Description

Skin care composition for repairing skin barrier and application thereof
Technical Field
The invention belongs to the technical field of cosmetics, and particularly relates to a skin care composition for repairing skin barrier and application thereof.
Background
Defective skin barriers can allow more moisture to be lost from the skin and the ingress of external stimuli (e.g., allergens, toxins) to trigger dryness, redness, itching, etc. of the skin. Factors that cause skin to fail in completing the barrier function are various, such as damage to the skin due to ultraviolet rays, or the regeneration of the barrier function of human skin after being stimulated becomes more and more problematic with age.
The barrier function of the skin is mainly represented by the stratum corneum, which is the outermost layer of the skin. The stratum corneum is composed of necrotic keratinocytes and intercellular lipids, and serves as a skin protective film that protects the skin from external stimuli, prevents evaporation of water from the inside, and is responsible for core functions. At the same time, the stratum corneum not only plays a barrier function, but also influences the function, and structure of the cell layers present in the interior, i.e., the epidermis layer or dermis layer.
The lysate of the fermentation product of the saccharomyces cerevisiae, the English name is BIFIDA FERMENT LYSATE, is a generic name of a metabolite, a cytoplasmic fragment, a cell wall component and a polysaccharide complex which are obtained by culturing, inactivating and decomposing bifidobacteria, comprises small molecules such as vitamin B group, mineral substances, amino acids and the like which are beneficial to skin care, and is a high-quality yeast essence which is very suitable for skin care. The lysate of the fermentation product of the saccharomyces cerevisiae can strengthen the metabolism of horny layer, has strong anti-immunosuppression activity, can promote DNA repair, can effectively protect skin from being damaged by ultraviolet rays, and has the effects of enhancing skin immunity and repairing skin barriers.
However, the lysate of the fermentation product of the saccharomyces cerevisiae is difficult to obtain raw materials with high effective content due to factors such as raw material stability, and the like, so that the lysate is high in addition amount during application, and the economic cost cannot be ignored.
Disclosure of Invention
It is an object of the present invention to provide a skin care composition which is capable of repairing the skin barrier, thereby improving the skin moisturization.
In order to achieve the above object, the present invention provides the following technical solutions:
a skin care composition comprising a combination of active ingredients for repairing a skin barrier, the combination of active ingredients comprising a saccharomyces cerevisiae fermentation product lysate, sialic acid, and a pH adjusting agent, wherein the pH adjusting agent is used in an amount such that the pH of the resulting skin care composition is not less than 4.5.
As the known technology, the lysate of the fermentation product of the saccharomyces cerevisiae is also called as lysate of the fermentation product of the saccharomyces cerevisiae, and some cases can be abbreviated as the saccharomyces cerevisiae, which contains vitamin B group, mineral substances, amino acid and other skin-friendly small molecules, can strengthen metabolism of horny layer, has strong anti-immunosuppression activity and can promote DNA repair, thereby having the effects of enhancing skin immunity and repairing skin barrier.
Compared with the method that the yeast fermentation product lysate is used as the active ingredient for repairing the skin barrier, the method has the advantages that the yeast fermentation product lysate is compounded with sialic acid and then used together, the obtained active ingredient combination can obviously improve the effect of repairing the skin barrier, but the yeast fermentation product lysate and the sialic acid are both partial acidic substances, skin injury can be caused when the yeast fermentation product lysate is directly used for normal skin, especially for people with thin horny layer, in order to avoid skin problems such as skin redness and inflammation and poor skin feel problems such as stinging and burning, and the pH regulator is added to balance the skin use feeling. The sialic acid is also called N-acetylneuraminic acid, nidus Collocaliae acid and nidus Collocaliae essence.
The pH regulator is mainly alkaline substances such as arginine, betaine, triethanolamine and the like commonly used in cosmetics, can achieve the aim of the invention, and the dosage of the pH regulator is not excessively limited. However, in one embodiment of the present invention, it is shown that when the basic substance is selected to be arginine, it has a better effect than other pH adjusting agents.
In a preferred embodiment, the mass ratio of lysate to sialic acid of the fermentation product of Saccharomyces cerevisiae is 1 (0.1-3).
In a further preferred embodiment, the mass ratio of lysate of the fermentation product of Saccharomyces cerevisiae, sialic acid is 1 (0.6-1.5).
In the most preferred embodiment, the mass ratio of the lysate of the fermentation product of the Saccharomyces cerevisiae to sialic acid is 1 (0.8-1.2).
Experiments show that in the proportion range, the lysate of the fermentation product of the saccharomyces cerevisiae and sialic acid can improve the effect of repairing the skin barrier more remarkably.
Since the lysate of the fermentation product of the Saccharomyces cerevisiae currently commercially available is mostly an aqueous solution or other mixture, the mass or weight content of the lysate of the fermentation product of the Saccharomyces cerevisiae can be calculated according to the identified effective content.
In a preferred embodiment, the skin care composition comprises:
0.1 to 20% by weight of said active ingredient combination;
wherein each weight% is relative to the total skin care composition.
In the skin care composition, the conventional amount of the active ingredient combination for repairing the skin barrier is 0.1 wt% or more, and when the amount of the active ingredient combination exceeds 20 wt%, harmful irritating side effects may be generated to the human body, so that it is not preferable.
In a preferred embodiment, the skin care composition described above, the skin barrier is characterized by reduced moisture loss from the skin and improved skin moisturization. The stratum corneum, which is the first protective film of the skin, is not only vulnerable to the external environment, but also the skin barrier function of the stratum corneum is not successfully achieved or is damaged if the skin is a dry person. In order to impart moisture retention to dry skin, the problem is generally solved temporarily by applying a moisturizer formulation, but it is difficult to solve the fundamental problem, and further, the skin barrier damaged by ultraviolet rays cannot be improved by simply applying a moisturizer formulation.
In another preferred embodiment, in the above skin care composition, the skin barrier is characterized by inhibiting uv-induced skin damage.
The invention is to prevent skin injury caused by ultraviolet rays, and is understood to be the effect of preventing further deterioration, damage and denaturation of the skin and repairing the occurred photoinjury by the active ingredient combination in the irradiation process of the skin which is damaged or is to be damaged by ultraviolet rays; the meaning of inhibition can be synonymous as slowing down, relieving, alleviating, repairing, treating and the like; skin injury refers to the phenomenon of increased dandruff, desquamation, local hardening, roughness, thickening, dryness and the like on the surface of skin under the toxic action of ultraviolet light, or the phenomenon of various cell swelling, enlargement and the like in the epidermis and dermis of the skin.
Further, the inhibition of skin damage caused by ultraviolet rays is to slow down abnormal keratosis of photoaged skin; or, inhibiting photoaging skin thickening; or, the malondialdehyde content in blood is reduced.
The slowing and repairing of the invention is understood to be that when the active ingredient combination acts on the slightly or severely damaged parts of the skin such as dander, dryness, hardening and the like in the irradiation process of excessive light or daily light, the phenomena such as softness, dander reduction and the like of the damaged skin can be recovered, and the slowing or repairing is synonymous as alleviation, treatment, alleviation, inhibition, repairing and the like.
The abnormal keratosis of the skin refers to the phenomena that the epidermis is obviously thickened, keratinocytes are enlarged, keratinization is excessive and the like after phototoxic skin is formed.
The content of malondialdehyde measured in the invention can reflect the peroxidation degree of body lipid based on the content characteristics of malondialdehyde, and the content can be used as one of indexes for examining the severity of stress on cells.
In a preferred embodiment, the skin care composition is in the form of a facial cleanser, essence, skin lotion, cream, ointment, mask, cosmetic lotion or foundation. Meanwhile, it may appropriately contain components commonly used in cosmetic products or skin external preparations, for example, moisturizers, antioxidants, surfactants, oily components, UV absorbers, emulsifiers, thickeners, aqueous components, water, and various skin application agents, etc., as needed, within a range not impairing the effects of the present invention.
In the present invention, unless otherwise specified:
as used herein, "comprising" is intended to mean "including but not necessarily consisting of … …". In other words, the listed choices are not necessarily exhaustive.
As used herein, "skin care composition" is meant to include compositions for topical application to the skin of a mammal, particularly a human. Such compositions can generally be classified as leave-on or rinse-off, preferably leave-on.
"skin" as used herein is meant to include skin on the face and body (e.g., neck, chest, back, arms, hands, legs, etc.), preferably facial skin.
The beneficial effects obtained by the invention are as follows:
the invention provides a skin care composition which comprises an active ingredient combination for repairing skin barriers, wherein the active ingredient combination adopts the combination of a lysate of a fermentation product of saccharomyces cerevisiae, sialic acid and a pH regulator, so that the effect of repairing the skin barriers can be improved, and the risk of irritation to the skin can be reduced.

50 ICR female mice were adapted for 1 week. The hair on the back of the mice was removed by the rosin paraffin dehairing method, and the area was 2 cm. Times.3 cm. After dehairing, 40 mice are taken as Sup>A model set, and after 0.1ml of methoxsalen solution is smeared on the exposed part of the back skin for 1 hour, the mice are fixedly arranged in Sup>A skin phototoxicity experiment instrument by Sup>A transparent mouse fixer, and the mice are irradiated at Sup>A position about 15cm away from Sup>A light source (the energy measured by an ultraviolet radiometer UV-A is 2.25 mW/cm) 2 ) The method comprises the steps of carrying out a first treatment on the surface of the Another 10 mice served as normal controls without methoxsalen solution and without irradiation. The mice for making the modules are coated with the methoxsalen solution for 1 hour every day and then irradiated once for 15 minutes each time for 2 weeks continuously; the methoxsalen is not smeared for direct irradiation once every day, and each time lasts for 15 minutes and 3 weeks. All mice were shaved every two days with animals shaved Mao Qiti Mao Yici to shave back bare skin hair.
2.2 grouping and administration of animals
40 mice were successfully modeled (randomly divided into 3 groups according to the degree of back skin photodamage: example 1 group (10), comparative example 2 group (10), comparative example 3 group (10) and model control group (10).
Example 1 and comparative examples 2-3 were prepared by diluting the skin care composition 10 times and then applying 0.2mL, the model control group was given the same volume of purified water, and after 30 minutes, the skin care composition was fixed in a skin phototoxicity tester with a transparent mouse holder and irradiated at a distance of about 15cm from the light source, once a day for 15 minutes each time. The same procedure was continued for 5 weeks. All mice were shaved every two days with animals shaved Mao Qiti Mao Yici to shave back bare skin hair. The normal control group was not irradiated.
2.3 experimental observations and materials
The skin smoothness, color and softness and elasticity of each group of mice were visually observed during the experiment. 24 hours after the last administration irradiation, the administration group and model group mice take blood of about 1.0ml from the orbital venous plexus, centrifuge at 3000rpm for 10min, take serum, remove cervical vertebra and sacrifice, take the whole skin of the back dehairing area, fix in 10% neutral formalin solution, and make histopathological section; serum was used to detect MDA content and SOD activity. Normal control mice were operated in the same manner.
3. Test results
3.1 visual inspection during the test
The back skin of the normal control group mice is smooth, tender and elastic; skin of the mice in the model control group gradually appears dander, dryness and partial desquamation along with the increase of irradiation time, and the skin is dark in color, locally hardened and rough and hypertrophic; after the sample group mice are irradiated and coated with the medicines, skin injury is relieved to different degrees, and the phenomena of light color, reduced dander and the like are caused.
3.2 influence of nidus Collocaliae acid on MDA content in serum of photoaged mice
