2. A preparation method of a composition with constipation preventing effect comprises the following steps:
(1) Mixing fructus Lycii 4 parts, semen Phaseoli 8 parts, poria 8 parts, semen Armeniacae amarum 6 parts, cowberry fruit 4 parts, and Raphani semen 2 parts, adding 10 times of water, decocting for 2 times each for 1.5 hr, filtering, and mixing extractive solutions; vacuum concentrating under vacuum degree less than or equal to 50Pa and concentration temperature less than or equal to 50deg.C to obtain mixed concentrated solution;
(2) Spray drying the mixed concentrated solution obtained in the step (1) at the drying temperature of less than or equal to 60 ℃ to obtain plant composite extract powder such as medlar;
(3) Mixing 6 parts of chicory, 3 parts of fructus cannabis and 4 parts of black sesame, freezing and crushing at the temperature of minus 8 ℃, sieving with a 80-mesh sieve, and uniformly mixing with 6 parts of konjak powder to obtain plant composite powder such as konjak;
(4) Mixing taurine 0.05 parts, bifidobacterium adolescentis 0.5 parts and lactobacillus plantarum 0.5 parts with the plant composite extract powder obtained in the step (2) and the plant composite powder obtained in the step (3) uniformly to obtain the composition with constipation preventing effect.
Example 2
1. A composition with constipation prevention effect is prepared from the following raw materials in parts by weight:
5 parts of medlar, 9 parts of red bean, 7 parts of tuckahoe, 6 parts of sweet almond, 2 parts of cowberry fruit, 3 parts of radish seed, 4 parts of chicory, 4 parts of hemp seed, 2 parts of black sesame, 8 parts of konjak powder, 0.06 part of taurine, 0.8 part of bifidobacterium adolescentis and 0.8 part of lactobacillus plantarum.
2. A preparation method of a composition with constipation preventing effect comprises the following steps:

(4) Mixing taurine 0.06 parts, bifidobacterium adolescentis 0.8 parts and lactobacillus plantarum 0.8 parts with the plant composite extract powder obtained in the step (2) and the plant composite powder obtained in the step (3) uniformly to obtain the composition with constipation preventing effect.
Example 3
1. A composition with constipation prevention effect is prepared from the following raw materials in parts by weight:
2 parts of medlar, 10 parts of red bean, 10 parts of poria cocos, 3 parts of sweet almond, 2 parts of cowberry fruit, 1 part of radish seed, 8 parts of chicory, 5 parts of fructus cannabis, 5 parts of black sesame, 8 parts of konjak powder, 0.02 part of taurine, 0.2 part of bifidobacterium adolescentis and 0.2 part of lactobacillus plantarum.

(3) Mixing 8 parts of chicory, 5 parts of fructus cannabis and 5 parts of black sesame, freezing and crushing at the temperature of minus 6 ℃, sieving with a 100-mesh sieve, and uniformly mixing with 8 parts of konjak powder to obtain plant composite powder such as konjak;
(4) Mixing taurine 0.02 parts, bifidobacterium adolescentis 0.2 parts and Lactobacillus plantarum 0.2 parts with the plant composite extract powder obtained in step (2) and the plant composite powder obtained in step (3) uniformly to obtain the composition with constipation preventing effect.
Example 4A method for preparing a concentrated paste of a composition having constipation preventing effect comprises the steps of:
(1) Dissolving the composition obtained in example 1 in appropriate amount of water, adding Mel, heating, and mixing to obtain concentrated extract;
(2) Taking the prepared concentrated paste liquid obtained in the step (1), and filling the concentrated paste liquid into glass bottles by a paste filling machine, wherein each bottle contains 250g or 500g; filling into aluminum plastic strip bags, wherein each bag is 10g or 20g.
Example 5 a method for preparing a solid particulate beverage composition having constipation preventing effect comprising the steps of:
(1) Taking the composition obtained in the example 2, adding soluble starch and spice, uniformly mixing, and granulating by a dry granulating machine;
(2) And (5) taking the particles obtained in the step (1), and subpackaging the particles in aluminum-plastic composite bags by using a particle packaging machine.
Example 6 a method for preparing a pressed candy of a composition having constipation preventing effect comprising the steps of:
(1) Taking the composition obtained in the example 1, adding lactose, compressible starch and appropriate amount of essence and spice, and mixing uniformly; making into granule by wet granulating machine, oven drying at below 60deg.C, and grading;
(2) And (3) taking the granules obtained in the step (1), placing the granules into a mixer, adding a tabletting lubricant, mixing for 15 minutes, and pressing the granules into tabletting candies by using the tablet press.
EXAMPLE 7 Constipation prevention function evaluation
1. Instrument and reagent

The compositions prepared in example 1, example 2 and example 3 were dissolved in water to prepare the desired concentrations.
2. Instrument and reagent
Instrument and equipment: analytical balance (Mettler Toledo), ophthalmic forceps, syringe, activated carbon powder, gum arabic, compound diphenoxylate (vincristine red pharmaceutical company).
Preparing ink: accurately weighing 100g of gum arabic, adding 800mL of water, boiling until the solution is transparent, weighing 50g of active carbon powder, adding into the solution, boiling for three times, cooling the solution, adding water to 1000mL, keeping the volume at 4 ℃ in a refrigerator, and shaking uniformly before use.
Preparing a compound diphenoxylate suspension: taking 10 compound diphenoxylate tablets (each tablet contains 2.5mg of compound diphenoxylate), grinding with a mortar, adding distilled water, suspending, and fixing volume to 100mL to prepare 0.025% compound diphenoxylate suspension. Is prepared before use.
3. Test animal
Animal origin: ICR male mice, 40, 18-22g, were purchased from Shanghai Laek laboratory animal Co., ltd (license number: SCXK (Shanghai) 2017-0005). The conventional feeding temperature is kept at (22+/-2) DEG C, the relative humidity is kept at (60+/-2)%, and the illumination period is 12 hours.

2. Experimental method
1. Animal grouping treatment and dose setting
Animals were randomly divided into 5 groups (blank, model, and example 1, example 2, example 3) based on body weight after 3 days of adaptive feeding. The recommended dose is calculated according to the recommended dose (10 g/d) for human oral administration, and 5 times the recommended dose for human is taken as the administration dose. After the test drugs are prepared, the mice are irrigated with the normal saline with the same volume as the BW volume, and the blank control group and the model control group are irrigated with the normal saline with the same volume once a day. After 8 days of continuous gastric lavage administration, the compound diphenoxylate is used for molding, and the bowel relaxing function indexes such as the first defecation time, the defecation grain number, the fecal weight and the like are measured.
2. Experimental principle and experimental procedure for bowel relaxing function

The compound diphenoxylate of the stomach-infused drug is used for establishing a constipation model of the mice, and measuring the defecation time, the defecation grain number and the defecation weight of the mice in 5 or 6 hours for first grain defecation to reflect the defecation condition of the model mice.
(2) Experimental procedure
And (3) establishing a model: after 7d of administration of the test sample, each group of mice fasted without water for 16h. The model control group and each example administration group were respectively given the compound diphenoxylate (10 mg/kgBW) by gavage, and the blank control group was given the same volume of physiological saline by gavage.
The index measuring method comprises the following steps: after animals were given the compound diphenoxylate for 0.5h, mice in the blank control group and the model control group were lavaged with ink, and the administration group was lavaged with ink containing the test drug. Animals were kept in single cages and fed with normal drinking water.
Starting from the stomach-filling ink, the black stool discharge time of the first grains of each animal, the number of black stool discharge grains in 5 hours and the weight are recorded.
3. Data analysis and result determination
1. Influence on body weight
The initial body weight of the animals and the body weight measurement results after the end of the experiment are shown in Table 1. As can be seen from Table 1, the animals in each group had a good weight gain during the test period and were able to feed water normally. No significant differences in body weight were seen between the animals of each group, indicating that the compositions provided by the present invention had no significant effect on the body weight of the animals tested.

2. Influence on first bowel movement time
The results of the first bowel movement time measurements for each group of animals are shown in Table 2. As can be seen from table 2, the first grain of the mice in the model group had significantly longer bowel movement time than the blank group within 5h, indicating successful model replication. The mice of the administration groups of the example 1, the example 2 and the example 3 have the first time to discharge the black stool shorter than the model group, wherein the differences of the administration groups of the example 1 and the example 2 are significant compared with the model group. The above results demonstrate that the composition provided by the invention can shorten the first black stool discharge time of mice.

3. influence on the number of black excrement particles discharged within 5 hours
The results of the analysis of the number of black bowel movements discharged within 5 hours of each group of animals are shown in Table 2. Table 2 shows that the number of black bowel movements in 5h of mice in the model group is significantly less than that in the blank group, indicating successful model replication. The number of the black discharge particles in 5h of the mice in the administration groups of the example 1, the example 2 and the example 3 is higher than that of the mice in the model group, wherein the differences between the administration groups of the example 1 and the example 2 and the model group are significant, which indicates that the composition provided by the invention can increase the number of the black discharge particles of the mice in the constipation model.
4. Influence on total weight of black stool discharged within 5 hours
The total weight analysis results of the black bowel movement in 5h of each group of animals are shown in table 2. Table 2 shows that the total weight of the black bowel movement in 5h of the model mice is significantly lower than that of the blank group, indicating successful replication of the constipation model. The mice in the administration groups of example 1 and example 2 have significantly longer first-time defecation time than the model group, and have significant differences, which indicates that the composition provided by the invention can increase the total weight of the defecation of the constipation model mice.
5. Result determination
From the experimental results, the composition provided by the invention can obviously shorten the first defecation time of a constipation model mouse and increase the number of black bowel movement particles and the total weight of black bowel movement in 5 hours of the model mouse. Based on the experimental results, the composition provided by the invention has the function of relaxing the bowels and protecting health based on animal experiments according to the rules of the function experiments related to relaxing the bowels of the health food functional evaluation program and the inspection method.
The composition provided by the invention can obviously shorten the first black stool discharge time of a constipation animal model and increase the quantity and weight of black stools, and has the function of relaxing the bowels and protecting health, and can be used for preventing constipation. Compared with the existing medicines and health-care foods, the composition adopts natural traditional medicine and food homologous raw materials with multiple health-care effects, is safer and has no adverse reaction.

1 to 5 parts of medlar, 2 to 10 parts of red bean, 2 to 10 parts of tuckahoe, 3 to 8 parts of chicory, 3 to 8 parts of konjaku flour, 3 to 8 parts of sweet almond, 1 to 5 parts of hemp seed, 1 to 5 parts of cowberry fruit, 1 to 5 parts of radish seed, 1 to 5 parts of black sesame, 0.01 to 0.1 part of taurine, 0.2 to 2 parts of bifidobacterium adolescentis and 0.2 to 2 parts of lactobacillus plantarum.
