本发明公开了一种高产率的氮硫共掺杂荧光碳量子点及其制备方法和应用,属于碳纳米材料制备技术领域。所述的制备方法包括如下步骤:将甲基橙粉末和水混合,搅拌后得到混合液;将混合液进行水热碳化反应,得到产物A;将产物A过滤后,收集滤液得到氮硫共掺杂的荧光碳量子点水溶液;将获得的水溶液干燥后得到高产率的氮硫共掺杂荧光碳量子点。本发明公开的合成方法操作简单、绿色高效、成本低,具有大规模实际生产的巨大潜力;获得的碳量子点不仅有高的荧光量子产率又有高的产率75.6%,作为荧光探针能用于活细胞成像和检测水样中的痕量Hg2+。

技术领域
本发明属于荧光碳纳米材料制备技术领域,具体涉及一种高产率的氮硫共掺杂荧光碳量子点及其制备方法和应用。
背景技术
碳量子点是一种新兴的碳质纳米材料,由于其独特的发光特性、低毒性、良好的生物相容性、表面易于修饰和优异的水分散性,引起了研究人员的极大兴趣。自2004年首次意外发现以来,已经提出了许多基于不同碳源的碳量子点合成方法,例如碳材料(石墨烯、石墨、蜡烛烟灰和石墨棒),有机分子(柠檬酸、二苯胺等)和生物质(蔬菜、食物残渣和水果等)通过化学或物理切割处理、超声波/微波辐射、水热处理、热分解、等离子体法、磁热法、微流控法等来合成碳量子点。
目前,碳量子点的制备方法仍然存在以下主要问题:1)碳量子点的量子产率通常小于10%,这限制了其广泛的应用。为了提高碳量子点的荧光量子产率,通常需要在合成过程中进行额外的掺杂或表面改性,这导致合成后复杂的纯化步骤,并使得大规模生产具有挑战性和浪费性;2)尽管先前报道的方法可以合成高质量的碳量子点,但大多数碳量子点的产率仍然极低,约低于30%,这导致制备效率低下和生产成本高。上述问题严重阻碍了碳量子点的实际应用和大规模制备。不幸的是,之前的大多数技术都集中在如何提高碳量子点的量子产率,而低产率的问题往往被忽视。因此,迫切需要开发高效和简便的策略来合成既有高量子产率又有高产率的荧光碳量子点,以促进碳量子点的大规模合成和实际应用。
Hg2+作为一种典型的重金属,由于其对人类健康和生态环境的有害影响,引起了越来越多的关注。目前,包括原子吸收光谱法、等离子体发射光谱法、质谱法和比色法在内的几种技术已普遍用于检测水中的重金属。然而,这些方法需要精密的仪器和冗长的样品制备过程,这大大增加了测试成本并降低了检测效率。为了克服这些限制,基于碳量子点的荧光探针为Hg2+检测带来了更好的选择。因为其低成本、良好的荧光特性和易于操作等特点。
半导体量子点和有机荧光染料虽然具有高的荧光性能,但是其低的亲水性、高的毒性、复杂的合成工艺、高的成本限制了它在生物成像方面的实际应用。相比,碳量子点作为一种主要由碳元素制成的碳纳米材料,具有优异的荧光性能、较小的颗粒尺寸、良好的水溶性、低的毒性和低的制备成本。这使其在生物成像中具有巨大的应用价值。
发明内容
为了克服上述现有技术的缺点,本发明的目的在于提供一种高产率的氮硫共掺杂荧光碳量子点及其制备方法和应用,以解决现有的碳量子点制备方法存在的产率低和纯化工艺复杂的技术问题,为碳量子点在重金属离子检测和生物成像应用方面提供一种高质量的碳纳米材料。
为了达到上述目的,本发明采用以下技术方案予以实现:
本发明公开了一种高产率的氮硫共掺杂荧光碳量子点的制备方法,包括以下步骤:
S1:将甲基橙粉末和水混合后,得到混合溶液;将混合溶液搅拌后进行水热反应,得到反应产物A;
S2:将反应产物A进行过滤后,收集滤液,得到氮硫共掺杂的荧光碳量子点水溶液;将氮硫共掺杂的荧光碳量子点水溶液干燥后得到高产率的氮硫共掺杂荧光碳量子点。
进一步地,S1中,所述混合溶液的比例为(60~120)mL/g。
进一步地,S1中,所述水热反应的参数为:将混合溶液搅拌后,以5~10℃/min的升温速率从室温升至200~240℃,进行水热反应12~16h。
进一步地,S2中,所述反应产物A进行过滤的方式是将反应产物A采用纳米级的有机系滤膜过滤若干次。
进一步地,S2中,所述干燥的方式为:将氮硫共掺杂的荧光碳量子点水溶液转移到表面皿上,在80~90℃的烘箱中干燥12~15h后得到高产率的氮硫共掺杂荧光碳量子点。
本发明还公开了采用上述制备方法制备得到的氮硫共掺杂荧光碳量子点。
本发明还公开了上述氮硫共掺杂荧光碳量子点的应用,所述氮硫共掺杂荧光碳量子点用于细胞成像的荧光探针。
进一步地,所述氮硫共掺杂荧光碳量子点作为荧光探针用于选择性检测水样品中的Hg2+。
与现有技术相比,本发明具有以下有益效果:
本发明公开了一种高产率的氮硫共掺杂荧光碳量子点的制备方法,首次以甲基橙为碳源、同时为氮和硫源,不需要添加额外的掺杂剂;避免了先前技术中除了碳源外,还需添加额外掺杂剂的复杂工艺,降低了制备成本;同时,仅以水为溶剂绿色环保,避免了先前技术例如长时透析、高速离心等除去溶剂和掺杂剂的复杂工艺,减少了因复杂提纯而造成的样品损失,实现高产率的制备过程。
进一步地,本发明在制备时采用纳米级的有机系滤膜,可以实现生成物的高效纯化,避免了复杂耗时的纯化工艺。
本发明还公开了采用上述制备方法制备得到的高产率的氮硫共掺杂荧光碳量子点,根据相关实验结果,本发明合成的碳量子点不仅具有高的量子产率29.4%和超低的毒性,更重要的是具有高的产率75.6%大大超过了先前技术的结果,该合成方法具有大规模实际生产的巨大潜力。
本发明还公开了上述高产率的氮硫共掺杂荧光碳量子点的应用,作为细胞成像和选择性检测水样品中Hg2+的潜在荧光探针,其具有高的亲水性和抗干扰性、稳定的荧光、超低的细胞毒性、良好的生物相容性。

下列实施例中使用本领域常规的仪器设备。下列实施例中未注明具体条件的实验方法,通常按照常规条件,或按照制造厂商所建议的条件。下列实施例中使用各种原料,除非另作说明,都使用常规市售产品,其规格为本领域常规规格。在本发明的说明书以及下述实施例中,如没有特别说明,“%”都表示重量百分比,“份”都表示重量份,比例都表示重量比。
实施例1
一种高产率的氮硫共掺杂荧光碳量子点的制备方法,包括以下步骤:
S1:将1.0g甲基橙粉末与60mL纯净水混合,得到混合溶液;将混合溶液在1000r/min下连续磁力搅拌30min后,装入100mL聚四氟乙烯水热釜中,以10℃/min的速率升温至200℃,在该温度下水热反应16h后,自然冷却到室温,得到反应产物A;
S2:将反应产物A用50nm的有机系滤膜过滤5次,收集最终过滤的透明滤液,得到氮硫共掺杂的荧光碳量子点水溶液;将得到的氮硫共掺杂的荧光碳量子点水溶液转移到表面皿上,在90℃烘箱中干燥15h后,得到高产率的氮硫共掺杂荧光碳量子点。
实施例2
一种高产率的氮硫共掺杂荧光碳量子点的制备方法,包括以下步骤:
S1:将0.8g甲基橙粉末与60mL纯净水混合,得到混合溶液;将混合溶液在1000r/min下连续磁力搅拌20min后,装入100mL聚四氟乙烯水热釜中,以8℃/min的速率升温至220℃,在该温度下水热反应14h后,自然冷却到室温,得到反应产物A;
S2:将反应产物A用50nm的有机系滤膜过滤4次,收集最终过滤的透明滤液,得到氮硫共掺杂的荧光碳量子点水溶液;将得到的氮硫共掺杂的荧光碳量子点水溶液转移到表面皿上,在90℃烘箱中干燥12h后,得到高产率的氮硫共掺杂荧光碳量子点。
实施例3
一种高产率的氮硫共掺杂荧光碳量子点的制备方法,包括以下步骤:
S1:将0.5g甲基橙粉末与60mL纯净水混合,得到混合溶液;将混合溶液在1000r/min下连续磁力搅拌10min后,装入100mL聚四氟乙烯水热釜中,以5℃/min的速率升温至240℃,在该温度下水热反应12h后,自然冷却到室温,得到反应产物A;
S2:将反应产物A用50nm的有机系滤膜过滤3次,收集最终过滤的透明滤液,得到氮硫共掺杂的荧光碳量子点水溶液;将得到的氮硫共掺杂的荧光碳量子点水溶液转移到表面皿上,在80℃烘箱中干燥12h后,得到高产率的氮硫共掺杂荧光碳量子点。
实施例4
一种高产率的氮硫共掺杂荧光碳量子点的制备方法,包括以下步骤:
S1:将0.6g甲基橙粉末与60mL纯净水混合,得到混合溶液;将混合溶液在1000r/min下连续磁力搅拌15min后,装入100mL聚四氟乙烯水热釜中,以5℃/min的速率升温至240℃,在该温度下水热反应13h后,自然冷却到室温,得到反应产物A;
S2:将反应产物A用50nm的有机系滤膜过滤3次,收集最终过滤的透明滤液,得到氮硫共掺杂的荧光碳量子点水溶液;将得到的氮硫共掺杂的荧光碳量子点水溶液转移到表面皿上,在80℃烘箱中干燥15h后,得到高产率的氮硫共掺杂荧光碳量子点。
对实施例3制备得到的高产率的氮硫共掺杂荧光碳量子点的进行了TEM测试,结果如图1所示。可以看出碳量子点是近球形,完全分散没有任何团聚。对应的粒径分布柱状图如图2所示,尺寸分布范围是3.2~5.8nm,平均颗粒尺寸是4.2nm。
得到的高产率的氮硫共掺杂荧光碳量子点的HRTEM照片如图3所示,显示其有两种明显的晶格条纹,间距分布是0.21nm和0.32nm,说明合成的碳量子点具有很好的结晶性。
从图4的红外光谱图中可以看出,获得的碳量子中存在含O、N、S等官能团。进一步通过XPS技术分析了实施例3制备得到的氮硫共掺杂荧光碳量子点。结果如图5所示,可以看出碳量子点主要由C、O、N、S四种元素组成,其对应的含量分别为68.11%、21.41%、8.10%、2.38%。图6、7、8、9分别对应氮硫共掺杂荧光碳量子点的C1s、O1s、N1s、S2p的XPS精细谱,可以看出含C、O、N、S等基团存在于碳量子点的表面,充分说明了氮硫共掺杂的碳量子点已经被成功合成。
对实施例3制备得到的高产率的氮硫共掺杂荧光碳量子点进行了紫外可见吸收光谱的测试,结果如10所示,可以看出碳量子点的吸收光谱图与甲基橙的相比,已经发射了明显的变化,甲基橙的特征吸收峰在碳量子点的吸收光谱中已经完全消失,说明甲基橙已经被完全转化为氮硫共掺杂的碳量子点。
测试了实施例3制备得到的高产率的氮硫共掺杂荧光碳量子点的激发和发射光谱,结果如图11,可以看出合成的氮硫共掺杂荧光碳量子的最佳激发波长为330nm,对应的最佳发射波长约为447nm。利用绝对法测定了在激发波长330nm下,实施例3制备得到的氮硫共掺杂荧光碳量子点的荧光量子产率约为29.4%。
实施例3制备的高产率的氮硫共掺杂荧光碳量子点在不同激发波长下的发射光谱图参见图12,可以看出随着碳量子点的激发波长从310nm增加到440nm的时候,发射峰的中心明显发射了红移。说明合成的高产率的氮硫共掺杂荧光碳量子点具有激发依赖的荧光属性。
应用实施例1
将高产率的氮硫共掺杂荧光碳量子点用作细胞成像的荧光探针,包括如下步骤:
1)、将制备得到的氮硫共掺杂荧光碳量子点粉末配制成浓度为100μg/mL、200μg/mL、300μg/mL、400μg/mL、500μg/mL碳量子点溶液,分别与A549细胞在37℃培养24h。具体采用MTT法评估细胞的毒性,结果如图14所示,从图可以看出在碳量子点浓度高达500μg/mL时,细胞仍然可以保持90%以上的活性。同时采用相同的方法对比了甲基橙对A549细胞的毒性;从图13可以看出,与对照组相比,MO诱导的A549细胞活性以剂量依赖的方式降低,在浓度为500μg/mL,A549细胞的活性仅有65%,该结果表明,甲基橙被成功转化为具有超低毒性的氮硫共掺杂荧光碳量子点。
2)、通过A549细胞的成像评估制备得到的氮硫共掺杂荧光碳量子点在生物标记中的潜能,将400μg/mL氮硫共掺杂荧光碳量子点培养后的A549细胞,分别在364nm、405nm和458nm光激发下,使用共焦显微镜(Leica TCS SP8)采集图像,结果如图14所示,细胞周围均显示出了明亮的荧光。与对照组相比细胞的形态并没有明显变化,说明合成的氮硫共掺杂荧光碳量子点具有低的毒性和良好的生物相容性,在生物成像方面具有巨大的应用前景。
应用实施例2
采用高产率的氮硫共掺杂荧光碳量子点用于选择性检测水样品中的Hg2+,包括如下步骤:
1)、将制备得到的氮硫共掺杂荧光碳量子点粉末,用超纯水配制成浓度为60μg/mL碳量子点溶液。
2)、将浓度为100μM的各种金属离子(Ag+、Zn2+、Cu2+、Sr2+、Ca2+、Na+、Hg2+、La3+,Cd2+、Cr3+、Pb2+、Co2+、Fe2+、Mg2+、CO3 2ˉ、SO4 2ˉ、PO4 3ˉ、NO3 -、Cl-),分别加入到1mL浓度为60μg/mL的氮硫共掺杂碳量子点溶液中,在室温下静置5min;
3)、随后在荧光光谱仪上进行测试,在330nm光激发下测量每种混合物的荧光光谱,并记录其在447nm处的发射强度,从图15可以看出,在各种金属离子中只有Hg2+对氮硫共掺杂的荧光碳量子点的荧光产生了明显淬灭,说明合成的氮硫共掺杂荧光碳量子点可以选择性检测对水中的Hg2+。

Claims (5)
1.一种高产率的氮硫共掺杂荧光碳量子点的应用,其特征在于,高产率的氮硫共掺杂荧光碳量子点用于细胞成像的荧光探针或作为荧光探针用于选择性检测水样品中的Hg2+;
高产率的氮硫共掺杂荧光碳量子点的制备方法,包括以下步骤:
S1:将甲基橙粉末和水混合后,得到混合溶液;将混合溶液搅拌后进行水热反应,得到反应产物A;
S2:将反应产物A进行过滤后,收集滤液,得到氮硫共掺杂的荧光碳量子点水溶液;将氮硫共掺杂的荧光碳量子点水溶液干燥后得到高产率的氮硫共掺杂荧光碳量子点。
2.根据权利要求1所述的一种高产率的氮硫共掺杂荧光碳量子点的应用,其特征在于,S1中,所述混合溶液的比例为(60~120)mL/g。
3.根据权利要求1所述的一种高产率的氮硫共掺杂荧光碳量子点的应用,其特征在于,S1中,所述水热反应的参数为:将混合溶液搅拌后,以5~10 ℃/min的升温速率从室温升至200~240 ℃,进行水热反应12~16 h。
4.根据权利要求1所述的高产率的氮硫共掺杂荧光碳量子点的应用,其特征在于,S2中,所述反应产物A进行过滤的方式是将反应产物A采用纳米级的有机系滤膜过滤若干次。
5.根据权利要求1所述的一种高产率的氮硫共掺杂荧光碳量子点的应用,其特征在于,S2中,所述干燥的方式为:将氮硫共掺杂的荧光碳量子点水溶液转移到表面皿上,在80~90℃的烘箱中干燥12~15 h后得到高产率的氮硫共掺杂荧光碳量子点。
