Abstract

本发明提供一种创制萝卜彩叶新种质的方法,包括以下步骤:(1)测定所有萝卜种质材料的叶片PH值,建立萝卜叶片PH值数据库;(2)以萝卜紫红叶株系Red1为核心材料,该株系叶片富含花青素,呈紫红色,肉质根不发达;以其他肉质根性状优异的绿叶萝卜种质为亲本材料,进行杂交转育。(3)将萝卜紫红叶株系Red1与叶片PH值小于5.7的绿叶亲本杂交,得到出根型各异的红叶萝卜新种质;与PH值大于6.2的亲本杂交,得到出根型各异的紫叶萝卜新种质。本发明可以创制聚合叶色靓丽、肉质根品质好、产量高等多个优异性状的萝卜彩叶新种质材料。

一种创制萝卜彩叶新种质的方法
技术领域
本发明属于植物培育技术领域,本发明提供一种创制萝卜彩叶新种质的方法。
背景技术
目前萝卜的叶色遗传资源挖掘甚少,创制萝卜彩叶新种质非常有利于叶用萝卜、观赏萝卜等特色彩叶萝卜新品种的研发。
发明内容
针对上述技术问题,本发明提供一种创制萝卜彩叶新种质的方法,包括以下步骤:
(1)测定所有萝卜种质材料的叶片PH值,建立萝卜叶片PH值数据库;
(2)以萝卜紫红叶株系Red1为核心材料,该株系叶片富含花青素,呈紫红色,肉质根不发达;以其他肉质根性状优异的绿叶萝卜种质为亲本材料,进行杂交转育;
(3)将萝卜紫红叶株系Red1与叶片PH值小于5.7的绿叶亲本杂交,得到根型各异的红叶萝卜新种质;与PH值大于6.2的亲本杂交,得到根型各异的紫叶萝卜新种质。
经试验检测发现,在彩叶萝卜种质的叶片中,花青素含量增多是导致叶片呈现彩叶的原因,花青素含量的多少影响呈色深浅,而叶片PH值影响呈色种类。因此本发明可以创制聚合叶色靓丽、肉质根品质好、产量高等多个优异性状的萝卜彩叶新种质材料。这将为选育具有观赏兼食用双重功能的彩叶萝卜新品种奠定物质基础,对满足市场及消费者的多样化需求具有重要意义。
具体实施方式
结合实施例说明本发明的具体技术方案。
(1)同种环境条件下,种植Red1与其他萝卜种质材料;
(2)在萝卜肉质根膨大期,选择晴朗上午取样,测定萝卜叶片PH值;
(3)录入PH值数据,构建萝卜种质材料叶片PH值数据库。
(4)在萝卜花期,将Red1依次与不同叶片PH值的萝卜材料杂交,收种;
(5)播种获得的F1杂交种子,观察杂交F1代叶色表现,结果显示:Red1与叶片PH值小于5.7的亲本杂交,F1代叶色均呈现不同程度红色;Red1与叶片PH值大于6.2的亲本杂交,F1代叶色均呈现不同程度紫色;亲本叶片PH值在5.7-6.2时,F1代颜色呈红紫或紫红色。
由此获得结论:本材料库中,可以PH值为参照指数,定向杂交选育不同叶色的萝卜种质材料。
已将萝卜紫红叶株系Red1与绿叶萝卜YZ1492(PH值5.3)杂交,获得葫芦形肉质根的淡红叶萝卜新种质,经过多代自交,获得稳定遗传的葫芦形肉质根的红叶萝卜新种质;萝卜紫红叶株系Red1与绿叶萝卜FL2307(PH值6.4)杂交,获得短圆柱根的深紫叶萝卜新种质,经过多代自交,获得稳定遗传的短圆柱肉质根的紫叶萝卜新种质。

1.一种创制萝卜彩叶新种质的方法,其特征在于,包括以下步骤:
(1)测定所有萝卜种质材料的叶片PH值,建立萝卜叶片PH值数据库;
(2)以萝卜紫红叶株系Red1为核心材料,该株系叶片富含花青素,呈紫红色,肉质根不发达;以其他肉质根性状优异的绿叶萝卜种质为亲本材料,进行杂交转育;
(3)将萝卜紫红叶株系Red1与叶片PH值小于5.7的绿叶亲本杂交,得到根型各异的红叶萝卜新种质;与PH值大于6.2的亲本杂交,得到根型各异的紫叶萝卜新种质。
