Abstract

背景技术
外泌体(exosome)是由细胞分泌的小囊泡,并含有细胞表达的蛋白质、mRNA和miRNA,表现出细胞自身的特征。外泌体通过将物质递送至其它细胞来参与细胞-细胞通讯。特别是,癌细胞来源的外泌体在血液中以高浓度存在,并且已知通过移动到癌症转移发生的部位来帮助癌症转移。许多研究表明,外泌体从肿瘤细胞非常活跃地分泌,并在递送癌症转移和发展必需的物质到转移部位中发挥作用。
癌症是世界范围内最常见的疾病之一,目前应用的治疗包括手术、放射和化学疗法。尽管正在积极研究癌症的分子机制,但目前开发的大部分治疗都依赖于手术。然而,最近,各种靶向治疗剂(targeted therapeutic agent)(例如小分子抑制剂(small moleculeinhibitor)、单克隆抗体(monoclonal antibody)和靶向癌细胞的短靶向肽(shorttargeting peptide))已被开发并应用为治疗剂。特别是,短靶向肽组织渗透性高,并且毒性和免疫反应低,从而保证了作为有效的抗癌剂的高潜力。

技术方案

本公开的示例性实施方式提供了特异性结合癌细胞来源的外泌体的肽,所述肽具有由SEQ ID NO:1表示的氨基酸序列。
优选地,癌细胞可以是肺癌细胞、乳腺癌细胞或胰腺癌细胞,但不限于此。
本公开的示例性实施方式的肽可以通过本领域已知的化学合成容易地制备(Creighton,Proteins;Structures and Molecular Principles,W.H.Freeman and Co.,NY,1983)。典型方法可包括液相或固相合成、片段缩合、和F-MOC或T-BOC化学方法(Chemical Approaches to the Synthesis of Peptides and Proteins,Williams等编著,CRC Press,Boca Raton Florida,1997;A Practical Approach,Athert on&Sheppard,编著,IRL Press,Oxford,England,1989),但不限于此。

使用本公开的示例性实施方式的肽对癌症进行诊断可以通过以下来进行:使本公开的示例性实施方式的肽与通过血液、尿或活检直接获得的细胞或组织反应并检测其结合。
此外,为了容易地确认、检测和量化本公开的示例性实施方式的肽是否与癌症组织结合,本公开的示例性实施方式的肽可以以标记状态提供。换言之,它可以通过与可检测的标记连接(例如,共价键合或交联)来提供。可检测的标记可包括产色酶(chromogenicenzyme)(例如过氧化物酶、碱性磷酸酶),放射性同位素(例如124I、125I、111In、99mTc、32P、35S),发色团(chromophore),发光材料,或荧光材料(例如FITC、RITC、罗丹明(rhodamine)、花菁(cyanine)、德克萨斯红(Texas Red)、荧光素(fluorescein)、藻红蛋白(phycoerythrin)和量子点(quantum dot))。
相似地,可检测的标记可以是抗体表位、底物、辅因子(cofactor)、抑制剂或亲和配体。此类标记可以在合成本公开的示例性实施方式的肽的过程中进行,或者可以在已经合成的肽上另外进行。如果使用荧光物质作为可检测标记,可以通过荧光介导断层摄影(fluorescence mediated tomography,FMT)来诊断癌症。例如,用荧光物质标记的本公开的示例性实施方式的肽可以在血液中循环,以通过荧光断层摄影观察肽的荧光。如果观察到荧光,则诊断出癌症。
此外,本公开的示例性实施方式提供了用于药物递送的组合物,所述组合物包含所述肽作为活性成分。
优选地,所述药物可以是肽类药物或抗癌药物。更优选地,肽药物可以是具有诱导凋亡或坏死的活性的细胞毒性肽,但不限于此。
根据本公开的示例性实施方式的肽可用于选择性地将药物递送至癌细胞来源的外泌体的智能药物递送。通过将所述肽与传统的已知药物连接来使用本公开的示例性实施方式的肽治疗癌症,由于药物由本公开的示例性实施方式的肽选择性地递送至癌细胞来源的外泌体,可以提高药物的疗效,同时药物对正常组织的副作用可显著减少。

此外,本公开的示例性实施方式提供了用于检测癌细胞来源的外泌体的组合物,所述组合物包含所述肽作为活性成分。
实施例
在下文中,将详细描述示例性实施方式,以帮助理解本公开的内容。然而,以下示例性实施方式对于本公开内容仅仅是说明性的,而且本公开的范围不限于以下实施例。提供本公开的示例性实施方式是为了向本领域的普通技术人员更完整地解释本公开内容。
<实施例1>对从A549肿瘤细胞中分离的外泌体的特征的分析
使用超速离心机或Exoquick(一种商业试剂)从A549肺癌细胞培养基中分离外泌体。首先,为了通过蛋白质印迹分析分离的外泌体的特征,将外泌体提取物加载到SDS聚丙烯酰胺凝胶上。对凝胶进行电泳并转移至NC膜,用由含有5%脱脂牛奶和Tween-20的Tris缓冲液(TBST)成分组成的封闭液进行反应1小时,以阻断非特异性反应。此后,用TBST清洗涤膜3次,每次10分钟,与抗CD63和Alix的抗体(Abcam,USA)在4℃反应过夜。第二天,在室温下用TBST洗涤膜数次,用辣根过氧化物酶缀合的二抗在室温下进行反应1小时。此后,使用West Femto Maximum Sensitivity Substrate(ThermoFisher,美国)试剂和LAS进行分析。
作为结果,来源于A549、MDA-MB231和HEK293细胞系的外泌体与细胞提取物相比,显示出更高的外泌体标志物(例如CD63、Alix和Tsg 101)的定量水平,并且只在细胞提取物中钙连蛋白有表达(图1A)。在另一方面,根据细胞系,外泌体标志物的水平没有显著差异。
此外,为了测量分离的外泌体的尺寸,使用NanoSight仪器对外泌体进行了分析。作为结果,通过超速离心和Exoquick分离的外泌体的尺寸分别为193nm和103nm,其在外泌体的正常尺寸范围中(图1B)。
此外,通过CD63抗体标记的珠子(Invitrogen,USA)捕获的外泌体和通过生物素标记之后由亲和素磁珠捕获的外泌体以与上述相同的方式进行凝胶电泳并使用CD63抗体进行蛋白质印迹。作为结果,在两种情况下都很好地观察到了外泌体标志物CD63(图1C)。
<实施例2>通过噬菌体筛选对特异性结合A549肿瘤细胞来源外泌体的肽的发掘
使用T7噬菌体疏水氨基酸文库(1.3×1010pfu)进行噬菌体筛选。该文库具有CXXXXXXXC(X=随机序列)氨基酸序列,具有在两端的半胱氨酸和在中间的7个随机氨基酸,而7个随机氨基酸中的至少一个具有疏水性氨基酸。A549作为肺癌细胞系使用。使用超速离心机从肿瘤细胞培养基中分离外泌体。为了选择只选择性结合至A549来源的外泌体的T7噬菌体,首先进行了除去非特异性结合至珠子的噬菌体的过程,通过使噬菌体与CD63抗体标记的珠子反应,然后收集上清液中没有结合至珠子的噬菌体。然后,将上清液中的噬菌体与CD63抗体标记的珠子上捕获的外泌体反应(图2A)。
使用大肠杆菌(E.coli)作为宿主收集与外泌体结合的噬菌体,并且通过噬菌斑测定计算每轮中收集的噬菌体的滴度。用大肠杆菌扩增未经受噬菌斑测定的剩余噬菌体,以确保将在下一轮中使用的噬菌体。相同的过程被重复多至5轮。
作为结果,计算在每轮收集的噬菌体的滴度。当进行第一轮时,收集的噬菌体的滴度为约5.7×106,发现在5轮之后最终收集到了2.06×107个噬菌体。换言之,作为从第1轮进行到第5轮的结果,噬菌体的滴度增加了约4倍(图2B)。
为了对所选噬菌体上展示的肽进行氨基酸测序,从第4轮和第5轮用于滴度测量的噬菌斑测定的产物中收集50个克隆(共100个),并储存在10μL Tris缓冲液中。通过PCR扩增编码插入这些噬菌体中的肽的基因,分析核苷酸序列,并由此进行氨基酸测序。使用Clustal X程序对肽序列进行了比对和分析(图2C)。其中,选择了代表由7至9个氨基酸组成的肽的10个噬菌体克隆用于进一步研究。
<实施例3>评价噬菌体克隆对A549肿瘤细胞来源的外泌体和产生外泌体的细胞的选择性结合能力
为了研究噬菌体克隆与A549细胞来源的外泌体和产生外泌体的细胞的选择性结合,进行了噬菌体外泌体结合ELISA和噬菌体细胞结合ELISA。对于噬菌体外泌体结合ELISA,外泌体首先被生物素化并被单体的亲和素标记的磁珠固定(图3A)。用针对辣根过氧化物酶缀合的T7噬菌体的抗体检测与外泌体结合的噬菌体,并根据反应程度和使用酶的底物的显色来量化数量。
作为结果,发现展示具有序列CTDTKIK(4R-3)、CRLSKKS(5R-25)和CRKVAKG(5R-34)的肽的三个噬菌体克隆各自比其它克隆更多地与A549来源的外泌体结合(图3B)。
此外,为了通过噬菌体细胞结合ELISA测量噬菌体与产生外泌体的细胞的结合,将细胞在平板上进行培养,并如上所述使用抗体和底物对与细胞结合的噬菌体进行量化。作为结果,发现展示具有序列CRKRPAL(4R-12)和CAVRRKL(5R-47)的肽的两个克隆各自比其它克隆更多地与产生外泌体的A549细胞结合(图3C)。
<实施例4>对ExoPep肽与A549肿瘤细胞来源的外泌体的结合及其内化的分析
基于图3中示出的ELISA结果,合成了5种肽(CTDTKIK、CRLSKKS、CRKVAKG、CRKRPAL、CAVRRKL)。为了通过FACS方法研究候选肽的结合,请求Peptron Co.(Daegeon,Korea)合成其中异硫氰酸荧光素(FITC,一种荧光物质)与羧基末端缀合的肽。每个肽都通过标准的Fmoc方法合成,并通过质谱纯化。此外,FITC标记的NSSSVDK肽被用来作为对照。
为了观察肽与外泌体的结合,首先以与图3中描述的相同方式将生物素和外泌体复合,然后通过与亲和素珠结合将外泌体固定。通过在室温下用溶解在磷酸盐缓冲液中的1%的牛血清白蛋白(BSA)处理30分钟来阻断外泌体-珠子复合物的非特异性结合。洗涤之后,FITC标记的肽与外泌体-珠子复合物在4℃结合30分钟。反应之后,使用流式细胞仪分析肽与外泌体的结合。
另一方面,为了观察肽与产生外泌体的细胞的结合,首先准备1×106个细胞。每个细胞培养基在37℃用1% BSA处理30分钟。此后,将FITC标记的肽在4℃结合1小时。反应之后,使用流式细胞仪分析肽与细胞的结合。
结果如图4A所示,与每种产生外泌体的细胞相比,本公开的示例性实施方式的肽(序列:CRKVAKG,名称:ExoPep;SEQ ID NO:1)显示出对来源于A549肺癌细胞、H460肺癌细胞、MDA-MB231乳腺癌细胞和Panc-1胰腺癌细胞的外泌体的结合更高。另一方面,与每种产生外泌体的细胞相比,在来源于正常细胞(包括HEK293和MCF10A细胞)和其它类型的肿瘤细胞(包括LLC小鼠肺癌细胞、HT-29结直肠癌细胞和HepG2肝癌细胞的外泌体中显示出非常低的结合,或者没有观察到更高的结合。另一方面,由于与正常细胞的结合相对较高,其它类型的肽被排除在候选组之外。
图4B示出了外泌体和ExoPep肽的结合和内化实验的示意图。收集A549、HEK293和MDA-MB231细胞的培养基,通过超速离心分离出外泌体,用DiD荧光试剂(红色)标记外泌体,然后与ExoPep肽(FITC标记,绿色)反应。反应之后,将细胞固定,用DAPI(蓝色)将细胞核染色以用共聚焦显微镜进行分析。更具体而言,分离的外泌体和DiD(1:200稀释)在搅拌器上在37℃反应30分钟。在添加Exoquick试剂后,在4℃保持状态30分钟,然后通过在13000rpm离心3分钟来沉淀DiD标记的外泌体。此后,将10μM FITC缀合的肽和DiD标记的外泌体的混合物与A549细胞在37℃反应1小时。洗涤三次后,使用2%多聚甲醛(PFA)进行固定。再次洗涤之后,用4'-6-二脒基-2-苯基吲哚(DAPI)处理细胞以染色细胞核。最后,封固之后,使用共聚焦显微镜(Zeiss,Oberkochen,德国)进行观察。
作为结果,在A549细胞中,ExoPep肽与外泌体一起被观察到,但没有观察到对照肽(c.p)(图4C)。这表明,对照肽没有与A549细胞来源的外泌体结合,但ExoPep肽结合了并有效地内化到细胞中。另一方面,以与上述相同的方式处理来源于正常HEK293细胞的外泌体,并在共聚焦显微镜(Zeiss,Oberkochen,德国)下观察。作为结果,外泌体到HEK293细胞内的内化有效地发生,而在细胞中没有观察到对照肽或ExoPep肽(图4C)。这表明ExoPep肽没有结合至正常细胞的外泌体。
此外,对来源于MDA-MB231细胞的外泌体进行了实验,制备所述细胞以分泌通过表达CD63-GFP显示绿色荧光的外泌体。GFP外泌体(绿色)和10μM TAMARA缀合的肽(红色)与MDA-MB231细胞在37℃反应1小时。此后,在进行如上所述的固定、DAPI细胞核染色和封固之后,使用共聚焦显微镜(Zeiss,Oberkochen,德国)进行观察。作为结果,ExoPep肽与MDA-MB231细胞来源的外泌体结合,然后与外泌体一起内化进入细胞,与对照肽相比,在细胞中观察到更多所述肽(图4D)。
<实施例5>使用ExoPep肽与A549肿瘤细胞来源的外泌体的结合来抑制外泌体内化进入受体细胞
为了进一步确认ExoPep肽与A549细胞来源的外泌体的结合特异性,用DID荧光染料标记外泌体,与生物素标记的ExoPep肽反应,然后通过使标记的亲和素与磁性颗粒反应而使其与生物素缀合。通过反应制备的外泌体/生物素-肽/亲和素-磁性颗粒的复合物被捕获并使用磁铁除去,将剩余的外泌体处理至受体细胞(图5A)。
作为结果,与将细胞来源的外泌体直接处理至受体细胞或将外泌体和ExoPep肽进行反应而不经历除去过程的情况相比,通过用ExoPep肽预处理A549细胞来源的外泌体然后去除同样的外泌体,外泌体进入受体细胞的内化受到显著抑制(图5B)。在另一方面,用对照肽(序列:NSSSVDK)进行预处理没有减少外泌体进入受体细胞的内化(图5B)。此外,当对来源于正常HEK293细胞的外泌体进行类似的实验时,外泌体进入受体细胞的内化没有被本公开的示例性实施方式的ExoPep肽以及对照肽抑制(图5C)。
<实施例6>由于抑制A549肿瘤细胞来源的外泌体的释放,ExoPep肽的内化和外泌体结合减少
为了进一步确认ExoPep与外泌体的结合特异性,将GW4869(鞘磷脂抑制剂,Sigma-Aldrich)(一种抑制外泌体释放到细胞外的药物)以不同浓度(2.5μM、5μM和10μM)处理细胞。此后,分离外泌体,并以与图1中所述相同的方式进行电泳和用针对CD63、Alix、Tsg101和钙连蛋白(Abcam)的抗体进行的蛋白质印迹。
作为结果,发现与浓度为2.5μM和5μM相比,在浓度为10μM的GW4869抑制剂下,外泌体标志物(例如CD63、Alix和Tsg101)极大地减少(图6A)。
在另一方面,为了确认GW4869抑制剂对细胞活力的影响,将A549细胞(在96孔细胞培养容器中每孔5×103个细胞)在无血清培养基中与不同浓度的GW4869在37℃培养3小时。此后,将培养基用含有10%牛血清(FBS)的培养基替换,培养细胞24小时,然后使用CCK-8测定(Dojindo,Japan)测量细胞毒性。作为结果,甚至在高浓度,GW4869也没有显示出毒性(图6B)。基于上述结果,使用10μM GW4869抑制剂,分析了ExoPep肽与外泌体的结合及其内化。作为免疫荧光染色的结果,当用10μM GW4869处理时,由于外泌体释放减少,外泌体和与其结合的肽的内化显著减少(图6C)。
<实施例7>对ExoPep肽与来源于A549肿瘤小鼠血液和正常小鼠血液的外泌体的结合和内化的分析
将A549细胞注射进入BALB/c裸鼠,在肿瘤生长之后收集血液,并使用外泌体分离试剂盒分离外泌体。为了通过流式细胞仪测量肽与外泌体的结合,用CD63抗体珠标记外泌体。作为对照组,使用从正常小鼠血液中分离的外泌体。通过与1% BSA/PBS在室温下反应30分钟来封闭用CD63珠子标记的外泌体-珠子复合物以减少非特异性结合。洗涤之后,FITC标记的ExoPep肽与外泌体-珠子复合物在4℃结合30分钟。再次用PBS洗涤之后,使用流式细胞仪(ThermoFisher Scientific,USA)分析肽与外泌体-珠子复合物的结合(图7A)。
作为结果,发现相比正常小鼠血液来源的外泌体,ExoPep肽与A549肿瘤小鼠血液来源的外泌体更好地结合(图7B和图7C)。
为了观察肽内化进入细胞的情况,用DiD标记外泌体,与FITC标记的肽反应,并处理至A549细胞。用4%多聚甲醛固定细胞5分钟,用DAPI染色,然后在显微镜下观察。
作为结果,发现与正常小鼠血液来源的外泌体相比,本公开的示例性实施方式的ExoPep肽与肿瘤小鼠血液来源的外泌体结合并更多地被内化进入受体细胞A549。此外,在细胞中一起观察到肽(绿色)和肿瘤来源的外泌体(红色)的荧光(图7E)。
<实施例8>对ExoPep肽与在A549肿瘤小鼠血液中循环的外泌体的结合的分析
为了检查肽与小鼠血液中循环的外泌体的结合,将生物素标记的肽静脉内注射进入A549肿瘤小鼠和健康小鼠并循环。然后收集血清,在收集后30分钟分别用CD63抗体珠和链霉亲和素珠进行反应。使由CD63抗体珠处理的血清反应过夜,第二天用磁铁将珠子沉淀。使用RIPA裂解液裂解与珠子结合的外泌体,然后使用CD63和ALIX抗体进行蛋白质印迹分析。使由链霉亲和素珠处理的血清反应1小时,然后使用磁铁沉淀。使用洗脱缓冲液(PBS中2mM D-生物素)洗脱与珠子结合的外泌体,然后使用CD63和ALIX抗体进行蛋白质印迹分析(图8A)。
作为结果,与健康小鼠相比,当用CD63抗体珠在肿瘤小鼠的血液中分离时,检测到高得多的外泌体标志物(图8B,左)。甚至当用链霉亲和素珠分离时,在肿瘤小鼠的血液中检测到较高的外泌体标志物,但外泌体标志物的数量相对低于用CD63珠子分离时(图8B,右)。这表明,由于CD63抗体与血液中的所有外泌体结合,而生物素-ExoPep肽选择性地与肿瘤来源的外泌体结合,分离的外泌体数量相对较少。上述结果表明,本公开的示例性实施方式的肽与循环血液中的外泌体结合,并且特别是选择性地与肿瘤来源的外泌体结合。
然后,进行了体外溶血测定。体外溶血测定,作为在暴露于药物或药剂之后红血细胞溶血的指标,通过测量血浆中的血红蛋白释放来进行。这是一种预测药物溶血活性的准确、灵敏的方法。新鲜血液在500×g离心10分钟,将红血细胞沉淀洗涤3次并重新悬浮在pH7.4的10mM PBS中。通过在37℃搅拌1小时将等体积的红血细胞与不同浓度的ExoPep肽反应。然后在4℃以500×g离心样品10分钟。根据不同的肽浓度,通过分析OD540nm处的吸光度来测量RBC裂解。考虑到用作对照的1%Triton X 100样品显示100%的溶血,测量了溶血百分比。使用以下方程,以百分比计算肽的溶血活性:

因此,在高至200μM的终浓度的各种浓度的肽中,只显示出低水平的溶血活性(图8C)。
<实施例9>根据ExoPep肽的结合,A549肿瘤小鼠血液来源的外泌体和A549细胞来源的外泌体在A549肿瘤小鼠中的体内分布
将DiD标记的A549肿瘤小鼠血液来源的外泌体和A549细胞来源的外泌体与ExoPep肽反应,并对体内分布进行图像监测。通过将A549细胞悬浮液(5×106个细胞)与PBS一起皮下植入5周大雌性BALB/c裸鼠的右胁腹以制备肿瘤异种移植小鼠。当肿瘤尺寸达到约100-200mm3的体积时,麻醉小鼠。向A549肿瘤小鼠静脉内施用DiD标记的A549肿瘤小鼠血液来源的外泌体(mEXO)和通过将外泌体(mEXO)与肽反应获得的产物(mEXO+ExoPep,n=3),以及DiD标记的A549细胞来源的外泌体(cEXO)和通过将外泌体(cEXO)与肽反应获得的产物(cEXO+ExoPep,n=3)。本公开的发明人使用FITC标记的对照肽和ExoPep(n=3)作为对照。使用IVIS成像系统(Caliper Life Sciences,Massachusetts,USA)在施用DiD标记的外泌体之后的不同时间点(分别为2、4、8和24小时)分析体内的荧光图像。施用后24小时,收集切除的肿瘤和器官的体外荧光图像。将所有具有肿瘤组织的主要器官(肝、肾、脾、心脏和肺)分离,用PBS洗涤,并进行体外荧光分析(n=3)。
作为结果,与mEXO和cEXO相比,观察到mEXO+ExoPep和cEXO+ExoPep的组合在注射后4小时在肿瘤组织中更多积累(图9A)。在分析整个身体中的目标区域(ROI)、特别是在肿瘤部位的荧光强度的结果中也获得了统计学显著性(图9C)。注射之后24小时,移除器官并对荧光进行拍照(图9B)。作为测量强度的结果,与mEXO相比,在肿瘤组织中mEXO+ExoPep的积累没有差异,但在肝中观察到较少的积累(图9D)。另一方面,与cEXO相比,显示在肿瘤组织和肝中cEXO+ExoPep积累更多(图9D)。FITC标记的对照和ExoPep肽本身显示出低信号(图9A-图9D)。
<实施例10>由ExoPep肽和凋亡诱导肽组成的ExoPep-KLA肽的细胞毒性IC50分析
利用ExoPep肽与外泌体结合之后很好地内化进入细胞这一事实,制备了一种肽(名为ExoPep-KLA),与一种通过对细胞中的线粒体膜造成损坏而诱导凋亡的肽(KLA)融合。为了确认ExoPep-KLA在A549、MDA-MB231、HEK293、Panc-1、HT29和HepG2细胞中的细胞毒性(在96孔细胞培养容器中每孔5×103个细胞),将细胞在无血清培养基中与不同浓度的ExoPep-KLA在37℃培养3小时。此后,将培养基替换为含有10% FBS的培养基,将细胞培养24小时,然后使用CCK-8测定(Dojindo)测量细胞毒性。将不同浓度的ExoPep-KLA与从A549、MDA-MB231、Panc-1、HT29、HepG2和HEK293细胞中分离的各种外泌体(5μg)反应,然后处理至细胞。用作对照的是单独的ExoPep-KLA或通过先用ExoPep-KLA然后用外泌体处理细胞获得的产品。
作为结果,与单独用肽处理的组和用肽处理后用外泌体处理的组相比,在用肽和外泌体处理的组中表现出更有效的细胞毒性(即,较低的IC50值)。特别是,ExoPep-KLA肽在正常HEK293细胞、HT-29结直肠癌细胞和HepG2肝癌细胞中几乎不显示细胞毒性,但在A549肺癌细胞、MDA-MB231乳腺癌细胞和Panc-1胰腺癌细胞中特异性地显示出较高的细胞毒性(图10A-图10F)。另一方面,在将正常HEK293细胞的外泌体与ExoPep-KLA反应之后,当处理至HEK293细胞时,几乎没有观察到细胞毒性。另一方面,在将A549细胞外泌体与ExoPep-KLA反应并将其处理至HEK293细胞之后,与单独用肽处理的组和用肽处理之后用外泌体处理的组相比,当外泌体和ExoPep-KLA肽一起处理时,获得了对HEK293细胞的细胞毒性(图10G)。这表明ExoPep-KLA肽与来源于肿瘤细胞的外泌体的结合特异性。
<实施例11>ExoPep-KLA的凋亡诱导作用
为了研究ExoPep-KLA对凋亡的诱导,将磷光性的A549-luc和MDA-MB231-luc细胞以1×104个细胞/孔的密度接种在黑色96孔ELISA板上。在将细胞与ExoPep-KLA和每种细胞的外泌体(5μg)一起反应24小时之后,向每个孔添加3μL(3mg/mL)的D-荧光素,使用IVIS成像系统(PerkinElmer,Waltham,MA,USA)测量磷光强度(外排活性)。作为结果,与单独用ExoPep-KLA肽处理的组和肽处理之后用外泌体处理的组相比,随着ExoPep-KLA与外泌体反应的处理浓度和反应时间增加,凋亡效果增加,并且luc信号强度下降(图11)。
<实施例12>ExoPep-KLA(经历了与A549细胞来源的外泌体和A549肿瘤小鼠血液来源的外泌体的反应)对A549细胞的凋亡作用
为了确定ExoPep-KLA依赖于与A549细胞来源的外泌体结合的凋亡诱导作用,本公开的发明人用5μg与细胞来源的外泌体预反应的ExoPep-KLA或未反应的ExoPep-KLA处理A549细胞。用Annexin V-647进行染色,并使用流式细胞仪(ThermoFisher Scientific,USA)分析细胞,以量化凋亡细胞(Annexin+/PI-)。与单独使用ExoPep-KLA处理的组相比,与外泌体预反应的ExoPep-KLA组(cEXO+ExoPep-KLA)显示出更高的凋亡细胞比例,分别为62.9%和20.3%(p<0.05,图12A)。结果表明,与外泌体结合的ExoPep-KLA比单独的ExoPep-KLA更有效地诱导凋亡。
此外,为了测量依赖于与来源于皮下植入有A549肿瘤的小鼠血液的外泌体结合的ExoPep-KLA的凋亡诱导作用,本公开的发明人在经过或不经过与ExoPep-KLA预反应的情况下,用来源于植入有A549肿瘤的小鼠血液的外泌体(5μg)处理A549细胞。用Annexin V-647进行染色,用流式细胞仪(ThermoFisher Scientific,USA)分析细胞以量化凋亡细胞(Annexin+/PI-)。与外泌体预反应的ExoPep-KLA组(mEXO+ExoPep-KLA)显示出比单独用ExoPep-KLA处理的组更高的凋亡细胞比率,分别为19.1%和6.1%,但比率本身不高(p<0.05,图12B)。上述结果表明,与来源于植入肿瘤的小鼠血液的外泌体相比,更多数量的ExoPep-KLA结合至来源于肿瘤细胞的外泌体,诱发的凋亡比A549细胞中的高。
<实施例13>对ExoPep-KLA在血清中的稳定性的分析
本公开的发明人试图确认ExoPep-KLA肽在小鼠血清中的稳定性。在将ExoPep-KLA肽与小鼠血清反应24小时之后,分析在血清中剩余的肽的量。从血清的非特异性峰中分离肽峰,通过峰面积计算血清中肽的残留量。ExoPep-KLA肽在直到4小时时几乎没有分解,在8小时时部分分解,其中半衰期为约24小时(图13A和图13B)。作为每个肽峰的质谱分析的结果,发现该峰为ExoPep-KLA肽。结果表明,该肽在血清中相对稳定。
<实施例14>在A549肿瘤小鼠模型中通过ExoPep-KLA肽抑制肿瘤的生长和转移
为了测试在A549肿瘤裸鼠模型中使用ExoPep-KLA的抗癌效果,ExoPep-KLA肽或者单独或者在体外与外泌体进行预反应,然后在方案中指示的时间点系统性静脉内施用3周,每周施用一次阿霉素,持续3周(图14A)。
作为结果,当施用磷酸盐缓冲液和肿瘤小鼠血液来源的外泌体(仅mEXO)时,没有抑制肿瘤生长。然而,在施用阿霉素、ExoPep-KLA、和通过将外泌体(mEXO)和ExoPep-KLA预反应制备的mEXO+ExoPep-KLA的组中,肿瘤生长受到显著抑制(图14B)。特别是,即使当单独注射ExoPep-KLA时,与体外预反应后注射外泌体的情况相比,肿瘤生长也被以相似水平抑制。这表明,ExoPep-KLA肽与在血液中循环的来源于肿瘤的外泌体(mEXO)结合,并有效地迁移至肿瘤组织。在治疗期间,即使伴随治疗组的连续施用,小鼠的体重也没有变化(图14C)。另一方面,当进行阿霉素、ExoPep-KLA和mEXO+ExoPep-KLA处理时,肿瘤的重量减少(图14D)。
当将A549肺癌细胞在裸鼠的皮肤下异种移植时,由肺转移导致的肿瘤结节数量低(图14E)。此外,在mEXO+ExoPep-KLA组和单独施用ExoPep-KLA的组中,与磷酸盐缓冲液和阿霉素相比,肿瘤结节的数量略微减少,但没有显著差异(图14E)。此外,与其它组在肺重量上没有显著差异(图14F)。然而,当单独施用从肿瘤小鼠血液中分离的外泌体(仅mExo)时,与其它组相比,肺和转移至肺的结节的重量显著增加(图14E和图14F)。这是一个出乎意料的结果,表明当肿瘤小鼠血液中循环的外泌体在体外分离之后被施用时,促进了肿瘤的转移。
另一方面,作为测量肝重量的结果,在所有组之间没有显著差异(图14G)。与磷酸盐缓冲液、单独用mEXO处理的组和施用阿霉素的组相比,在用mEXO+ExoPep-KLA和ExoPep-KLA处理的组中的生存率明显延长(图14H)。结果表明,静脉内注射的ExoPep-KLA肽特异性地与肿瘤小鼠中的循环外泌体结合,促进内化进入肿瘤组织和肿瘤细胞,并随后,诱导癌细胞凋亡。这也表明,在外泌体与ExoPep结合的情况下,即使当施用血液来源的外泌体时,获得了相似的抗癌治疗效果。处理后,切除肝组织以检查外泌体的积累(mEXO-DiD标记),但在所有处理组中没有观察到积累(图14I)。
<实施例15>在A549肿瘤小鼠模型中施用ExoPep-KLA肽之后,对血液水平以及肝和肾功能的分析
为了研究由于ExoPep-KLA肽的处理导致的系统性副作用,在实施例14中的处理后收集小鼠血液,并分析血液水平以及肝和肾的功能水平。
作为结果,在用ExoPep-KLA或mEXO+ExoPep-KLA肽处理的组与健康小鼠之间,在血液水平(包括白血细胞计数)方面没有超出正常范围的显著差异(图15)。此外,作为处理组中肝和肾的功能测试结果,没有显示出特别的毒性(图15)。
<实施例16>在A549肿瘤小鼠模型中,通过阿霉素和ExoPep-KLA肽的共同施用抑制肿瘤的生长和转移
在A549肿瘤裸鼠模型中,在指定时间点每周三次施用ExoPep-KLA肽,并且每周一次施用阿霉素,持续三周。单一施用和共同施用的时间在处理方案中示出(图16A)。
作为结果,在仅用阿霉素(2.5mg/kg)、ExoPep-KLA(5mg/kg)和ExoPep-KLA(10mg/kg)处理的组中,与用磷酸盐缓冲液处理的组相比,肿瘤的尺寸略微减少。然而,没有显著差异(图16B)。另一方面,当共同施用阿霉素(5mg/kg)、阿霉素+ExoPep-KLA(2.5+5mg/kg)以及阿霉素+ExoPep-KLA(2.5+10mg/mg)时,肿瘤受到更强烈的抑制。尽管连续施用外泌体肽和与阿霉素的组合,在处理期间小鼠的体重没有变化(图16C)。此外,阿霉素+ExoPep-KLA(2.5+10mg/kg)和阿霉素+ExoPep-KLA(2.5+5mg/kg)的共同施用显著降低了肿瘤的重量。与其它组相比,这种效果在阿霉素+ExoPep-KLA(2.5+10mg/kg)组合的组中最明显(图16D)。此外,与缓冲液处理组相比,在用阿霉素(5mg/kg)、阿霉素+ExoPep-KLA(2.5+5mg/kg)和阿霉素+ExoPep-KLA(2.5+10mg/kg)处理组中的转移性结节和微小结节显著减少(图16E)。
此外,作为测量肺的重量的结果,在所有组之间没有显著差异(图16F)。在测量肝重量的结果中,在所有组之间没有显著差异(图16G)。
另一方面,与磷酸盐缓冲液和单一处理组相比,在阿霉素+ExoPep-KLA(2.5+5mg/kg)和阿霉素+ExoPep-KLA(2.5+10mg/kg)共同处理的组中生存率显著增加(图16H)。此外,在处理之后,对肿瘤组织进行切片,并在组织中观察TUNEL染色(绿色)。作为免疫组织化学分析的结果,阿霉素+ExoPep-KLA(2.5+5mg/kg)和阿霉素+ExoPep-KLA(2.5+10mg/kg)的处理显著增加了肿瘤组织中TUNEL阳性凋亡细胞的比例(图16I)。以上结果表明,当ExoPep-KLA和阿霉素共同施用时,阿霉素的治疗效果可能增加,并且其副作用可能通过之后减少剂量而减弱。
<实施例17>在A549肿瘤小鼠模型中共同施用ExoPep-KLA肽和阿霉素之后对血液水平以及肝和肾的功能水平的分析
在图16中的处理后,收集血液以分析血液水平以及肝和肾功能。作为结果,在处理组和健康小鼠之间,在血液水平(包括白血细胞计数)方面没有超出正常范围的显著差异,这支持了没有由于ExoPep-KLA或阿霉素+ExoPep-KLA(2.5+10mg/kg)的共同处理导致的系统性副作用。此外,作为肝和肾的功能测试的结果,与健康小鼠相比,ExoPep-KLA或阿霉素+ExoPep-KLA(2.5+10mg/kg)的共同处理没有显示出毒性(图17)。
<实施例18>在Panc-1胰腺癌小鼠模型中,通过ExoPep-KLA肽的单独施用和与吉西他滨的共同施用抑制肿瘤的生长和转移
在Panc-1肿瘤模型中,确认了ExoPep-KLA的单独施用和与吉西他滨的共同施用的抗肿瘤效果(图18A)。ExoPep-KLA和吉西他滨的系统性共同施用显著抑制了肿瘤的生长,ExoPep-KLA施用组和PBS组没有显著抑制肿瘤的生长(图18B)。为了确认外泌体肽和共同施用组的生物安全性,本公开的发明人观察了施用期间体重的变化。在外泌体肽的施用期间,没有观察到小鼠体重的变化(图18C)。重要的是,ExoPep-KLA和吉西他滨共同施用的组减少了原发肿瘤的生长。
与单独施用ExoPep-KLA的组和PBS组相比,观察到在共同施用组中肿瘤重量下降(图18D)。与PBS组相比,ExoPep-KLA和吉西他滨共同施用的组几乎没有显示转移性结节或微小结节(图18E)。此外,在施用结束时测量了肺的重量(图18F)。在另一方面,在共同施用组中原发肿瘤的重量减少,并且寿命增加(图18G)。在所有组中没有观察到由于施用导致的系统性副作用(图18H)。
