背景技术

传统地,针对酮的定点照护测试是基于发射系数的。由于反应膜的电抗,膜倾向于变黄。这种变黄可能修改所测量到的光学信号。变黄可能导致低分析物浓度下的信号增大(过恢复)和高分析物浓度下的信号减小(欠恢复)。变黄是至少部分由于某种不稳定载色体而产生的。为了避免与不稳定载色体工作,可以进行酮试验作为电化学测试。通过针对电化学试验交换中介物的载色体,试验将要稳定得多。
电化学酮测试条试验的创建提供了:
1.酮测试条的增大稳定性。
2.施加到测试条的血液较少。通过进行安培测试,血液量可以从15μL下降到1.2μL或更少。
3.更快的反应时间。由于电化学测试通常不是端点测试,反应时间可以较快。
4.安培测试的精确度一般较好。
5.具有糖尿病面板的能力——在单个电化学测试条中的葡萄糖和酮。
这种测试条不存在于市场上。
电化学酮测试条的附加优点包括:
1.测试条具有较长的产品有效期并且不会被对光或氧气敏感的载色体损害。
2.施加到测试条的血液较少。通过进行安培测试,血液量可以从15μL下降到1.2μL或更少。
3.更快的反应时间。由于电化学测试通常不是端点测试,反应时间可以较快。
4.安培测试的精确度一般较好。
5.具有糖尿病面板的能力——在单个电化学测试条中的葡萄糖和酮。
酮反射系数测试条试验的实施例使用不稳定且感光的四唑染料。这使得反应膜变黄,从而引起低分析物浓度下过恢复和高分析物浓度下的欠恢复。由于这种载色体问题,需要以更好的方式测量β-羟基丁酸酯(酮)水平。由于反应机制,可以去除有问题的载色体,用中介物代替以创建电化学酮测试。这个概念已经被示出针对电化学测试以制造模型的方式工作。可以另外优化反应物。该技术的变化件提供更可靠的测试条,其具有更长的保存期限并具有安培测试的所有附加优点——更小样本提及、更快速的反应时间、以及更好的精确度。
除了葡萄糖测试以外,有时糖尿病主要检查β-羟基丁酸酯(酮)水平。AmericanDiabetes Assocation(美国糖尿病协会)推荐酮测试应当在生病期间以及葡萄糖水平持续高的任何时候执行。(参见American Diabetes Assocation,Test of Glycemia inDiabetes Care,2004;27(S1):S91-93)。此外,推荐在血液中完成酮测试,因为血液中β-羟基丁酸酯的出现指示酮症的开始比尿中检测到乙酰乙酸更早。优选的是具有针对糖尿病人群的酮和葡萄糖两者的单个测试条或具有可以进行这两种测试的计量器。电化学葡萄糖和酮的糖尿病面板在商业上不可由任何诊断公司使用。然而,由于本发明,允许安培糖尿病/葡萄糖组(葡萄糖和酮)的第一商业开发的大门被打开。
以下反应是针对创建电化学酮测试所提出的反应。当前市场上出售的基于反应的测试使用四唑盐化物Nitro-TB代替产生甲染料的中介物铁氰化物。替代颜色变化,反应现在测量所产生的电流(nA)。反应路径如下所示。
在制造用于酮检测的电化学测试条的一种方法中,空的碳电极条涂覆有酮反应物,其包含与反射系数实验相同比例和浓度的缓冲剂、表面活性剂、羟丁酸脱氢酶、NAD、铁氰化物、以及心肌黄酶。反应物在对流式烤箱中的测试条上烘干。
酮条在具有操纵培养时间、所施加的电势等的能力的稳压器上测试。电势被设置为300mV并且测试不同的培养期。最初,仅测试血清样本。附加的优化可用于电化学试验,但最初的测试已经表明试验是可行的。结果如图1a-1d中所示。从附图中可清楚看出,R正方形良好并且另外截距处于合理水平。测量中的延迟似乎呈现更线性的趋势线。
在另一轮测试中,CardioChek版本1.05用于确定受非培养期、400mV电势和最大20秒测试时间约束的电化学酮的性能。与稳压器相比,CardioChek/>中的参数是相当固定的。试验可以进一步优化反应物、所施加的电势和测试时间。下面的图2显示了使用全血和CardioChek />计量器的结果。截距和R正方形值示出了合理的电势,其考虑到进一步的优化是可用的。
电化学酮的实施例的附加优点中的一个是制作用于测试葡萄糖和酮两者安培糖尿病面板的能力。图3示出了描绘这样的测试条可能如何出现的的附图的一个实施例。每个测试将独立的操作但仅需要一小滴血液。
在图3中,测试条310包括酮区段320和葡萄糖区段330。在酮区段320中,可以提供工作电极335和对电极345。另外,可以提供两个填充检测电极340。从工作电极335、345通过这些填充检测电极340的电流确保血液已经填充了腔室从而指示样本尺寸是充足的。在葡萄糖区段320中,可以提供附加的工作电极345和对电极350。在操作中,可以将血液样本施加到测试条的样本入口360。可以针对每个电极和填充检测器提供测试条的接触区365。另外,可以针对工作接触部375中的一个提供测试条检测电极370,使得可以建立分析电路的电路独立以指示测试条在计量器中的插入。
校准曲线和测试协议可以作为计量器和测试条系统的部分被提供。除了使用计量器以外,各种设备可以被配置为接收测试条并执行测试。可以远程地或在向测试条提供电流的电子设备处完成酮水平和葡萄糖水平的计算。
总之,我们示出了产生电化学酮测试条的能力。这个试验应当显示更好的精确度并且具有更长的保存期限以及更小的样本体积和更快的反应时间。这样的电化学测试还开启了创建用于测试葡萄糖和酮的新型糖尿病面板的可能性。
在用于葡萄糖检测系统的一些替代方案中,不使用心肌黄酶并且替代地使用1-甲氧基PMS。
在许多实施例中,在包括微处理器的设备中提供了系统的部分。本文所述的系统和方法的各种实施例可以完全或部分地用软件和/或固件来实施。这个软件和/或固件可以采用包含在非暂时性计算机可读存储介质上或中的指令的形式。这些指令继而可以被一个或多个处理器读取或执行以实现本文所述的操作的性能。指令可以采用任何适合的形式,例如但不限于:源代码、编译代码、解译代码、可执行代码、稳态代码、动态代码等。这样的计算机可读介质可以包括用于以可由一个或多个计算机读取的形式(例如但不限于只读存储器(ROM)、随机存取存储器(RAM)、磁盘储存介质、光学储存介质、闪速存储器等)存储信息的任何有形非暂时性介质。
本文所述的系统和方法的实施例可以用各种系统来实施,系统包括但不限于智能电话、平板电脑、膝上型电脑以及计算设备与云计算资源的组合。例如,操作的部分可以出现在一个设备中,并且其它操作可以出现在远程位置,例如远程服务器或服务器。例如,数据的收集可以出现在智能电话,并且数据分析可以出现在服务器或云计算资源中。任何单个计算设备或计算设备的组合可以执行所述方法。

测试条,所述测试条包括第一电极和第一对电极以及第二电极和第二对电极,所述第一电极和所述第一对电极位于第一样本接收区中,并且所述第二电极和所述第二对电极位于第二样本接收区中,所述第一样本接收区和所述第二样本接收区经由入口互连,所述第一样本接收区和所述第二样本接收区中的每一个是独立的;
第一涂层,所述第一涂层位于所述第一电极和所述第一对电极中的一个上,所述第一涂层包括酮的中介物;其中,所述酮的中介物为铁氰化物,所述第一涂层另外包括羟丁酸脱氢酶和烟酰胺腺嘌呤二核苷酸(NAD),其中,所述第一涂层另外包括心肌黄酶并且所述第一涂层包括1-甲氧基-5-甲基吩嗪甲基硫酸盐;
第二涂层,所述第二涂层位于所述第二电极和所述第二对电极中的一个上,所述第二涂层包括葡萄糖的中介物;其中,所述第二涂层包括1-甲氧基-5-甲基吩嗪甲基硫酸盐并且所述第二涂层不包括心肌黄酶;以及
与所述测试条接合的计量器,所述计量器提供测量中的延迟以针对酮呈现更线性的趋势线,所述更线性的趋势线由高R正方形值表示,其中,R正方形是表示回归模型中由自变量解释的因变量方差的比例的统计测量结果。
2.根据权利要求1所述的系统,其中,所述第一涂层包括缓冲剂和表面活性剂。
