Abstract
本发明提供氧甾醇结合蛋白OSBPL2作为结直肠癌分子标志物及其治疗靶点的应用,涉及结直肠癌技术领域,提出了氧甾醇结合蛋白OSBPL2作为结直肠癌分子标志物的应用,提出了氧甾醇结合蛋白OSBPL2在制备检测结直肠癌的试剂、组织芯片、患者血清中的应用,还提出了OSBPL2基因或者OSBPL2蛋白的激活剂在制备预防和/或治疗结直肠癌的药物中的用途。OSBPL2作为该结直肠癌分子标志物的发现,可以用于预测、诊断和检测结直肠癌,为进一步研究结直肠癌的发病机理,探索结直肠癌的治疗靶点提供了新的实验理论基础和新的方向,并可应用在制备预测、诊断、检测结直肠癌患者试剂或结直肠癌治疗药物的领域,丰富了结直肠癌的诊断、检测和治疗手段。
Description
技术领域
本发明涉及结直肠癌技术领域,具体涉及到一种氧甾醇结合蛋白OSBPL2作为结直肠癌分子标志物及其治疗靶点的应用。
背景技术
结直肠癌是一种高发病率、高致死率的癌症。在临床上,主要采取粪便隐血试验和结肠镜作为结直肠癌的筛查诊断方法,但该方法并不能完全有效确认患者实际情况,且覆盖人群有限。结直肠癌的治疗目前以手术切除为主,放疗、化疗为辅。现有的技术手段可将结直肠癌患者的生存率提高一倍,但由于该疾病的潜伏期较长,通常情况下一经发现癌症病灶就已有远端转移。迄今为止,结直肠癌仍在探索有效的肿瘤诊断标志物及治疗靶点。
目前有公告号为CN111411155B的中国专利公开了一种lncRNA IGFL2-AS1作为结肠癌诊断标志物的应用,里面提出认为敲低结肠癌细胞中的lncRNA IGFL2-AS1,能够明显抑制SW620细胞的增殖、迁移、侵袭能力和恶性程度,表明lncRNA IGFL2-AS1在结肠癌发病过程中发挥重要作用,可以作为诊疗结直肠癌的新的分子标记和药物靶点。又如公告号为CN111592487B的中国专利公开了一类含羟肟酸基团的二芳基乙烯类LSD1_HDACs双靶点抑制剂、其制备方法及应用,里面提出的LSD1/HDACs双靶点抑制剂对人结肠癌HCT-116细胞株和人胃癌MGC-803细胞株显示出较好的体外抗肿瘤活性,为LSD1/HDACs双靶点抑制剂药物的研发提供了基础。
肿瘤转移是结直肠癌致死的重要因素,而上皮间充质转化(Epithelial–mesenchymal transition,EMT)在肿瘤转移中尤为关键。上皮间充质转化是细胞失去上皮特征,例如细胞骨架重塑、细胞结合和极性的缺失等,并获得间充质特性的过程,使细胞迁移、侵袭能力增强。氧甾醇结合蛋白(OSBPL2,Oxysterol binding protein like 2)是细胞内的转运蛋白,负责运输甾醇及磷脂。有文献报道,OSBPL2缺失会重塑肝癌细胞骨架,并抑制细胞迁移、粘附及生长。然而目前还未提出过OSBPL2在结直肠癌转移中的功能。
针对上述内容,本发明提供一种氧甾醇结合蛋白OSBPL2作为结直肠癌分子标志物及其治疗靶点的应用。
发明内容
针对现有技术所存在的不足,本发明目的在于提出一种氧甾醇结合蛋白OSBPL2作为结直肠癌分子标志物及其治疗靶点的应用,具体方案如下:
氧甾醇结合蛋白OSBPL2作为结直肠癌分子标志物的应用。
氧甾醇结合蛋白OSBPL2在制备预测、诊断或者检测结直肠癌的试剂、组织芯片、患者血清中的应用。
OSBPL2基因或者OSBPL2蛋白的激活剂在制备预防和/或治疗结直肠癌的药物中的用途,OSBPL2基因的激活剂包括OSBPL2基因的表达载体和/或结合于OSBPL2基因启动子区域的转录因子表达载体、和/或OSBPL2基因表观修饰的激活剂或抑制剂、和/或OSBPL2基因突变体的激活剂或抑制剂,OSBPL2蛋白的激活剂包括OSBPL2蛋白翻译后修饰、促进蛋白稳定性以及活性的激活剂或抑制剂、和/或OSBPL2蛋白突变体的激活剂或抑制剂。
一种预防和/或治疗结直肠癌的药物,包括载体以及活性成分,活性成分包括OSBPL2基因的激活剂和/或OSBPL2蛋白的激活剂。
进一步的,OSBPL2基因的激活剂包括OSBPL2基因的表达载体和/或结合于OSBPL2基因启动子区域的转录因子表达载体、和/或OSBPL2基因表观修饰的激活剂或抑制剂、和/或OSBPL2基因突变体的激活剂或抑制剂。
OSBPL2基因的激活剂是指一些可以促进OSBPL2基因表达的物质。这些激活剂包括:
OSBPL2基因的表达载体:这是一种可以将OSBPL2基因导入细胞中并促进其表达的载体,类似于“基因注射器”。该种表达载体是一种DNA分子,其中包含OSBPL2基因的完整序列并带有启动子、终止子和调节元件等区域,可以在细胞内转录成mRNA分子,再经过翻译成OSBPL2蛋白质。因此,使用OSBPL2基因的表达载体可以实现在细胞中大量表达OSBPL2蛋白质的目的。
结合于OSBPL2基因启动子区域的转录因子表达载体:这是一种可以结合于OSBPL2基因启动子区域并促进基因表达的载体,类似于“转录调节剂”。转录因子是一类能够结合到基因启动子区域的蛋白质,能够启动或者抑制该基因的转录过程。当一个转录因子结合到基因启动子区域时,它能够引导RNA聚合酶转录出相应的mRNA分子,从而促进基因的表达。
OSBPL2基因表观修饰的激活剂或抑制剂:这是一种可以改变OSBPL2基因表观修饰状态(如DNA甲基化)从而促进或抑制基因表达的物质。OSBPL2基因的表观修饰是指影响基因表达而不改变DNA序列的化学修饰。表观修饰包括DNA甲基化、组蛋白修饰、非编码RNA等。OSBPL2基因表观修饰的激活剂或抑制剂是指能够影响OSBPL2基因表观修饰状态的化合物或物质,以实现对OSBPL2基因表达的调控。例如,可以使用DNA甲基转移酶抑制剂或组蛋白去乙酰化酶激活剂来减少OSBPL2基因的DNA甲基化或者增加组蛋白的去乙酰化,从而促进OSBPL2基因的表达。相反,使用DNA甲基化酶或组蛋白乙酰化酶抑制剂则可以增加OSBPL2基因的DNA甲基化或者组蛋白乙酰化,从而抑制OSBPL2基因的表达。
OSBPL2基因突变体的激活剂或抑制剂:这是一种可以激活或抑制OSBPL2基因突变体(如突变导致基因功能异常激活或失活)的表达的物质。OSBPL2基因突变体的激活剂或抑制剂并不是直接促进OSBPL2基因表达的,它们实际上是能够激活或抑制突变体的活性。突变体是指基因发生了某些不正常的变化,导致它的功能受到影响。有些突变体可能会导致OSBPL2基因的表达升高、异常激活或降低、功能失调,因此使用突变体的激活剂或抑制剂可以恢复或改善OSBPL2基因的表达。
总的来说,这些激活剂或抑制剂可以通过不同的途径促进OSBPL2基因的表达及功能。
进一步的,OSBPL2蛋白的激活剂包括OSBPL2蛋白翻译后修饰、促进蛋白稳定性以及活性的激活剂或抑制剂、和/或OSBPL2蛋白突变体的激活剂或抑制剂。
OSBPL2蛋白的激活剂是指能够促进OSBPL2蛋白发挥作用的化合物或因素。这些激活剂包括:
OSBPL2蛋白翻译后修饰、促进蛋白稳定性的激活剂或抑制剂:OSBPL2蛋白翻译后修饰的激活剂是指能够促进OSBPL2蛋白发生化学修饰的物质,从而改变OSBPL2蛋白的结构和功能,进而促进其活性和稳定性的物质。化学修饰是指生物分子在特定条件下与其他生物分子结合,从而改变其物理性质和功能的过程,包括磷酸化、甲基化、乙酰化、泛素化等等。OSBPL2蛋白翻译后修饰的激活剂可以通过促进OSBPL2蛋白的磷酸化、乙酰化等化学修饰来提高OSBPL2蛋白的活性和稳定性,从而发挥治疗作用。OSBPL2蛋白翻译后修饰的抑制剂指的是一类可以抑制OSBPL2蛋白翻译后修饰的化合物或药物。蛋白质翻译后的修饰包括磷酸化、甲基化、乙酰化、泛素化等多种类型,这些修饰可以影响蛋白质的功能、稳定性和互作等。OSBPL2蛋白的翻译后修饰可以被抑制,进而影响OSBPL2蛋白的功能和稳定性。
OSBPL2蛋白突变体的激活剂或抑制剂:这是一种能够抑制某些突变体形成或降低其活性的化合物或物质。在某些情况下,OSBPL2蛋白可能会发生突变,导致其结构或功能的改变,从而影响其在细胞内的作用。OSBPL2蛋白突变体的激活剂或抑制剂的具体作用取决于突变体引起的影响。如果突变体导致OSBPL2蛋白的稳定性降低或降解加速,则抑制剂可能通过阻止这种降解或加速稳定化蛋白,从而促进OSBPL2蛋白的表达。如果突变体引起的是OSBPL2蛋白功能上的改变,则抑制剂可能通过其他途径来促进OSBPL2蛋白的表达。反之亦然。
进一步的,所述载体采用水、盐水、缓冲液、甘油、乙醇、脂质体、脂质、蛋白、蛋白-抗体缀合物、肽类物质、纤维素、纳米凝胶中一种或者多种的组合。
活性成分为药学上可接受的成分,“药学上可接受的”的成分是适用于人和/或哺乳动物而无过度不良副反应(如毒性、刺激和变态反应)的,即具有合理的效益/风险比的物质。活性成分的含量达到有效量或者有效剂量即可,“有效量”或“有效剂量”是指可对人和/或动物产生功能或活性的且可被人和/或动物所接受的量。
本发明所述的活性成分的有效量可随给药的模式和待治疗的疾病的严重程度等而变化。优选的有效量的选择可以由本领域普通技术人员根据各种因素来确定(例如通过临床试验)。所述的因素包括但不限于:所述的活性成分的药代动力学参数例如生物利用率、代谢、半衰期等;患者所要治疗的疾病的严重程度、患者的体重、患者的免疫状况、给药途径等。
载体设置为药学上可接受的载体,“药学上可接受的载体”指用于治疗剂给药的载体,包括各种赋形剂和稀释剂。载体的选择应与给药方式相匹配,这些都是本领域的普通技术人员所熟知的。
通常药物制剂应与给药方式相匹配,本发明的药物组合物的剂型为注射剂、口服制剂(片剂、胶囊、口服液)、透皮剂、缓释剂。例如用生理盐水或含有葡萄糖和其他辅剂的水溶液通过常规方法进行制备。所述的药物组合物宜在无菌条件下制造。
与现有技术相比,本发明的有益效果如下:
(1)氧甾醇结合蛋白是一种含有酸性磷酸残基的糖蛋白,其在细胞外基质中发挥重要的生物学作用,与细胞黏附、迁移、增殖以及骨代谢等过程密切相关。氧甾醇结合蛋白OSBPL2作为该结直肠癌分子标志物的发现,可以用于预测、诊断和检测结直肠癌,为进一步研究结直肠癌的发病机理提供了新的实验理论基础和新的方向,并可应用在制备预测、诊断或检测结直肠癌患者试剂的领域,丰富了结直肠癌的预测、诊断和检测手段。
(2)本发明提供了结直肠癌的新的治疗靶点,针对这些靶点开发的药物可以干预或调节其活性,从而达到治疗疾病的目的。该靶点可有效用于结直肠癌发展判断、治疗方案选择和/或预后评估,从而为本领域提供了一种新颖的结直肠癌诊断剂和/或治疗剂,具有临床应用前景。

具体实施方式
下面结合实施例及附图对本发明作进一步的详细说明,但本发明的实施方式不仅限于此。
在本实施例中,使用到一个P值的概念。P值是一个统计学指标,用于衡量观察结果与假设之间的差异程度,P值越小,差异越显著,通常P值小于0.05被认为是具有显著性差异的阈值,其中,*代表P值小于0.05,**代表P值小于0.01,***代表P值小于0.001,*P<0.05表示差异显著,**P<0.01表示差异非常显著,***P<0.001表示差异极其显著,*P<0.05、**P<0.01、***P<0.001均是常用的统计学符号,用来表示数据之间的显著性差异。
(1)结直肠癌患者样本及组织芯片
研究中所收集的结直肠癌患者样本来自于同济大学附属杨浦医院,样本的采集及使用均获得病人同意。
将石蜡包埋的结直肠癌病人组织制成组织芯片后,进行免疫组化染色实验,具体如下:
(2)免疫组化染色实验
上述(1)中的组织芯片在4%多聚甲醛中固定过夜后,用PBS清洗。之后,进行石蜡包埋并切片。将切片水化、抗原修复后,进行一抗、二抗染色。苏木精染色后,脱水,封片。样本的免疫组化评分标准为抗体着色程度(0-3),阳性细胞比例(0-4)。
实验结果:图1A为IV期结直肠癌病人中OSBPL2表达的免疫组化代表图,为便于观看着色程度,图1采用彩色效果,OSBPL2低表达抗体着色程度1,阳性细胞比例4,综合评分4;OSBPL2高表达抗体着色程度3,阳性细胞比例4,综合评分12;图1B为OSBPL2表达在IV期结直肠癌病人的Kaplan-Meier生存曲线,OSBPL2Low的曲线在下。如图1B,组织芯片结果显示在IV期结直肠癌中OSBPL2表达越少,患者生存率越低,预后越差,OSBPL2表达越高,患者生存率越高,且*P<0.05,表示两种OSBPL2表达程度不同时,患者生存率差异显著。可知,OSBPL2高表达的IV期结直肠癌患者预后会较好。
由此,发现OSBPL2低表达与IV期结直肠癌患者的恶性预后相关,OSBPL2可作为远处转移结直肠癌的一个良性的结直肠癌肿瘤标记物。
(3)构建质粒及验证稳定细胞株
为了验证OSBPL2在结直肠癌中的生物学功能,本发明利用shRNA慢病毒在结直肠癌细胞中构建OSBPL2敲低(KD)的稳定细胞株。
需要说明的是,在基因敲低实验中,可以通过RNA干扰技术(如使用shRNA)降低目标基因的表达水平,然后观察细胞的生物学行为,如增殖、凋亡、分化等的变化。通过检测敲低后的目标基因的mRNA水平的变化,可以确定目标基因的表达是否被成功抑制。因此,在基因敲低实验中,检测目标基因的mRNA水平是一个常用的指标。
在本发明中,检测OSBPL2 mRNA level的变化可以用于基因敲低实验中的指标之一,OSBPL2 mRNA level指的是细胞或组织中OSBPL2基因的mRNA水平,也称为OSBPL2的mRNA表达水平。mRNA是messenger RNA(信使RNA)的缩写,是由DNA转录成的一类RNA分子,具有指导蛋白质合成的功能。OSBPL2 mRNA level表示在特定条件下,OSBPL2基因转录成mRNA的数量,通常使用实时荧光定量PCR(RT-qPCR)等技术进行检测和分析。通过检测OSBPL2 mRNAlevel的变化,可以研究与OSBPL2基因表达相关的生物学问题,如基因调控、信号转导、细胞增殖、凋亡、分化等。因此,OSBPL2 mRNA level的检测也是用于确认osbpl2基因被成功敲低的指标之一。
具体验证过程如下:
(a)用含有10%胎牛血清FBS(Gibco,10091148)的DMEM无菌培养液(Invitrogen,11995065)或1640培养基(Gibco,11875093)培养HT29和LoVo细胞,并加入1%的青霉素和链霉素(Gibco,15140122)。培养皿或培养瓶置于含5% CO2的37℃细胞培养箱中培养。
(b)首先将OSBPL2的短发夹RNA(shRNA)构建到pLKO.1-PURO载体上。shRNA序列如下:
OSBPL2:5’-GGATTACTTTGAGCGGAATTT-3’;
OSBPL2:5’-GGGAGAAACGTATGAATTAAT-3’;
OSBPL2:5’-GAAGATTTAGGATTCAGATTT-3’;
Scramble:5’-CCTAAGGTTAAGTCGCCCTCG-3’.
其次将慢病毒包装质粒(psPAX2/pMD2.G)与病毒载体通过转染试剂Lipofectamine 3000(Invitrogen)一起转染到HEK293T细胞中。48小时之后,收集细胞的上清液并用0.45μM的过滤器过滤。将病毒液直接感染结直肠癌细胞48小时,去除病毒液,加入筛选药物puromycin(Invivogene)去筛选出稳定细胞株。
(c)RNA提取及qPCR检测、western blot检测
RNA提取及qPCR检测是指用Trizol(诺唯赞)提取上述(4)中的细胞株中的RNA。将提取的RNA进行逆转录并通过检测SYBR绿色荧光进行qPCR实验。qPCR的引物如下:
OSBPL2:
5’-AGAGGTGACCACCTGAGAAAGG-3’;
5’-GTTGATCCTCCAGAGCAGCTTG-3’.
β-actin:
5’-CACCATTGGCAATGAGCGGTTC-3’;
5’-AGGTCTTTGCGGATGTCCACGT-3’.
实验结果:图2使用shRNA,通过慢病毒感染构建HCT116、HT29、LoVo和SW620的稳定敲低细胞株。
需要说明的是,使用shRNA(short hairpin RNA),通过慢病毒感染,构建HCT116、HT29、LoVo和SW620的稳定敲低细胞株,指的是在实验室中进行的一种基因敲低实验。具体来说,通过设计和合成shRNA,将其引入慢病毒质粒中,并将该质粒转染至HCT116、HT29、LoVo和SW620等肠癌细胞系中,使其细胞内表达shRNA。shRNA可特异性地与目标基因的mRNA结合并诱导其降解,从而抑制目标基因的表达。通过使用慢病毒作为载体,实现了目标基因稳定的敲低。HCT116、HT29、LoVo和SW620作为常用的肠癌细胞系,因此该实验用于研究目标基因OSBPL2对肠癌发生和发展的作用。通过检测细胞中目标基因OSBPL2的表达水平,以及观察细胞生长、增殖、凋亡等生物学行为的变化,可以进一步揭示目标基因OSBPL2的功能和作用机制。
具体的,图2A为realtime-PCR检测结直肠癌细胞中OSBPL2的mRNA敲低情况,shcontrol作为对照组分别与OSBPL2 shRNA的1、2、3组形成对比,从HCT116、HT29、LoVo和SW620的四种情况可以看出,OSBPL2 shRNA的1、2、3组的OSBPL2 mRNA level数值均远小于shcontrol对照组的数值,可知,当目标基因OSBPL2的mRNA水平降低时,可以说明目标基因OSBPL2的转录受到了抑制,从而使该基因的蛋白质表达量降低。
western blot检测是指蛋白免疫印迹实验,试验过程为收集细胞后,用PBS清洗并用RIPA裂解液(碧云天)裂解。加入蛋白酶抑制剂及磷酸酶抑制剂。裂解后加入上样缓冲液,煮样。将样品加入SDS-PAGE胶后,转膜。一抗、二抗孵育后,ECL显色。
需要说明的是,ECL作为一种生物分子检测技术,主要使用辐射性同位素标记的抗体与待检测蛋白质结合,然后使用酶促荧光底物将蛋白质检测结果转化为荧光信号。荧光信号的强度可通过特定的检测设备来定量测量,但本身并没有颜色表现。通常,ECL显色的结果会以数字、强度值或灰度图像的形式呈现。本发明的western blot检测中的抗体:OSBPL2(来源为Proteintech),β-actin(来源为Abclonal)。
图2B为western blot检测结直肠癌细胞中OSBPL2的蛋白敲低情况,通过同时检测OSBPL2与Actin的表达水平,shcontrol作为对照组分别与OSBPL2shRNA的1、2、3组形成对比,从HCT116、HT29、LoVo和SW620的四种情况可以看出,OSBPL2的蛋白表达水平明显下降,同时Actin的蛋白表达水平没有明显变化,可知,目标基因OSBPL2的表达被成功抑制。
需要说明的是,Actin是一种细胞骨架蛋白,它是细胞内最主要的蛋白质成分之一。Actin蛋白在细胞内具有多种生物学功能,如维持细胞形态、参与细胞运动、细胞分裂、细胞黏附等。Actin蛋白还在细胞内调节多种信号传导途径,包括细胞增殖、凋亡、分化等生物学过程。在细胞内,actin蛋白通过组装成微丝(微小的纤维状结构),并参与形成细胞骨架,从而保持细胞的稳定性和形态。Actin蛋白在许多生物学研究中都扮演着重要的角色,包括癌症、肌肉疾病、神经系统疾病等的研究。
Actin用作Western blot分析中的内参对照,用于校正样品间蛋白质含量的差异,保证分析结果的准确性。因此,在上述Western blot检测中,除了检测目标蛋白OSBPL2外,同时检测Actin作为对照。在分析实验结果时,实验者通常会将目标蛋白的表达水平与Actin的表达水平进行比较,计算相对表达量,以评估目标蛋白的表达水平变化。
而且,在上述shRNA敲低实验中,Mean±SEM代表每组数据的平均值和标准误差。其中Mean表示平均值,SEM表示标准误差,标准误差是衡量平均值精度的一种统计量,它表示平均值与真实值之间的误差范围。结合图2,Mean±SEM,LoVo和SW620两种情况对应的是**P<0.01,HCT116、HT29两种情况对应的是***P<0.001。因此,**P<0.01表示差异非常显著,***P<0.001表示差异极其显著。可知,在realtime-PCR检测以及Western blot检测中,shcontrol对照组与OSBPL2 shRNA的1、2、3组之间所代表的目标基因OSBPL2的表达量降低以及被抑制的差异的确极大。
图2A通过real-time PCR检测了OSBPL2 mRNA的敲低效率;图2B通过western blot检测了OSBPL2蛋白的敲低效率。
(4)裸鼠成瘤实验
为了验证OSBPL2在结直肠肿瘤生长中的功能,将shcontrol对照组及OSBPL2敲低稳定株进行裸鼠成瘤实验,具体如下:
订购6周的雄性裸鼠(BALB/cA-nu/nu),并饲养在无菌环境中。将稳定细胞系皮下注射到裸鼠背部。一周后开始测量肿瘤大小(每3-5天),并观察肿瘤生长。肿瘤体积按照如下公式计算:V(mm3)=0.5×length×width2。一段时间后,处死裸鼠并收集肿瘤,称量,拍照。
实验结果:图3A将相同数目的shcontrol和OSBPL2敲低(KD)肿瘤细胞注射到裸鼠皮下,22天后处死HCT116和HT29组小鼠,29天后处死LoVo和SW620组小鼠,取出肿瘤后拍照;
图3B统计取出肿瘤的重量,Mean±SEM之后,HCT116、HT29、LoVo三种情况对应的是*P<0.05,SW620这组对应的是ns,*P<0.05表示差异显著,ns表示无显著统计学差异。
图3C裸鼠皮下接种肿瘤细胞后,第8天开始测量肿瘤大小,随后每隔3-4天测量一次,统计肿瘤体积,shcontrol对照组为下方的折线,OSBPL2 KD为上方的折线。显示,Mean±SEM之后,HCT116、HT29两组对应的是**P<0.01,LoVo这组对应的是***P<0.001,SW620这组对应的是ns,*P<0.05表示差异显著,**P<0.01表示差异非常显著,***P<0.001表示差异极其显著,ns表示无显著统计学差异。
结果显示,可知,当敲低OSBPL2时,目标基因OSBPL2的表达量降低后,可显著的加速结直肠肿瘤生长,除了SW620细胞并无差异(图3)。
(5)transwell细胞迁移及侵袭实验
为了验证OSBPL2在结直肠癌转移中的功能,我们首先利用transwell细胞迁移及侵袭实验检测OSBPL2对结直肠癌细胞迁移及侵袭的影响。
将稳定细胞株(5×105/孔,无血清稀释)种在transwell小室(Corning)中的上腔,下腔加入800μL完全培养基。侵袭实验需先用Matrigel基底膜基质包被小室。培养48-72h后,取出小室用多聚甲醛(赛维尔)固定后,用结晶紫(碧云天)进行染色拍照。
图4A transwell小室中铺相同数目的shcontrol和OSBPL2 KD HCT116、HT29、LoVo、SW620细胞,培养2-3天后收获细胞,用结晶紫染色,并用光学显微镜拍照。
图4B统计迁移细胞数目。shcontrol对照组与OSBPL2 shRNA的1、2、3组对比之后,可以看出,Mean±SEM,HT29这组对应为*P<0.05,*P<0.05表示差异显著;HCT116、LoVo、SW620这三组对应为**P<0.01,**P<0.01表示差异非常显著。通过HCT116、HT29、LoVo、SW620四种细胞的实验结果,可得,敲低OSBPL2后,显著促进结直肠癌细胞的迁移能力。
图4C统计侵袭细胞数目。shcontrol对照组与OSBPL2 shRNA的1、2、3组对比之后,可以看出,HT29这组对应为*P<0.05,*P<0.05表示差异显著;LoVo、SW620这三组对应为**P<0.01;HCT116这组对应为**P<0.01,***P<0.001表示差异极其显著。通过HCT116、HT29、LoVo、SW620四种细胞的实验结果,可得,OSBPL2敲低后,显著促进结直肠癌细胞的侵袭能力。
(6)小鼠肝转移模型
为了验证OSBPL2在结直肠癌转移中的体内功能,我们利用shcontrol对照组及OSBPL2敲低稳定株构建小鼠肝转移模型。
需要说明的是,小鼠肝转移模型是指将肿瘤细胞移植到小鼠的肝脏中,观察肿瘤细胞是否能够成功生长、侵袭和转移。这种模型适用于研究肝癌、胰腺癌、结直肠癌等肿瘤细胞在肝脏中的转移和侵袭机制。具体如下:
使用麻醉箱麻醉小鼠,待小鼠翻正反射消失后可进行手术,转移到桌面后用面罩维持麻醉药物吸入。用眼科剪或组织剪剪开皮肤,暴露肌层,用眼科剪剪开肌肉,暴露脾脏。将脾脏稍拖出,镊子固定脾脏,吸取一定量细胞(2x106个/只,50ul),用胰岛素针推入脾脏。退针后迅速用酒精棉签按压住注射部位,加压按压,防止细胞悬液渗出及出血。计时5分钟后将小鼠脾脏提起,沿脾脏下缘分离脾脏,有细小出血点电凝止血,分离至脾动脉和脾静脉处时结扎血管,直至完全离断脾脏。观察是否存在活动性出血,还纳腹腔内容物,肌层视切口大小用圆针间断缝合1-2针,皮肤视切口大小用三角针间断/连续缝合数针。将小鼠放置于加热垫上待其复苏后放回鼠笼。
图5A为敲低OSBPL2后小鼠肝转移模型的肝脏对比图;图5B为敲低OSBPL2后小鼠肝转移模型的肝脏HE染色图。结果显示,OSBPL2表达的减少显著促进小鼠肝转移、肝脏的转移瘤体积增大、数量显著增多(图5A)。挑选每组具有代表性的肝脏进行HE染色,结果显示敲低OSBPL2后肝脏内浸润的肿瘤面积显著增多,如图5B。
图6A为western blot检测结直肠癌细胞中OSBPL2的蛋白升高情况图;图6B为OSBPL2过表达后治疗小鼠结直肠癌肝转移的肝脏对比图;图6C为OSBPL2过表达后治疗结直肠癌小鼠肝转移的肝脏HE染色图。将OSBPL2的全长CDS序列克隆到pCDH载体上并通过慢病毒构建OSBPL2过表达(OE)稳定细胞,western blot检测OSBPL2表达情况(图6A)。过表达OSBPL2可显著抑制结直肠癌细胞引起的肝转移,具体表现为肝脏的转移瘤体积变小、数量显著减少(图6B)。挑选每组具有代表性的肝脏进行HE染色,结果显示OSBPL2过表达后肝脏内浸润的肿瘤面积显著减少(图6C)。
通过敲低OSBPL2抑制OSBPL2蛋白表达,发现抑制OSBPL2蛋白表达诱导结直肠肿瘤生长,促进结直肠癌细胞迁移、侵袭及肿瘤转移,而过表达OSBPL2可以显著治疗小鼠结直肠癌肝转移。因此OSBPL2蛋白是一个潜在的结直肠癌治疗新靶点,在此基础上,研究其在结直肠癌中的治疗靶点,可以为结直肠癌相关疾病提供治疗思路和理论依据,为筛选和制备治疗结直肠癌的药物提供新靶点。
以上所述仅是本发明的优选实施方式,本发明的保护范围并不仅局限于上述实施例,凡属于本发明思路下的技术方案均属于本发明的保护范围。应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理前提下的若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (7)
1.氧甾醇结合蛋白OSBPL2作为结直肠癌分子标志物的应用。
2.氧甾醇结合蛋白OSBPL2在制备预测、诊断或者检测结直肠癌的试剂、组织芯片、患者血清中的应用。
3.OSBPL2基因或者OSBPL2蛋白的激活剂在制备预防和/或治疗结直肠癌的药物中的用途,其特征在于,OSBPL2基因的激活剂包括OSBPL2基因的表达载体和/或结合于OSBPL2基因启动子区域的转录因子表达载体、和/或OSBPL2基因表观修饰的激活剂或抑制剂、和/或OSBPL2基因突变体的激活剂或抑制剂,OSBPL2蛋白的激活剂包括OSBPL2蛋白翻译后修饰、促进蛋白稳定性以及活性的激活剂或抑制剂、和/或OSBPL2蛋白突变体的激活剂或抑制剂。
4.一种预防和/或治疗结直肠癌的药物,包括载体以及活性成分,活性成分包括OSBPL2基因的激活剂和/或OSBPL2蛋白的激活剂。
5.根据权利要求4所述的预防和/或治疗结直肠癌的药物,其特征在于,OSBPL2基因的激活剂包括OSBPL2基因的表达载体和/或结合于OSBPL2基因启动子区域的转录因子表达载体、和/或OSBPL2基因表观修饰的激活剂或抑制剂、和/或OSBPL2基因突变体的激活剂或抑制剂。
6.根据权利要求2所述的预防和/或治疗结直肠癌的药物,其特征在于,OSBPL2蛋白的激活剂包括OSBPL2蛋白翻译后修饰、促进蛋白稳定性以及活性的激活剂或抑制剂、和/或OSBPL2蛋白突变体的激活剂或抑制剂。
7.根据权利要求6所述的预防和/或治疗结直肠癌的药物,其特征在于,所述载体采用水、盐水、缓冲液、甘油、乙醇、脂质体、脂质、蛋白、蛋白-抗体缀合物、肽类物质、纤维素、纳米凝胶中一种或者多种的组合。
