The multi-effect traditional Chinese medicine antibacterial agent comprises, by mass, 5-30 parts of mugwort leaf, 10-40 parts of giant knotweed, 10-40 parts of spanishneedles herb, 5-25 parts of pennisetum hydridum, 20-45 parts of beet, 5-20 parts of weeping forsythiae capsule and 5-15 parts of glaucescent fissistigma root. The multi-effect traditional Chinese medicine antibacterial agent can prevent and treat streptococcus infection of fish, improve the immune function and the breeding seedling rate of fish bodies, reduce the use of chemical medicines and antibiotics, has the characteristics of high efficiency, low toxicity, yield increase, environmental protection, low cost, difficult generation of drug resistance of pathogenic bacteria and the like, and has good application prospect.
Description
Technical Field
The invention belongs to the technical field of antibacterial agents, and particularly relates to a multi-effect traditional Chinese medicine antibacterial agent, and a preparation method and application thereof.
Background
With the continuous improvement of the intensive degree of fishery culture, the occurrence probability of various infectious diseases is increased year by year, wherein streptococcal diseases become the greatest threat for healthy culture of tilapia. Streptococcus is a gram positive bacterium of Streptococcus genus, is a typical pathogenic bacterium commonly suffered by human, livestock and fish, and can cause various suppurations and septicemia, and also can cause local infection. Epidemiological investigation of streptococcicosis in the main tilapia farmed areas of asia and latin america has found that the main pathogenic bacteria are streptococcus agalactiae and streptococcus ragus. At present, related research at home and abroad shows that streptococcus agalactiae and streptococcus iniae can infect more than 30 fishes including tilapia, so that the fish is ill. Because streptococcus can avoid the immune defense of phagocytes, further penetrate through the blood brain barrier, destroy brain nervous system, damage organs such as liver, kidney and the like, the death rate of the fish with disease is more than 80%, the disease speed is high, the epidemic range is wide, and once the streptococcus disease is exploded, huge economic loss is easily caused.
The occurrence of the fish streptococcicosis is the result of the combined action of environmental factors, fish body resistance and pathogenic bacteria, and the fish streptococcicosis is mainly prevented and controlled. At present, the prevention and treatment of streptococcicosis of fishes in the aquaculture industry mainly uses various chemical medicines and antibiotics, and the serious residue of forbidden medicines of aquatic products is easily caused by a large amount of use, so that the quality of cultured tilapia products is seriously influenced, and the problems of enhanced drug resistance of streptococcicosis, ecological environment pollution and the like are caused.
The Chinese herbal medicine as natural material has the advantages of wide resource, less toxic side effect, less possibility of drug resistance of pathogenic bacteria, multi-target and multi-path synergistic pharmacological action, etc., and is a better antibiotic substitute because of the rich active substances such as flavone, alkaloid, organic acid, immune polysaccharide, etc. In order to reduce the harm of diseases to fish bodies, ensure the healthy development of fishery and improve the economic benefit of farmers, the development of a multi-effect traditional Chinese medicine antibacterial agent is a problem to be solved urgently at present.

The invention provides a multi-effect traditional Chinese medicine antibacterial agent, which comprises, by mass, 5-30 parts of mugwort leaf, 10-40 parts of giant knotweed, 10-40 parts of spanishneedles herb, 5-25 parts of pennisetum hydridum, 20-45 parts of beet, 5-20 parts of weeping forsythiae capsule and 5-15 parts of glabrous greenbrier rhizome.
In one embodiment of the invention, the Chinese herbal medicine comprises, by mass, 10-20 parts of mugwort leaf, 10-25 parts of giant knotweed, 10-25 parts of spanishneedles herb, 10-20 parts of pennisetum hydridum, 20-30 parts of beet, 5-15 parts of weeping forsythia and 5-10 parts of meadowrue-leaf.
In one embodiment of the invention, the Chinese herbal medicine comprises 20 parts of folium artemisiae argyi, 20 parts of polygonum cuspidatum, 20 parts of sticktight, 10 parts of pennisetum hydridum, 20 parts of beet, 8 parts of fructus forsythiae and 7 parts of thalictrum spanishmani.
In one embodiment of the invention, the Chinese medicinal composition comprises 20 parts of folium artemisiae argyi, 20 parts of polygonum cuspidatum, 10 parts of sticktight, 10 parts of pennisetum hydridum, 20 parts of beet, 8 parts of fructus forsythiae and 7 parts of thalictrum spanishmani.
The second object of the invention is to provide a preparation method of the multi-effect traditional Chinese medicine antibacterial agent, which comprises the following steps,
(1) Pulverizing folium Artemisiae Argyi, rhizoma Polygoni Cuspidati, herba Bidentis Bipinnatae, herba Phyllostachydis Henonis, beet, fructus forsythiae and herba Thalictri Pratentis to 180-200 mesh, mixing, and mixing according to mass volume ratio of 1:14-16, soaking in distilled water for 20-24 hr, boiling for 3-4 hr, and filtering to obtain primary filtrate and primary residue;
(2) And (3) adding the raw materials into the primary filter residue in the step (1) according to the mass volume ratio of 1:9-11 adding distilled water, heating and boiling for 2-3 h, and filtering to obtain secondary filtrate and secondary filter residue;
(3) Combining the primary filtrate in the step (1) and the secondary filtrate in the step (2), concentrating under reduced pressure to obtain a relative density of 1.20-1.40 at 60 ℃, and drying under reduced pressure and crushing to obtain the multi-effect traditional Chinese medicine antibacterial agent.
In one embodiment of the invention, the method comprises the following steps,
(1) Pulverizing folium Artemisiae Argyi, rhizoma Polygoni Cuspidati, herba Bidentis Bipinnatae, herba Phyllostachydis Henonis, beet, fructus forsythiae and herba Thalictri Pratentis to 200 mesh respectively, mixing, and mixing according to mass volume ratio of 1:15 adding distilled water, soaking for 24 hours, heating and boiling for 3 hours, and filtering to obtain primary filtrate and primary filter residue;
(2) And (3) adding the raw materials into the primary filter residue in the step (1) according to the mass volume ratio of 1:10 adding distilled water, heating and boiling for 2 hours, and filtering to obtain secondary filtrate and secondary filter residue;
(3) Combining the primary filtrate in the step (1) and the secondary filtrate in the step (2), concentrating under reduced pressure until the concentration is 0.5-1 g/mL, centrifuging at 2800-3200 rpm for 20-25 min, drying under reduced pressure, and pulverizing to obtain the multi-effect traditional Chinese medicine antibacterial agent.
The third purpose of the invention is to provide an application of the multi-effect traditional Chinese medicine antibacterial agent in preparing a medicine for treating fish streptococcosis.
In one embodiment of the invention, the pathogen of fish streptococcosis is Streptococcus.
In one embodiment of the invention, the streptococcus is streptococcus agalactiae and/or streptococcus iniae.
In one embodiment of the invention, the medicament is for killing and/or inhibiting the growth and reproduction of pathogens.
The fourth object of the invention is to provide an application of the multi-effect traditional Chinese medicine antibacterial agent in fish feed.
The traditional Chinese medicine used in the invention has the following properties:
Mugwort leaf: pungent, bitter and warm nature, enter liver, spleen and kidney meridians. The main chemical components of the mugwort leaf comprise volatile oil, flavone, eudesmane, triterpenes and trace elements, the mugwort extract has pharmacological activities such as bacteriostasis, antivirus, antioxidation and the like, and the mugwort as a feed additive component can enhance the immunity of organisms, improve the activity of digestive enzymes, improve the intestinal mucosa morphology, promote the metabolism rate of nutrient substances and the like, and promote the growth of organisms.
Giant knotweed rhizome: slightly bitter and slightly cold, enter liver, gallbladder and lung meridians. Contains stilbenes such as polydatin and anthraquinones such as large Huang Shu, and has liver protecting, antibacterial and antiviral pharmacological activities. Researches prove that the giant knotweed can relieve fatty liver of tilapia caused by high-fat feed, and the giant knotweed alcohol extract has bacteriostatic activity of inhibiting streptococcus mutans and the like.
Herba Bidentis Bipinnatae: sweet, light and bitter. It enters liver, lung and large intestine meridians due to its slightly cold nature. Researches show that the principal components of the spanishneedles herb are flavonoid compounds, polyacetylene compounds, triterpenes and sterols compounds, organic acids and lipids, alkaloids, sugar, vitamins, volatile oil, choline and the like. The herba Bidentis Bipinnatae extract can inhibit antibacterial activity of Staphylococcus aureus, bacillus subtilis, etc.
Pennisetum hydridum: is grass plants of the grass family and the pennisetum genus perennially upright cluster, and has appearance and growth similar to sugarcane. Pennisetum hydridum contains abundant amino acids, is typical of high protein pasture, and contains up to 18.46% of crude protein in dry matter.
Beet: beet is cool in nature, sweet and bitter in taste, and has the functions of clearing heat and detoxicating, removing blood stasis and stopping bleeding. In the diet spectrum of the following-rest, it is recorded that "clearing pathogenic fire, expelling wind, killing parasites, removing toxic substances, removing turbid pathogen, diluting acne, stopping leukorrhagia, regulating menstruation, treating stranguria and dysentery". The extract contains betaine and organic acid, and betaine helps to prevent cell, protein and enzyme from environmental stress, and has antiinflammatory effect.
Fructus forsythiae: the fructus forsythiae extract contains active ingredients such as limonene, cumyl alcohol, phytosterol, benzoic acid, alkaloid, forsythol, phytosterol, alkaloid and the like, and polyphenols in the ingredients contain phenolic hydroxyl groups, have oxidation resistance and antibacterial effect.
Grass of Thalictrum: tender She Jingfu contains multiple vitamins and various microelements, is one of wild vegetables, and can also be used for medicine: it has mild nature and bitter taste, and is suitable for eating, and has effects of clearing heat and detoxicating, invigorating stomach, making acid, and inducing sweat, and the extract contains berberine.
Compared with the prior art, the technical scheme of the invention has the following advantages:
(1) Organic acid and the like can be extracted from beet and the like adopted by the multi-effect traditional Chinese medicine antibacterial agent, so that the stability of other components is improved. The multi-effect traditional Chinese medicine antibacterial agent is added into fish feed, can regulate pH value in stomach, has the effects of activating digestive enzyme and improving amino acid digestion capability, and is helpful for intestinal epithelium growth. In addition, organic acid and the like can inhibit the growth of microorganisms and pathogenic bacteria, improve the feed conversion efficiency and effectively improve the weight of fish.
(2) The multi-effect traditional Chinese medicine antibacterial agent provided by the invention can extract berberine from the thalictrum spani, and has antibacterial property. The berberine and the organic acid have the functions of inhibiting and destroying bacterial biomembrane, so that the combined use of the berberine and the organic acid and other extracts can reduce the use concentration of the berberine and the organic acid, reduce the toxicity and the effective dose of the antibacterial drug, further shorten the acting time of the drug, accelerate the sterilization speed and furthest reduce the drug resistance of bacteria.

The preparation method specifically comprises the following steps:
(1) Pulverizing folium Artemisiae Argyi, rhizoma Polygoni Cuspidati, herba Bidentis Bipinnatae, herba Phyllostachydis Henonis, beet, fructus forsythiae and herba Thalictri Pratentis to 200 mesh respectively, mixing, and mixing according to mass volume ratio of 1:15 adding distilled water, soaking for 24 hours, heating and boiling for 3 hours, and filtering to obtain primary filtrate and primary filter residue;
(2) Adding the mixture into the primary filter residue in the step (1) according to the mass volume ratio of 1:10 adding distilled water, heating and boiling for 2 hours, and filtering to obtain secondary filtrate and secondary filter residue;
(3) And (3) combining the primary filtrate in the step (1) and the secondary filtrate in the step (2), concentrating under reduced pressure until the concentration is 1g/mL, centrifuging at 3000rpm for 20min, taking supernatant, and drying under reduced pressure and crushing to obtain the multi-effect traditional Chinese medicine antibacterial agent.

Test example 1A traditional Chinese medicine antibacterial agent inhibits Streptococcus growth
(1) Preparation of bacterial suspension: 100. Mu.L of Streptococcus agalactiae and Streptococcus iniae bacterial solutions were inoculated into 10mL of BHI liquid medium, and cultured in a shaker at 37℃and 180rpm for 24 hours, with the bacterial solutions having a concentration of about 10 7 cfu/mL.
(2) And (3) bacteriostasis circle experiment: sucking 100. Mu.L of bacterial suspension (Streptococcus agalactiae and Streptococcus iniae) by adopting an agar diffusion method, and adding 900. Mu.L of ultrapure water for dilution by 10 times; 100. Mu.L of the diluted bacterial suspension was pipetted onto a petri dish. Each experiment was repeated 3 times and statistical analysis was performed, and the results are shown in table 1:

As can be seen from Table 1, the materials of the comparative example and the example have bacteriostatic action on streptococcus agalactiae and streptococcus iniae for inhibiting growth, but under the same extraction mode and concentration range, the bacteriostatic effect of the materials of the example on streptococcus agalactiae and streptococcus iniae is more obvious, so that the materials of the invention have better synergistic effect among various Chinese herbal medicines, and the bacteriostatic effect of the materials of the example in combination use is more obvious.
Test example 2 Effect on growth Rate of Tilapia
(1) Healthy tilapia is purchased from a fry factory, the weight of the tilapia is 25g-28g, and experimental feeding is performed after one week of adaptive feeding.
(2) The random groups were divided into four groups, i.e. test group (materials prepared in example 1, comparative examples 1-5) and base group (commercially available Tongwei 130), 3 replicates of each group, 30 tails. The special circulating water system is adopted for oxygen-dissolving feeding, feeding is carried out twice a day in the morning and at the evening, the feeding amount is 5% of the weight each time, the water temperature is regulated and controlled to be 22+/-2 ℃, feces and water are regularly cleaned, the water is changed, the tilapia is weighed after 4 weeks of feeding, the average weight of each group of tilapia is calculated, and the result is shown in Table 2:

As can be seen from Table 2, the growth rate of tilapia in the traditional Chinese medicine antibacterial agent group is obviously increased relative to the weight of the fish in the basic feeding group and the comparative example group, which shows that the traditional Chinese medicine antibacterial agent can obviously improve the growth rate of tilapia, and shows that the traditional Chinese medicine antibacterial agent has better synergistic effect among various Chinese herbal medicines, and the effect of compound use on the growth of fish body is more obvious.
It is apparent that the above examples are given by way of illustration only and are not limiting of the embodiments. Other variations and modifications of the present invention will be apparent to those of ordinary skill in the art in light of the foregoing description. It is not necessary here nor is it exhaustive of all embodiments. And obvious variations or modifications thereof are contemplated as falling within the scope of the present invention.
Claims (9)
1. The multi-effect traditional Chinese medicine antibacterial agent is characterized by being prepared from the following traditional Chinese medicines in parts by mass: 10-20 parts of mugwort leaf, 10-25 parts of giant knotweed, 10-25 parts of spanishneedles herb, 10-20 parts of pennisetum hydridum, 20-30 parts of beet, 5-15 parts of weeping forsythiae capsule and 5-10 parts of meadowrue-leaved sweetflag; the multi-effect traditional Chinese medicine antibacterial agent is used for preparing medicines for treating fish streptococcicosis.
2. The multi-effect traditional Chinese medicine antibacterial agent according to claim 1, wherein the multi-effect traditional Chinese medicine antibacterial agent is prepared from the following traditional Chinese medicines in parts by mass: 20 parts of mugwort leaf, 20 parts of giant knotweed, 20 parts of spanishneedles herb, 10 parts of pennisetum hydridum, 20 parts of beet, 8 parts of weeping forsythiae capsule and 7 parts of largehead atractylodes rhizome.

The multi-effect traditional Chinese medicine antibacterial agent according to claim 1, wherein the multi-effect traditional Chinese medicine antibacterial agent is prepared from the following traditional Chinese medicines in parts by mass: 20 parts of mugwort leaf, 20 parts of giant knotweed, 10 parts of spanishneedles herb, 10 parts of pennisetum hydridum, 20 parts of beet, 8 parts of weeping forsythiae capsule and 7 parts of largehead atractylodes rhizome.
4. The preparation method of the multi-effect traditional Chinese medicine antibacterial agent according to any one of claims 1 to 3, which is characterized by comprising the following steps,
(1) Pulverizing folium Artemisiae Argyi, rhizoma Polygoni Cuspidati, herba Bidentis Bipinnatae, herba Phyllostachydis Henonis, beet, fructus forsythiae and herba Thalictri Pratentis to 180-200 mesh, mixing, and mixing according to mass volume ratio of 1:14-16, soaking in distilled water for 20-24 hr, boiling for 3-4 hr, and filtering to obtain primary filtrate and primary residue;
(2) And (3) adding the raw materials into the primary filter residue in the step (1) according to the mass volume ratio of 1:9-11 adding distilled water, heating and boiling for 2-3 h, and filtering to obtain secondary filtrate and secondary filter residue;

5. Use of a multi-effect traditional Chinese medicine antibacterial agent according to any one of claims 1-3 in the preparation of a medicament for treating streptococcicosis in fish.
6. The use according to claim 5, wherein the pathogen of streptococcicosis in fish is streptococcus.
7. The use according to claim 6, wherein the streptococcus is streptococcus agalactiae and/or streptococcus iniae.
8. Use according to claim 5, wherein the medicament is for killing and/or inhibiting the growth and reproduction of pathogens.
9. Use of the multi-effect traditional Chinese medicine antibacterial agent according to any one of claims 1-3 in preparing fish feed.
