EcoRI内切酶切割,琼脂糖凝胶电泳,胶回收载体片段。
3、连接sgRNA编码片段和lentiCas9-Blast载体,合成目的表达载体lentiCas9-Blast-HLA-A/HLA-B,随后转化,挑菌和质粒提取进行酶切鉴定,酶切体系如表3所示,37℃酶切1 h,琼脂糖凝胶鉴定条带大小,鉴定结果如图1所示,选择正确质粒进行扩增及后续细胞转染。

4、lentiCas9-Blast-HLA-A/HLA-B载体质粒扩增。
1)取干净1.5 mL EP管加入20 μL DH5α感受态细胞,加入100 ng lentiCas9-Blast-HLA-A/HLA-B载体质粒,冰上孵育30 min;放于水浴锅42℃热激;随后至于冰上冰浴2min,加入1 mL不含氨苄霉素的液体LB培养基,液体LB培养基如表4所示,混匀后,放于37 ℃摇床40 min;取出后3000 rpm/min离心2 min,弃上清,保留100 μL左右上清,吹打混匀后取40 μL进行涂板,先正置于37 ℃培养基中1 h,随后进行倒置培养。

2)挑取单克隆,连同枪头一起打入预先加入5 mL含氨苄霉素的液体LB培养基中,37 ℃小摇后取1 mL菌液进行质粒抽提。
3)将上一步筛选出来符合预期的克隆,取1 mL鉴定正确的菌液加入含有预先加入氨苄霉素的200-300 mL液体LB培养基的锥形瓶中,放置于37 ℃摇床中过夜培养。
4)取全部菌液进行质粒抽提。
实施例2:靶向敲除HLA-A/HLA-B双基因表达载体慢病毒的制备
1、HEK293T细胞培养,培养HEK293T细胞至70-90 %汇合度时进行慢病毒质粒制备。
2、慢病毒制备前2-4 h更换新鲜的含10% FBS的DMEM培养基,将含有表达载体lentiCas9-Blast-HLA-A/HLA-B和包装质粒混合配置成质粒包装体系。
3、24 h后更换10 mL新鲜的培养基,72 h后收集培养基,使用0.22 μm滤膜进行过滤,即可获得含有表达载体lentiCas9-Blast-HLA-A/HLA-B的慢病毒。
实施例3:含有lentiCas9-Blast-HLA-A/HLA-B双基因敲除表达载体的具有多血系分化潜能永生化细胞的构建
1、取对数生长期的永生化的髓系祖细胞,按照2.5×105/mL种至12孔板,每孔细胞添加800 μL慢病毒进行侵染,8 h后洗病毒,并更换为新鲜培养基,按照2 μg/mL添加嘌呤霉素进行药筛处理。
2、取药筛处理3天后的细胞,进行RNA提取和反转录,获得cDNA,反转体系如表5所示。

3、反应完成后,得到的cDNA溶液于-20 ℃保存,并可直接或稀释后用于RT-qPCR扩增反应,按照SYBR green荧光染料:Primer-F:Primer-R=5:0.5:0.5的比例配制混合体系;按照cDNA:无RNA酶水 =0.5:3.5稀释cDNA。取384孔板,避光,每孔加入6 μL混合体系,4 μL稀释的cDNA。放入高通量荧光定量PCR仪中进行扩增检测。RT-qPCR反应使用引物序列如表6所示(总体积为10 μL)。

4、检测结果
如图2所示,图2代表经过嘌呤霉素筛选,在转染基因敲除质粒(即基因打靶后)的永生化的髓系祖细胞中检测HLA-A及HLA-B的表达水平。以管家基因GAPDH为内参,通过高通量荧光定量PCR检测HLA-A及HLA-B表达水平。其中图2横坐标分别代表空载组和敲除组,纵坐标代表基因打靶后多血系分化潜能细胞中的相对表达水平。从图2的结果可以看出,相较于空载组,敲除组中HLA-A及HLA-B基因表达量均有显著性降低,表明人类白细胞主抗原基因HLA-A及HLA-B同时敲除成功。
实施例4:敲除HLA-A/HLA-B双基因的多血系分化潜能细胞分化为红细胞/血小板/巨噬细胞的能力
1、取药筛处理3天后的细胞,按照2×105/mL种至24孔板,加入40 μM Hemin,诱导红系分化,在第0天、第2天、第4天检测红细胞分化比例。
2、取药筛处理3天后的细胞,按照2.5×105/mL种至12孔板,加入1 nM TPA,诱导血小板产生及巨噬系方向分化,在第0天、第2天、第4天检测血小板产生情况,在第0天、第2天、第4天检测巨噬细胞分化比例。
3、收集孔板中细胞,吹打混匀后吸取50 μL至干净1.5 mL离心管中,加入50 μL预先配置的血小板染色抗体,室温避光染色30min,随后补加150 μL PBS使用流式细胞仪检测血小板生成比例。
4、收集孔板中细胞,500 g离心5min后弃上清,分别加入100 μL预先配置的红细胞、血小板和巨噬细胞表面标志抗体,室温避光染色30min,随后补加随后补加150 μL PBS使用流式细胞仪检测红细胞、血小板、巨噬细胞生成比例。
5、检测结果
如图3至图5所示,分别显示了敲除HLA-A/HLA-B的多血系分化潜能细胞经诱导后向红细胞、血小板和巨噬细胞的分化能力。空载组、敲除组和空白组分别表示空载质粒重组细胞、敲除质粒重组细胞和未处理的细胞。纵坐标代表流式细胞仪检测诱导分化后各类细胞分化比例,横坐标显示诱导分化的第0天、第2天和第4天。从图3至图5的结果可以看出,敲除组具有分化为红细胞、血小板和巨噬细胞的能力。图3显示,与空白组相比,敲除组向红细胞的分化能力略有增强。与空载组相比,敲除组向红细胞分化的能力无显著变化。图4显示敲除向血小板分化的能力与空白组或空载组相仿。图5显示敲除组向巨噬细胞分化的能力略低于空白组或空载组。说明敲除HLA-A/HLA-B后,多血系分化潜能细胞保持了向红细胞、血小板的分化能力,也保留了向巨噬细胞分化的能力。

Claims (6)
1.一种HLA-A和HLA-B共敲除的永生化的髓系祖细胞,其特征在于,所述细胞在永生化的髓系祖细胞稳定转染了一种包含靶向敲除人源HLA-A基因和HLA-B基因的共敲除gRNA表达载体;其中,靶向敲除人源HLA-A基因的gRNA序列如SEQ ID NO.1所示;靶向敲除人源HLA-B基因的sgRNA序列如SEQ ID NO.2所示。
2.一种权利要求1所述HLA-A和HLA-B共敲除的永生化的髓系祖细胞的制备方法,其特征在于,包括如下步骤:
S1、将靶向敲除人源HLA-A基因和HLA-B基因的sgRNA构建到整合了CRISPR/Cas9的载体上得到表达载体,将表达载体和包装质粒混合液加入病毒包装细胞中进行培养,培养后收集细胞上清液,过滤得到含有表达载体的慢病毒;
S2、采用慢病毒对处于对数生长期的永生化的髓系祖细胞进行侵染,侵染后洗病毒,并筛选得到所述永生化的髓系祖细胞。
3.根据权利要求2所述制备方法,其特征在于,所述载体为pLentiCas9-Blast、pLentiCas9-BFP、pLentiCas9-GFP、pLentiCas9-T2A-GFP、dCas9 plasmid、pLenti_dCas9-2xAM、lenti-dCas9-VP64-BFP、pLentiV2-dCas9-VP64、LentiV_Cas9_puro、lenti-SpCas9puro或lenti-SpCas9 hygro。
4.根据权利要求2所述制备方法,其特征在于,所述病毒包装细胞为293T细胞、293T/17细胞、293T/17 SF细胞、293H细胞、293-6E细胞、293F细胞、293FT细胞或MEF细胞。
5.根据权利要求2所述制备方法,其特征在于,在S1步骤中,培养时间为45-60 h。
6.权利要求1所述HLA-A和HLA-B共敲除的永生化的髓系祖细胞在制备成熟红细胞、巨噬细胞、血小板产品中的应用。
