Abstract
一种包含植物萃取液的弹力凝胶状组合物,其为作为医药品、化妆品、保养品、香氛或人体清洁用品等产品,其中该弹力凝胶状组合物包含含有硅聚合物的油体的上层与含有胶体的液体的下层,借由先加入液体后加入油体,并经过一定时间的静置,使得油液出现明显分层,使该上层能够保护下层所包含的植物萃取液,使其免于变质或受污。此外,该弹力凝胶状组合物用于肌肤时形成薄膜,于皮肤外用品上能保护肌肤。

技术领域
本发明是关于一种拥有强度与柔软性,具有高弹性的弹力凝胶状组合物,其包含植物萃取液。此外,本发明特别是用于化妆材料,可作为医药品、化妆品、保养品、香氛或人体清洁用品等产品的原料。
背景技术
中国台湾地区发明专利申请号103141497提出一种水中油型乳化化妆料,其于油相中高调配紫外线吸收剂及紫外线散射剂,达成高SPF(Sun Protection Factor,防晒系数),同时于涂布于皮肤时具有独特的娇嫩的使用触感。其特征在于:其含有0.05~1质量%的疏水改性烷基纤维素、5~40质量%的油分、2.5~30质量%的具有疏水性表面的紫外线散射剂以及耐盐性较低的水相增黏剂,且上述紫外线散射剂分散于油相中。
中国台湾地区发明专利申请号106114234提出一种水中油型固形化妆料,其是水中油型制剂,且能够于维持由增黏剂产生的弹性的状态下固化为稳定的固形状,具有所期望的弹力感,指蘸取性良好,且水润的使用感优异。其特征在于:包含疏水改性聚醚胺基甲酸酯0.1重量%~5重量%、亲水性板状粉末0.01重量%~5重量%、疏水性粉末、油分以及水。
日本特许公告号2013-82686,揭示一种水中油型乳化皮肤外用剂,含有具有疏水基的水溶性增黏剂、具有HLB值14以上的非离子性界面活性剂、具有HLB3至5的聚醚改性有机硅及非乳化性的交联型有机硅。
日本特许公告号2006-213730,揭示一种化妆料,含有三-三甲基硅氧基异丁基硅烷15.0重量%、为聚甘油改性有机聚硅氧烷的非离子性界面活性剂1.0重量%、于交联构造中具有聚甘油部分的交联型有机聚硅氧烷1.5重量%及水。
发明内容
本发明的一目的在于提供一种包含植物萃取液的弹力凝胶状组合物,其可用为医药品、化妆品、保养品、香氛、冻凝露或人体清洁用品等产品。
为达成上述目的的功效,本发明提供一种弹力凝胶状组合物,其具有上层以及下层,该下层包含含有胶体的液体,该上层包含含有硅聚合物的油体。
更佳者,该上层的比重介于0.7~0.95之间,该下层的比重介于1.0~1.5之间。
更佳者,该油体与该液体为分别制备。
更佳者,以该油体总重量计,该油体的组成包含:低黏度硅油(1~20cst):1重量%~5重量%、中黏度硅油(50~5000cst):1重量%~5重量%、聚硅氧化合物:15重量%~30重量%、有机硅胶联共聚物:5重量%~15重量%、有机硅弹体乳化剂:3重量%~6重量%、植物油:1重量%~5重量%、合成酯:1重量%~10重量%。
更佳者,该有机硅弹体乳化剂和该有机硅胶联共聚物的重量比例为1:1.5~1:10之间。
更佳者,以该液体总重量计,该液体的组成包含:水:10重量%~40重量%、甘油:5重量%~20重量%、植物萃取液:1重量%-10重量%、多元醇:1重量%~10重量%、玻尿酸钠:0.01重量%~5重量%、聚丙烯酸:0.5重量%~5重量%、海藻酸钠:0.5重量%~5重量%、三仙胶:0.5重量%~5重量%、小核菌胶:0.5重量%~2重量%、羟乙基纤维素:0.04重量%~2重量%、瓜尔胶:0.04重量%~2重量%。
更佳者,该弹力凝胶状组合物的制法包含:先加入液体后加入油体,并将其混合物摇晃混合均匀然后静置。
更佳者,该油体与液体的重量比例为1:1~1:10之间。

本发明的弹力凝胶状组合物可实现快速分层的效果,使得位于下层液体中的植物萃取液能被上层的油体保护而不会与空气、光线或灰尘直接接触,由此,其不易产生质变,可以提升植物萃取液的存放时间,进而长时间地维持植物萃取液的生物活性。
此外,含有硅聚合物的油体的上层,除了能保护下层所包含的植物萃取液,使其免于变质或受污,也可用于肌肤时形成薄膜,具保护皮肤,防止水分散失、滋润修护等防护功效。
此外,本发明在不受外力的情况、倒置或者轻微晃动后不会使油液混合。

图2A为本发明弹力凝胶状组合物(30)与第一对照品(40)倒置前的状态,其中,本发明弹力凝胶状组合物(30)与第二对照品(50)倒置前的状态也相同;图2B为本发明弹力凝胶状组合物(30)与第一对照品(40)倒置后的状态;图2C为本发明弹力凝胶状组合物(30)与第二对照品(50)倒置后的状态。
更佳者,本发明为先加入水相后加入油相,其中,有机硅弹体乳化剂和有机硅胶联共聚物的重量比例为1:1.5~1:10之间,油液的重量比例为1:1~1:10之间,再经过静置一小时来达成油液分层,并且在无外力作用下、倒置或轻微晃动不会使油液混合;如图2B所示,该第一对照品(40)为先加入油相后加入水相,其中,有机硅弹体乳化剂和有机硅胶联共聚物的重量比例为1:1.5~1:10之间,油液的重量比例为1:11,再经过静置一小时,由于油相与水相加入的顺序错误,该油液的重量比例也错误,因此该油体无法凝固,其再倒置后无法产生与本发明相同的功效;如图2C所示,该第二对照品(50)为先加入油相后加入水相,其中,有机硅弹体乳化剂和有机硅胶联共聚物的重量比例为1:1.5~1:10之间,油液的重量比例为1:0.8,再经过静置一小时,由于油相与水相加入的顺序错误,该油液的重量比例也错误,因此该油体无法凝固,其再倒置后无法产生与本发明相同的功效。具体而言,本发明的油液分层中,油体是形成于液体的上层,并且本发明于轻微摇晃、或倒置时,液体无法穿过油体而与空气接触,因此该油体可以有效地保护液体内所含的有效成分。其中可以了解地,本发明于适当地摇晃后,油液可以均匀混合,而便于涂抹于肌肤表面。
更佳者,该植物萃取液为一山刺番荔枝叶萃取物。于一较佳实施例中,该山刺番荔枝叶萃取物的制备方法包含:首先,将山刺番荔枝叶子洗净,阴干约3至5天,再将山刺番荔枝叶子剪成小碎片约0.5cm2大小以进行萃取。其中,本发明的萃取率公式为:(萃取物重量/样品原总重)×100%。再来,将剪碎的山刺番荔枝叶进行一浸泡步骤,于此步骤中包含两种实施态样,其中,第一实施态样为“超音波低温水萃法”,具体而言,其实施方式为:将山刺番荔枝叶子秤取约100g于4℃的环境下浸泡于900ml的去离子水中,并以超音波振荡萃取设备,于低温下以总能量300~600w的能量连续式萃取。于一较佳实施例中,该连续式萃取以震荡3分钟,休息2分钟的方式一共震荡60分钟。于另一较佳实施例中,于连续式萃取完成后,在4℃温度下以15,000rpm高速离心30分钟,收集上清液,再经抽真空过滤器过滤,获得澄清过滤液,利用减压浓缩机及冷冻干燥机将半成品萃取液进行浓缩,制成山刺番荔枝叶水萃取物。于又一较佳实施例中,山刺番荔枝叶水萃取物的冻干物储存于-80℃;第二实施态样为“超音波低温乙醇萃法”,具体而言,其实施方式为:将山刺番荔枝叶子秤取约100g浸泡于五倍体积的95%乙醇,于4℃的环境中避光三天,取出后于低温下以总能量300~600w的能量连续式萃取。于一较佳实施例中,该连续式萃取以震荡3分钟,休息2分钟的方式一共震荡60分钟。于另一较佳实施例中,于连续式萃取完成后,在4℃温度下以15,000rpm高速离心30分钟,收集上清液,再经抽真空过滤器过滤,获得澄清过滤液,利用减压浓缩机及冷冻干燥机将半成品萃取液进行去除溶剂作业,制成山刺番荔枝叶乙醇萃取物。于又一较佳实施例中,该山刺番荔枝叶乙醇萃取物的冻干物储存于-80℃。以下将山刺番荔枝叶水萃萃取物简称为AMLW:将山刺番荔枝叶乙醇萃萃取物简称为AMLE。
如图3所示,为山刺番荔枝叶乙醇萃取物以50~1000μg/ml作用于黑色毒瘤细胞(B16F10细胞)时,B16F10细胞的存活度,其中:AMLE于50μg/ml时,即对老鼠黑色毒瘤细胞(B16F10)细胞具有显著毒性,其细胞存活度只剩下30%,表示山刺番荔枝叶乙醇萃取物具有毒杀癌细胞的作用,可用于制备治疗皮肤癌的医药品、化妆品、保养品、人体清洁用品、或香氛等产品的原料。
如图4A至4B所示,为经过UVB照射的DNA pUC119试验,当DNA受到破坏时,DNA会从环形环绕结构(supercoiled-form,S-form)断裂成直线结构(linear-form,L-form),利用不同分子量的核酸在胶体孔径中移动速度的差异特性,将pUC19 DNA以H2O2(1mM)诱导及UVB(20mJ/cm2)照射,若DNA能受到保护作用时,则能呈现至S-form的位置,而当受到破坏时,DNA便会呈现在L-form的位置。如图4A至4B所示,可发现控制组未受到伤害可以保持完整的DNA型态并呈现环绕结构,未加药物的UVB伤害组别,DNA结构受到损伤而呈现直线形结构,而先添加AMLS或AMLW再经UVB照射的状态下,可保持较完整的DNA环绕结构,因此可发现山刺番荔枝叶萃取物具有DNA保护的作用,并且山刺番荔枝叶的水萃萃取物对于DNA的保护效果较乙醇萃萃取物为佳。
更佳者,该植物萃取液为刺葱茎萃取物。于一较佳实施例中,该刺葱茎萃取物的制备方法与该山刺番荔枝叶萃取物的制备方法相同,在此不多做赘述,其中以该“超音波低温水萃法”制成刺葱茎水萃萃取物;以该“超音波低温乙醇萃法”制成刺葱茎乙醇萃萃取物。于另一较佳实施例中,该植物萃取物为刺葱叶萃取物。于又一较佳实施例中,该刺葱叶萃取物的制备方法与该山刺番荔枝叶萃取物的制备方法相同,在此不多做赘述,其中以该“超音波低温水萃法”制成刺葱叶水萃萃取物;以该“超音波低温乙醇萃法”制成刺葱叶乙醇萃萃取物。以下将刺葱叶水萃萃取物简称为ZLLW:将刺葱叶乙醇萃萃取物简称为ZLLE;将刺葱茎水萃萃取物简称为ZLSW:将刺葱茎乙醇萃萃取物简称为ZLSE。
如图5所示,为刺葱萃取物对于DNA保护效果的实验,其中:于仅受H2O2、FeSO4及UVB伤害的组别,其DNA结构受到损伤,呈现大量的直线型的结构,而经刺葱茎水萃萃取物作用的组别,能保持DNA的完整型态呈现环形缠绕结构,具有优异的DNA保护的作用。
如图6所示,为刺葱萃取物以20~1000μg/ml作用于黑色毒瘤细胞(B16F10细胞)时,B16F10细胞的存活度,其中:提供一控制组,其B16F10细胞活性为100%,当添加ZLLE在100μg/ml及以上的浓度作用下,B16F10细胞存活度低于80%;当添加ZLSE在250μg/ml及以上的浓度作用下,B16F10细胞存活度低于80%,显示其对B16F10细胞具有毒性。表示刺葱叶乙醇萃萃取物或刺葱茎乙醇萃萃取物具有毒杀癌细胞的作用,可用于制备治疗皮肤癌的医药品、化妆品、保养品、人体清洁用品、或香氛等产品的原料。
如图7所示,ZLSE、ZLLE及ZLLW明显促进酪胺酸酶能力,导致黑色素生成量增加,且其增加比率有随浓度增高而增强的趋势,故推测ZLSE、ZLLE及ZLLW具备促进黑色素生成的能力,可用于制备促进黑色素生成或强化发根的组合物,并且所制成的组合物可做为仿晒制剂、头皮养护产品、洗发精、口服制剂、食品或生发水等产品的原料。
更佳者,该植物萃取液为牡丹花蕊生长点组织萃取物。于一较佳实施例中,该牡丹花蕊生长点组织萃取物包含牡丹花蕊生长点组织水萃萃取物或牡丹花蕊生长点组织乙醇萃萃取物。于另一较佳实施例中,该牡丹花蕊生长点组织水萃萃取物的制备方法包含:采取牡丹花新鲜花蕊,将牡丹花蕊组织以急速-195.79℃(77K)液态氮冷冻固化,并以冷冻研磨器研磨牡丹花蕊组织,将牡丹花蕊组织磨成粉末;提取重量比为9:1的纯水以及牡丹花蕊组织粉末,并将牡丹花蕊组织粉末浸泡于纯水中,于温度条件4℃~50℃的情况下浸泡72~120小时;将浸泡完成的牡丹花蕊组织粉末以及纯水的混合物,于温度条件-4℃~25℃的情况下,以总能量500~800w的超音波能量连续快速振荡萃取,其中,以每震动5分钟休息2分钟的方式连续震荡,直到总震荡时间达60~120分钟;于温度条件-20℃下以15,000g高速离心10分钟以去除底部沉淀物并收集上清液;将该上清液以孔径0.22~0.45μM的多微孔结构滤膜后,利用真空筛检器的程序过滤,以获得澄清过滤液;利用减压浓缩机及真空冷冻干燥机将该澄清过滤液冻干,获得牡丹花蕊生长点组织萃取物(PSB)。于又一较佳实施例中,该牡丹花蕊生长点组织乙醇萃萃取物的制备方法包含:采取牡丹花新鲜花蕊,将牡丹花蕊组织以急速-195.79℃(77K)液态氮冷冻固化,并以冷冻研磨器研磨牡丹花蕊组织,将牡丹花蕊组织磨成粉末;提取重量比为9:1的酒精溶液(含50重量%~75重量%乙醇)以及牡丹花蕊组织粉末,并将牡丹花蕊组织粉末浸泡于酒精溶液(含50重量%~75重量%乙醇)中,于温度条件4℃~50℃的情况下浸泡72~120小时;将浸泡完成的牡丹花蕊组织粉末以及酒精溶液(含50重量%~75重量%乙醇)的混合物,于温度条件-4℃~25℃的情况下,以总能量500~800w的超音波能量连续快速振荡萃取,其中,以每震动5分钟休息2分钟的方式连续震荡,直到总震荡时间达60~120分钟;于温度条件-20℃下以15,000g高速离心10分钟以去除底部沉淀物并收集上清液;将该上清液以孔径0.22-0.45μm的多微孔结构滤膜后,利用真空筛检器的程序过滤,以获得澄清过滤液;利用减压浓缩机及真空冷冻干燥机将该澄清过滤液冻干,获得牡丹花蕊生长点组织萃取物(PSB)。以下将牡丹花蕊生长点组织萃取物简称为PSB。
PCNA(proliferating cell nuclear antigen)是一种增殖细胞核抗原,可作为DNA钳,帮助DNA聚合酶活性(DNA pol)然后募集FEN1(Flap内切核酸酶1)以去除悬垂的5'端,是DNA修复的最后一步涉及DNA连接酶,该酶将最终的DNA链以磷酸二酯键结合在一起。有许多机制可以修复受损的双链。如图8所示,未经过UVB伤害的控制组,PCNA表现定量(1.0),而经过UVB照射后,其表现明显降低(0.2)。而经牡丹花蕊生长点组织萃取物(PSB)作用的细胞则可明显提升PCNA的表现(0.8)。显示牡丹花蕊生长点组织萃取物可有效调控修护皮肤细胞免于UV的伤害。
更佳者,该液体更包含活性成分,其中该活性成分是为牡丹花蕊复合微晶囊体组合物。于一较佳实施例中,该牡丹花蕊复合微晶囊体组合物的制备方法包含:将牡丹花蕊生长点组织萃取物(PSB)与载体原料混合,使载体原料分散形成胶体复合物。具体而言,将载体原料溶解于溶剂中,利用旋转减压浓缩仪去除溶剂以形成脂质薄膜,之后加入牡丹花蕊生长点组织萃取物(PSB)至脂质薄膜使脂质分散成胶体复合物,所获得的胶体复合物即为牡丹花蕊复合微晶囊体组合物。以下将牡丹花蕊复合微晶囊体组合物简称为PSBL。
核纤层蛋白Lamin A基因变异可损坏DNA修复功能,而令基因组变得不稳定,导致早老症。抗衰老启动机制发现Lamin A蛋白质能刺激长寿基因SIRT1,使细胞自我保护、自我更新及推迟衰老。评估包覆型牡丹花蕊微囊(PSBL)调控抗老化蛋白Lamin A的表现,进而推迟细胞衰老。人类皮肤角质株化细胞(HaCaT细胞)(1.5×105cell/mL)培养于直径10cm的培养皿(dish),在37℃及5%CO2培养箱中培养至少24小时,加入包覆型牡丹花蕊微囊(PSBL),置入培养箱作用24小时。当达反应时间,利用RIPA buffer进行蛋白质萃取,之后进行SDS-PAGE电泳直到目标蛋白分离。加入0.5%脱脂牛奶2分钟后,再加入1级抗体反应10分钟,以0.1%磷酸盐缓冲生理盐水(PBST)冲洗3次,再加入2级抗体反应10分钟,再以0.1%PBST冲洗3次。最后加入ECL冷光感应试剂,置入冷光照相是统拍照并使用image J定量分析蛋白质表现。如图9所示,人类皮肤角质株化细胞(HaCaT细胞)经包覆型牡丹花蕊微囊(PSBL)作用后,分析细胞内Lamin A蛋白质表现量。细胞骨架蛋白β-actin作为定量控制组。结果显示,与控制组(Lamin A表现为1.0)相比,细胞经包覆型牡丹花蕊微囊(PSBL)作用后Lamin A表现增加至3.8,显示包覆型牡丹花蕊微囊(PSBL)可活化Lamin A蛋白质,推测其具抗老化效果。
惟以上所述者,仅为本发明的较佳实施例,但不能以此限定本本发明专利保护范围;故,凡依本发明申请专利保护范围及说明书内容所做的简单的等效改变与修饰,皆仍落入本专利保护范围内。
Claims (10)
1.一种弹力凝胶状组合物,其特征在于:具有上层以及下层,该下层为含有胶体的液体,该上层为含有硅聚合物的油体。
2.根据权利要求1所述的弹力凝胶状组合物,其特征在于,该上层的比重介于0.7~0.95之间,该下层的比重介于1.0~1.5之间。
3.根据权利要求1或2所述的弹力凝胶状组合物,其特征在于,该油体与该液体为分别制备。
4.根据权利要求1所述的弹力凝胶状组合物,其特征在于,以该油体总重量计,该油体的组成包含:低黏度硅油(1~20cst):1重量%~5重量%、中黏度硅油(50~5000cst):1重量%~5重量%、聚硅氧化合物:15重量%~30重量%、有机硅胶联共聚物:5重量%~15重量%、有机硅弹体乳化剂:3重量%~6重量%、植物油:1重量%~5重量%、合成酯:1重量%~10重量%。
5.根据权利要求4所述的弹力凝胶状组合物,其特征在于,该有机硅弹体乳化剂和该有机硅胶联共聚物的重量比例为1:1.5~1:10之间。
6.根据权利要求1所述的弹力凝胶状组合物,其特征在于,以该液体总重量计,该液体的组成包含:水:10重量%~40重量%、甘油:5重量%~20重量%、植物萃取液:1重量%-10重量%、多元醇:1重量%~10重量%、玻尿酸钠:0.01重量%~5重量%、聚丙烯酸:0.5重量%~5重量%、海藻酸钠:0.5重量%~5重量%、三仙胶:0.5重量%~5重量%、小核菌胶:0.5重量%~2重量%、羟乙基纤维素:0.04重量%~2重量%、瓜尔胶:0.04重量%~2重量%。
7.根据权利要求1所述的弹力凝胶状组合物,其特征在于,以该油体总重量计,该油体的组成包含:低黏度硅油(1~20cst):1重量%~5重量%、中黏度硅油(50~5000cst):1重量%~5重量%、聚硅氧化合物:15重量%~30重量%、有机硅胶联共聚物:5重量%~15重量%、有机硅弹体乳化剂:3重量%~6重量%、植物油:1重量%~5重量%、合成酯:1重量%~10重量%,而以该液体总重量计,该液体的组成包含:水:10重量%~40重量%、甘油:5重量%~20重量%、植物萃取液:1重量%-10重量%、多元醇:1重量%~10重量%、玻尿酸钠:0.01重量%~5重量%、聚丙烯酸:0.5重量%~5重量%、海藻酸钠:0.5重量%~5重量%、三仙胶:0.5重量%~5重量%、小核菌胶:0.5重量%~2重量%、羟乙基纤维素:0.04重量%~2重量%、瓜尔胶:0.04重量%~2重量%。
8.根据权利要求1所述的弹力凝胶状组合物,其特征在于,其制备方法先加入液体后加入油体,并将其混合物摇晃混合均匀然后静置。
9.根据权利要求8所述的弹力凝胶状组合物,其特征在于,该油体与液体的重量比例为1:1~1:10之间。
10.根据权利要求9所述的弹力凝胶状组合物,其特征在于,该静置时间至少为五分钟。
